An experimental study of inner ear pathology due to NaCl on the round window.

An animal experiment was performed to evaluate structural and ultrastructural changes to the inner ear as a result of placing 3-4 crystals of reagent grade sodium chloride (NaCl) on the round window membrane. Chinchillas were sacrificed at 8 and 24 hours after treatment and the cochlear and vestibular tissues were examined by light microscopy and scanning electron microscopy. Inner ear pathology consisted of destruction of both sensory and supporting cells in the basal turn of the organ of Corti, atrophy of the stria vascularis and alterations to the otoconia and the maculae and ampullae of th

Axonal and cellular alterations in the inner ear of rats treated with chlorphentermine or iprindole.

An electron-microscopic study was carried out on the inner ear of rats, which had been treated with the anorectic drug chlorphentermine and the antidepressant drug iprindole, two cationic amphiphilic compounds known to induce a generalized lipidosis. After chronic drug treatment the following vestibular and cochlear alterations were observed: a) numerous lamellated and crystalloid cytoplasmic inclusion bodies in various cell types, typical of drug-induced lipidosis; b) axonal balloonings predominantly affecting preterminal sensory endings which were filled with masses of coarse osmiophilic inc

Monitoring pressure changes in the inner ear induced by middle ear pressure therapy with an EFET01 device in guinea pigs.

BACKGROUND: As a low-cost, portable, handheld air pressure generation tool not requiring a ventilation tube, the EFET01 device has shown clinical effectiveness for intractable Ménière's disease (MD) patients in Japan. However, no animal studies have investigated changes in inner ear pressure (PI) when applying this device. OBJECTIVE: To determine the PI properties in response to middle ear pressure therapy (MEPT) induced by the EFET01 in guinea pigs. MATERIAL AND METHODS: In seven healthy guinea pigs, bi-phasic pressure pulses from -5 to 12 cm H2O were delivered to the external ear canal and

Expression of macrophage migration inhibitory factor and CD74 in the inner ear and middle ear in lipopolysaccharide-induced otitis media.

CONCLUSION: Significant expression of macrophage migration inhibitory factor and its receptor (CD74) was observed in both the middle ear and inner ear in experimental otitis media in mice. Modulation of macrophage migration inhibitory factor and its signaling pathway might be useful in the management of inner ear inflammation due to otitis media. OBJECTIVES: Inner ear dysfunction secondary to otitis media has been reported. However, the specific mechanisms involved are not clearly understood. The aim of this study is to investigate the expression of macrophage migration inhibitory factor and

Morphologic changes in the inner ear of Chinchilla laniger after middle ear administration of gentamicin in a sustained-release vehicle.

The use of transtympanic gentamicin has become a popular method of treating Meniere's disease; nevertheless, many questions still remain regarding this therapy. Until investigators can control the exact amount of medicine that is administered to the ear and have an understanding of the kinetics of gentamicin, therapy will continue to rely on empirical data. Previously we described the use of a fibrin-based sustained-release vehicle impregnated with gentamicin in the middle ears of chinchillas. With this model a kinetics curve of gentamicin was defined. The inner ears of these animals were subm

Kinetics of gentamicin uptake in the inner ear of Chinchilla langier after middle-ear administration in a sustained-release vehicle.

The search for a safe, effective treatment for the vertigo associated with Meniere's disease has long been an important topic in otolaryngology. In recent years many groups have begun using intratympanic gentamicin to treat this vertigo. Although reported cure rates are as high as 90%, many questions remain regarding this type of treatment. Current limitations are the necessity for repeated treatments and a lack of clear dosing guidelines. In addition, the gentamicin must be delivered in a manner that allows maximal vestibulotoxic effect without injury to hearing. Until investigators can contr

Inner Ear Conductive Hearing Loss and Unilateral Pulsatile Tinnitus Associated with a Dural Arteriovenous Fistula: Case Based Review and Analysis of Relationship between Intracranial Vascular Abnormalities and Inner Ear Fluids.

While pulsatile tinnitus (PT) and dural arteriovenous fistula (DAVF) are not rarely associated, the finding of a conductive hearing loss (CHL) in this clinical picture is unusual. Starting from a case of CHL and PT, diagnosed to be due to a DAVF, we analyzed relationship between intracranial vascular abnormalities and inner ear fluids. DAVF was treated with endovascular embolization. Following this, there was a dramatic recovery of PT and of CHL, confirming their cause-effect link with DAVF. We critically evaluated the papers reporting this association. This is the first case of CHL associated

Isolated Temporal Bone Metastasis With Inner Ear Invasion as Manifestation of Rectal Cancer.

