Recognition DNA sequence of a novel putative transcription factor, BCL6.

The BCL6 gene involved in the 3q27 translocation associated with B-cell lymphomas encodes a novel Cys2-His2 zinc finger protein. We generated a fusion protein of glutathione S-transferase and zinc finger domain of BCL6 to determine recognition sequences of BCL6 with polymerase chain reaction using random oligonucleotides of 26 bases as a ligand. A consensus of 14 nucleotides consisting of (T/A)NCTTTCNAGG(A/G)AT was identified in the recognition sequences. In a gel mobility shift assay, the probe containing the 14-nucleotide recognition sequence formed a complex with the fusion protein and nucl

Erythrocyte membrane protein deficiencies in paroxysmal nocturnal hemoglobinuria.

Paroxysmal nocturnal hemoglobinuria (PNH) is an acquired disorder characterized by intermittent hemolytic anemia. Membrane abnormalities of blood cells from patients with PNH are the reason for the unusual sensitivity to lysis by autologous plasma complement. A patient with typical clinical disease consistent with PNH is described together with a few strategies and pitfalls for treatment. Commonly used in vitro assays are discussed that document the complement-mediated lysis of aberrant PNH erythrocytes. Membrane-associated proteins that are abnormal in PNH cells, the characteristics of these

Mass Spectrometry-Based Proteomics of Intestinal Organoids.

Proteomics, the large-scale study of proteins, enables the identification, quantification, and functional characterization of proteins, revealing post-translational modifications and protein interactions that are not apparent from transcriptomic data. Human organoids, which recapitulate the structural and functional complexity of native epithelial tissues, provide powerful tools to study disease mechanisms and personalize therapies. However, their culture poses challenges for efficient protein extraction and reproducible analysis. Here, we present a proteomics workflow optimized to maximize pr

Human serum proteins bind to Sporothrix schenckii conidia with differential effects on phagocytosis.

Serum is an important source of proteins that interact with pathogens. Once bound to the cell surface, serum proteins can stimulate the innate immune system. The phagocytosis of Sporothrix schenckii conidia by human macrophages is activated through human serum opsonisation. In this study, we have attempted to characterise human blood serum proteins that bind to the cell wall of S. schenckii conidia. We systematically observed the same four proteins independent of the plasma donor: albumin, serum amyloid protein (SAP), α-1 antitrypsin (AAT), and transferrin were identified with the help of tand

Cerebral organoids expressing mutant actin genes reveal cellular mechanism underlying microcephaly.

Actins are cytoskeletal proteins that are essential for multiple cellular processes. Mutations in the ACTB and ACTG1 genes, encoding the ubiquitous beta- and gamma-cytoskeletal actin isoforms, respectively, cause a broad spectrum of neurodevelopmental disorders, with microcephaly as the most frequent one. To investigate the pathogenesis underlying this cortical malformation, we studied patient-derived cerebral organoids from induced pluripotent stem cells of individuals with the Baraitser-Winter-CerebroFrontoFacial syndrome (BWCFF-S) carrying an ACTB/ACTG1 missense mutation. These organoids we

Clonal analysis of Salmonella-specific effector T cells reveals serovar-specific and cross-reactive T cell responses.

To tackle the complexity of cross-reactive and pathogen-specific T cell responses against related Salmonella serovars, we used mass cytometry, unbiased single-cell cloning, live fluorescence barcoding, and T cell-receptor sequencing to reconstruct the Salmonella-specific repertoire of circulating effector CD4+ T cells, isolated from volunteers challenged with Salmonella enterica serovar Typhi (S. Typhi) or Salmonella Paratyphi A (S. Paratyphi). We describe the expansion of cross-reactive responses against distantly related Salmonella serovars and of clonotypes recognizing immunodominant antige

Peptides representing the specific variable regions but not the full porin proteins can be useful for antibody based diagnosis of typhoid fever.

Antibody response to major porin proteins of S. Typhi (OmpC and OmpF) was evaluated in sera of typhoid patients (culture positive, n = 28; Widal positive, n = 16). Sera from fever patients (n = 6) having etiology other than Salmonella, and normal healthy human controls (n = 18) were also included. No significant difference between the anti-OmpC and anti-OmpF antibodies (Ab) of typhoid patients and controls was observed. The amino acid sequences of OmpC (and OmpF) porin of enterobacteria was aligned and searched for the variable regions specific to S. Typhi. Two regions, each representing one s

Inhibition of the magnesium-sensitive TRPM7-like channel in cardiac myocytes by nonhydrolysable GTP analogs: involvement of phosphoinositide metabolism.

BACKGROUND/AIMS: A magnesium-inhibited, transient receptor potential melastatin 7 (TRPM7)-like channel is expressed in cardiac cell membranes. The role and regulation of this channel by intracellular nucleotides and membrane components remain unclear. METHODS: We used the whole-cell voltage-clamp technique in pig isolated ventricular myocytes to investigate the effect of non-hydrolysable guanine nucleotides. RESULTS: The TRPM7-like current, induced by intracellular dialysis with low [Mg(2+)], remained stable when the intracellular solution contained GTP. Substituting GTP by GTP-gamma-S or Gp

Loss of mRor1 enhances the heart and skeletal abnormalities in mRor2-deficient mice: redundant and pleiotropic functions of mRor1 and mRor2 receptor tyrosine kinases.

