Clonal expansion of the germinal center B cells of human reactive lymph nodes was analyzed. By micromanipulation, 28 germinal centers were microdissected from three nonneoplastic lymph nodes that had been fixed with formalin. Immunoglobulin heavy chain variable (V) region gene rearrangement was examined by seminested polymerase chain reaction (PCR) using two sets of primers (FR2-J and FR3A-J). An oligoclonal development (one to five clones) was found in each germinal center. Depending on the primer used, four or five (16%) of the germinal centers showed a single rearrangement band. The average

A study was made of 29 patients with monoclonal gammopathies to detect aberrations in immunoglobulin (Ig) light chain isotype expression in lymphocytes at various levels of B-cell differentiation, namely, circulating surface Ig positive (SIg+) cells, Ig-secreting cells (plaque forming cells, PFC) and mitogen-induced PFC. By using kappa-lambda analysis, two major phenotypes of aberrant Ig light chain isotype expression were found in circulating B cells at these three levels of differentiation: an absolute increase in B cells bearing the same Ig light chain isotype as that of monoclonal protein

We studied the effects of exogenous melatonin (administrated ad libitum at a concentration of 4 mg/liter in drinking water for 4 weeks) on the differentiation of B lymphocytes (PAX-5+) and apoptosis (Casp3+) of spleen cells in outbred laboratory mice kept under conditions of natural photoperiod, constant darkness, or constant illumination. Constant darkness increased the number of B-lymphocyte precursors and caspase-3 lymphocytes in the lymphoid follicles of the spleen. Constant light exposure reduced the number of B lymphocyte precursors, increased the number of Casp3+ lymphocytes and the con

The evolutionary origin of vertebrate adaptive immunity has been a longstanding problem in evolutionary biology. The diversified proteins that orchestrate this complex system are present throughout the jawed vertebrates but are absent in extant jawless vertebrates and invertebrates. From work initially carried out in the sea lamprey (Petromyzon marinus) and subsequently extended to other lampreys and hagfishes, we now know that, instead of immunoglobulin domain-based receptors, jawless vertebrates have leucine-rich repeat-based variable lymphocyte receptors (VLRs). Like immunoglobulins (Igs) a

CD22 (also known as Siglec-2) is a member of the Siglec family of glycan-recognition proteins and functions as a negative regulator of the B-cell receptor-mediated calcium signaling. Although the low level of CD22 expression on the B cells in patients with chronic lymphocytic leukemia (CLL) has been documented, CD22's role and its downregulation mechanism in CLL are yet to be fully studied. In this study, we confirmed that the surface CD22 protein and its mRNA are downregulated in the B cells of CLL patients. We analyzed a public transcriptomic dataset and found that the CD22 mRNA level is neg

Tuberculosis (TB), an infectious disease caused by the bacterium Mycobacterium tuberculosis (Mtb), is a major cause of morbidity and mortality worldwide. Annually, millions of new cases of tuberculosis are documented. Research in tuberculosis-immune has hitherto focused predominantly on the role of T cells in Mtb infection. Although there have been studies in progress which have supported the notion that B cells are crucial players in combating infectious diseases, the role of B cells in TB is still not fully understood. There is a paucity of in-depth analysis of various B lymphocyte subpopula

Aberrant signal transduction through the B cell receptor (BCR) plays a critical role in the pathogenesis of chronic lymphocytic leukemia (CLL)/small lymphocytic lymphoma (SLL). BCR-dependent signaling is necessary for the growth and survival of neoplastic cells, making inhibition of down-stream pathways a logical therapeutic strategy. Indeed, selective inhibitors against Bruton's tyrosine kinase (BTK) and phosphoinositide 3-kinase (PI3K) have been shown to induce high rates of response in CLL and other B cell lymphomas. In particular, the development of BTK inhibitors revolutionized the treatm

BACKGROUND: Parkinson's disease (PD) is associated with peripheral inflammation and abnormal peripheral blood lymphocyte immune responses. Peripheral blood B-lymphocyte subset distributions and whether they are associated with PD are unclear.
METHODS: Sixty-one PD patients and sixty-one one-to-one paired healthy controls (HCs) were enrolled. We used flow cytometry to perform immunophenotyping of peripheral B-lymphocyte, in vitro stimulation and measured serum cytokine. The relationship between variables and PD were assessed.
RESULTS: The percentage of naive B cells in blood of PD patients wa

