Effects of Several Therapeutic Agents on Mammalian Vestibular Function: Meclizine, Diazepam, and JNJ7777120.

Management of vestibular dysfunction may include treatment with medications that are thought to act to suppress vestibular function and reduce or eliminate abnormal sensitivity to head motions. The extent to which vestibular medications act centrally or peripherally is still debated. In this study, two commonly prescribed medications, meclizine and diazepam, and a candidate for future clinical use, JNJ7777120, were evaluated for their effects on short latency compound action potentials generated by the peripheral vestibular system and corresponding central neural relays (i.e., vestibular senso…

Benzodiazepine modulation of homomeric GABAAρ1 receptors: differential effects of diazepam and 4'-chlorodiazepam.

GABA(A) receptors (GABA(A)Rs) are ligand-gated ion channels that mediate inhibitory neurotransmission in the central nervous system (CNS). T … ecorded in the presence or absence of BDZs. GABA(A)ρ1 receptor-mediated responses were modulated by diazepam and 4'-chlorodiazepam in the micromolar range, in a concentration-dependent, voltage-independent and reversible manner. Diazepam produced potentiating effects on GABA-evoked Cl(-) currents and 4'-Cl diazepam induced biphasic effects depending on the …

Salvianolic acid A exerts antiamnesic effect on diazepam-induced anterograde amnesia in mice.

Benzodiazepine was known to produce amnesia. Salvianolic acid A extracted from Salvia miltiorrhiza was an effective antioxidant. The objective of the study was to evaluate the effect of salvianolic acid A on diazepam-induced amnesia in mice. C57BL/6 mice were treated with salvianolic acid A at doses of 10, 20 and 40 mg/kg following administration with diazepam at a dose of 3 mg/kg. Morris water maze was performed to evaluate the effect of salvianolic acid A on amnesia. The antioxidative parameters in hippocampus were measured. The results showed that salvianolic acid A decreased the mean escap…

Midazolam or diazepam administration during electroencephalography helps to diagnose subacute sclerosing panencephalitis (SSPE).

Although diagnostic contribution of intravenous diazepam administration during electroencephalography (EEG) recording in subacute sclerosing panencephalitis has been known, no another drug with less potential side effects has been studied in this procedure. In this study, diazepam is compared with midazolam in 25 subacute sclerosing panencephalitis-diagnosed children and 10 children with subacute sclerosing panencephalitis-suggesting symptoms, normal EEG findings and no certain diagnosis. Neither midazolam nor diazepam affected typical periodic slow-wave complexes. However, in the patients wit…

Molecularly imprinted solid-phase extraction of diazepam and its metabolites from hair samples.

An anti-diazepam, molecularly imprinted polymer (MIP) has been synthesized and used to extract diazepam and other benzodiazepines from hair samples via a molecularly imprinted solid-phase extraction (MISPE) protocol. Optimum retention of diazepam on the MIP columns was achieved using an apolar solvent, and the binding capacity of the polymer toward diazepam was found to be 110 ng of diazepam/mg of polymer. The recovery of a 50 ng diazepam standard spiked into blank hair was 93%, with good precision (RSD = 1.5%). The LOD and LOQ of diazepam in spiked hair samples were 0.09 and 0.14 ng/mg, respe…

Lidocaine or diazepam can decrease fasciculation induced by succinylcholine during induction of anesthesia.

Succinylcholine is used during induction of anesthesia, and it may induce fasciculations. In this study we demonstrated that intravenous diazepam (1 mg/kg) or lidocaine (1.5 mg/kg) can decrease fasciculations induced by succinylcholine. There is no significant difference between these two drugs in reducing fasciculations moreover, these drugs can also prevent raised blood pressure and heart rate during intubation.

Analysis of multicomponent formulations containing phenylpropanolamine hydrochloride, caffeine and diazepam by using LC.

A reverse phase high performance liquid chromatography assay was carried out for the simultaneous determination of three active principles present in tablets of different origin and wide commercial use in the Province of Córdoba (Argentina). Prescriptions, commercially available as appetite suppressants, very often include the active principles Phenylpropanolamine Hydrochloride (I), Caffeine (II) and Diazepam (III). Simultaneous determination of these three drugs: anorexic, central nervous stimulant and tranquilizer, respectively, in pharmaceutical dosage forms has not been reported. In this s…

Corticosterone increases severity of acute withdrawal from ethanol, pentobarbital, and diazepam in mice.

It has been suggested that withdrawal from several subclasses of central nervous system (CNS) depressants involves common underlying mechanisms. For example, mice genetically selected for severe ethanol withdrawal convulsions (Withdrawal Seizure Prone or WSP) have also been found to express severe withdrawal following treatment with barbiturates and benzodiazepines. Corticosteroids appear to modulate severity of withdrawal from CNS depressants. Therefore, it was hypothesized that corticosterone would enhance withdrawal convulsions following acute ethanol, pentobarbital, and diazepam in WSP mic…

Transduction of a discriminative stimulus through a diazepam-insensitive gamma-aminobutyric acidA receptor.

