Gain-of-function p53 mutations enhance alteration of the T-cell receptor following X-irradiation, independently of the cell cycle and cell survival.

Missense mutations are by the far the most common types of mutations found in p53 of human tumors, suggesting that mutant p53 proteins function either by abrogating wild-type function or by gaining new oncogenic functions. To distinguish between the dominant-negative effect and gain of new function of p53 missense mutants, we measured the ability of transfected missense mutant p53s in p53-null Jurkat cells to alter T-cell receptor (TCR) surface expression. The TCR is a key signal transduction moiety common to T lymphocytes and is one of the major sites for aberrations in T-cell leukemias/lymph

Restricted mean survival time-based network meta-analysis of first-line therapies for metastatic clear cell renal cell carcinoma.

To compare hazard ratio (HR)- and difference in restricted mean survival time (dRMST)-based network meta-analyses of first-line immune checkpoint inhibitor-based regimens for metastatic clear-cell renal cell carcinoma. We also introduce a RMST-based framework contextualizing survival benefit alongside toxicity, corticosteroid burden, and drug-acquisition cost. Five phase III trials comparing ICI-based combinations with sunitinib in mccRCC were included. Published trial-level HRs informed HR-based NMAs, while reconstructed individual patient data were used to estimate dRMST for OS at 40 to 64 m

PANoptosis-Related Molecular Subtyping and a Subtype-Derived Prognostic Signature for Survival Prediction in Clear Cell Renal Cell Carcinoma.

IntroductionPANoptosis is an inflammatory form of programmed cell death integrating pyroptosis, apoptosis, and necroptosis, but its prognostic and immunological significance in clear cell renal cell carcinoma (ccRCC) remains unclear.MethodsIn this retrospective study, candidate PANoptosis-related genes (candidate PRGs) were identified from GeneCards and TCGA databases. Prognosis-related candidate PRGs were screened using copy number variation and Cox regression analyses, followed by consensus clustering to establish PANoptosis-related molecular subtypes. Differentially expressed genes (DEGs) b

Immune checkpoint profiling of B7-H proteins predicts survival and treatment response in metastatic clear cell renal cell carcinoma.

The B7-H family of immune checkpoint proteins plays a central role in tumor immune regulation, yet their prognostic and predictive significance in metastatic clear cell renal cell carcinoma (ccRCC) remains incompletely defined. We analyzed a retrospective cohort of primary tumor specimens from 145 patients with synchronous or metachronous metastatic ccRCC to evaluate the expression of B7-H3, B7-H4, B7-H5, and B7-H7 and their association with tumor aggressiveness, treatment response, and long-term survival. Immunohistochemistry revealed marked intratumoral heterogeneity and distinct compartment

T-cell dynamics, CD8/NK ratio and tumor survival hallmarks determine response to T-cell bispecific forimtamig in myeloma.

T-cell bispecific (TCB) therapies have demonstrated promising therapeutic efficacies for patients with relapsed/refractory multiple myeloma. However, responses are transient and essentially all patients eventually relapse. Cytokine release syndrome and other on-target off-tumor toxicities are common. Together, these challenges ask for new approaches to optimize treatment and to eventually understand resistance mechanisms. This phase 1 study (NCT04557150) investigated mode of action and resistance mechanisms of forimtamig, a novel GPRC5DxCD3 TCB. Biomarker analyses included deep phenotyping of

LAC-TME classifier: machine learning-driven model predicts survival and prioritizes targeted therapy in clear cell renal cell carcinoma.

BACKGROUND: Clear cell renal cell carcinoma (ccRCC) is a major type of kidney cancer, making up about 80% of cases, with advanced stages showing low survival rates. Current treatments face challenges like toxicity and drug resistance. Studies indicate lactate, through the Warburg effect, promotes an immune-suppressive tumor microenvironment (TME), prompting the development of the LAC-TME classifier using machine learning to predict outcomes and personalize treatment. METHODS: The study used data from TCGA-KIRC set and E-MTAB-1980 set, analyzing gene expression, mutations, and clinical data. I

Effects of neoadjuvant zoledronate and radiation therapy on cell survival, cell cycle distribution, and clinical status in canine osteosarcoma.

