Potential role of host autophagy in Clonorchis sinensis infection.

An in vivo mouse model of Clonorchis sinensis (C. sinensis) infection with or without the administration of autophagy inhibitor chloroquine (CQ) stimulation was established to assess the possible involvement of autophagic response during C. sinensis infection. Abnormal liver function was observed at 4, 6, and 8 weeks post-infection, as indicated by elevated levels of ALT/GPT, AST/GOT, TBIL, and α-SMA in the infected groups. These findings indicated that C. sinensis infection activated autophagy, as shown by a decreased LC3II/I ratio and accumulated P62 expression in infected mice. Interestingl

[Experimental study progress on tribendimidine, artemether and artesunate against Clonorchis sinensis and other trematodes].

Currently praziquantel is one of the major drugs used in treatment of schistosomiasis and other trematode infections. Recent experimental studies indicate that a new anthelmintic, tribendimidine, is used in the treatment of intestinal nematodes, also possesses effect against several species of trematodes including Clonorchis sinensis, Opisthorchis viverrini and Echinostoma caproni. Tribendimidine is even more effective against C. sinensis in rats that a single 300 mg/kg oral dose cures almost all of the animals treated, a lower cure dose than praziquantel (375-500 mg/kg). The anti-malarial dru

Combination chemotherapy against Clonorchis sinensis: experiments with artemether, artesunate, OZ78, praziquantel, and tribendimidine in a rat model.

Caused by the Chinese liver fluke Clonorchis sinensis, clonorchiasis is of growing public health importance. Treatment and control of the disease rely on a single drug, praziquantel, and little information regarding combination chemotherapy is available. Here, we evaluated the in vivo efficacy of praziquantel combined with artemether, artesunate, OZ78, and tribendimidine, as well as an artesunate-tribendimidine combination against C. sinensis, in a rat model. Data from previous experiments were included, and negative binomial regression analyses were carried out to determine dose-response rela…

Susceptibility of experimental animals to reinfection with Clonorchis sinensis.

The present study observed the resistance to reinfection with Clonorchis sinensis in various experimental animals including mice, guinea pigs, rabbits, and dogs, as well as rats and hamsters. The resistance rates to reinfection in rats, mice, hamsters, guinea pigs, rabbits, and dogs were 79.7%, 58.0%, -12.6%, 54.8%, 62.6%, and 6.0%, respectively. Worms recovered from reinfected rats and mice were immature, and significantly smaller than those from the primarily infected (P < 0.01), whereas those from other animals were fully matured to adults. These findings indicate that the protective respon

Attempts to transfer immunity against Clonorchis sinensis in nude and DS mice.

The effects of peritoneal exudate cells (PEC) and sera of athymic nude and DS mice infected with Clonorchis sinensis metacercariae or sensitized by injection of metabolic products into footpad on transfer of immunity against the fluke to the syngeneic mice were studied. There was no significant difference in eggs per gram pattern between the sensitized and control groups, and between nude and DS mice. However, the worm burdens were slightly greater in nude mice than in DS mice. Also, a few plaque forming cells were found in only DS mice given PEC and serum from Group II DS mice. In the light o

High prevalence of Clonorchis sinensis infections and coinfection with hepatitis virus in riverside villages in northeast China.

In China, the prevalence of Clonorchis sinensis (C. sinensis) infections is only evaluated at the provincial level by national sampling surveys, and data from villages and counties are still lacking. In this study, we conducted a cross-sectional survey in 10 villages located along the Lalin River in northeast China. Clonorchiasis was diagnosed using a modified Kato-Katz method that detects the C. sinensis egg in stools. A total of 3,068 persons were screened and 2,911 were recruited for the study. Overall, the prevalence of C. sinensis infection was 29.3%. Among 175 participants who were cured…

Metacercariae in fishes of Sun Moon lake which is an endemic area for Clonorchis sinensis in Taiwan.

The Sun Moon lake in Central Taiwan is a known endemic area for clonorchiasis. Of the 45 fresh water fish, Hemiculter leucisculus, from the lake that were examined by artificial gastric juice digestion in October 1995, all were found to harbor metacercariae in their muscle. The number of metacercariae isolated from each fish ranged from 2 to 2,185, with an average of 254. A total of 11,443 metacercariae was collected from the 45 fish. Of the 4,223 metacercaria that were examined under light microscope, 4,064 (96.23%) were found to belong to Haplorchis taichui, 90(2.13%) to H. pumilio, 2(0.05%)…

Therapy of fluke infections in the past. A review.

Chemotherapy of trematode infections has been studied intensively in the past. The compounds so far used for the treatment of those parasitic infections were not satisfactory, however, because of their poor tolerability and/or their limited anthelmintic efficacy. Since the introduction of tartar emetic in 1918, a number of antimonial as well as non-antimonial preparations have been used for the treatment of schistosomiasis. In the treatment of clonorchiasis with antimony preparations, gentian violet, emetine HCl, chloroquine diphosphate and dithiazanine iodide, only temporary clinical improvem…

Paleoparasitological report on the stool from a Medieval child mummy in Yangju, Korea.

Previous studies have successfully shown evidence for parasitic infections in human remains from various archaeological sites. However, in the case of Korea, since there have been very few paleoparasitological reports published, pre-20th century parasitic infection patterns remain obscure. Therefore, in order to partly fill this gap, we are reporting on a case of paleoparasitic infection from the feces of a 15th century child mummy from Yangju, Korea. In the course of the present study, we found the eggs of Clonorchis sinensis, Ascaris lumbricoides, and Trichuris trichiura in the feces of the
