Improving the extraction efficiency on the determination of aflatoxin B1 and other mycotoxins in corn gluten feed - analysis by LC-ID-MS/MS.

Corn gluten feed (CGF) is a side-product from the manufacture of corn starch from maize grain by wet-milling. This process accumulates the mycotoxins in the CGF, moreover it gives an unique composition to CGF distinct from any other regular feed matrix for which mycotoxin analytical methods have been standardized. In this study, we report an optimised extraction medium for the determination of aflatoxin B1 (AFB1) and other mycotoxins in naturally contaminated CGF samples. Central composite experimental designs (CCDs) were utilised to find the appropriate extraction solution composition for the

Two birds with one stone: A hollow MnO2nanozyme immunosensor for simultaneous accurate detection and highly efficient removal of aflatoxin B1in peanuts.

Aflatoxin B1(AFB1), a highly toxic mycotoxin, is widely prevalent in mold-contaminated grain crops and poses serious health risks to humans and animals, even at trace levels. Owing to its high toxicity and stable chemical structure, AFB1 is difficult to specifically detect and remove using conventional methods, leading to substantial annual losses of grain crops due to contamination. Therefore, there is an urgent need to develop sensitive and on-site strategies for the early warning and effective decontamination of AFB1. To address this challenge, we synthesized a dual-functional hollow mesopo

A dual-functional magnetic-fluorescent nanobead-based multi-color lateral flow immunoassay for simultaneous detection of aflatoxin B1 and zearalenone.

Herein, we developed a lateral flow immunoassay (LFIA) based on dual-functional magnetic-fluorescent nanobeads for the simultaneous detection of aflatoxin B1 (AFB1) and zearalenone (ZEN). Magnetic quantum dot nanobeads (MQBs) were synthesized by encapsulating CdSe/ZnS quantum dots and iron oxide nanoparticles into polystyrene nanobeads. These MQBs exhibit excellent superparamagnetic and fluorescent properties, enabling the dual functions of target purification and signal output in the LFIA, effectively reducing matrix interference. Further, a multi-color LFIA was developed with red and green M

Fe3O4@Au magnetic SERS nanotags-based vertical flow immunoassay for simultaneous detection of fumonisin B1, aflatoxin B1 and deoxnivalenol.

Agricultural products are usually contaminated by various mycotoxins, the rapid and sensitive detection of mycotoxins at the early stage is significant for food safety and human health. Here, we develop a rapid and ultrasensitive surface-enhanced Raman scattering-based vertical flow assay (VFA) for simultaneous detection of multiple mycotoxins by using Fe3O4@Au magnetic SERS nanotags. In this competitive immunoassay, three test zones are designed on the sensing membrane, and modified with multiple capture antigens. Moreover, three kinds of specific monoclonal antibodies conjugated Fe3O4NBA@Au,

Spatial transcriptomics unravels the interactive network of aflatoxin B1-driven gastric cancer: Multi-omics integration of transcriptome and mendelian randomization.

The escalating threat of widespread exposure to environmental pollutants on human health has drawn increasing attention, particularly regarding mycotoxin contaminants with potent carcinogenic properties. As a prototypical environmental toxin, the molecular mechanisms underlying aflatoxin B1-induced gastric carcinogenesis remain poorly defined. This study systematically elucidates the molecular network through which aflatoxin B1 (AFB1) synergistically drives the malignant progression of gastric cancer by specifically targeting five core regulatory genes, exemplified by its upregulation of cycli

Novel ultrafine Pt@Fe-MIL-101 nanozyme enables robust aflatoxin B1 immunoassay in diverse marine and agricultural systems.

BACKGROUND: Aflatoxin B1 (AFB1) stands among the most toxic naturally occurring substances, with its acute toxicity characterized by the induction of acute hepatic necrosis, hemorrhage, and even fatal outcomes, thereby posing a profound threat to human health. Contamination of AFB1 in food commodities can arise at multiple stages throughout the production cycle, including cultivation, storage, and processing. This contamination cascade permeates the entire food supply chain, encompassing primary agricultural products as well as a diverse range of processed food items. Consequently, the develop

Hydroalcoholic Extract of Saffron Petals, Yeast Cell Wall and Bentonite Reduce Contamination Effects With Aflatoxin B1 and Ochratoxin A in Exposed Broilers.

BACKGROUND: Contamination is not surprising in light of the ubiquitous nature of the fungi that produce aflatoxin and ochratoxin A. The presence of these toxins in the broiler diet leads to increased losses, an increased feed conversion ratio, and decreased productivity. OBJECTIVES: This study aimed to investigate the effects of the use of hydroalcoholic extracts of saffron petals, yeast cell walls and bentonite in the diets of broiler chickens contaminated with aflatoxin B1 and ochratoxin A. METHODS: In a completely randomised design, 350 one-day-old Ross 308 broilers were allocated into se

Aflatoxin B1-induced hepatotoxicity through mitochondrial dysfunction, oxidative stress, and inflammation as central pathological mechanisms: A review of experimental evidence.

Aflatoxin B1 (AFB1) is a class of mycotoxin known to contaminate agricultural products, animal feed and animal food products, subsequently causing detrimental effects on human and animal health. AFB1 is the most common and potent aflatoxin found in food and contributes significantly to liver injury as well as the development of hepatocellular carcinoma. Although the liver is a primary target organ for AFB1 toxicity and biotransformation, underlying mechanisms implicated in liver injuries induced by these mycotoxins remain to be fully elucidated for therapeutic purposes. This review aims to dis

Exploring the Impact of Efavirenz on Aflatoxin B1 Metabolism: Insights from a Physiologically Based Pharmacokinetic Model and a Human Liver Microsome Study.

