The clinical manifestations of COVID-19 vary broadly, ranging from asymptomatic infection to acute respiratory failure and death. But the predictive biomarkers for characterizing the variability are still lacking. Since emerging evidence indicates that extracellular vesicles (EVs) and extracellular RNAs (exRNAs) are functionally involved in a number of pathological processes, we hypothesize that these extracellular components may be key determinants and/or predictors of COVID-19 severity. To test our hypothesis, we collected serum samples from 31 patients with mild COVID-19 symptoms at the tim

Noninvasive prenatal diagnosis (NIPD) has become a common, safe, and effective procedure for detection of inherited diseases early in pregnancy. It is based on the analysis of fetal cell-free DNA (cffDNA) derived from the placenta, circulating in the maternal plasma. De novo mutations, although rare, cause a considerable number of dominant genetic disorders. Due to the sparse representation of fetal-derived sequences in the blood, the challenge of detecting low frequency fetal de novo mutations becomes preponderant. Hence, this detection type requires deep genome-wide sequencing of cffDNA from

Non-invasive prenatal testing (NIPT) is performed worldwide to detect common chromosomal aneuploidies. The analysis of cell-free DNA (cfDNA) in maternal blood for NIPT is highly accurate for the detection of the main fetal trisomies: 21,18, and 13. However, false-positive, false-negative, and non-reportable results can occur, and these can have biological causes. Understanding the causes of unexpected NIPT results is essential to enable clinicians and genetic counselors to counsel patients comprehensively and appropriately, both prior to testing as well as after receiving the test results. BACKGROUND: The 5-hydroxymethylcytosine (5hmC) DNA modification is an epigenetic marker involved in a range of biological processes. Its function has been studied extensively in tumors, neurodegenerative diseases, and atherosclerosis. Studies have reported that 5hmC modification is closely related to the phenotype transformation of vascular smooth muscle cells and endothelial dysfunction. However, its role in coronary artery disease (CAD) has not been fully studied.
RESULTS: To investigate whether 5hmC modification correlates with CAD pathogenesis and whether 5hmC can be used as a biomarker,

Despite an increase in the rates of survival in patients suffering myocardial infarction, as yet there is no therapy specifically targeting ischaemia and reperfusion injury of the myocardium. With a greater understanding of immune activation during infarction, more potential treatment targets are now being identified. The innate immune system is believed to play an important role in the myocardium after ischaemia-driven cardiomyocyte death. The release of intracellular contents including DNA into the extracellular space during necrosis and cell rupture is now believed to create a pro-inflammat

Over one million people are living with cystic echinococcosis (CE) and alveolar echinococcosis (AE). For CE, long-term albendazole treatment is often needed, which requires regular follow-up. Follow-up is mainly through imaging which is insensitive to subtle changes and subjective to experience. We investigated the changes of Echinococcus granulosus (Eg) cell-free DNA (cfDNA) in plasma of CE patients before and after albendazole treatment to evaluate its potential as an objective marker for treatment follow-up. Plasma samples of nine CE patients were collected before and after treatment. We id

BACKGROUND AND OBJECTIVES: Quantification of serum Epstein-Barr virus (EBV) DNA and/or cell-free total DNA (cf-DNA) may become valuable sources for prognosis evaluation and monitoring treatment response in lymphomas. We aimed to investigate their roles as potential markers in pediatric Hodgkin (HL) and non- Hodgkin lymphomas (NHL).
METHOD: Between the years 2005-2008, 34 patients with HL and 45 with NHL were prospectively included. Serum samples were collected upon diagnosis, after 1-3 and 4-6 months and at the end of treatment, or at disease recurrence. RT-PCR determination of cf-DNA and EBV

Despite the essential role of Human Papillomavirus (HPV) in cervical carcinogenesis, other factors are required for cancer establishment, like miRNAs. Such molecules present a complex biogenesis, being diversely distributed across tissues and biological fluids, as cell-free miRNAs or miRNAs present in extracellular vesicles (EV). After HPV infection, an interplay between HPV and the miRNA network occurs in cervical cells. As the virus persists and cellular transformation occurs, specific patterns of miRNA expression are found in different stages of cervical disease. Thus, defining promising mi