Temporal bone metastasis is a rare find. The most common sites of origin of temporal bone metastases are breast, lungs, and prostate. The main route of dissemination appears to be hematogenic. We present a case report of an 81-year-old male patient with locally advanced rectal cancer, without evidence of distant metastasis on CT of the chest, abdomen, and pelvis, presenting with rapidly progressive hearing loss, facial palsy, and loss of balance. Initial imaging is inconclusive, with meningioma being the leading hypothesis. However, histological analysis confirms the presence of carcinoma of c

The Blood-Labyrinth Barrier: Non-Invasive Delivery Strategies for Inner Ear Drug Delivery.

The inner ear is a relatively isolated organ, protected by the blood-labyrinth barrier (BLB). This barrier creates a unique lymphatic fluid environment within the inner ear, maintaining a stable physiological state essential for the mechano-electrical transduction process in the inner ear hair cells while simultaneously restricting most drugs from entering the lymphatic fluid. Under pathological conditions, dysfunction of the stria vascularis and disruption in barrier structure can lead to temporary or permanent hearing impairment. This review describes the structure and function of the BLB, a

Invisible saccule in 3T MRI: insights into inner ear vulnerability and fluid dynamics.

BACKGROUND: The trabecular mesh of the inner ear plays a critical role in maintaining the structure of the vestibular organs, particularly within the pars superior. Cases have been reported in which the saccule cannot be visualized in the pars inferior lacking this structure. OBJECTIVES: To investigate the clinical features and endolymphatic hydrops (EH) and perilymphatic enhancement (PE) on MRI in ears with an invisible saccule (IS) and to clarify the mechanism of IS. MATERIALS AND METHODS: Retrospective study of 89 IS ears and 71 contralateral visible saccule (VS) ears from 406 patients wi

Intracochlear Pressure Measurements Quantify Overall Noise Exposure to the Inner Ear During Mastoidectomy.

INTRODUCTION: The acoustic exposure patients endure during otologic surgery is unclear, as previous studies have not captured the effects of vibrational noise on the inner ear. To fill this knowledge gap, we examined inner ear noise directly via intracochlear pressure changes during mastoidectomy and cochleostomy. METHODS: Inner ear stimulation was measured using fiber optic pressure sensors inserted into the cochlea of cadaveric heads using a transcanal approach. A tympanomeatal flap was elevated, cochleostomies were made into scala vestibuli and tympani, sensors were placed and secured to t

Surgical procedure of intratympanic injection and inner ear pharmacokinetics simulation in domestic pigs.

Introduction: One major obstacle in validating drugs for the treatment or prevention of hearing loss is the limited data available on the distribution and concentration of drugs in the human inner ear. Although small animal models offer some insights into inner ear pharmacokinetics, their smaller organ size and different barrier (round window membrane) permeabilities compared to humans can complicate study interpretation. Therefore, developing a reliable large animal model for inner ear drug delivery is crucial. The inner and middle ear anatomy of domestic pigs closely resembles that of humans

Auckland hearing science discovery and translation in purinergic signaling and inner ear therapeutics.

The inner ear is a complex sensory organ responsible for hearing and balance. It is deeply embedded in the temporal bone with challenging access for diagnostic and therapeutic purposes. Stress and injury to the peripheral hearing organ (cochlea) lead to temporary or permanent sensorineural hearing loss (SNHL), which is the most common form of hearing loss resulting from cellular and molecular damage to the sensory hair cells and primary auditory neurons in the spiral ganglion. These cells cannot regenerate, and their loss leads to hearing disability. Hearing aids can amplify sound and improve

Superparamagnetic iron oxide nanoparticle regulates microbiota-gut-inner ear axis for hearing protection.

Noise-induced hearing loss (NIHL) is a highly prevalent form of sensorineural hearing damage that has significant negative effects on individuals of all ages and there are no effective drugs approved by the US Food and Drug Administration. In this study, we unveil the potential of superparamagnetic iron oxide nanoparticle assembly (SPIOCA) to reshape the dysbiosis of gut microbiota for treating NIHL. This modulation inhibits intestinal inflammation and oxidative stress responses, protecting the integrity of the intestinal barrier. Consequently, it reduces the transportation of pathogens and in
