Influence of long-term temperature stress on respiration frequency, Na+/K+-ATPase activity, and lipid metabolism in common carp (Cyprinus carpio).

Temperature is a key factor that influences fish metabolism. Therefore, it is important to understand how temperature variation affects lipid metabolism and energy consumption in fish. We determined respiration frequency, Na+/K+-ATPase activity, and lipid metabolism in common carp (Cyprinus carpio) after 18 days of temperature stress (5 °C and 30 °C, with 17 °C as the control). We found that respiratory frequency was positively correlated with water temperature (p < 0.01), whereas red blood cell count and mean corpuscular hemoglobin content and concentration were negatively correlated with tem

Molecular cloning, characterization and expression of immunoglobulin D on pathogen challenge and pathogen associated molecular patterns stimulation in freshwater carp, Catla catla.

The primordial immunoglobulin class, IgD, was the first non-IgM isotype discovered in teleosts. The crucial roles of IgM and IgZ in imparting systemic and mucosal immunity, respectively, in various fish species have been widely established. However, the putative function of a unique IgD isotype during pathogenic invasions has not been well explored. The present study reports the existence of an IgD ortholog in freshwater carp, Catla catla, and further evaluates its differential expression profile in response to bacterial, parasitic and viral antigenic exposure and pathogen associated molecular

Tyrosine hydroxylase in the olfactory system, forebrain and pituitary of the Indian major carp, Cirrhinus cirrhosus: organisation and interaction with neuropeptide Y in the preoptic area.

Dopamine (DA) inhibits, whereas gonadotrophin-releasing hormone (GnRH) stimulates, luteinisiing (LH) cells in the pituitary of some but not all teleosts. A reduction in the hypophysiotropic dopaminergic tone is necessary for the stimulatory effect of GnRH on LH cells. Neuropeptide Y (NPY) has emerged as one of the potent, endogenous agent that modulates LH secretion directly or indirectly via GnRH. Involvement of NPY in the regulation of hypophysiotropic DA neurones, however, is not known, but there is good evidence suggesting an interaction in the mammalian hypothalamus. DA neurones, identifi

Anti-CarP antibodies in two large cohorts of patients with rheumatoid arthritis and their relationship to genetic risk factors, cigarette smoking and other autoantibodies.

INTRODUCTION: In rheumatoid arthritis (RA), several genetic risk factors and smoking are strongly associated with the presence of anticitrullinated protein antibodies (ACPA), while much less is known about risk factors for ACPA-negative RA. Antibodies against carbamylated proteins (anti-CarP) have been described in both ACPA-positive and ACPA-negative RA patients. In this study, we have analysed the relationships among anti-CarP antibodies, ACPA, genetic risk factors (HLA-DRB1 alleles and PTPN22) and smoking in RA. METHODS: Presence of antibodies to carbamylated fetal calf serum (CarP-FCS) an

Molecular characterization of cholecystokinin in grass carp (Ctenopharyngodon idellus): cloning, localization, developmental profile, and effect of fasting and refeeding on expression in the brain and intestine.

Cholecystokinin (CCK) is a multi-functional brain-gut peptide in fish and mammals. To investigate the role of CCK in appetite regulation in fish, a 770-bp full-length cDNA sequence of CCK gene was obtained by RT-PCR and rapid amplification of cDNA ends methods in grass carp Ctenopharyngodon idellus. Homology analysis showed that the CCK cDNA sequence of grass carp had the highest similarity (90 %) to that of goldfish Carassius auratus and a higher similarity (>70 %) to those of other teleosts than to mammals. The PCR amplification using genomic DNA identified that the CCK gene of grass carp wa

Application of hazard analysis critical control point (HACCP) as a possible control measure for Opisthorchis viverrini infection in cultured carp (Puntius gonionotus).