The mammalian Ror family of receptor tyrosine kinases consists of two structurally related proteins, Ror1 and Ror2. We have shown that mRor2-deficient mice exhibit widespread skeletal abnormalities, ventricular septal defects in the heart, and respiratory dysfunction, leading to neonatal lethality (S. Takeuchi, K. Takeda, I. Oishi, M. Nomi, M. Ikeya, K. Itoh, S. Tamura, T. Ueda, T. Hatta, H. Otani, T. Terashima, S. Takada, H. Yamamura, S. Akira, and Y. Minami, Genes Cells 5:71-78, 2000). Demonstration of the target molecule of a protective IgE antibody in secretory glands of Schistosoma japonicum larvae.

We have demonstrated that a mouse monoclonal IgE antibody, SJ18 epsilon.1, recognizes a 97 kDa surface molecule (Sj97) of Schistosoma japonicum larvae and that the antibody induces partial but significant protection against the skin to lung-stage of S. japonicum infection. The antibody stimulates eosinophil- and macrophage-mediated killing of schistosomula in vitro. In the present study, we isolated the putative full-length cDNA of Sj97 by screening a lambda gt11 cDNA library from S. japonicum adult worms with SJ18 epsilon.1. The predicted amino acid sequence of the cDNA showed highly signific

A phase 1 clinical trial of a multi-antigen SARS-CoV DNA vaccine as a booster after dose three spike-based mRNA vaccinations.

The COVID-19 pandemic was controlled by active vaccination, but emergence of viral mutations in the receptor-binding domain (RBD) of the spike (S) protein supported the need for vaccines targeting genetically stable viral domains. We developed a unique vaccine (OC-007) combining WH1-, Alpha-, and Beta-variant RBD loops; the membrane (M; WH1); and nucleoproteins (N; WH1). In a first-in-human, placebo-controlled, randomized, double-blind, dose-escalation phase 1 clinical trial, the OC-007 DNA vaccine was evaluated as one booster dose delivered intramuscularly by in vivo electroporation (EP) in 1…

Molecular diversity and evolutionary dynamics of phage receptor binding proteins in Staphylococcus aureus: implications for phage therapy and resistance.

Phage-host interactions are defined by the molecular specificity of receptor-binding proteins (RBPs), which determine the efficacy of infection and the host range. The genetic and structural variability of bacterial receptor molecules is essential for designing effective phage therapeutics and mitigating resistance. This study investigated four receptor-binding proteins, tarS, tarL, tagO, and tagG, in multiple Staphylococcus aureus isolates. Single-nucleotide polymorphism (SNP) analysis was performed to assess receptor diversity and evolutionary conservation. Molecular docking was conducted be

Immunoproteomic Identification of Scedosporium boydii Antigens with Potential Diagnostic Interest in Cystic Fibrosis Patients.

Fungal infections caused by Scedosporium/Lomentospora species are a significant threat to patients with cystic fibrosis (pwCF), ranking as the second most common filamentous fungi in their airways after Aspergillus. Current serodiagnostic methods, such as counterimmunoelectrophoresis and ELISA using crude extracts, lack standardization and commercial availability and often show cross-reactivity with other fungi. This study aimed to identify specific Scedosporium boydii antigens with potential for serodiagnosis in pwCF. An immunoproteomic approach combining two-dimensional polyacrylamide gel el

NARFL Knockout Triggers Ferroptosis-Driven Vascular Endothelial Dysfunction.

Nuclear prelamin A recognition factor-like (NARFL) is a core component of the cytosolic iron-sulfur (Fe-S) protein assembly (CIA) system. Yet, its role in vascular pathophysiology remains poorly defined. This study demonstrates that NARFL deficiency triggers ferroptosis, leading to severe vascular endothelial dysfunction across species. In zebrafish, narfl knockout causes embryonic lethality, accompanied by neurovascular defects, blood-brain barrier disruption, and aberrant hemodynamics. Similarly, knockout of the murine ortholog (Ciao3) results in mid-gestational embryonic lethality due to im

A New Paradigm of Transcriptional Regulation by the SufR-Like Iron-Sulfur Transcription Factors.

SufR is an iron-sulfur ([4Fe-4S]) cluster-containing transcription factor belonging to an uncharacterized domain family (COG2345). It has been shown to negatively regulate the sulfur utilization factor (SUF) Fe-S biogenesis system in several Gram-negative and Gram-positive bacteria including Cyanobacteria, Mycobacteria and Streptomyces. The structural basis for its DNA recognition and transcriptional regulation by the SufR-like proteins remains enigmatic. In this study, we present the cryo-EM structure of Mycobacterium tuberculosis SufR bound to its promoter, revealing a new domain architectur

CEP112 coordinates translational regulation of essential fertility genes during spermiogenesis through phase separation in humans and mice.

Spermiogenesis, the complex transformation of haploid spermatids into mature spermatozoa, relies on precise spatiotemporal regulation of gene expression at the post-transcriptional level. The mechanisms underlying this critical process remain incompletely understood. Here, we identify centrosomal protein 112 (CEP112) as an essential regulator of mRNA translation during this critical developmental process. Mutations in CEP112 are discovered in oligoasthenoteratospermic patients, and Cep112-deficient male mice recapitulate key phenotypes of human asthenoteratozoospermia. CEP112 localizes to the

Structural Evaluation of the Spike Glycoprotein Variants on SARS-CoV-2 Transmission and Immune Evasion.

The emergence of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) presents significant social, economic and political challenges worldwide. SARS-CoV-2 has caused over 3.5 million deaths since late 2019. Mutations in the spike (S) glycoprotein are of particular concern because it harbours the domain which recognises the angiotensin-converting enzyme 2 (ACE2) receptor and is the target for neutralising antibodies. Mutations in the S protein may induce alterations in the surface spike structures, changing the conformational B-cell epitopes and leading to a potential reduction in vacci