In recent years significant progress has been made in the clinical management of chronic lymphocytic leukemia (CLL) as well as other B-cell malignancies; targeting proximal B-cell receptor signaling molecules such as Bruton Tyrosine Kinase (BTK) and Phosphoinositide 3-kinase (PI3Kδ) has emerged as a successful treatment strategy. Unfortunately, a proportion of patients are still not cured with available therapeutic options, thus efforts devoted to studying and identifying new potential druggable targets are warranted. B-cell receptor stimulation triggers a complex cascade of signaling events t

A 77-year-old man was referred to our department with a 6-month history of a painless, rapidly enlarging tumor on the left leg. Fourteen years ago, he was diagnosed with chronic lymphocytic leukemia (CLL), and he had presented with lymphocytosis and multiple lymphadenopathies (Rai stage II). Both bone marrow aspiration and peripheral blood immunophenotyping identified a monoclonal B-cell population expressing surface CD5, CD20, CD23, CD43, and IgG kappa. At that time, he underwent eight courses of treatment with oral fludarabine, followed by complete remission, and he had been clinically stabl

Chronic lymphocytic leukaemia (CLL) has long been thought to be an immunosuppressive disease and abnormalities in T-cell subset distribution and function have been observed in many studies. However, the role of T cells (if any) in disease progression remains unclear and has not been directly studied. This has changed with the advent of new therapies, such as chimeric antigen receptor-T cells, which actively use retargeted patient-derived T cells as "living drugs" for CLL. However complete responses are relatively low (~26%) and recent studies have suggested the differentiation status of patien

UNLABELLED: In this review, we discuss the intricate roles of the Wnt signalling network in the development and progression of mature B-cell-derived haematological malignancies, with a focus on chronic lymphocytic leukaemia (CLL) and related B-cell lymphomas. We review the current literature and highlight the differences between the β-catenin-dependent and -independent branches of Wnt signalling. Special attention is paid to the role of the non-canonical Wnt/planar cell polarity (PCP) pathway, mediated by the Wnt-5-receptor tyrosine kinase-like orphan receptor (ROR1)-Dishevelled signalling axi

Haemophagocytic lymphohistiocytosis (HLH) is a rare, life-threatening syndrome characterised by defective cytotoxic function and hypercytokinaemia leading to macrophage expansion and haemophagocytosis. Patients often present with unexplained fevers, hepatosplenomegaly and pancytopenia, with elevation in serum ferritin and triglyceride. Acquired forms are triggered by infection, malignancy or rheumatological disorders. HLH in the setting of chronic lymphocytic leukaemia is rarely reported, however, and is usually associated with infection or as a consequence of chemotherapy. We present a case o

BACKGROUND: B lymphocyte stimulator (BLyS/BAFF) is an endogenous protein that plays an important role in the differentiation and maduration of B lymphocytes. Enhanced levels of BLyS have been reported in lupus and other rheumatic diseases.
METHODS: Serum samples from 92 lupus patients (94% females, median age 35.5 years) and 106 controls (50 healthy donors, 38 with rheumatoid arthritis, 18 with scleroderma) were analyzed for BLyS. The cutoff of BLyS ˃1.98 ng/ml corresponds to the 95th percentile from the healthy donors. Antibodies against native DNA and disease activity also were evaluated in

Survival and progression of patients with mature B-cell malignancies, including chronic lymphocytic leukemia and non-Hodgkin's lymphoma, depend on signals from the B -cell receptor, and Bruton's tyrosine kinase(BTK) is a critical signaling kinase in this pathway. Ibrutinib is a novel and selective inhibitor of BTK, inactivating the kinase irreversibly. It has shown exciting results in early clinical trials. In this review, the application and molecular mechanism of Ibrutinib in the treatment of B-cell malignancies are discussed.

B-cell chronic lymphocytic leukemia (B-CLL) is a low-grade lymphoproliferative disease. Cutaneous involvement of B-CLL is limited and, in most cases, it represents non-specific manifestations related to an impaired immune system. Leukemic skin infiltrates (leukemia cutis) occur in 4-20% of patients. Herein we report the case of a 65-year-old woman with B-CLL presenting with papular, nodular, and plaque skin infiltrates affecting the nose, mimicking granulomatous rosacea. We discuss several aspects of rare cutaneous manifestations of B-CLL involving the face.

B-cell receptor (BCR) signaling is a central pathologic mechanism in B-cell malignancies, including chronic lymphocytic leukemia (CLL), in which it promotes leukemia cell survival and proliferation, and modulates CLL cell migration and tissue homing. BCR signaling now can be targeted with new, small molecule inhibitors of the spleen tyrosine kinase (Syk), Bruton's tyrosine kinase (Btk), or phosphoinositide 3'-kinase (PI3K) isoform p110δ (PI3Kδ), which have recently entered the clinical stage and show promising results in patients with CLL. During the first weeks of therapy, these agents charac
