Selected physico-chemical properties of succinylated legumin from pea (Pisum sativum L.).

Selected physico-chemical properties of pea legumin before and after succinylation have been investigated using isoelectric focusing, PAGE, SDS-PAGE, hydrophobicity measurements, SE-HPLC and RP-HPLC. Exhaustive succinylation shifted the I.P. of legumin from 4.75 to 3.5. The stepwise dissociation of legumin by increasing succinylation has been confirmed both by means of PAGE in a nondenaturing system, and by SE-HPLC. The results of SDS-PAGE provided evidence for the exposure of alpha-polypeptide chains in the native legumin. High succinylation resulted in a decrease of the surface hydrophobicit

Is islet amyloid polypeptide a significant factor in pathogenesis or pathophysiology of diabetes?

Islet amyloid polypeptide (IAPP) or amylin, a recently discovered minor secretory peptide of the beta-cell related to calcitonin gene-related peptide (CGRP), is a constituent of amyloid deposits in the islets of many non-insulin-dependent (type II) diabetic individuals and some elderly nondiabetic subjects. IAPP is synthesized as a small precursor at a level of approximately 1% that of insulin and is processed, amidated, stored in beta-granules, and released along with insulin and C-peptide. Analysis of its gene (located on chromosome 12) supports an evolutionary relationship to calcitonin and

Cyclosporin A slows collagen triple-helix formation in vivo: indirect evidence for a physiologic role of peptidyl-prolyl cis-trans-isomerase.

Peptidyl-prolyl cis-trans-isomerase accelerates otherwise slow, rate-limiting isomerization steps during folding of proteins in vitro, but is not yet securely identified with any specific physiologic role. Peptidyl-prolyl cis-trans-isomerase and the cyclosporin A (CsA)-binding protein cyclophilin are identical, and peptidyl-prolyl cis-trans-isomerase activity is inhibited by the immunosuppressive drug CsA in vitro. To establish a possible physiologic role of peptidyl-prolyl cis-trans-isomerase, we have studied the folding of procollagen I in suspended chick embryo tendon fibroblasts. Folding o

G protein diversity and the regulation of signaling pathways.

The subunits of the heterotrimeric guanine nucleotide-binding (G) proteins mediate the transfer of information from receptor to effector molecules. Biochemical studies and recent molecular cloning efforts have revealed a rich but largely unexpected diversity in G protein subunit structure and function. Extreme specificity in signaling pathways could be accommodated by the combinatorial association of individual subunits. Alternatively, this wealth of diversity may allow for the simultaneous activation of related G alpha subunits and the modulation of several effector systems. In tissues where

Calcium(II) site specificity: effect of size and charge on metal ion binding to an EF-hand-like site.

The molecular mechanisms by which protein Ca(II) sites selectively bind Ca(II) even in the presence of high concentrations of other metals, particularly Na(I), K(I), and Mg(II), have not been fully described. The single Ca(II) site of the Escherichia coli receptor for D-galactose and D-glucose (GGR) is structurally related to the eukaryotic EF-hand Ca(II) sites and is ideally suited as a model for understanding the structural and electrostatic basis of Ca(II) specificity. Metal binding to the bacterial site was monitored by a Tb(III) phosphorescence assay: Ca(II) in the site was replaced with

Lymphocytic choriomeningitis virus. X. Demonstration of nucleoprotein on the surface of infected cells.

Of a total of 17 monoclonal antibodies (MAb) directed against structural proteins of the lymphocytic choriomeningitis (LCM) virus, 3 were specific for the viral nucleoprotein (p63) and attached to the plasma membrane of infected cells, as disclosed by the indirect immunofluorescence procedure and complement-mediated cytolysis. We had previously demonstrated that a portion of the nucleoprotein (p63E) was part of the envelope of the intact virion (M. Bruns, W. Zeller, H. Rohdewohld, and F. Lehmann-Grube (1986) Virology 151, 77-85), and we now show that after external iodination of virions follow

Electron paramagnetic resonance detectable states of cytochrome P-450cam.