Immunotherapy has transformed the management of hepatocellular carcinoma (HCC), yet its efficacy is frequently limited by the immunosuppressive tumor microenvironment (TME). Building on a systematic overview of current immunotherapeutic strategies, including immune checkpoint inhibitors, adoptive cell therapy, bispecific antibodies, therapeutic vaccines, and cytokines, we identify extracellular vesicles (EVs) as pivotal intercellular signaling hubs that underpin resistance across these diverse modalities. This review delineates the dual role of EVs in HCC immunotherapy, characterizing them as

ZnSe nanodisks:Ti3C2 MXene complex was prepared for the first time. Based on its remarkable photoelectrochemical performance, combined with the enzyme-free toehold-mediated strand displacement reaction, a photoelectrochemical biosensor for the detection of the non-small-cell cancer biomarker ctDNA KRAS G12D was developed. ZnSe nanodisks were in situ grown on Ti3C2 MXene surface by two-step hydrothermal method. The high conductivity and adjustable band gap of MXene significantly enhanced the photoelectric response of ZnSe. Subsequently, the photoelectrochemical biosensor was prepared by combini

The biogenesis and biological roles of extracellular vesicles (EVs) in the progression of liver diseases have attracted considerable attention in recent years. EVs are membrane-bound nanosized vesicles found in different types of body fluids and contain various bioactive materials, including proteins, lipids, nucleic acids, and mitochondrial DNA. Based on their origin and biogenesis, EVs can be classified as apoptotic bodies, microvesicles, and exosomes. Among these, exosomes are the smallest EVs (30-150 nm in diameter), which play a significant role in cell-to-cell communication and epigeneti

We describe a virucidal small molecule, PD 404,182, that is effective against hepatitis C virus (HCV) and human immunodeficiency virus (HIV). The median 50% inhibitory concentrations (IC(50)s) for the antiviral effect of PD 404,182 against HCV and HIV in cell culture are 11 and 1 μM, respectively. The antiviral activity of PD 404,182 is due to the physical disruption of virions that is accompanied to various degrees (depending on the virus and exposure temperature/time) by the release of viral nucleic acids into the surrounding medium. PD 404,182 does not directly lyse liposomal membranes even

Droplette (US 9,700,686 B2 and PCT/US2016/035695) is the first portable and contact-free transdermal technology comprising the unique combination of a piezoelectric transducer and a pneumatic diaphragm pump to deliver large biomolecules including nucleic acid therapeutics (NATs) deep into cells, and into skin and soft tissue for effective delivery over short timescales. The droplets that come out of the device are 10-50× smaller upon impact than what is created through other commercial atomizers, such as the piezoelectric transducer alone. This device has been tested extensively in vitro, in v

A 52 year-old man with Erdheim-Chester Disease (ECD) (a non-Langerhans polyostotic sclerosing histiocytosis) had next-generation sequencing (NGS) performed as part of his diagnostic workup. In addition to the tissue BRAF V600E mutation that is found in over 50% of ECD cases, he was also found to have a JAK2 V617F alteration in cell-free circulating tumor DNA (ctDNA) (liquid biopsy). The latter was thought to be an "incidental" finding, perhaps due to clonal hematopoiesis (though this usually occurs in older individuals), as his blood counts were normal and he had no splenomegaly. Approximately

L-carnitine plays a central role in mitochondrial fatty acid transport and cellular energy regulation; effects on the biochemical phenotype of brain cancer cells remain insufficiently characterized. Here, we applied confocal Raman spectroscopy and imaging to investigate the biochemical alterations induced by L-carnitine supplementation-administered as its tartrate salt-in human astrocytoma cells. Raman spectral analysis revealed distinct changes in lipid-, protein-, nucleic acid-, and cytochrome-associated vibrational features following 24 h of treatment, suggesting alterations in mitochondria

OBJECTIVES: Cell-free DNA (cfDNA) is a highly regarded noninvasive biomarker in cancer diagnosis, but analytical variability may hinder its practical application. This study aims to assess the analytical performance of various cfDNA measurement techniques using plasma samples from children with acute lymphoblastic leukemia (ALL).
METHODS: Plasma was obtained from 1500 pediatric patients with ALL between the ages of 4 and 13 years. Standardized methods for obtaining double-centrifuged plasma were used to reduce the possibility of contaminating genomic DNA. The extraction of cfDNA was done by a