INTRODUCTION: Zoledronic acid (ZOL) is a third-generation bisphosphonate with a higher affinity for bone resorption areas than earlier bisphosphonates (i.e., pamidronate, PAM). In human medicine, ZOL provides improved bone pain relief and prolonged time to skeletal-related events compared to its older generational counterparts. Preclinical studies have investigated its role as an anti-neoplastic agent, both independently and synergistically, with radiation therapy (RT). ZOL and RT act synergistically in several neoplastic human cell lines: prostate, breast, osteosarcoma, and fibrosarcoma. Howe

In vivo measurement of RBC survival in patients with sickle cell disease before or after hematopoietic stem cell transplantation.

Stable, mixed-donor-recipient chimerism after allogeneic hematopoietic stem cell transplantation (HSCT) for patients with sickle cell disease (SCD) is sufficient for phenotypic disease reversal, and results from differences in donor/recipient-red blood cell (RBC) survival. Understanding variability and predictors of RBC survival among patients with SCD before and after HSCT is critical for gene therapy research which seeks to generate sufficient corrected hemoglobin to reduce polymerization thereby overcoming the red cell pathology of SCD. This study used biotin labeling of RBCs to determine t

Survival of Patients With Head and Neck Merkel Cell Cancer: Findings From the Pan-Canadian Merkel Cell Cancer Collaborative.

IMPORTANCE: Merkel cell carcinoma (MCC) is an aggressive cutaneous neuroendocrine carcinoma. Due to its relatively low incidence and limited prospective trials, current recommendations are guided by historical single-institution retrospective studies. OBJECTIVE: To evaluate the overall survival (OS) of patients in Canada with head and neck MCC (HNMCC) according to American Joint Committee on Cancer 8th edition staging and treatment modalities. DESIGN, SETTING, AND PARTICIPANTS: A retrospective cohort study of 400 patients with a diagnosis of HNMCC between July 1, 2000, and June 31, 2018, was

Long survival of patients with metastatic clear cell renal cell carcinoma. Results of real life study of 344 patients.

The treatment landscape in metastatic renal cell carcinoma has changed fundamentally over the last decade by the development of antiangiogenic agents, mammalian target of rapamycin inhibitors and immunotherapy. Outside of the context of a clinical trial, the treatments are used sequentially. We describe results under real-life conditions of a sequential treatment strategy, before the era of immunotherapy. All patients were treated according to their prognostic score (either Memorial Sloan Kettering Cancer Center or International Metastatic Renal Cell Carcinoma Database Consortium) for advanced

Elucidation of tonic and activated B-cell receptor signaling in Burkitt's lymphoma provides insights into regulation of cell survival.

Burkitt's lymphoma (BL) is a highly proliferative B-cell neoplasm and is treated with intensive chemotherapy that, because of its toxicity, is often not suitable for the elderly or for patients with endemic BL in developing countries. BL cell survival relies on signals transduced by B-cell antigen receptors (BCRs). However, tonic as well as activated BCR signaling networks and their relevance for targeted therapies in BL remain elusive. We have systematically characterized and compared tonic and activated BCR signaling in BL by quantitative phosphoproteomics to identify novel BCR effectors and…

The T-cell Receptor Repertoire Influences the Tumor Microenvironment and Is Associated with Survival in Aggressive B-cell Lymphoma.