Cytochrome P-450cam is a low-spin Fe3+hemoprotein (g = 2.45, 2.26, and 1.91) which is made 60% high spin (g = 7.85, 3.97, and 1.78) at 12 K by the addition of 1 mol of substrate per mol of enzyme. Low-temperature EPR spectra show that the low-spin fraction of substrate-bound P-450cam contains two magnetic species. The majority species has an unusual EPR spectrum (g = 2.42, 2.24, and 1.97) which connot be simulated by using the range of crystal field parameters known for other heme proteins. The minority species has the same g values as substrate-free enzyme. Both low-spin species show Curie la

Catalytically impaired TrpA subunit of tryptophan synthase from Chlamydia trachomatis is an allosteric regulator of TrpB.

Intracellular growth and pathogenesis of Chlamydia species is controlled by the availability of tryptophan, yet the complete biosynthetic pathway for l-Trp is absent among members of the genus. Some representatives, however, preserve genes encoding tryptophan synthase, TrpAB - a bifunctional enzyme catalyzing the last two steps in l-Trp synthesis. TrpA (subunit α) converts indole-3-glycerol phosphate into indole and glyceraldehyde-3-phosphate (α reaction). The former compound is subsequently used by TrpB (subunit β) to produce l-Trp in the presence of l-Ser and a pyridoxal 5'-phosphate cofacto

Interfacial engineering of alkyl-chain-tailored Pickering foams for quinoa protein fractionation: mechanisms and functional upgrading.

The continuous foam fractionation of low-surface-activity plant proteins, such as those from quinoa, is fundamentally hindered by the recovery-enrichment paradox driven by unstable foam dynamics. Herein, we engineered tunable Pickering stabilizers by in-situ functionalizing calcium carbonate nanoparticles (CNPs) with homologous alkyl-chain modifiers (C4, C8, C12) to precisely regulate interfacial drainage. Obeying Traube's rule, the long-chain laurate modified nanoparticles (SL-CNPs) optimized interfacial wettability (θ ≈ 90o) and maximized detachment energy at a minimal dosage (100 mg/L), ach

How a Single-Chain Polystyrene Nanoparticle Dictates α-Synuclein Adsorption and Intramolecular β-Sheet Nucleation.

A growing number of studies have detected nanoplastics, particularly polystyrene (PS) nanoparticles, in human blood and various critical tissues. Nevertheless, the molecular mechanisms by which their physical size modulates the aggregation of disease-related proteins remain poorly understood. Here, using well-tempered metadynamics simulations, we demonstrate that single-chain PS nanoparticles with a degree of polymerization N collapse into spherical globules in water, following a scaling law of radius of gyration Rg ∼ N1/3, and that their size critically determines the conformational fate of a

Multi-omics network pharmacology and computational validation reveal amodiaquine and desethylamodiaquine as apoptosis-regulating multi-target candidates in colon adenocarcinoma.

One of the biggest causes of cancer-related mortality worldwide is still colon adenocarcinoma (COAD). Therefore, it is essential to investigate new therapy strategies. This study presents a multi-scale, hypothesis-generating framework that integrates network pharmacology with multi-omics data and quantum chemical analyses to systematically explore the repositioning potential of amodiaquine and desethylamodiaquine in colon adenocarcinoma. Eleven gene targets that were found to be shared across medication, gene expression, and illness databases included several computationally significant centra

Condensate-Like Organization in Respiratory Aerosols Modulates the Dynamics of an Airborne Virus.

