METTL3 Alleviates Liver Abscess by Enhancing Macrophage Efferocytosis via m6A Methylation of Nrf2 mRNA.

BACKGROUND: Persistent inflammation and impaired macrophage efferocytosis contribute to liver injury during bacterial liver abscesses (BLA). This study was conducted to investigate whether METTL3 alleviates BLA by promoting macrophage efferocytosis through m6A methylation of Nrf2 mRNA. MATERIALS AND METHODS: METTL3 was overexpressed in the liver of a Klebsiella pneumoniae-induced BLA mouse model. Liver injury, serum cytokine levels, Nrf2 signaling, m6A enrichment of Nrf2 mRNA, TAM receptor expression, and macrophage-associated Mertk and Nrf2 expression were evaluated. Nrf2 knockdown rescue ex

Mitochondrial DNA 6 mA methylation by METTL4 drives neuroinflammation via cGAS-STING activation in vascular cognitive impairment.

BACKGROUND: Vascular cognitive impairment (VCI) is strongly associated with mitochondrial dysfunction, yet the underlying molecular mechanisms connecting mitochondrial impairment to neuroinflammation remain elusive. While mitochondrial epigenetic modifications are emerging as key regulators of cellular metabolism, the role of mitochondrial DNA (mtDNA) N6-methyladenine (6 mA) modification and its writer enzyme METTL4 in VCI pathogenesis has not been established. METHODS: Using complementary in vitro (oxygen-glucose deprivation, OGD) and in vivo (chronic cerebral hypoperfusion, CCH) models of V

p53 hotspot mutants attenuate CTL-mediated tumor cell killing through a novel ALKBH5-YTHDF3-PD-L1 pathway.

TP53 is a well-documented tumor suppressor gene frequently mutated in malignancies. It has been demonstrated that the gain-of-function (GOF) mutation of p53 promotes the development and progression of cancers; however, its extrinsic oncogenic mechanisms are still poorly understood. Herein, we found that the oncogenic effect of mutant p53 in vivo is dependent on the tumor immune microenvironment. Importantly, our data demonstrated a significant increase in PD-L1 expression in cells expressing mutant p53. Consistently, subsequent investigations indicated that PD-L1 mRNA is subject to N6-methylad

Integrating Methylation Signature Modifications and Subtype Classification into a Prognostic Risk Model for Survival Prognosis and Predicting Immunotherapy Response in Colorectal Cancer.

BACKGROUND: The prognosis for colorectal cancer (CRC) is poor, and the disease is marked by high rates of morbidity and death, highlighting the need for reliable biomarkers. RNA methylation modifications play important roles in cancer biology. However, the integrated role of m6A/m5C/m1A/m7G modifications in CRC has not been fully characterized and requires further investigation. METHODS: Data from The Cancer Genome Atlas colon adenocarcinoma cohort (TCGA-- COAD) were used as the training set, and GSE17536 served as the validation cohort. We further investigated the probable biological process

BALF editome profiling reveals A-to-I RNA editing associated with severity and complications of Mycoplasma pneumoniae pneumonia in children.

Mycoplasma pneumoniae is an important human respiratory pathogen that causes mild-to-moderate community-acquired M. pneumoniae pneumonia (MPP), particularly in children. RNA editing plays a vital role in pathogen infection and host immune response, but it remains largely unknown how it could be involved in the epigenetic regulation of host response to M. pneumoniae infection. In the present study, we performed an epitranscriptomic analysis of adenosine to inosine (A-to-I) editing in 39 bronchoalveolar lavage fluid (BALF) samples from the severe side (SS) and the opposite side (OS) of patients

AVCAPIR: A Novel Procalcific PIWI-Interacting RNA in Calcific Aortic Valve Disease.

BACKGROUND: Calcification of the aortic valve leads to increased leaflet stiffness and consequently results in the development of calcific aortic valve disease (CAVD). However, the underlying molecular and cellular mechanisms of calcification remain unclear. Here, we identified a novel aortic valve calcification-associated PIWI-interacting RNA (piRNA; AVCAPIR) that increases valvular calcification and promotes CAVD progression. METHODS: Using piRNA sequencing, we identified piRNAs contributing to the pathogenesis of CAVD that we termed AVCAPIRs. High-cholesterol diet-fed ApoE-/- mice with AVC

Intranuclear paraspeckle-circular RNA TACC3 assembly forms RNA-DNA hybrids to facilitate MASH-related hepatocellular carcinoma growth in an m6A-dependent manner.

