Intermolecular histone H4 interactions in core nucleosomes.

Chicken histone H4, labeled at methionine-84 with 1-N-pyrenyliodoacetamide, has been incorporated into a nucleosome-like particle with core length DNA and unmodified histones H2A, H2B, and H3. These synthetic nucleosomes exhibit properties very similar to those displayed by native particles and those labeled with other fluors. The emission spectrum of the pyrene-labeled nucleosome was characteristic of excited dimer (excimer) fluorescence, indicating that the single pyrene groups on the two H4 molecules are in close proximity in the reconstituted particle. Histone H4 was also labeled randomly

Autoantibodies against the insulin receptor recognize the insulin binding subunits of an oligomeric receptor.

125I-insulin was specifically cross-linked to membranes of human cultured lymphocytes (IM-9 line) using disuccinimidyl suberate. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis under reducing conditions and autoradiography of this preparation revealed a major 125I-labeled band with an apparent mol wt of 130,000 and minor bands of about 300,000 and 95,000. Labeling of these bands was inhibited by incubation of membranes with either unlabeled insulin or autoantibodies to the insulin receptor. The bands were also observed after the 125I-insulin cross-linked preparation was solubilized a

A model for the structure of the yeast mitochondrial adenosine triphosphatase complex.

Monodisperse, Triton-solubilized ATPase complex was treated with the reversible protein-protein cross-linker dithiobis(succinimidyl)propionate under conditions where only intracomplex cross-links were formed. The resulting products were analyzed by two-dimensional sodium dodecyl sulfate-acrylamide slab gel electrophoresis under reducing and oxidizing conditions. Using the observed major subunit-subunit cross-links and the previously published subunit stoichiometries (Todd, R., Griesenbeck, T., and Douglas, M. (1980) J. Biol. Chem. 255, 5461-5467), a model of the ATPase complex was constructed.

Dupuytren's contracture. An electron microscopic, biochemical, and clinical correlative study.

Fascial specimens were obtained from twenty-four patients treated operatively for Dupuytren's contracture. The nodules and cords were examined by electron microscopic and biochemical techniques. The clinical course and response to operative treatment were then correlated with the tissue findings. Electron microscopic analysis revealed myofibroblasts in the nodules of seven of twenty-four patients. Fibroblasts containing prominent microtubules were found in ten patients. The fascia contained type-III collagen, increased amounts of collagen per unit of dry weight, and an increase in reducible cr

Cross-linking of immunoglobulin E-receptor complexes induces their interaction with the cytoskeleton of rat basophilic leukemia cells.

Bridging of immunoglobulin E (IgE)-receptor complexes on rat basophilic leukemia cells by polyclonal anti-IgE antibodies induces a detergent-resistant association of these complexes with the cellular cytoskeleton. In dose-response curves the extent of the cytoskeletal association appears to follow the extent of bridging, continuing to increase beyond where stimulated degranulation is maximal. This stable association is maintained after the aggregated IgE-receptor complexes have been internalized by the cell. Multivalent antigen and trimeric IgE cause less extensive receptor cross-linking than

Preparation of dextran glassy particles through freezing-induced phase separation.

This report demonstrates a novel method to prepare fine polysaccharide glassy particles of uniform sizes under a condition without water/oil and water/air interfacial tension and cross-linking reagents. When a co-solution of dextran and polyethylene glycol (PEG) was frozen gradually, phase separation occurred during which dextran formed the dispersed phase and PEG remained in the continuous part. Fine dextran glassy particles were harvested after lyophilizing this frozen sample, followed by re-dissolving the continuous phase (PEG) in dichloromethane or acetonitrile. Desired mean particle diame…

Preparation and characterization of bifunctional biopolymers for receptor-based liposomal immunosensing.

In this study, we prepared bifunctional biopolymers for development of a novel liposomal immunosensing element. These biopolymers were produced such that a rat monoclonal antibody fragment Fab' was linked to a cardiac protein Troponin I (TnI) peptide by a cross-linking reagent, o-phenylenedimaleimide (o-PDM) or N-sucinimidyl 3-(2-pyridyldithio)propionate (SPDP). The biopolymer formation yields were approximately 10% for Fab-TnIMal and 30% for Fab-TnISPDP. Molar ratios of Fab' to SPDP or o-PDM and conjugated Fab' to TnI peptide and conjugation pH have considerable effects on the biopolymer yiel

Affinity-dependent cross-linking to neurotoxin sites of the acetylcholine receptor mediated by catechol oxidation.

