Reconstruction of historical malaria transmission in Senegal using multiplex serocatalytic models.

The advent of multiplexing technologies, allowing antibodies to hundreds of antigens to be measured in a single test, has led to enormous increases in the amount of data generated by serological surveys. New modelling methods are required to exploit this data. This study extends serocatalytic models to consider up to three antibody responses targeting the same pathogen simultaneously. These models were fitted to data from cross-sectional serological surveys of Plasmodium falciparum malaria in the Senegalese villages of Dielmo and Ndiop, and model predictions were validated against 22 years of

Determine neighboring region spatial effect on dengue cases using ensemble ARIMA models.

The state of Selangor, in Malaysia consist of urban and peri-urban centres with good transportation system, and suitable temperature levels with high precipitations and humidity which make the state ideal for high number of dengue cases, annually. This study investigates if districts within the Selangor state do influence each other in determining pattern of dengue cases. Study compares two different models; the Autoregressive Integrated Moving Average (ARIMA) and Ensemble ARIMA models, using the Akaike Information Criterion (AIC) and Bayesian Information Criterion (BIC) measurement to gauge t

Exploring the link between PFASs exposure and gestational diabetes risk within the TRAEC framework.

Per- and polyfluoroalkyl substances (PFASs) are environmentally persistent industrial substances with potential health impacts during pregnancy. However, evidence linking PFASs exposure to gestational diabetes mellitus (GDM) remains inconclusive. This study systematically evaluated the PFASs-GDM association using the Toxicological Risk Assessment for Environmental Chemicals (TRAEC) strategy. A systematic literature search was conducted using PubMed and Web of Science to identify epidemiological studies, animal models, and cell-based experiments. To validate the evidence, we performed targeted

Evaluation of bi-directional causal association between periodontitis and benign prostatic hyperplasia: epidemiological studies and two-sample mendelian randomization analysis.

Background: Periodontitis and benign prostatic hyperplasia (BPH) are all common chronic diseases with higher incidence in middle-aged and old men. Several studies have indicated a potential association between periodontitis and BPH, although the findings remain inconclusive. However, there is no mendelian randomization (MR) studies to assess this association. Methods: The 40 men who had received health check-ups were included in an epidemiological study. Genetic data of BPH (13118 cases and 72799 controls) and periodontitis (3046 cases and 195395 controls) from FinnGen project was used to perf

Validation, calibration and refitting of a familial breast cancer model in sisterships: a case study in the Swedish sisters data.

In Sweden, a unique data set has been compiled with breast cancer incidence in all sisterships with at least two sisters born between 1932 and 2001, and the effect of family history has been analyzed by standard epidemiological methods. Such data are ideal to explore the validity of existing models for familial breast cancer. This paper explores the validity of the Jonker model that adds a hypothetical gene to the well-known BRCA1 and BRCA2 genes. The validity of the model for the Swedish data is checked by using a calibration model for breast cancer incidence given the (retrospective) family

The role of aquatic reservoir fluctuations in long-term cholera patterns.

We propose and analyze an important extension of standard cholera epidemiological models, explicitly accounting for fluctuations of water availability to the human community under study. The seasonality of water input in the reservoir drives the variation of concentration of Vibrio cholerae. Two compartments are added to the Susceptible-Infected-Bacteria model. First, the recovered individuals, which, over many seasons, lose their immunity to the disease and replenish the Susceptible group. Second, the water volume of the reservoir, which determines bacterial dilution and, consequently, the pr…

[Allergic respiratory diseases and outdoor air pollution].

INTRODUCTION: After having increased for some time, the prevalence of allergic diseases may have reached a plateau. During this increase, considerable concomitant changes in air pollution have occurred. Photo-oxidant air pollution, related to traffic, has become preponderant. The implication of air pollution in the epidemic of allergies is still debated. BACKGROUND: Experimental studies have suggested that the effect of air pollutants, including particulates and ozone, on the worsening and even the induction of allergies is biologically plausible. In addition, epidemiological studies have sho

The carcinogenicity of dietary acrylamide intake: a comparative discussion of epidemiological and experimental animal research.

Since 2002, it is known that the probable human carcinogen acrylamide is present in commonly consumed carbohydrate-rich foods, such as French fries and potato chips. In this review, the authors discuss the body of evidence on acrylamide carcinogenicity from both epidemiological and rodent studies, including variability, strengths and weaknesses, how both types of evidence relate, and possible reasons for discrepancies. In both rats and humans, increased incidences of various cancer types were observed. In rats, increased incidences of mammary gland, thyroid tumors and scrotal mesothelioma were

A real-time forecasting framework for emerging infectious diseases affecting animal populations.

Infectious disease forecasting has become increasingly important in public health. However, forecasting tools for emergency animal diseases, particularly those offering real-time decision support when parameters governing disease dynamics are unknown, remain limited. We introduce a generalised modelling framework for near-real-time forecasting of the temporal and spatial spread of infectious livestock diseases using data from the early stages of an outbreak. We applied the framework to the 2007 equine influenza outbreak in Australia, generating forecasts at three timepoints across four regiona

On the predictive ability of mechanistic models for the Haitian cholera epidemic.