Opisthorchiasis due to Opisthorchis viverrini and transmitted through infected freshwater cyprinoid fish (carps) affects more than 8 million people in Thailand, People's Democratic Republic of Lao, and Vietnam. The Hazard Analysis Critical Control Point (HACCP)-concept has been recommended by FAO and WHO to be included in programs to control foodborne trematode infections (FBT). HACCP is a multifactorial approach to control food hazards through surveillance of diseases, foods, and operations and education. This study describes the first attempt to apply HACCP to the prevention and control of O…

Phenotype-Anchored Multi-Omics Reveals Mechanisms Underlying Starvation-Induced Body, Liver, and Gallbladder Discoloration in Rock Carp (Procypris rabaudi).

Visible changes in body and internal organ coloration can serve as practical indicators of nutritional and metabolic stress in aquaculture, but their physiological meaning remains poorly understood. Here, we used starvation as a controlled nutritional stress model to investigate the physiological and molecular changes associated with body color fading and liver and gallbladder discoloration in rock carp (Procypris rabaudi). To assess the effects of food deprivation (0-35 days), fish were examined using growth and biochemical assays, intestinal histology, as well as integrated hepatic transcrip

Chitosan‑coated Green‑synthesized ZnO Nanoparticles Attenuate Heavy Metal Co‑accumulation in Common Carp (Cyprinus carpio, Linnaeus, 1758).

The co-occurrence of engineered nanoparticles and heavy metals in aquatic environments poses critical unknowns. Common carp (Cyprinus carpio) were exposed for 28 days to a Cu/Pb/Cd/Cr mixture alone, with commercial ZnO nanoparticles, or with a chitosan-coated ZnO nanocomposite (CZN). Metal bioaccumulation in gill and liver was quantified. Commercial ZnO markedly enhanced gill‑specific accumulation of Pb, Cr, and Zn versus metals alone. Conversely, CZN substantially lowered these elements and reduced Cd below the detection limit in both tissues. Although tissue‑dependent trends were noted, mult

Dietary Prickly Ash Seed Meal Suppresses Hepatic Lipogenesis and Improves Fillet Fatty Acid Profiles in Grass Carp (Ctenopharyngodon idella).

Fishmeal shortage requires replacing fishmeal with plant proteins in aquafeeds. Over-fatness of viscera degrades the efficiency and quality of the fillet in farmed grass carp. Prickly ash seed meal (PASM) provides both high levels of PUFAs and bioactives, but its potential is still unevaluated for lipid regulation and fillet nutrition of this species. We hypothesised that dietary PASM would reduce liver lipid production and enhance fillet fatty acid composition without compromising growth. Five isonitrogenous and isolipidic diets were formulated with PASM at graded inclusion level: PAS 0 (cont

An aquatic probiotic alleviates lead toxicity in grass carp (Ctenopharyngodon idella) via improved intestinal barrier function and reduced Pb bioaccumulation.

Probiotics have been used to mitigate heavy metals (HMs) toxicity, however, the mechanisms by which they alleviate HMs toxicity by regulating intestinal microbiota remain unclear in aquatic animals. In this study, the aquatic probiotic Rhodobacter sphaeroides SC01 (RSSC01) was supplemented in the diet of grass carp to investigate its roles in alleviating lead (Pb) toxicity. The results showed that RSSC01 significantly reduced Pb content in the intestine, gills, and kidneys by 25.27%, 55.83%, and 47.42%, respectively, compared with Pb exposure alone. Additionally, it effectively conferred Pb-in

Precision silver carp allergy profiling and rapid IgE epitope mapping by a microfluidic chip-MALDI-TOF MS system and prediction models.

Food allergies are increasing at an alarming rate, and it has become necessary to identify patient-specific allergens for personalized allergy immunotherapy treatments. Herein, a precision fish allergy test was established for patient-specific allergen identification and IgE epitope mapping in silver carp by employing an integrated microfluidic chip-MALDI-TOF MS system. Tosyl-activated magnetic beads (MGBs) were functionalized with mouse monoclonal anti-human immunoglobulin E (IgE) antibodies and patient serum to form IgE-MGBs. Integrated Transcriptomic and Proteomic Analyses Reveal Stage-Dependent Gill Remodeling Under Chronic Salinity Stress in Grass Carp (Ctenopharyngodon idella).

