Reducing small intestinal permeability attenuates colitis in the IL10 gene-deficient mouse.

BACKGROUND: Defects in the small intestinal epithelial barrier have been associated with inflammatory bowel disease but their role in the causation of disease is still a matter of debate. In some models of disease increased permeability appears to be a very early event. The interleukin 10 (IL10) gene-deficient mouse spontaneously develops colitis after 12 weeks of age. These mice have been shown to have increased small intestinal permeability that appears early in life. Furthermore, the development of colitis is dependent upon luminal agents, as animals do not develop disease if raised under g

A novel mechanism for acute colonic pseudo-obstruction revealed by high-resolution manometry: A case report.

BACKGROUND: Acute colonic pseudo-obstruction (ACPO) is a severe form of colonic dysmotility and is associated with considerable morbidity. The pathophysiology of ACPO is considered to be multifactorial but has not been clarified. Although colonic motility is commonly assumed to be hypoactive, there is little direct pathophysiological evidence to support this claim. METHODS: A 56-year-old woman who developed ACPO following spinal surgery underwent 24 h of continuous high-resolution colonic manometry (1 cm resolution over 36 cm) following endoscopic decompression. Manometry data were analyzed a

Comparison of outcomes of pedicled jejunal and colonic conduit for esophageal reconstruction.

BACKGROUND: At present, the gastric tube is the first choice for esophageal reconstruction after esophagectomy for various benign and malignant diseases. However, when the stomach is not available, a pedicled jejunum or colon is used to reconstruct the esophagus. The present study aimed to compare the postoperative outcomes and quality of life of patients receiving jejunal and colonic conduits. METHODS: In the present retrospective study, the clinical data of 71 patients with esophageal carcinoma, who received jejunal reconstruction (jejunum group, n = 34) and colonic reconstruction (colon gr

Semaphorin 3A expression in the colon of Hirschsprung disease.

BACKGROUND: Hirschsprung disease (HSCR) is a congenital disorder characterized by an absence of intrinsic ganglion cells in the nerve plexuses of the lower colon. The Semaphorin 3A (SEMA3A) gene is involved in the migration of enteric neural precursors (ENPs). To analyze the function of SEMA3A in HSCR, the SEMA3A expression in different colon segments in HSCR was examined. METHODS: The expression levels of SEMA3A in both ganglionic and aganglionic colon tissues of 32 patients with HSCR and in colon tissue of 5 newborn unaffected individuals were examined by real-time RT-PCR, Western-blot, and

Biomaterial-based microbiota strategies for targeting the intestinal immune microenvironment in inflammatory bowel disease.

Inflammatory bowel disease (IBD) is a chronic inflammatory disorder of the gastrointestinal tract with an unclear etiology. Its onset and progression are closely associated with gut microbiota dysbiosis and immune dysfunction. Microbiota-based therapeutic strategies can alleviate intestinal inflammation to some extent. However, their clinical efficacy remains limited due to challenges during delivery, including degradation by gastrointestinal components, insufficient exposure of active agents, and unstable colonization in the colon. This review systematically summarizes various biomaterial-bas

Human enteric viruses autonomously shape inflammatory bowel disease phenotype through divergent innate immunomodulation.

Altered enteric microorganisms in concert with host genetics shape inflammatory bowel disease (IBD) phenotypes. However, insight is limited to bacteria and fungi. We found that eukaryotic viruses and bacteriophages (collectively, the virome), enriched from non-IBD, noninflamed human colon resections, actively elicited atypical anti-inflammatory innate immune programs. Conversely, ulcerative colitis or Crohn's disease colon resection viromes provoked inflammation, which was successfully dampened by non-IBD viromes. The IBD colon tissue virome was perturbed, including an increase in the enterovi

Control of inflammation by stromal Hedgehog pathway activation restrains colitis.