The choline homologue 3-[(trimethylammonio)methyl]catechol (TMC) has been synthesized, and the controllable features of its complex oxidation have been examined spectroscopically and correlated with its toxin binding inactivating reactions with the acetylcholine receptor (AcChR) from Torpedo californica electroplax. Affinity-dependent reactions of early intermediates in the oxidation of TMC are suggested to intercede covalently in this inactivation. At pH 7.4, where the oxidative polymerization of catechols proceeds spontaneously, pyrocatechol produced no effect on the toxin binding function o

Dual-crosslinked chitosan nanofibres - Balancing mechanical enhancement with cytotoxic risk.

This study evaluated the effects of dual-crosslinking strategies on the mechanical performance and biocompatibility of chitosan-norbornene (Chi-NB) nanofibres for tissue reconstruction. Chi-NB, bearing 28.6 ± 1.4 mol% alkene groups, was processed into nanofibres via solution blow spinning. Dual crosslinking was conducted by photo-initiated thiol-ene crosslinking with dithiothreitol (DTT), achieving >95% norbornene conversion with minimal thiol oxidation, followed by exposure to tannic acid (TA). Structural and chemical validation was performed using SEM, NMR, XPS, FT-IR, and tensile testing. I

One-pot dual-crosslinked gelatin/alginate hydrogels for microstructure and texture regulation in 3D-printed meat analogues.

3D printing technology can precisely simulate the complex structure of natural meat, but balancing the viscosity and mechanical properties of printing inks remains challenging. This study proposes a one-pot dual-crosslinking strategy. CaCO3/GDL-mediated ionic crosslinking provides immediate structural support for 3D printing, while temperature regulation delays the TG enzyme crosslinking rate. This enables the interpenetration of covalent and ionic networks to form a composite network structure, ensuring long-term product stability. The effects of crosslinking sequence and gelatin concentratio

Photo-crosslinkable quaternized chitosan-based hydrogel for immunomodulation and functional recovery in spinal cord injury.

Spinal cord injury (SCI) causes severe neurological deficits and remains difficult to treat with current strategies such as surgical decompression or corticosteroids, which offer limited benefits. Hydrogels represent a promising alternative because they provide structural support and serve as therapeutic carriers. Quaternized chitosan (QCS), a derivative of chitosan, shows improved solubility and anti-inflammatory properties but lacks crosslinking ability and degrades rapidly, restricting its application in SCI. To overcome these limitations, we introduced methacrylate groups into QCS (QCS-MA)

Engineering a Dual-Regenerative Cascade: Injectable Hydrogel with Zn2+/l-Arg Cross-Linkers and Macrophage-Targeting Carbon Dots for Synergistic CLI Therapy.

Critical limb ischemia (CLI) remains refractory to current revascularization strategies due to its complex pathology. Here, we report an injectable alginate hydrogel that orchestrates a dual-pathway regenerative cascade by simultaneously reprogramming the immune microenvironment and activating direct tissue repair. The system is rationally designed by integrating two functional modules: (i) immunomodulatory carbon dots (PNS-AA@CQDs-COS) that selectively target M1 macrophages and drive them to the M2 phenotype, and (ii) intrinsically bioactive cross-linkers (Zn2+/l-Arg) that serve as sustained-

Kinetics of Lignin/Epigallocatechin Gallate Co-Release by Multiscale Structures of Dual-Dynamic Cross-Linked Hydrogels.

The controlled-release hydrogel with dual-dynamic-bond networks provides superior tunability and stability through collaborative exchange mechanisms, which are often used for drug release. However, the impact of its multiscale structures involving various orientations and cross-linking densities on release kinetics is still poorly understood. Herein, a dual-dynamic-bond cross-linked hydrogel with tunable structures (orientation and cross-linking density) was developed, where the cross-linked network is formed by imine bonds between aminated lignin and oxidized carboxymethyl cellulose, as well

Amide-Forming Ligations at Physiological pH for the Encapsulation of Human Mesenchymal Stem Cells.

