A Pilot Proteomics Analysis of Mycobacterium tuberculosis Lacking a Putative Short-Chain Dehydrogenase Rv0148.

Mycobacterium tuberculosis (Mtb) is an intracellular pathogen that survives in host cells by resisting hostile antimicrobial defenses. However, the molecules and mechanisms that contribute to Mtb's intracellular survival are not fully understood. We have previously reported that Rv0148, a putative short-chain dehydrogenase/reductase, plays a significant role in Mtb stress response and virulence in in vitro and in vivo models. To further understand the role of Rv0148 in regulating global functions of Mtb, we performed comparative pilot proteomic analysis between Δrv0148 mutant, and wild-type (W

Investigating POLG-driven modulation of cancer stemness: a pilot study in breast cancer cells.

Mitochondria have emerged as key regulators of breast cancer stem cell (CSC) biology. Mitochondrial metabolic pathways, including oxidative phosphorylation (OXPHOS) and mitochondrial biogenesis, are frequently altered during tumorigenesis, highlighting their role in breast cancer pathogenesis. Since breast CSCs are highly dependent on mitochondrial metabolism, targeting mitochondrial DNA (mtDNA) replication may represent a strategy to impair CSC maintenance. Mitochondrial DNA polymerase-γ (POLG), composed of a catalytic subunit encoded by POLG1 and an accessory subunit encoded by POLG2, is ess

Retrospective analysis of neurofilament-light chain in patients with inflammatory bowel disease - A pilot study.

BACKGROUND: Chronic inflammatory bowel diseases, encompassing Crohn's disease and ulcerative colitis, are characterized by persistent inflammation of the gastrointestinal tract. While traditionally regarded as confined to the gut, the systemic nature of inflammatory bowel disease has been increasingly recognized. The nervous system has garnered particular attention due to molecular and clinical evidence suggesting a potential interplay between inflammatory bowel disease and neurodegenerative diseases. Inflammatory bowel disease patients have a higher risk of developing neurological disorders s

Benzodiazepine-Sparing Alcohol Withdrawal Protocol Pilot in Postsurgical Head and Neck Cancer Patients.

BACKGROUND: Alcohol use disorder is associated with high rates of morbidity and mortality in hospitalized patients. Patients with head and neck cancer have rates of comorbid alcohol use as high as 30% to 60%. Traditionally, withdrawal is managed by assessing symptoms with the Clinical Institute Withdrawal Assessment (CIWA) and treating with benzodiazepines. Benzodiazepines perpetuate neurotransmitter dysregulation in alcohol withdrawal, but valproic acid can correct dysregulations of glutamic and GABAergic systems. METHODS: We conducted an IRB-approved retrospective chart review of patients a

Modified Unified Wilson's Disease Rating Scale - scale presentation and pilot clinimetric testing.

AIM OF THE STUDY: The Unified Wilson's Disease Rating Scale (UWDRS) was first introduced in 2007 to assess neurologic status and impairment in patients with Wilson's disease (WD). However, major issues have been raised, including the lack of assessment of mental and psychiatric status, the lack of instructions for the assessment of certain motor functions, difficulties with gait and posture assessment, and heterogenous response options for questions across the scale. CLINICAL RATIONALE FOR THE STUDY: The development of a modified version of the UWDRS (mUWDRS) was proposed. MATERIALS AND METH

Establishing a methodological framework for evaluating gait symmetry through muscle synergies: A pilot study.

Gait symmetry is vital for stable locomotion, yet conventional metrics often yield inconsistent results. Muscle synergy analysis provides a more precise understanding of neural mechanisms behind symmetric gait. A clear methodology is essential to distinguish true inter-limb differences from natural variability. This study aimed to refine the analysis of muscle synergy symmetry in healthy individuals by incorporating intra-limb similarity as a baseline for comparison, while also including muscle weightings within each synergy to explore muscle synchrony. EMG signals were collected from both low

Cross sectional pilot study on clinical review generation using large language models.

As the volume of medical literature accelerates, necessitating efficient tools to synthesize evidence for clinical practice and research, the interest in leveraging large language models (LLMs) for generating clinical reviews has surged. However, there are significant concerns regarding the reliability associated with integrating LLMs into the clinical review process. This study presents a systematic comparison between LLM-generated and human-authored clinical reviews, revealing that while AI can quickly produce reviews, it often has fewer references, less comprehensive insights, and lower log

Association of Cerebellar Peduncle Morphometry with Hypertrophic Olivary Degeneration: A Pilot Case-Control Study.

BACKGROUND AND PURPOSE: Hypertrophic olivary degeneration (HOD) is a rare neurologic condition resulting from disruption of the dentato-rubro-olivary pathway (DROP). Nearly one-half of HOD cases, however, lack detectable lesions in the DROP, and fewer than 10% of patients develop the condition after surgical resection of posterior fossa tumors. Based on these observations, we hypothesized that inherent anatomic variations in components of the DROP, specifically the inferior cerebellar peduncle (ICP) and superior cerebellar peduncle (SCP), may predict susceptibility to HOD.