The molecular behavior of viruses within respiratory aerosols plays a critical role in airborne disease transmission yet remains largely inaccessible to experimental characterization. Here, we use a billion-atom all-atom molecular dynamics simulation of a virus-laden respiratory aerosol to uncover how respiratory proteins, lipids, ions, and water collectively assemble around SARS-CoV-2, giving rise to structured microenvironments that influence viral stability and spike dynamics. We find that respiratory components rapidly evolve into heterogeneous networks characterized by protein-rich aggreg

Solution structures of glycosaminoglycan-bound CXCL8 complexes determined by small-angle X-ray scattering (SAXS).

Glycosaminoglycans (GAGs) play diverse and fundamental roles in physiology by regulating the function of large classes of proteins. Despite their importance, knowledge of how GAGs are organized in protein-bound complexes remains limited. This can be attributed to the linear structure, conformational flexibility, and high negative charge of GAGs, all of which disfavor structure determination by crystallography or NMR spectroscopy. A hybrid approach based on GAG-binding-induced changes in NMR protein chemical shifts, computational docking, and molecular dynamics simulations has proven to be valu

The mutational landscape of STING-induced immunity.

Stimulator of interferon genes (STING) is an evolutionary conserved immune signalling protein with key roles in host defence, cancer, senescence and inflammation1-3. Downstream of STING, type I interferon, inflammatory cytokine signalling and non-canonical autophagy are governed by a multilayered mechanism integrating ligand-induced structural transitions, protein-protein interactions and coordinated intracellular trafficking4-13. Despite its central role in immunity and relevance as therapeutic target14, the sequence elements that govern STING (in)activation in cells remain incompletely under

Integrated transcriptomic and in silico structural analysis identifies NFS1 and HSPA9 as potential regulators associated with mitochondrial ferroptosis and cell death resistance in colorectal cancer.

Colorectal cancer (CRC) exhibits extensive metabolic reprogramming and resistance to regulated cell death, contributing to tumor progression and therapeutic failure. Ferroptosis, an iron-dependent and mitochondria-associated form of cell death, has emerged as a potential therapeutic vulnerability; however, its integrated molecular regulation in CRC remains poorly understood. In this study, an integrative transcriptomic and in silico structural analysis was performed using two GEO datasets (GSE290002 and GSE65632). Differential expression analysis combined with curated mitochondrial and ferropt

Local entropy in proteins.

Proteins populate dynamic ensembles, yet how temperature and mutations reshape these ensembles remains poorly understood. We introduce a local entropy metric that assigns each residue a Shannon entropy based on a graph-derived map of accessible substates, providing a continuous measure of structural complexity across folded, unfolded, and intrinsically disordered states. In molecular dynamics simulations of the fast-folding gpW protein, the average local entropy exhibits a sharp transition near the melting point. Residue-specific entropy curves cluster into distinct unfolding categories and re

Valosin-Containing Protein as a therapeutic target in CAG repeat-driven Spinocerebellar ataxias: Integrative transcriptomic and computational insights.

Spinocerebellar ataxias (SCAs) are progressive neurodegenerative disorders caused by abnormal CAG repeat expansions in genes such as ATXN1, ATXN2, and ATXN3, with no effective therapeutic options currently available. To identify key molecular drivers of disease pathology, transcriptomic datasets GSE75249 and GSE151276 were analyzed. Differentially expressed genes were determined, and overlapping upregulated genes from both datasets were extracted for downstream analysis. Functional enrichment revealed significant biological processes and pathways related to protein homeostasis, cellular stress

Facile Control of a Tailed Virus Surrogate by Iron Conventional Coagulation and Electrocoagulation.

Both FeCl3 conventional coagulation and Fe(0) electrocoagulation were highly effective in mitigating the long-tailed somatic phage P1. We targeted enterobacterial coliphages because they are better than fecal indicator bacteria in tracking environmental persistence of viral pathogens and their fate in wastewater unit operations. Cryogenic electron microscopy (cryo-EM) of intact/damaged P1 and enumeration of infective virions by plaque assays demonstrated control via both removal and inactivation. Cryo-single particle analysis was coupled with cryo-electron tomography to generate 3-dimensional
