The Ca2+-sensing receptor activates cytosolic phospholipase A2 via a Gqalpha -dependent ERK-independent pathway.

The Ca(2+)-sensing receptor (CaR) stimulates a number of phospholipase activities, but the specific phospholipases and the mechanisms by which the CaR activates them are not defined. We investigated regulation of phospholipase A(2) (PLA(2)) by the Ca(2+)-sensing receptor (CaR) in human embryonic kidney 293 cells that express either the wild-type receptor or a nonfunctional mutant (R796W) CaR. The PLA(2) activity was attributable to cytosolic PLA(2) (cPLA(2)) based on its inhibition by arachidonyl trifluoromethyl ketone, lack of inhibition by bromoenol lactone, and enhancement of the CaR-stimul…

Thrombin-stimulated increases in cytosolic Ca2+ level and gonadotropin-releasing hormone release in GT1-7 neurons.

The effects of thrombin on cytosolic calcium levels ([Ca2+]cyt), and on gonadotropin-releasing hormone (GnRH) release, were characterized in cultured GT1-7 neurons. GnRH release from GT1-7 neurons was pulsatile with an average pulse amplitude of 14.3+/-5.8 pg x min x ml(-1) and an average pulse duration of 21.3+/-4.2 min. The [Ca2+]cyt response to 0.005 to 0.2 U/ml thrombin was saturable and concentration dependent (EC50 = 0.0268 U/ml). Ethyleneglycotetraacetic acid (EGTA) chelation of extracellular Ca2+ resulted in an approximately 70% attenuation of thrombin-stimulated increase in [Ca2+]cyt.…

Cytosolic pH variations in perfused rat liver at 4 degrees C: role of intracellular buffering power.

The effect of low temperature on cytosolic pH regulation and buffering capacity was evaluated in the isolated rat liver. The pH changes were followed by phosphorus-31 nuclear magnetic resonance. Cooling from 37 to 4 degrees C, with Krebs-Heinseleit perfusion at an external pH of 7.35, induced an alkaline shift in cytosolic pH (pHcyt) of 0.13 or 0.75 pH units in the presence of bicarbonate, respectively (dpH cys/dT values were 0.004 and 0.022 unit/degrees C. With 4 degrees C perfusion, in the presence or absence of bicarbonate, acute changes of external pH (from 7.40 to 5.90) did not affect pHc…

Characterization of human immune responses to the cytosolic superoxide dismutase and glutathione S-transferase from Onchocerca volvulus.

In onchocerciasis patients and in O. volvulus-exposed individuals without signs of onchocericiasis, T- and B-cell responses to two recombinantly expressed O. volvulus enzymes were analysed and compared to responses to total protein extract of adult parasites. The cytosolic enzymes Cu/Zn superoxide dismutase 1 (OvSOD1) and glutathione S-transferase 2 (OvGST2) represent 2 detoxifying molecules which may play an important role in parasite defense against host-induced oxidative stress. The T-cell response to the two recombinant proteins was analysed by investigating the cytokine responses of perip…

Identification of promoter elements involved in the cytosolic Ca(2+)-mediated photoregulation of maize cab-m1 expression.

Changes in cytoplasmic Ca2+ levels are involved in the regulation of several plant genes. However, to our knowledge, no regions of genes or specific cis elements have been shown to be involved in the regulation of plant gene expression by cytosolic Ca2+ signaling. The maize (Zea mays) gene cab-m1, which encodes a light-harvesting chlorophyll a/b-binding apoprotein, is positively photoregulated in mesophyll cells (MC) but not in bundle-sheath cells (BSC). This gene is highly preferentially expressed in maize MC versus BSC. In situ transient expression assays have revealed that exposure of tissu…

Cytosolic Na+ controls and epithelial Na+ channel via the Go guanine nucleotide-binding regulatory protein.

In tight Na+-absorbing epithelial cells, the fate of Na+ entry through amiloride-sensitive apical membrane Na+ channels is matched to basolateral Na+ extrusion so that cell Na+ concentration and volume remain steady. Control of this process by regulation of apical Na+ channels has been attributed to changes in cytosolic Ca2+ concentration or pH, secondary to changes in cytosolic Na+ concentration, although cytosolic Cl- seems also to be involved. Using mouse mandibular gland duct cells, we now demonstrate that increasing cytosolic Na+ concentration inhibits apical Na+ channels independent of c…

[A study on the effect of cortisol and progesterone on cytosolic arachidonic and palmitic acid concentrations in cultured human myometrial cells].