Inflammation disrupts tissue architecture and function, thereby contributing to the pathogenesis of diverse diseases; the signals that promote or restrict tissue inflammation thus represent potential targets for therapeutic intervention. Here, we report that genetic or pharmacologic Hedgehog pathway inhibition intensifies colon inflammation (colitis) in mice. Conversely, genetic augmentation of Hedgehog response and systemic small-molecule Hedgehog pathway activation potently ameliorate colitis and restrain initiation and progression of colitis-induced adenocarcinoma. Within the colon, the Hed

Laparoscopic colon surgery: past, present and future.

Since its first described case in 1991, laparoscopic colon surgery has lagged behind minimally invasive surgical methods for solid intra-abdominal organs in terms of acceptability, dissemination, and ease of learning. In colon cancer, initial concerns over port site metastases and adequacy of oncologic resection have considerably dampened early enthusiasm for this procedure. Only recently, with the publication of several large, randomized controlled trials, has the incidence of port site metastases been shown to be equivalent to that of open resection. Laparoscopic surgery for colon cancer has

Commensal bacteria play a fundamental role in maintaining gut immune homeostasis.

The intestinal microbiome, which is a key factor in the maintenance of host gut homeostasis, enhances intestinal mucosal barrier function and immune tolerance (Rooks and Garrett, 2016; Skelly et al., 2019). However, the specific immunomodulatory functions of microbiota-derived metabolites in mucosal inflammatory responses remain largely unknown. The effects of microbial metabolites may vary across different immune cell types and host homeostasis (Hu et al., 2023; Zhao et al., 2023). Hence, it is fundamental to understand how specific intestinal microbes and their metabolic small molecules caus

Macrophage SELENOP controls neutrophil homeostasis through redox stress regulation in Crohn's disease.

BACKGROUND: Neutrophil accumulation and oxidative injury are prominent features of Crohn's disease (CD). This study aimed to investigate whether macrophage-derived SELENOP functions as a redox checkpoint in macrophage-neutrophil crosstalk during CD. METHODS: Inflamed and non-inflamed colon tissues from CD patients were examined by single-cell RNA sequencing. SELENOP function was assessed in macrophages, macrophage-neutrophil co-cultures and inflammation models. Mechanistic studies included loss- and gain-of-function assays, co-immunoprecipitation, ubiquitination assays, and rescue experiments

Evaluation of probiotic properties and genome analysis of the new Pediococcus acidilactici strain 46A isolated from Chinese young adults: in vitro and in vivo studies.

BACKGROUND: The human gut microbiota plays a key role in intestinal homeostasis and inflammatory diseases, prompting the search for novel probiotic strains with strain-specific functional properties. Differences in dietary habits and cultural backgrounds represent a valuable source for isolating previously uncharacterized gut bacteria. Given the increasing use of probiotics, rigorous safety assessment, including antimicrobial resistance (AMR) evaluation, is required, and the integration of whole-genome sequencing with phenotypic characterization is recommended. OBJECTIVES: This study evaluate

Feed restriction minimally affects nursery pig colonic function.

Reduced feed intake is a hallmark following weaning, enteric diseases, such as Escherichia coli, and numerous environmental conditions, such as heat stress and has been shown to cause intestinal barrier dysfunction. Currently, several different markers and assays are utilized to evaluate intestinal barrier function, and these methods have not been thoroughly validated. These restrictions may present a potential model to validate and compare multiple in vivo, ex vivo, and tissue markers of intestinal integrity. Forty-eight barrows (9.7 kg initial body weight) were fed for 7 d at 100%, 75%, 50%,

Smart capsule for targeted proximal colon microbiome sampling.

The gastrointestinal (GI) tract, particularly the colon region, holds a highly diverse microbial community that plays an important role in the metabolism, physiology, nutrition, and immune function of the host body. Accumulating evidence has revealed that alteration in these microbial communities is the pivotal step in developing various metabolic diseases, including obesity, inflammatory bowel disease (IBD), and colorectal cancer. However, there is still a lack of clear understanding of the interrelationship between microbiota and diet as well as the effectiveness of chemoprevention strategie

Competitive fungal commensalism mitigates candidiasis pathology.

