Enhanced osteogenic differentiation and bone regeneration of poly(lactic-co-glycolic acid) by graphene via activation of PI3K/Akt/GSK-3β/β-catenin signal circuit.

The reconstruction of bone defects by guiding autologous bone tissue regeneration with artificial biomaterials is a potential strategy in the area of bone tissue engineering. The development of new polymers with good biocompatibility, favorable mechanical properties, and osteoinductivity is of vital importance. Graphene and its derivatives have attracted extensive interests due to the exceptional physiochemical and biological properties of graphene. In this study, poly(lactic-co-glycolic acid) (PLGA) films incorporated by graphene nanoplates were fabricated. The results indicated that the inco

Conjunctival-limbal autograft.

PURPOSE OF REVIEW: To provide an overview of conjunctival-limbal autografting (CLAU) for ocular surface rehabilitation with emphasis on more recent literature detailing outcomes of the procedure over the last 2 decades as well as technique variations and adjuvant techniques. RECENT FINDINGS: Limbal autografting initially described in 1964 by Barraquer and Strampelli and later popularized by Kenyon and Tseng is considered the best option for restoration of corneal phenotype in unilateral limbal stem-cell deficiency. Although there have been developments in alternative limbal epithelial stem-ce

Dual-acting biofunctionalised scaffolds for applications in regenerative medicine.

Off the shelf scaffolds for replacing ultra-small diameter vascular grafts are valuable for reconstruction of diseased or damaged vessels. The limitations for such grafts include optimal handling with ready availability of varied lengths of grafts, graft patency with the ability to replace the function of active cellular mechanisms and adequate mechanical properties to maintain physicochemical function. We used a well-established, solvent casting method for potential tissue replacement scaffold fabrication with incorporated bioactive molecules, which we have previously explored to confer haemo

Differential cell-matrix responses in hypoxia-stimulated aortic versus mitral valves.

Tissue oxygenation often plays a significant role in disease and is an essential design consideration for tissue engineering. Here, oxygen diffusion profiles of porcine aortic and mitral valve leaflets were determined using an oxygen diffusion chamber in conjunction with computational models. Results from these studies revealed the differences between aortic and mitral valve leaflet diffusion profiles and suggested that diffusion alone was insufficient for normal oxygen delivery in mitral valves. During fibrotic valve disease, leaflet thickening due to abnormal extracellular matrix is likely t

Microsphere-Incorporated Hybrid Thermogel for Neuronal Differentiation of Tonsil Derived Mesenchymal Stem Cells.

Neuronal differentiation of tonsil-derived mesenchymal stem cells (TMSCs) is investigated in a 3D hybrid system. The hybrid system is prepared by increasing the temperature of poly(ethylene glycol)-poly(l-alanine) aqueous solution to 37 °C through the heat-induced sol-to-gel transition, in which TMSCs and growth factor releasing microspheres are suspended. The in situ formed gel exhibits a modulus of 800 Pa at 37 °C, similar to that of brain tissue, and it is robust enough to hold the microspheres and cells during the 3D culture of TMSCs. The neuronal growth factors are released over 12-18 d,

Recent advances in bone regeneration using adult stem cells.

Bone is a highly vascularized tissue reliant on the close spatial and temporal association between blood vessels and bone cells. Therefore, cells that participate in vasculogenesis and osteogenesis play a pivotal role in bone formation during prenatal and postnatal periods. Nevertheless, spontaneous healing of bone fracture is occasionally impaired due to insufficient blood and cellular supply to the site of injury. In these cases, bone regeneration process is interrupted, which might result in delayed union or even nonunion of the fracture. Nonunion fracture is difficult to treat and have a h

Model mice for 15q11-13 duplication syndrome exhibit late-onset obesity and altered lipid metabolism.

Copy number variations on human chromosome 15q11-q13 have been implicated in several neurodevelopmental disorders. A paternal loss or duplication of the Prader-Willi syndrome/Angelman syndrome (PWS/AS) region confers a risk of obesity, although the mechanism remains a mystery due to a lack of an animal model that accurately recreates the obesity phenotype. We performed detailed analyses of mice with duplication of PWS/AS locus (6 Mb) generated by chromosome engineering and found that animals with a paternal duplication of this region (patDp/+) show late-onset obesity, high sensitivity for high

Sustained inflammation induces degeneration of the temporomandibular joint.

The temporomandibular joint (TMJ) undergoes degenerative changes among patients who suffer from arthritis, and yet the pathogenesis of TMJ osteoarthritis and rheumatoid arthritis is poorly understood. We hypothesized that sustained inflammation in the TMJ induces structural abnormalities, and accordingly characterized the disc and synovium in a novel model with double injections of complete Freund's adjuvant (CFA), using behavioral, morphological, cellular, and molecular assessments. Thirty-five days following double CFA injections in seven-week-old female Sprague-Dawley rats, the disc in the

Neurovirulent flavivirus can be attenuated in mice by incorporation of neuron-specific microRNA recognition elements into viral genome.

Engineering viruses by inserting microRNA (miRNA) recognition elements (MREs) into the 3'-untranslated region (3'-UTR) of viral RNA can efficiently restrict viral tissue tropism. We used the mosquito-borne Japanese encephalitis virus (JEV) to investigate whether endogenous neuron-specific microRNA-124 (miR-124) could be used to restrict viral neurotropism and, consequently, diminish the neurovirulence of JEV in mice. To recover a neuron-restricted JEV, we inserted 2 copies of a perfectly matched MRE specific to miR-124 into the 3'-UTR to create infectious JEV recombinant RP-124PT (rRP-124PT).