Glucocorticoids inhibit prostaglandin synthesis in several cell types, presumably by inhibiting arachidonic acid deacylation from phospholipids. We studied the effects of cortisol (F) and progesterone (P) on fatty acid release from cultured human myometrial cells. Confluent monolayer cultures of myometrial cells were adapted to steroids containing medium for 24 hours and the intracellular arachidonic acid and palmitic acid concentrations were determined. In the presence of fetal calf serum (FCS), the palmitic acid concentrations significantly decreased after the addition of 10(-7)M F. In the a

Demonstration of specific insulin binding to cytosolic proteins in H35 hepatoma cells, rat liver and skeletal muscle.

We previously demonstrated that internalized insulin enters the cytoplasm before accumulating in nuclei of H35 rat hepatoma cells. This finding raises the possibility that insulin may interact with cytosolic proteins in addition to insulin-degrading enzyme (IDE). In the present study, cytosol from H35 hepatoma cells, rat liver or muscle was incubated with A14- or B26-125I-insulin at 4 degrees C for 5-120 min in the absence or presence of 25 micrograms/ml unlabelled insulin. 125I-insulin was cross-linked to cytosolic proteins by disuccinimidyl suberate and analysed by reducing or non-reducing S…

Increased platelet cytosolic calcium responses to low density lipoprotein in type II diabetes with and without hypertension.

Non-insulin dependent diabetes mellitus (NIDDM) and hypertension are common diseases which are independently associated with insulin resistance/hyperinsulinemia, dyslysidemia, abnormalities of platelet function, and accelerated atherogenesis. The interaction of these independent risk factors is poorly understood. Recently, low density lipoprotein (LDL) receptors have been described, in platelets, and LDL elevates [Ca2+]i in these cells. In this study we have evaluated platelet [Ca2+]i responsiveness to LDL and arginine vasopressin (AVP) in NIDDM patients with (n = 28) and without (n = 13) conc

Glycochenodeoxycholate-induced lethal hepatocellular injury in rat hepatocytes. Role of ATP depletion and cytosolic free calcium.

Chenodeoxycholate is toxic to hepatocytes, and accumulation of chenodeoxycholate in the liver during cholestasis may potentiate hepatocellul … digitonin-permeabilized cells in a dose-dependent manner. After ATP depletion, a sustained rise in cytosolic free calcium (Cai2+) was observed. Removal of extracellular Ca2+ abolished the rise in Cai2+, decreased cellular proteolysis, and protected against cell killing by glycochenodeoxycholate. The results suggest that glycochenodeoxycholate cytotoxicit …

Cell-free activation of phagocyte NADPH-oxidase: tissue and differentiation-specific expression of cytosolic cofactor activity.

We examined a variety of tissues for the presence of cytosolic cofactor activity that would support arachidonate-dependent cell-free activation of NADPH-oxidase in isolated human neutrophil membranes. Cofactor activity was not found in cytosol isolated from erythrocytes, lymphocytes, placenta, brain, liver, or the human promyelocytic leukemic cell line HL-60. Induction of differentiation in HL-60 cells led to expression of cytosolic cofactor activity. In dimethylsulphoxide-induced HL-60 cells the level of cytosolic cofactor activity was closely correlated with phorbol myristate acetate-stimula…

Correlation of extracts obtained by high efficiency gel chromatography of lactic dehydrogenase virus infected mouse serum and cytosol from human tumors using leukocyte adherence inhibition assay.

Immunochemically active fractions were obtained using Separon Hema-300-glc(R) from serum of lactic dehydrogenase virus (LDV) infected mice and from homogenates of human tumors. The mixture of proteins of tumorous origin from the cytosol giving a positive reaction in the leukocyte adherence inhibition (LAI) test was found in the same fractions showing maximum of absorbance at 340 nm in the spectrophotometer and a corresponding peak in the refractometer. Analogous peaks were not proved in material obtained from healthy controls, but they were found in some human placentas and fetal organs. The L…

A Cytosolic Arabidopsis Homologue of Bacterial GppA/PPX Retains Polyphosphate and pppGpp Phosphatase Activities and Regulates Photosynthetic Acclimation.