The mycobiota are a critical part of the gut microbiome, but host-fungal interactions and specific functional contributions of commensal fungi to host fitness remain incompletely understood. Here, we report the identification of a new fungal commensal, Kazachstania heterogenica var. weizmannii, isolated from murine intestines. K. weizmannii exposure prevented Candida albicans colonization and significantly reduced the commensal C. albicans burden in colonized animals. Following immunosuppression of C. albicans colonized mice, competitive fungal commensalism thereby mitigated fatal candidiasis.

Phenolic Ethanolic Extracts of Specialty Sorghum Ameliorate Intestinal Colitis and Inflammation Induced by Dextran Sulfate Sodium in Mice.

Recently, sorghum [Sorghum bicolor (L.) Moench] has been given great attention as an excellent source of polyphenols that exhibit protective effects against multiple chronic disease models. Ulcerative colitis (UC) is strongly linked to the incidence of colon cancer and other intestinal chronic diseases. This study aimed to determine the ability of sorghum ethanolic phenolic extracts (SEPEs) to mitigate intestinal colitis and inflammation induced by dextran sulfate sodium (DSS) in a mice model. Forty C57BL/6 male mice were randomly assigned to one of the experimental groups: negative control, p

The Multifaceted Nature of Streptococcal Antigen I/II Proteins in Colonization and Disease Pathogenesis.

Streptococci are Gram-positive bacteria that belong to the natural microbiota of humans and animals. Certain streptococcal species are known as opportunistic pathogens with the potential to cause severe invasive disease. Antigen I/II (AgI/II) family proteins are sortase anchored cell surface adhesins that are nearly ubiquitous across streptococci and contribute to many streptococcal diseases, including dental caries, respiratory tract infections, and meningitis. They appear to be multifunctional adhesins with affinities to various host substrata, acting to mediate attachment to host surfaces a

One-stage transanal surgical technique for Hirschsprung's disease: is the age of a patient a limiting factor?

Hirschsprung's disease in the adolescent is a rare and often misdiagnosed cause of lifelong refractory constipation. In this particular case, a male patient spent 13 years with an unrecognized illness that was eventually diagnosed as Hirschsprung's disease. The age of the patient is considered a limiting factor due to over-distention and hypertrophy of the rectosigmoid colon that has to be pulled through a muscular cylinder in a limited operative field. This report of a successfully treated teenage boy using a one-stage transanal endorectal approach without conversion to laparotomy, his uneven

Segmental aganglionosis (zonal aganglionosis or "skip" lesions) in Hirschsprungs disease: a report of 2 unusual cases.

INTRODUCTION: There is accumulating evidence that "skip" lesions or zonal aganglionosis do occur in HSCR disease, albeit rarely. They are of interest because it may cause confusion in interpreting surgical margins as well as understanding the pathophysiology of HSCR disease. Normally described as "a skip area" of normally ganglionated bowel, surrounded proximally and distally by aganglionosis with variations may occur. CASE REPORTS: We report two cases of infants with unusual types of "skip lesions", identified within the last 5 years. RESULTS: One patient had an area of zonal aganglionosis

Gut commensal Bacteroides-derived pantothenic acid alleviates metabolic syndrome.

Pantothenic acid (PA), or vitamin B5, can be synthesized by gut commensals, but the contribution of microbial PA to metabolic health remains unclear. Here, we find that microbial PA supply is reduced in individuals with metabolic syndrome (MetS) and is associated with impaired gut barrier function and disease severity. Tracing microbial PA identifies Bacteroides fragilis as a key contributor, with panC required for PA biosynthesis, as confirmed by isotope tracing, bacterial culture, and germ-free colonization. In MetS models, colonization with wild-type, but not ΔpanC B. fragilis, restores PA,