Predictive models of epidemic cholera need to resolve at suitable aggregation levels spatial data pertaining to local communities, epidemiological records, hydrologic drivers, waterways, patterns of human mobility and proxies of exposure rates. We address the above issue in a formal model comparison framework and provide a quantitative assessment of the explanatory and predictive abilities of various model settings with different spatial aggregation levels and coupling mechanisms. Reference is made to records of the recent Haiti cholera epidemics. Our intensive computations and objective model

[Combating infectious disease using mathematical modelling].

When determining interventions against threatening infectious diseases such as HIV-infection, severe acute respiratory syndrome (SARS), smallpox and pandemic influenza, the use of mathematical models of the spread of infectious diseases is becoming increasingly popular. These models contribute to the structuring of the knowledge already available in various disciplines, to finding epidemiological connnections, to demonstrating lacunas within the pool of knowledge and to the comparison of the expected effects and costs of preventative and intervention measures. The use of models leads to a 'mad

Transmission patterns and the epidemiology of hookworm infection.

BACKGROUND: This paper presents a suite of models of hookworm transmission dynamics which vary the mixing patterns and rates of contamination and infection between children and adults. In this context mixing refers to the degree of epidemiological communication between children and adults, for example, whether adults are likely to get infected from infective material passed by children. METHODS: Three models are described which represent random mixing, no mixing and restricted mixing respectively. Child, adult and population targeted chemotherapy programmes are examined and compared between t

Analysis of an asymmetric two-strain dengue model.

In this paper we analyse a two-strain compartmental dengue fever model that allows us to study the behaviour of a Dengue fever epidemic. Dengue fever is the most common mosquito-borne viral disease of humans that in recent years has become a major international public health concern. The model is an extension of the classical compartmental susceptible-infected-recovered (SIR) model where the exchange between the compartments is described by ordinary differential equations (ode). Two-strains of the virus exist so that a primary infection with one strain and secondary infection by the other stra

Prediction of overexertion injuries using biomechanical and psychophysical models.

This paper summarizes a three year epidemiological study conducted in five large industrial plants in order to evaluate the validity of two alternative modeling approaches to overexertion injury prediction. Detailed biomechanical and psychophysical job evaluations were performed for 55 industrial jobs comprised of 2934 potentially stressful manual materials handling tasks. The medical experiences of 6912 incumbent workers were monitored retrospectively for two years and prospectively for one year to establish a data base for comparison of the different models. The results show that each of the

Peripheral Insulin Resistance and Alzheimer's Disease: Possible Mediators Including Extracellular Vesicles.

Epidemiological studies consistently indicate that type 2 diabetes mellitus increases the risk of Alzheimer's disease (AD). Accumulating evidence suggests that insulin resistance, rather than hyperglycemia per se, is the principal metabolic factor associated with AD development. Population-based longitudinal studies show that insulin resistance is strongly linked to earlier amyloid β (Aβ) accumulation, whereas its association with tau pathology remains inconsistent. Importantly, insulin resistance-related cognitive decline and earlier disease onset cannot be fully explained by the extent of Aβ

Measuring scientific coherence between global neglected tropical disease research and population health indicators: a 25-year meta-research study.

INTRODUCTION: Despite major investments in neglected tropical diseases (NTDs) research over the past two decades, it remains unclear whether global scientific production aligns with population-level health indicators. To address this gap, we designed and implemented a comprehensive meta-research analysis pipeline to quantify the scientific coherence between global NTDs research activity and epidemiological indicators across World Health Organization (WHO) regions from 2000-2024. METHODS: Using this pipeline, we linked 1,07,251 NTDrelated publications to 23 disease-specific indicators from WHO

Relationship between PFAS and attention-deficit/hyperactivity disorder or autism spectrum disorder: Epidemiological and experimental evidence.

Per- and polyfluoroalkyl substances (PFAS) are widespread environmental contaminants found in various consumer products, such as drinking water, infant formula, and food, as well as in human biospecimens, including blood, urine, breastmilk, and cerebrospinal fluid. Previous evidence indicates that PFAS exposure poses significant neurotoxic risks, owing to their ability to penetrate the blood-brain barrier and bioaccumulate in neural tissue. The developing brains of fetuses and infants are particularly susceptible to the harmful effects of environmental pollutants because of their incomplete ba

Infectivity and fatality of influenza in pre- and post-COVID-19 pandemic year.

The COVID-19 pandemic and related non-pharmaceutical interventions (NPIs) significantly alter the transmission dynamics of non-SARS-CoV-2 infectious diseases, with respiratory infections such as influenza being disproportionately affected. We aim to compare influenza's epidemiological characteristics between pre-pandemic and post-pandemic periods to inform public health responses. We develop two influenza transmission models incorporating age structure and multi-strain dynamics, featuring time-varying transmission and mortality rates. Using publicly available U.S. data, we calibrate these mode