Here, we performed biochemical and genetic characterization of a GppA/PPX-family phosphohydrolase from Arabidopsis thaliana (AtGPPA/PPX). Sixteen splice variants of AtGPPA/PPX are deposited in public databases and can be classified into two types based on differences in their N-terminal sequences. Green fluorescent protein fusions with either N-terminus indicated that neither possesses a transit peptide, suggesting that all AtGPPA/PPX splice variants localize to the cytosol. Biochemical analysis of the long splice variant of AtGPPA/PPX demonstrated phosphatase activity toward guanosine 5'-trip…

The Cytosolic Iron-Sulfur Cluster Assembly Targeting Complex Is Hijacked by Magnaporthe Effector AvrPi9 to Suppress Rice Resistance.

The cytosolic iron-sulfur (Fe-S) cluster assembly (CIA) targeting complex mediates the maturation of cytosolic and nuclear Fe-S proteins and governs core eukaryotic cellular processes. However, whether this complex contributes to plant disease resistance remains unclear. Here, we show that the Magnaporthe oryzae avirulence effector AvrPi9 targets the host CIA targeting complex to suppress rice blast resistance. AvrPi9 physically interacts with and stabilizes OsAE7, a core subunit of the CIA targeting complex. Using biochemical and genetic approaches, we demonstrate that OsAE7 facilitates Fe-S …

The cation/H+ exchanger ChCAX3 fine-tunes fruit ripening by modulating cytosolic Ca2+ homeostasis in Cerasus humilis.

Cerasus humilis is a calcium-rich fruit crop with high nutritional value, but its synchronized ripening causes concentrated harvest and shor … ening period by approximately 14 days. ChCAX3-mediated vacuolar Ca2+ sequestration may maintain low cytosolic Ca2+ levels, thereby potentially attenuate the expression and activity of a presumptive Ca2+ sensor ChCBL10 and its interacting kinase ChCIPK3 at the vacuolar membrane. This attenuation of the ChCBL10-ChCIPK3 module was accompanied by reduced expr …

AKNA drives neuroblastoma cell fate transition through chromatin modifications of stemness factors and differentiation by cytosolic retention.

Neuroblast cells play a pivotal role in adult neurogenesis. However, the detailed mechanisms underlying the acquisition of pluripotency or the process of differentiation remain unknown. Herein, the role of the AT-hook protein AKNA in regulating pluripotency and stemness in neuroblastoma cells is demonstrated through gene knockdown, immunofluorescence, chromatin immunoprecipitation (ChIP), localization of AKNA, signaling interactions, and transcriptional activity. AKNA was detected primarily in the nucleus during the induction of pluripotency and was retained in the cytosol by FAK signaling dur…

Demonstration of beat-to-beat, on-demand ATP synthesis in ventricular myocytes reveals sex-specific mitochondrial and cytosolic dynamics.

The energetic demands on ventricular myocytes imposed by the transport of ions and cross-bridge cycling are well known, yet the spatiotemporal dynamics of ATP supply and demand remain poorly understood. Here, using confocal microscopy and genetically encoded fluorescent sensors targeted to mitochondria and cytosol, we visualized beat-to-beat ATP dynamics in ventricular myocytes from male and female mice. These probes showed fluctuations in mitochondrial ATP levels with each contraction, revealing two distinct, spatially localized waveforms - ATP 'gain' and ATP 'dip' - representing transient in…

Extracellular pneumolysin enhances the activation of cytosolic pattern-recognition receptor NOD2 through its pore-forming activity.

Streptococcus pneumoniae is the leading cause of community-acquired pneumonia, meningitis, and sepsis. This bacterium produces pneumolysin, a cholesterol-dependent cytolysin that forms oligomeric transmembrane pores in the host cells. Although pneumolysin is known to exhibit proinflammatory properties, the mechanisms by which it activates innate immune responses remain to be investigated. Here, we show that extracellular pneumolysin enhances the activation of nucleotide-binding oligomerization domain 2 (NOD2), a cytosolic receptor that recognizes bacterial peptidoglycans. Experiments using HEK…