Bone formation and neovascularization mediated by mesenchymal stem cells and endothelial cells in critical-sized calvarial defects.

Bone represents a highly dynamic tissue whose development is strongly dependent on vasculogenic and angiogenic processes. Neovascularization also plays an important role in fracture healing and in tissue engineering applications aiming at restoring bone function. We have previously shown in a heterotopic subcutaneous implantation model of severe combined immunodeficiency (SCID) mice that implanted human umbilical vein endothelial cells (HUVECs) gave rise to the formation of a complex functional human neovasculature. In this study, we investigated the effect of HUVEC coimplantation on mesenchym

Collagen nerve conduits--assessment of biocompatibility and axonal regeneration.

Bridging of nerve gaps is still a major problem in peripheral nerve surgery. Alternatively to autologous nerve grafts tissue engineering of peripheral nerves focuses on biocompatible conduits to reconstruct nerves. Such non-neural conduits fail to support regeneration over larger gaps due to lacking viable Schwann cells that promote regeneration by producing growth factors and cell guiding molecules. This problem may be overcome by implantation of cultivated Schwann cells into suitable scaffolds. In the present experiments we tested a collagen type I/III tube as a potential nerve guiding matri

Development of a stable chemically defined surface for the culture of human keratinocytes under serum-free conditions for clinical use.

Within the field of tissue engineering there is a need to develop new approaches to achieve effective wound closure in patients with extensive skin loss or chronic ulcers. This article exploits the well-known interdependency of epithelial keratinocytes and stromal fibroblasts in conjunction with plasma surface technology. The aim was to produce a chemically defined surface, which with the aid of a feeder layer of lethally irradiated dermal fibroblasts would improve the attachment and proliferation of the keratinocyte cell from which subconfluent cells can be transferred to wound bed models. Anatomically Accessible Local mRNA Therapeutics: From Anatomical Access to Disease-Focused Applications.

Messenger RNA (mRNA) therapeutics have expanded from clinically validated prophylactic vaccines toward protein replacement and supplementation. Yet mRNA delivery remains centered on intramuscular vaccination and intravenous administration, largely optimized for systemic adaptive immunity or circulation-mediated biodistribution. Many localized diseases, however, are confined to tissues or compartments accessible by topical, transdermal, inhaled, oral, catheter-based, ocular, or intra-articular administration without initial reliance on circulation. For these diseases, the therapeutic objective

25+ Years of TRPV4: From Discovery to Translational Horizons.

Transient receptor potential vanilloid member 4 (TRPV4) is a calcium-permeable, nonselective cation channel first described in 2000. Polymodally-activated, also by osmotic, mechanical, thermal, actinic, and chemical stimuli, TRPV4 has emerged as a versatile molecular integrator across a remarkable diversity of cell lineages and vertebrate organ systems. Re orthology to invertebrate TRPV channels, TRPV4 rescued osmotic and mechanical sensing in ASH nociceptor neurons of C. elegans mutant for OSM-9 TRPV channels. This chapter surveys the arc of TRPV4 research from foundational cloning to transla

Vascularized composite xenotransplantation: Where we have been and where we could go?

Vascularized composite allotransplantation (VCA) enables functional and aesthetic reconstruction after complex tissue loss but remains limited by donor scarcity and immunosuppression. Xenotransplantation, using tissues from other species, offers a potential solution. While progress in solid organ xenografts has accelerated, vascularized composite xenotransplantation (VCX) remains largely experimental. This study presents the first systematic review of VCX, tracing its development over the past century and preclinical efforts to overcome immunologic and technical barriers. A systematic review f

Simulation-Based Trajectory for Non-Planar Scaffold Printing on Irregular Patches Using Robotic Arm.

This study proposes a reproducible and accessible methodological framework for non-planar path generation to enable scaffold biofabrication on irregular anatomical surfaces replicating the native morphology of human tissue. By integrating a simulation-based trajectory optimization system with a robotic arm, lattice paths are generated using an intersection-based method with parallel planes. This method is processed by intersecting the anatomical object with orthogonal planes, allowing for the creation of paths that conform to complex geometries. The proposed approach relies on widely available

Plant Regeneration: Influencing Factors, Regulatory Networks, and Epigenetic Mechanisms.

Plant tissue culture is a crucial part of biotechnology that supports crop improvement, plant conservation, and other related fields. Although tissue culture has been successfully used in many plants, low regeneration and transformation rates still exist in some species. Agrobacterium-mediated transformation is commonly used in genetic engineering, but its effectiveness depends heavily on establishing a reliable in vitro regeneration system through organogenesis or somatic embryogenesis. Over the past decade, substantial progress has been made in understanding the molecular basis of regenerati

Micro/Nanorobots (MNRs) at the Intersection of Mechanobiology and Artificial Intelligence: An Emerging Frontier for Precision Nanomedicine.

Micro/nanorobots (MNRs) are emerging as active precision-nanomedicine platforms that convert physical energy into propulsion and localized cellular or tissue mechanotransduction. Yet, the ways in which MNR-mediated forces and flows reprogram cell behavior, activate mechanotransduction pathways, or drive tissue remodeling remain poorly understood. Unlike passive nanocarriers that rely on diffusion and vascular permeability, MNRs enable directional transport, barrier penetration, and spatiotemporal control. Beyond locomotion, propulsion-induced stresses act as mechanical cues that engage cellula
