Cinnamon for Metabolic Diseases and Their Cardiovascular and Hepatic Complications: A Mechanistic Review.

Cinnamon is one of the world's oldest and most popular spices, and is derived from the inner bark of several tree species from the genus Cinnamomum. During the last two decades, cinnamon has demonstrated beneficial metabolic effects not only in animal experiments but also in clinical trials. Even recent meta-analyses have shown the protective effects of cinnamon on different components of metabolic syndrome and their complications. In the last 5 years, several experimental studies have unraveled the intricate molecular mechanisms underlying the antihypertensive, antihyperglycemic, lipid-loweri

Antimicrobial activity of cinnamaldehyde against multidrug-resistant Klebsiella pneumoniae: an in vitro and in vivo study.

The emergence and spread of multidrug-resistant (MDR) Klebsiella pneumoniae strains have increased worldwide, posing a significant health threat by limiting the therapeutic options. This study aimed to investigate the antimicrobial potential of cinnamaldehyde against MDR-K. pneumoniae strains in vitro and in vivo assays. The presence of resistant genes in MDR- K. pneumoniae strains were evaluated by Polymerase Chain Reaction (PCR) and DNA sequencing. Carbapenem-resistant K. pneumoniae strains show the blaKPC-2 gene, while polymyxin-resistant K. pneumoniae presented blaKPC-2 and alterations in

Rapid elimination of foodborne and environmental fungal contaminants by benzo analogs.

BACKGROUND: Contamination of food or the environment by fungi, especially those resistant to conventional fungicides or drugs, represents a hazard to human health. The objective of this study is to identify safe, natural antifungal agents that can remove fungal pathogens or contaminants rapidly from food and / or environmental sources. RESULTS: Fifteen antifungal compounds (nine benzo derivatives as candidates; six conventional fungicides as references) were investigated. Three benzo analogs, namely octyl gallate (OG), trans-cinnamaldehyde (CA), and 2-hydroxy-5-methoxybenzaldehyde (2H5M), at

Prevalence of antibiotic-resistant E. coli in broilers challenged with a multi-resistant E. coli strain and received ampicillin, an organic acid-based feed additive or a synbiotic preparation.

The aim of this study was to evaluate the effect of ampicillin, an organic acid-based feed additive and a synbiotic preparation on the prevalence of antibiotic-resistant E. coli in the ceca of broilers. A total of 2000 broiler chickens (Ross 708) were randomly assigned to 5 groups with 8 replicates. The negative control group was the only group that was not subjected to avian pathogenic E. coli challenge, while all the other 4 groups received a multi-resistant E. coli strain that was resistant to ampicillin, cephalexin, and nalidixic acid as an oral challenge. The second group served as a chal

Further development of LLNA:DAE method as stand-alone skin-sensitization testing method and applied for evaluation of relative skin-sensitizing potency between chemicals.

To date, there has been no well-established local lymph node assay (LLNA) that includes an elicitation phase. Therefore, we developed a modified local lymph node assay with an elicitation phase (LLNA:DAE) to discriminate true skin sensitizers from chemicals that gave borderline positive results and previously reported this assay. To develop the LLNA:DAE method as a useful stand-alone testing method, we investigated the complete procedure for the LLNA:DAE method using hexyl cinnamic aldehyde (HCA), isoeugenol, and 2,4-dinitrochlorobenzene (DNCB) as test compounds. We defined the LLNA:DAE proced

Effects of strain differences and vehicles on results of local lymph node assays.

The Local Lymph Node Assay (LLNA) is now regarded as the worldwide standard. The analysis of accumulated LLNA data reveals that the animal strains and vehicles employed are likely to affect LLNA results. Here we show that an obvious strain difference in the local lymph node response was observed between DMSO-treated CBA/CaOlaHsd and CBA/CaHsdRcc mice. We also show that a vehicle difference in the response was observed when CBA/CaHsdRcc mice were exposed to 6 vehicles; 4:1 v/v acetone/olive oil (AOO), ethanol/water (70% EtOH), N,N-dimethylformamide (DMF), 2-butanone (BN), propylene glycol (PG),

Conformational preferences of methacrolein in Diels-Alder and 1,3-dipolar cycloaddition reactions.