Physiologically based pharmacokinetic (PBPK) models were utilized to investigate potential interactions between aflatoxin B1 (AFB1) and efavirenz (EFV), a non-nucleoside reverse transcriptase inhibitor drug and inducer of several CYP enzymes, including CYP3A4. PBPK simulations were conducted in a North European Caucasian and Black South African population, considering different dosing scenarios. The simulations predicted the impact of EFV on AFB1 metabolism via CYP3A4 and CYP1A2. In vitro experiments using human liver microsomes (HLM) were performed to verify the PBPK predictions for both sing

New insights into the combined effects of aflatoxin B1 and Eimeria ovinoidalis on uterine function by disrupting the gut-blood-reproductive axis in sheep.

BACKGROUND: Sheep coccidiosis is an infectious parasitic disease that primarily causes diarrhea and growth retardation in young animals, significantly hindering the development of the sheep breeding industry. Cereal grains and animal feeds are frequently contaminated with mycotoxins worldwide, with aflatoxin B1 (AFB1) being the most common form. AFB1 poses a serious threat to gastrointestinal health upon ingestion and affects the function of parenteral organs, thus endangering livestock health. However, the impact of the combined effects of coccidia and AFB1 on the reproductive system of sheep

A Novel Multicellular Placental Barrier Model to Investigate the Effect of Maternal Aflatoxin B1 Exposure on Fetal-Side Neural Stem Cells.

Ingestion of food toxins such as aflatoxin B1 (AFB1) during pregnancy may impair fetal neurodevelopment. However, animal model results may not be accurate due to the species' differences, and testing on humans is ethically impermissible. Here, we developed an in vitro human maternal-fetal multicellular model composed of a human hepatic compartment, a bilayer placental barrier, and a human fetal central nervous system compartment using neural stem cells (NSCs) to investigate the effect of AFB1 on fetal-side NSCs. AFB1 passed through the HepG2 hepatocellular carcinoma cells to mimic the maternal

Ex Vivo and In Vitro Studies Revealed Underlying Mechanisms of Immature Intestinal Inflammatory Responses Caused by Aflatoxin M1 Together with Ochratoxin A.

Aflatoxin M1 (AFM1) and ochratoxin A (OTA), which are occasionally detected in milk and commercial baby foods, could easily enter and reach the gastrointestinal tract, posing impairment to the first line of defense and causing dysfunction of the tissue. The objective of this study was to investigate the immunostimulatory roles of individual and combined AFM1 and OTA on the immature intestine. Thus, we used ELISA assays to evaluate the generation of cytokines from ex vivo CD-1 fetal mouse jejunum induced by AFM1 and OTA and explored the related regulatory pathways and pivot genes using RNA-seq

Aflatoxin B1 Induces Intestinal Barrier Dysfunction by Regulating the FXR-Mediated MLCK Signaling Pathway in Mice and in IPEC-J2 Cells.

Aflatoxin B1 (AFB1) is a widespread mycotoxin in food and feed. Although the liver is the main target organ of AFB1, the intestine is the first exposure organ to AFB1. However, the mechanism by which AFB1 induced intestinal barrier dysfunction via regulating the farnesoid X receptor (FXR)-mediated myosin light chain kinase (MLCK) signaling pathway has rarely been studied. In vivo, AFB1 exposure significantly decreased the small intestine length and increased the intestinal permeability. Meanwhile, AFB1 exposure markedly suppressed the protein expressions of FXR, ZO-1, occludin, and claudin-1 a

Route exposure and adverse effects monitoring of Aflatoxin B1 in the workers of wet waste management, the role of body redox system modulation.

Exposure to dust, containing different fungi metabolites such as aflatoxins is a risk factor for developing liver and kidney health abnormalities. Occupational evaluation of the aflatoxin's exposure-induced health abnormalities should include the monitoring of bioaerosols in the workplace and personal air, and applying of appropriate blood biomarkers to assess Aflatoxin B1 (AFB1) detrimental effects on a worker's health. However, to the best of our knowledge, these appropriate methods, especially determining the associated-adverse effects on health, following exposure, haven't been well docume…

Development of a ZnCdS@ZnS quantum dots-based label-free electrochemiluminescence immunosensor for sensitive determination of aflatoxin B1 in lotus seed.

In this study, we designed a ZnCdS@ZnS quantum dots (QDs)-based label-free electrochemiluminescence (ECL) immunosensor for sensitive determination of aflatoxin B1 (AFB1). A Nafion solution assembled abundant QDs on the surface of a Au electrode as ECL signal probes, with specially coupled anti-AFB1 antibodies as the capturing element. As the reduction reaction between S2O82- in the electrolyte and QDs on the electrode led to ECL emission, the decreased ECL signals resulting from target AFB1 in the samples were recorded for quantification. We evaluated electrochemical impedance spectroscopy and

Temporal Variation and Association of Aflatoxin B₁ Albumin-Adduct Levels with Socio-Economic and Food Consumption Factors in HIV Positive Adults.

The association between aflatoxin exposure and alteration in immune responses observed in humans suggest that aflatoxin could suppress the immune system and work synergistically with HIV to increase disease severity and progression to AIDS. No longitudinal study has been conducted to assess exposure to aflatoxin (AF) among HIV positive individuals. We examined temporal variation in AFB₁ albumin adducts (AF-ALB) in HIV positive Ghanaians, and assessed the association with socioeconomic and food consumption factors. We collected socioeconomic and food consumption data for 307 HIV positive antire
