Fibroblasts as local immune modulators in ocular allergic disease.

Vernal keratoconjunctivitis (VKC), a severe form of ocular allergic disease, is characterized by the formation of giant papillae at the upper tarsal conjunctiva and corneal lesions that threaten vision. Recent evidence indicates that resident fibroblasts function as immune modulators in the pathogenesis of the chronic allergic inflammation associated with VKC. The T helper 2 (Th2) cell-derived cytokines interleukin (IL)-4 and IL-13 stimulate the migration and proliferation of conjunctival fibroblasts as well as protecting these cells from apoptotic cell death, effects that likely underlie the

Endocrinological masqueraders of allergy.

There are many endocrine conditions that can present with allergic symptoms and signs. Thyroid conditions ranging from fatigue to orbitopathy associated with Grave's disease can be confused with allergic conjunctivitis and angioedema. Autoimmune thyroid disease is commonly associated with idiopathic urticaria. Symptoms of orthostatic hypotension and intolerance often present when least expected and should be considered ahead of time to avoid confusion in treating possible systemic allergic reactions. Flushing is a frequent sign and differentiating from complaints commonly associated with aller

Cellular mechanisms and effects of IL-4 receptor blockade in experimental conjunctivitis evoked by skin inflammation.

Ocular surface diseases, including conjunctivitis, are recognized as common comorbidities in atopic dermatitis (AD) and occur at an increased frequency in patients with AD treated with biologics targeting IL-4 receptor α (IL-4Rα) or IL-13. However, the inflammatory mechanisms underlying this pathology are unknown. Here, we developed a potentially novel mouse model of skin inflammation-evoked conjunctivitis and showed that it is dependent on CD4+ T cells and basophils. Blockade of IL-4Rα partially attenuated conjunctivitis development, downregulated basophil activation, and led to a reduction i

Predictors of childhood food allergy: significance and implications.

BACKGROUND: Food allergy is common in children, and its occurrence is strongly associated with other allergies including anaphylaxis. Both genetic (e.g., CD14, STAT6, IL-10, SPINK5, and FOXP3 genes) and environmental (e.g., early exposure to highly allergic food) factors appear to contribute to food allergy. METHOD: Cross-sectional study involved children in public primary schools in Al-Ain city (United Arab Emirates). 660 students from the chosen classes were provided with 35 questions to be answered by their parents with a response rate 60.2%. The objective of the study was to determine pre

[Serum immunosuppressive acidic protein (IAP) in inflammatory ocular disease].

The titer of serum immunosuppressive acidic protein (IAP) was determined by the method of single radial immunodiffusion (Matsuda et al) in Behçet's disease, sarcoidosis, tuberculosis, toxoplasmosis, etiology-unknown anterior uveitis (granulomatous, non-granulomatous), allergic conjunctivitis and scleritis. The titer of IAP in these diseases except allergic conjunctivitis was significantly higher than in normal controls. In all the diseases studied, the titer during the active stage was significantly higher than that during the inactive stage. The titer in Behçet's disease was significantly hig

Does tobacco smoke prevent atopic disorders? A study of two generations of Swedish residents.

BACKGROUND: Earlier studies have given conflicting results regarding the effect of exposure to tobacco smoke on atopic sensibilization. MET … who smoked at least 15 cigarettes a day tended to have lower odds for suffering from allergic rhino-conjunctivitis, allergic asthma, atopic eczema and food allergy, compared to children of mothers who had never smoked (ORs 0.6-0.7). Children of fathers who had smoked at least 15 cigarettes a day had a similar tendency (ORs 0.7-0.9). CONCLUSIONS: This study demonstrates a …

Predicting allergic disease at age four using an atopy predisposition score at age two: the application of item response theory.

When defining allergic outcomes in epidemiology studies, results of the skin prick test (SPT) panel are often dichotomized as positive/negative or categorized based on the number of positive responses. Item response theory (IRT) models, however, may prove to be a better alternative with the ability to generate scores that account for both type and number of positive SPTs. IRT was applied to SPT responses administered to 537 children at age two to determine predictability of allergic disease at age four. The children received SPTs to 15 aeroallergens and two foods. Atopy predisposition scores w

Treating the ocular component of allergic rhinoconjunctivitis and related eye disorders.

CONTEXT: Allergy symptoms that affect the eyes are common in adults and children worldwide, and are often associated with nasal allergy symptoms, prompting the term 'rhinoconjunctivitis' to describe the condition. However, this condition has not always been recognized, and earlier literature reported allergic conjunctivitis only within a subset of nasal allergy patients. EVIDENCE ACQUISITION: To assess the current state of ocular allergy epidemiology, pathophysiology, and currently available treatment options, we performed a MEDLINE search for articles regarding ocular allergy, rhinoconjuncti