A B3LYP/6-31G* study has been carried out for the reactions of methacrolein with cyclopentadiene, parent nitrone, 1-pyrroline-1-oxide, and (Z)-C,N-diphenylnitrone, in which the coordination of a Lewis acid (borane) and the solvent polarity (dichloromethane) have been taken into account. Calculated activation parameters, regioselectivities (for 1,3-dipolar cycloaddition reactions), and endo/exo stereoselectivities show good agreement with available experimental data. Gas-phase calculations show a varied behavior of the s-cis/s-trans TS stability for noncatalyzed reactions (from the systematic s…

Perfume dermatitis.

The most common reaction to fragrance materials seen by practicing dermatologists is allergic contact dermatitis. Photodermatitis is occasionally seen, as is contact urticaria, irritation, and depigmentation. Fragrances are the leading cause of allergic contact dermatitis due to cosmetics. The fragrance mixture can cause false-positive reactions; therefore, it is more desirable to test with a separate series of fragrance materials.

The effect of quenching agents on contact urticaria caused by cinnamic aldehyde.

Inhibition (quenching) of contact sensitization by cinnamic aldehyde (CA) reportedly occurs from eugenol (E) and d-limonene (d-L). Experimentally the former inhibited nonimmunologic contact urticaria (NICU) from CA in seven of eleven test subjects, which prompted a search for possible mechanisms, including chemical interaction, altered absorption, and anti-inflammatory activity, and competitive inhibition. Mixtures of CA and E and CA and d-L showed no chemical changes or intermolecular bonding. Absorption was not increased by cellophane tape stripping, and neither E nor d-L inhibited urticaria

New Insights into the Pharmacological Effects and Potential Health Benefits of Volatile Phenylpropenes.

Phenylpropenes are a significant category of plant volatile compounds that play a crucial role in enhancing the flavor of a variety of herbs and spices. They are a subclass of phenylpropanoid compounds, being important constituents in several plant species like basil, clove, cinnamon, anise, sassafras, nutmeg, and fennel. The most common molecules from the phenylpropene group are represented by anethole, chavicol, cinnamaldehyde, estragole, eugenol, myristicin (methoxy-safrole), and safrole. Beyond their culinary and cosmetic applications, phenylpropenes have been found to have multiple pharma

Proteomics-Driven Molecular Mechanism of Cinnamaldehyde against Methicillin-Resistant Staphylococcus aureus Infection: Covalent Modifications of Sortase A and Aconitase.

Cinnamaldehyde (CA) from Cinnamomum cassia shows strong antimicrobial activity against MRSA. This study used proteomics and high-resolution mass spectrometry with an MRSA-infected cell model to explore CA's antimicrobial mechanisms. CA covalently modifies Sortase A's active site cysteine (Cys184), a key enzyme for anchoring virulence factors to the MRSA cell wall. This modification inhibits Sortase A's activity, blocking virulence factor anchoring and impairing MRSA's adhesion, infection, and immune evasion. CA also covalently modifies Aconitase's [4Fe-4S] cluster active sites (Cys509, Cys512)

Antimicrobial efficacy of cinnamaldehyde combined with polymyxin B against carbapenem- polymyxin-resistant Klebsiella pneumoniae.

The global spread of multi-drug-resistant bacteria, including strains resistant to both polymyxin and carbapenems, poses a significant threat to public health, highlighting the need to develop new antimicrobial agents. This study assessed the antimicrobial potential of cinnamaldehyde, alone and in combination with antibiotics, against carbapenem-polymyxin-resistant K. pneumoniae strains (CPR-Kp). The antimicrobial activity was assessed through minimum inhibitory concentration (MIC), checkerboard assay, and survival curve analysis. Cinnamaldehyde showed inhibitory effects (MIC 281 µg/mL), and w

Adaptation to cinnamaldehyde shapes Pseudomonas aeruginosa resistance to major antibiotics.