Although there appear to be a large number of possible isoforms of the gamma-aminobutyric acid (GABA)A receptor, the relationship of a particular isoform to a specific behavior or behavioral process has not been identified. In the present study we describe a functional role for a diazepam-insensitive (DI) isoform of the GABAA receptor in the control of a complex discrimination. Pigeons were trained to discriminate the benzodiazepine antagonist flumazenil (100 micrograms/kg) from vehicle by requiring 30 keypeck responses on one key when flumazenil was given and on a different key when vehicle w…

Basolateral amygdala lesions block diazepam-induced anterograde amnesia in an inhibitory avoidance task.

This experiment examined the effects of diazepam (DZP) on acquisition and retention of an inhibitory avoidance response by rats with excitotoxic-induced lesions of central (CE), lateral (LAT), or basolateral (BL) amygdala nuclei. Sham-operated and lesioned rats received i.p. injections of DZP (2.0 mg per kg of body weight) 30 min before training in a continuous multiple-trial inhibitory avoidance task. Retention was tested 48 h later. Acquisition was not impaired by the lesions or the DZP. Retention was impaired in animals with CE and LAT lesions in comparison with sham-operated controls. DZP …

Diazepam-insensitive [3H]Ro 15-4513 binding in intact cultured cerebellar granule cells.

[3H]Ro 15-4513, a partial inverse agonist at the benzodiazepine receptor, binds to two sites in the rat cerebellum, only one of which is sensitive to diazepam. The diazepam-insensitive component, whose identity is unknown, is unique to this brain area. We studied the binding of [3H]Ro 15-4513 in cultured cerebellar granule cells to characterize its binding sites in a specified neuronal cell population and to determine the effects of ethanol on the binding. We also compared the properties of [3H]Ro 15-4513 binding in washed membranes of cultured cells and 14-day-old rat cerebella. [3H]Ro 15-451…

[Double-blind study of clobazam (Odipam) and diazepam in non-psychotic anxiety states].

Clobazam (Odipam) and Diazepam were investigated by the double blind study method in patients with nonpsychotic anxiety. It was found out that both drugs had equal anxiolytic and tranquilizing effect with the same adverse effects. Greater frequency of excellent marks concerning the final therapeutic effect of Odipam in respect to other marks pointed to its better clinical tolerance, so that this drug was recommended as daily tranquilizer.

Diazepam binding inhibitor gene expression: location in brain and peripheral tissues of rat.

Diazepam binding inhibitor (DBI), an endogenous 10-kDa polypeptide was isolated from rat and human brain by monitoring displacement of radioactive diazepam bound to specific recognition sites in brain synaptic and mitochondrial membranes. The cellular location of DBI mRNA was studied in rat brain and selected peripheral tissues by in situ hybridization histochemistry with a 35S-labeled single-stranded complementary RNA probe. DBI mRNA was heterogeneously distributed in rat brain, with particularly high levels in the area postrema, the cerebellar cortex, and ependyma of the third ventricle. Int…

Home cage pretreatment with diazepam: effects on subsequent conflict testing and rotorod assessment.

In various anxiolytic tests, benzodiazepines produce a maximal anxiolytic response only upon repeated treatment. The question arises as to whether repeated treatment produces an increase in anxiolytic efficacy or a decrease in another benzodiazepine-mediated effect (i.e., muscle relaxation/sedation) that functions to attenuate the full expression of the anxiolytic response. To address this issue, rats were pretreated in their home cages with either vehicle, diazepam or CGS 9896 (a nonsedating, non-muscle relaxant partial benzodiazepine agonist with anxiolytic efficacy) and challenged, 7 days l…

Up-regulation of brain [3H]diazepam binding sites in chronic caffeine treated rats.

Brain [3H]diazepam and [3H]L-phenylisopropyladenosine binding sites in caffeine treated (75 mg/kg/day, i.p. 12 days) and caffeine withdrawn (30 days) rats were examined. Treatment with caffeine (75 mg/kg/day) for 12 days increases the Bmax (maximum binding capacity) for [3H]diazepam binding by 30.9% whereas the same treatment increases the Bmax for [3H]L-PIA binding by 165%. The Bmax for [3H]diazepam binding sites returns to slightly below control levels but [3H]L-PIA binding sites remain elevated after 30 days of caffeine withdrawal. These findings suggest that the up-regulation of [3H]diazep…

Age-dependent enhancement of diazepam sensitivity is accelerated in New Zealand Black mice.

Separate age groups of C57BL/6 and autoimmune New Zealand Black (NZB) mice were compared for diazepam-induced ataxia and barbiturate-induced loss of righting reflex. Between 1 and 3 months of age, both strains showed a similar age-related decrease in ED50 for diazepam-induced ataxia. However, between 3 and 12 months the decrease in ED50 was markedly greater in NZB mice. In contrast, age-related increases in the durations of loss of righting reflex following hexobarbital or barbital were similar in both strains. The results suggest that NZB mice show relatively accelerated age-related increases…

Benzodiazepine antagonist Ro 15-1788. Antagonism of diazepam sedation in outpatients undergoing gastroscopy.

In a double-blind, randomised trial the efficacy and safety of Ro 15-1788, a new benzodiazepine antagonist, was assessed in forty adults undergoing gastroscopy under diazepam sedation. Criteria of efficacy were the degree of sedation and anterograde amnesia. There was a significantly faster recovery of the patients after the injection of 0.6-1.0 mg of Ro 15-1788 than after placebo. Patients were awake shortly after Ro 15-1788, but remained drowsy or asleep after placebo administration. There were no side effects of note.