The development of rapid, chemoselective covalent bond-forming reactions enables the assembly of hydrogel scaffolds suitable for applications in cellular encapsulation. We previously reported that amide-forming ligations between potassium acyltrifluoroborates (KATs) and hydroxylamines produce robust hydrogels that have excellent cytocompatibility, but the requirement for somewhat acidic conditions for efficient hydrogel assemblies limited their application to robust cell types. To overcome this constraint, we have recently found that quinolinium acyltrifluoroborates (QATs) serve as highly effi

Improved biostability of collagen in sound and caries-affected dentin using an integrated crosslinking-remineralization treatment.

This study investigated a combined strategy using collagen crosslinking and polymer-induced liquid precursors (PILP)-based remineralization to enhance collagen biostability and resistance to collagenase degradation in sound (SD), denatured (DD), and caries-affected dentin (CAD). Ultra-thin demineralized collagen films and acid-etched dentin beams with demineralized layers (DL) were treated with 1% cranberry-extract (CR) collagen crosslinker for 30 s, followed by PILP remineralization for 1-7 days. Biostability was assessed by collagenase-induced weight loss and hydroxyproline release; structur

Assessing Effects of Riboflavin/UV-A Cross-Linking on Aqueous Outflow Facility, Corneal Biomechanics and Their Association With Intraocular Pressure.

Riboflavin/UV-A corneal collagen cross-linking (CXL) is a standard treatment for early-stage keratoconus. However, quantitative CXL outcomes remain limited. This study evaluated CXL effects on outflow facility and corneal biomechanics at intraocular pressures (IOPs) in ex vivo porcine eyes. Ocular rigidity and outflow facility were derived from pressure-volume and IOP decay curves using a direct manometric technique, while corneal elasticity (Young's modulus) was measured via non-contact air-pulse optical coherence elastography. CXL increased ocular rigidity (0.0066 ± 0.0001 μL-1 vs. 0.0060 ±

Bioprinting of transparent and adhesive corneal patches: Integrating photo-crosslinkable dopamine-conjugated silk fibroin and decellularized cornea matrix for sutureless tissue integration and regeneration.

Corneal injuries, a leading cause of visual impairment, are traditionally addressed through tissue transplantation. However, challenges such as donor shortages, graft rejection, and complications from suturing often limit their effectiveness. Current corneal adhesives frequently fall short in both adhesion strength and biocompatibility. We present an innovative solution: a photocurable hydrogel that integrates dopamine-conjugated methacrylated silk fibroin (d-MSF) with a decellularized corneal matrix (DCM). This hydrogel combines advanced materials to create a bioadhesive system that offers su

Multi-step functionalization of hydrogels through mechano- and photo-responsive linkages.

Patterning soft materials with cell adhesion motifs can be used to emulate the structures found in natural tissues. While patterning in tissue is driven by cellular assembly, patterning soft materials in the laboratory most often involves light-mediated chemical reactions to spatially control the presentation of cell binding sites. Here we present hydrogels that are formed with two responsive crosslinkers-an anthracene-maleimide adduct and a disulfide linkage-thereby allowing simultaneous or sequential patterning using force and UV light. Hydrogels were formed using poly(ethylene glycol)-based

Mechanically enhanced chitosan hydrogels based on micellar cross-linking for wound hemostasis.

Excessive blood loss caused by severe trauma is often life-threatening, so a kind of hemostatic material with superior performance is urgently needed to achieve rapid hemostasis of wounds. Here, we prepared CDCS/PF/PVA/Agar hydrogels by blending chitosan modified by catechol groups, PF127-NH2 micelles, PVA and agarose. The hydrogel showed adjustable swelling and degradation properties. The CDCS/PF/PVA/Agar hydrogel exhibited exceptional mechanical properties with an elastic modulus of 351.4 kPa after four freeze-thaw cycles, surpassing traditional polysaccharide hydrogels by 7-20 fold. Its tis