Purpose: To investigate the relationship between the intra-tumoral T-cell receptor (TCR) repertoire and the tumor microenvironment (TME) in de novo diffuse large B-cell lymphoma (DLBCL) and the impact of TCR on survival.Experimental Design: We performed high-throughput unbiased TCRβ sequencing on a population-based cohort of 92 patients with DLBCL treated with conventional (i.e., non-checkpoint blockade) frontline "R-CHOP" therapy. Key immune checkpoint genes within the TME were digitally quantified by nanoString. The primary endpoints were 4-year overall survival (OS) and progression-free sur

Nelarabine for T cell acute lymphoblastic leukemia relapsing after allogeneic hematopoietic stem cell transplantation: an opportunity to improve survival.

T-ALL relapsing after allogeneic stem cell transplantation is unusual but classically associated with poor outcome. Recently, encouraging results have been reported with Nelarabine in relapse or refractory cases. On behalf of the Group of Research in Adult ALL (GRAALL), we conducted a retrospective analysis of patients receiving Nelarabine following relapse after transplantation. Eleven patients received Nelarabine as salvage therapy in this situation. Most of them were transplanted in first Complete Remission (CR), and received a myeloablative conditioning regimen in 7 cases. Relapse occurred

Cell number and timing of transplantation determine survival of human neural stem cell grafts in stroke-damaged rat brain.

Neural stem cells (NSCs) derived from human fetal striatum and transplanted as neurospheres survive in stroke-damaged striatum, migrate from the implantation site, and differentiate into mature neurons. Here, we investigated how various steps of neurogenesis are affected by intrastriatal transplantation of human NSCs at different time points after stroke and with different numbers of cells in each implant. Rats were subjected to middle cerebral artery occlusion and then received intrastriatal transplants of NSCs. Transplantation shortly after stroke (48 hours) resulted in better cell survival …

Impact of CD34+ cell infusion dose on immune reconstitution and survival in multiple myeloma after autologous stem cell transplantation.

Autologous stem cell transplantation (ASCT) remains central in multiple myeloma, yet the optimal CD34+ infusion dose is uncertain. We retrospectively analyzed 176 newly diagnosed patients undergoing ASCT. Survival was estimated by Kaplan-Meier, and predictors were assessed by Cox regression. Among ASCT patients, a high CD34+ dose (≥5 × 106/kg) was associated with superior progression-free survival (PFS, p = 0.020), OS (p < 0.001), deeper responses (p = 0.034), and sustained minimal residual disease (MRD) negativity (p = 0.001). High dose correlated with oligoclonal bands formation (OB, p = 0.0

CONCLUSIONS: Altogether, these results point to a role of SF3B4 in neural crest cell survival, maintenance, and differentiation. We propose that the dysregulation of these processes may contribute to Nager/Rodriguez syndrome-associated craniofacial defects.

Endothelial cell signature in muscle stem cells validated by VEGFA-FLT1-AKT1 axis promoting survival of muscle stem cell.

Endothelial and skeletal muscle lineages arise from common embryonic progenitors. Despite their shared developmental origin, adult endothelial cells (ECs) and muscle stem cells (MuSCs; satellite cells) have been thought to possess distinct gene signatures and signaling pathways. Here, we shift this paradigm by uncovering how adult MuSC behavior is affected by the expression of a subset of EC transcripts. We used several computational analyses including single-cell RNA-seq (scRNA-seq) to show that MuSCs express low levels of canonical EC markers in mice. We demonstrate that MuSC survival is reg

Long-term survival with donor CD19 CAR-T cell treatment for relapsed patients after allogeneic hematopietic stem cell transplantation.

Chimeric Antigen Receptor T (CAR-T) cell therapy has significantly advanced in treating B-cell acute lymphoblastic leukemia (B-ALL) and has shown efficacy in managing relapsed B-ALL after allogeneic hematopoietic stem cell transplantation (allo-HSCT). Donor-derived CAR-T cell offer both high efficacy and rapid response. Although promising results exist, current research lacks definitive evidence of long-term survival benefits for patients treated with donor-derived CAR-T therapy. We report the long-term survival of 32 patients with post-transplant relapsed B-ALL treated with donor-derived CD19