The expansion of brackish-water aquaculture for freshwater species such as grass carp (Ctenopharyngodon idella) necessitates a mechanistic understanding of their physiological and molecular responses to elevated salinity. Here, we performed integrated histopathological, transcriptomic and proteomic analyses of gill tissues from grass carp chronically exposed to 7‰ and 14‰ salinity to elucidate stage-specific responses to prolonged salinity stress. At moderate salinity (7‰), gill responses were characterized by epithelial stabilization, activation of adhesion-related pathways, enhanced carbohydr

Novel Method for Preparing Pseudo-Male Common Carp by Inducing Secondary Female-to-Male Sex Reversal Under High Temperature Conditions.

For economically viable fish species that exhibit sexual dimorphism in growth performance, such as common carp, cultivating monosex populations is an effective approach to enhance aquaculture yields. This method for producing an all-female carp population involves inducing pseudo-male carp through exogenous androgen/estrogen inhibitor administration, followed by crossing these pseudo-males with wild-type female carp to generate all-female offspring. However, the sex reversal effect induced by exogenous androgen/estrogen inhibitors is unstable, making the maintenance of these pseudo-male broods

Chinese yam polysaccharide induces the differentiation and natural antibody secretion of IgM+ B cells to prevent Aeromonas hydrophila infection in grass carp.

Chinese yam polysaccharide (CYP) is an effective immunostimulant, however, its efficacy in grass carp, an important commercial fish species in Asia, remains untested. Here, our study evaluated the immunostimulatory effects of CYP on IgM+ B cells in vitro and on humoral immunity and immune defense against Aeromonas hydrophila infection in vivo. In vitro stimulation experiments showed that CYP could induce the secretion of IgM antibodies, because it could stimulate the proliferation and differentiation of head kidney IgM+ B cells. Moreover, CYP could also enhance the phagocytic activity of IgM+

Short-term fasting enhances the resistance of common carp (Cyprinus carpio) to Aeromonas hydrophila: Impacts on gut microbiota, glucose, and oxidative stress.

BACKGROUND AND AIM: Aeromonas hydrophila is a significant pathogen in freshwater aquaculture, contributing to high morbidity and mortality in common carp (Cyprinus carpio). Conventional reliance on antibiotics raises concerns about resistance and environmental impact. This study aimed to evaluate the effects of short-term fasting (1 or 2 days) on physiological, oxidative stress, and microbial responses in C. carpio infected with A. hydrophila. MATERIALS AND METHODS: Sixty C. carpio were divided into four groups (n = 15): negative control (uninfected), positive control (infected), T1 (1-day fa

Molecular mechanism of tea polyphenols in alleviating tetrabromobisphenol A (TBBPA)-induced PANoptosis in carp (Cyprinus carpio) livers based on network toxicology and pharmacology.

Tetrabromobisphenol A (TBBPA), characterized by extensive industrial utilization and low biodegradability, exhibits significant bioaccumulation potential in aquatic ecosystems, thereby posing substantial toxicological risks to aquatic organisms. Tea polyphenols (TP) are plant extracts with anti-inflammatory and antioxidant therapeutic potential. In this study, network toxicology and pharmacology approaches were employed to investigate the hepatotoxic mechanism of TBBPA and the hepatoprotective mechanism of TP in carp. Network analysis revealed that TBBPA-induced liver injury occurred primarily

Viral infections affect the transcription of biological clock components in lymphoid organs and lymphoid-associated tissues of common carp (Cyprinus carpio L.).

The circadian clock is a mechanism that allows organisms to adapt to the changing environment in a diurnal manner. It receives external cues (time givers), which synchronize the internal circadian clock with the external environment. A crucial aspect of the circadian clock involves the positive and negative feedback loops of clock genes. In the present study, we examined the diurnal changes in the expression of clock genes in the lymphoid organs (thymus, spleen, head kidney, trunk kidney), lymphoid-associated tissues (gills and gut), and peripheral blood leukocytes (PBLs) of common carp.