BACKGROUND: Metabolic dysfunction-associated steatohepatitis (MASH) is anticipated to become the leading cause of hepatocellular carcinoma (HCC). Accumulating evidence indicates that N6-methyladenosine (m6A)-modified circular RNAs (circRNAs) play key roles in tumor malignant progression. However, the precise molecular mechanisms by which circRNAs and their m6A modification regulatory networks respond to metabolic reprogramming, such as lipid overload stress, to drive malignant tumor progression in the context of MASH-related HCC remain unclear. This study aimed to investigate the role and regu

Methyltransferase METTL1 Upregulates USF2 in a m7G-Dependent Manner to Accelerate Septic Cardiomyopathy by Inactivating PINK1/Parkin-Mediated Mitophagy.

Septic cardiomyopathy (SCM) is a severe complication of sepsis, characterized by high mortality. The activation of mitophagy in cardiomyocytes is crucial for alleviating SCM. The N7-methylguanosine (m7G) modification mediated by methyltransferase 1 (METTL1) is known to negatively regulate mitophagy. This study investigated the mechanism of METTL1 in the mitophagy of cardiomyocytes in SCM. Human cardiomyocytes (HCMs) were stimulated with lipopolysaccharide (LPS) to establish the SCM model. C57BL/6 mice underwent cecal ligation and puncture (CLP) surgery to create a septic model. Mitophagy was a

METTL3-driven m6A modification promotes tumor progression and immune evasion in hypoxic pancreatic cancer by enhancing ADAM10-mediated MICA shedding.

BACKGROUND: Pancreatic cancer is characterized by asymptomatic early stages, fast progression, and dismal survival rates. Therefore, a better understanding of immune evasion is required to create more effective treatments. m6A methylation is linked to pancreatic cancer, particularly in hypoxia-induced immune escape. RESULTS: GEO microarray datasets identified pancreatic cancer differentially expressed genes (DEGs) and m6A modification regulators. Compared with controls, pancreatic cancer samples showed increased METTL3 levels. METTL3 levels are strongly associated with HIF1A, a key component

METTL14 inhibits atherogenesis by epigenetically activating PPAR-α/γ transcription and fatty acid oxidation in VSMCs.

AIMS: N6-methyladenosine (m6A) RNA modification can govern cell fate by co- or post-transcriptionally regulating gene expression. VSMCs can undergo phenotypic switching, contributing to other cells within atherosclerotic plaques, including foam cell- and macrophage-like cells. However, the role of VSMC m6A in atherosclerosis development remains unclear. While PPAR-α and PPAR-γ have been extensively studied in macrophages for their roles in atherosclerosis, the epigenetic regulation of these nuclear receptors under high cholesterol conditions remains poorly understood. METHODS AND RESULTS: We

METTL14-mediated m6A methylation orchestrates early NK cell development by maintaining metabolic fitness.

N6-methyladenosine (m6A) represents the most abundant mRNA modification, yet its role in natural killer (NK) cell development remains incompletely understood. Here we demonstrate that the m6A methyltransferase METTL14 plays an indispensable, stage-specific role in early NK cell development. Mettl14 deficiency at the progenitor stage caused severe NK cell lymphopenia by disrupting the NK progenitor to immature NK cell transition. Paradoxically, residual Mettl14-null NK cells exhibited a hypermetabolic state characterized by mTORC1 hyperactivation and enhanced mitochondrial function, which drove

METTL1-mediated m7G modification of ASNS mRNA regulates asparagine metabolism reprogramming to promote hepatocellular carcinoma progression.

Hepatocellular carcinoma (HCC) is a prevalent and aggressive malignancy, notorious for its high recurrence, substantial drug resistance, and poor prognosis. Although METTL1, a key regulator of RNA m7G modification, is implicated in the progression of various cancers, its role and underlying mechanisms in HCC remain poorly understood. This study identified that METTL1 was significantly upregulated in HCC tissues, correlating with advanced stages and poor survival. Knockdown of METTL1 inhibited cell proliferation, migration, and invasion, which was reversed by restoring METTL1 expression. Multi-