In France, the use of essential oils to treat bacterial infections is common, with approximately 40% of cystic fibrosis patients regularly using these natural products to control infections caused by Pseudomonas aeruginosa often in combination with their antibiotic treatments. Previous research has demonstrated that prolonged exposure of P. aeruginosa PA14 to cinnamaldehyde (CNA), the main component of cinnamon essential oil, can select for mutants resistant to β-lactams through overproduction of the MexAB-OprM efflux pump, some of which are also hypersusceptible to aminoglycosides and colisti

Cinnamaldehyde attenuates CCL4-induced liver fibrosis by inhibiting the CYP2A6/Notch3 pathway.

BACKGROUND: Hepatic stellate cells (HSCs) activation and hepatocyte injury contribute to liver fibrosis progression and subsequent cirrhosis. Literature showed that cinnamaldehyde (CA) could alleviate fibrosis procession and steatosis. However, its specific role in liver fibrosis remains largely unexplored. MATERIALS AND METHODS: Liver fibrosis was induced in vivo, and CA was administered for 4 weeks. Liver inflammation, fibrosis, apoptosis, and proliferation were evaluated using histological, western blotting, and immunohistochemistry. CYP2A6 and Notch3 expression levels were also measured.

Antimicrobial gelatin-based films with cinnamaldehyde and ZnO nanoparticles for sustainable food packaging.

Cinnamaldehyde (CIN), a harmless bioactive chemical, is used in bio-based packaging films for its antibacterial and antioxidant properties. However, high amounts can change food flavor and odor. Thus, ZnO nanoparticles (NPs) as a supplementary antimicrobial agent are added to gelatin film with CIN. The CIN/ZnO interactions are the main topic of this investigation. FTIR-Attenuated Total Reflection (ATR), X-ray diffraction (XRD), and scanning electron microscopy (SEM) were utilized to investigate CIN/ZnO@gelatin films. Transmission electron microscope (TEM) images revealed nanospheres morphology

Cinnamaldehyde and baicalin reverse colistin resistance in Enterobacterales and Acinetobacter baumannii strains.

PURPOSE: Colistin is used as a last resort antibiotic against infections caused by multidrug-resistant gram-negative bacteria, especially carbapenem-resistant bacteria. However, colistin-resistance in clinical isolates is becoming more prevalent. Cinnamaldehyde and baicalin, which are the major active constituents of Cinnamomum and Scutellaria, have been reported to exhibit antibacterial properties. The aim of this study was to evaluate the capacity of cinnamaldehyde and baicalin to enhance the antibiotic activity of colistin in Enterobacterales and Acinetobacter baumannii strains. METHODS: T

Novel chitosan-based nanocomposites as ecofriendly pesticide carriers: Synthesis, root rot inhibition and growth management of tomato plants.

Novel bio-based nanocomposites were developed as carriers for loading and sustained-release of vanillin (Van.) and cinnamaldehyde (Cinn.) antioxidants. The composites were obtained by intercalation of chitosan (CS) into sodium montmorillonite (CS/Mt), incorporation of chitosan with polyaniline (CS/PANI) and chitosan/polyaniline/exfoliated montmorillonite (CS/PANI/Mt). The structure and morphology of composites were characterized by FTIR, XRD, SEM and TEM. The release data of Van. and Cinn. from CS and CS/Mt obeyed well zero-order equation. However, Higuchi and Korsmeyer-Peppas models fitted we

Genomic and transcriptomic analysis of a coniferyl aldehyde-resistant Saccharomyces cerevisiae strain obtained by evolutionary engineering.

Phenolic inhibitors in lignocellulosic hydrolysates interfere with the performance of fermenting microorganisms. Among these, coniferyl aldehyde is one of the most toxic inhibitors. In this study, genetically stable Saccharomyces cerevisiae mutants with high coniferyl aldehyde resistance were successfully obtained for the first time by using an evolutionary engineering strategy, based on the systematic application of increasing coniferyl aldehyde stress in batch cultures. Among the selected coniferyl aldehyde-resistant mutants, the highly resistant strain called BH13 was also cross-resistant t
