Microbial co-occurrence network and spatial metabolism underlie wheat cultivar effects on medium-high temperature Daqu fermentation.

Although wheat cultivar is widely accepted to shape Daqu quality, it remains unclear which cultivar-linked microbial property - taxonomic composition, spatial niche differentiation, or the topology of cross-domain interactions - is the proximate driver of flavor-precursor accumulation. To address this gap, four wheat cultivars (GS19, QM725, QM838 and ZM) were used as natural perturbations, and depth-resolved (core/surface) 16S/ITS profiling, untargeted metabolomics and volatilomics were combined across the 90-day fermentation-maturation trajectory of medium-high-temperature Daqu. Of the four c

Multi-omics reveals metabolic division of labor in mixed-starter soymilk fermentation.

Fermentation by multiple microorganisms significantly improves the sensory quality and nutritional value of soymilk, yet the molecular mechanisms underlying the synergistic interactions, particularly regarding substrate utilization and nutrient exchange were not clear. Here, metatranscriptomics and non-targeted metabolomics were integrated to characterize the microbial succession and metabolic reprogramming of a mixed starter culture (Streptococcus salivarius subsp. thermophilus, Lactobacillus delbrueckii subsp. bulgaricus, and Lactiplantibacillus plantarum) during soymilk fermentation. Taxono

Heterogenetic mechanism in multidimensional pit mud with different fermentation years: From microbial structure dynamic succession to metabolism phenotypes.

Pit mud (PM) is fermenting agents in the strong-flavor baijiu (SFB) production. In this paper, the discrepancies in fermentation parameters, microbial community succession patterns and metabolic phenotypes were compared in multidimensional PMs. The results showed that pyruvic acid, succinic acid, S-Acetyldihydrolipoamide-E, glycerol and glyceric acid were the key metabolites responsible for the metabolic differences between the 2-, 30-,100- and 300-year multidimensional PMs, while the butanoic acid, heptyl, heptanoic acid, heptanoic acid ethyl ester, hexanoic acid and octanoic acid were the ke

Microalgal culture in animal cell waste medium for sustainable 'cultured food' production.

'Cultured food' has tremendous potential as a sustainable meat alternative. Increased cultured food production is increasing the amount of waste medium from cell culture. Nitrogen- and phosphorus-containing compounds in waste medium can cause eutrophication of water bodies. Currently, microalgae are used in energy production, environmental protection, agriculture and pharmaceutical and health food industries. Here, we used the microalgae, Chlorococcum littorale and Chlorella vulgaris and the waste medium of C2C12 cells for a case study. We found that 80% and 26% of ammonia and 16% and 15% of p

Reduction of cyanide levels in cassava during sequential sundrying and solid state fermentation.

The cyanide levels were followed during protein enrichment of cassava by the fungus Aspergilus oryzae. The total cyanogen level decreased by 158 mg/kg dry weight to 54.2 mg/kg dry weight as a result of the whole process including fermentation. The cyanogenic glucoside level decreased by 88% during the fermentation process while acetone cyanohydrin was retained in the cassava. The prefermentation processing which involved crushing, sundrying and milling the cassava into flour, reduced the total cyanogen levels by 40%. The process resulted in considerable reduction in the cyanogenic content of t

Prevalence and diversity of tetracycline resistant lactic acid bacteria and their tet genes along the process line of fermented dry sausages.

In order to study the prevalence and diversity of tetracycline resistant lactic acid bacteria (Tc(r) LAB) along the process line of two different fermented dry sausage (FDS) types, samples from the raw meat, the meat batter and the fermented end product were analysed quantitatively and qualitatively by using a culture-dependent approach. Both the diversity of the tet genes and their bacterial hosts in the different stages of FDS production were determined. Quantitative analysis showed that all raw meat components of both FDS types (FDS-01 and FDS-08) contained a subpopulation of Tc(r) LAB, and

Cloning and expression of the lysostaphin gene in Bacillus subtilis and Lactobacillus casei.

The lysostaphin structural gene was cloned in Bacillus subtilis DSM402 and in Lactobacillus casei 102S. The gene was expressed in both organisms and active lysostaphin was released into the medium. Lysostaphin produced by these organisms induced lysis of growing and heat inactivated staphylococci. Expression in a protective starter organism is a prerequisite to produce lysostaphin in situ in fermenting foods and hence, to reduce the hygienical risk of staphylococcal food poisoning.

Fermentation of Youlk, an Australian Native Root Vegetable, Using Defined Lactic Acid Bacterial Strains.

Youlk (Platusace deflexa) is a native root vegetable in Australia that is primarily consumed fresh, baked, or roasted. This study aimed to produce a fermented product using youlk with added lactic acid bacteria (LAB), in comparison with spontaneous fermentations. Three LAB strains were selected for their high in vitro antioxidant activity, leading to eight formulations that featured individual strains or a 1:1:1 combination. The strains were Latilactobacillus sakei F1, Lacticaseibacillus paracasei D2 and Lacticaseibacillus rhamnosus JL, all previously isolated from fermented vegetables. Two sa

Characterization of the angiotensin-converting enzyme inhibitory activity of fermented milks produced with Lactobacillus casei.

Our study assayed angiotensin-converting enzyme (ACE) inhibitory activity and fermentation characteristics of 41 food-originated Lactobacillus casei strains in fermented milk production. Twenty-two of the tested strains produced fermented milks with a high ACE inhibitory activity of over 60%. Two strains (IMAU10408 and IMAU20411) expressing the highest ACE inhibitory activity were selected for further characterization. The heat stability (pasteurization at 63°C for 30 min, 75°C for 25 s, and 85°C for 20 s) and resistance to gastrointestinal proteases (pepsin, trypsinase, and sequential pepsin/

Food preparation with mucoralean fungi: A potential biosafety issue?

Mucorales have been used for production of fermented food in Asia and Africa since time immemorial. Particularly Rhizopus species are rapidly growing, active producers of lipases and proteases and occur naturally during the first stages of soybean fermentation. Two biosafety issues have been raised in recent literature: (1) pathogenicity, Rhizopus species being prevalent opportunists causing erosive infections in severely compromised patients, and (2) toxicity, strains harbouring endosymbiotic Burkholderia producing toxic secondary metabolites. At the molecular level, based on different gene m

Influence of a Co-culture of Streptococcus thermophilus and Lactobacillus casei on the Proteolysis and ACE-Inhibitory Activity of a Beverage Based on Reconstituted Goat Whey Powder.

The potential for reusability of whey is of concern due to its substantial nutritional value and, second, in view of the need to reduce environmental impact. The use of goat whey powder in the production of a fermented dairy beverage using a Lactobacillus casei culture as adjunct shows a good prospect of a food product with hypotensive activity. This study investigated the microbial viability, proteolysis and angiotensin-converting enzyme (ACE) inhibitory activity of a fermented dairy beverage produced with goat whey powder and a probiotic culture of L. casei BGP93 co-cultured with Streptococc

Small Peptides Isolated from Enzymatic Hydrolyzate of Fermented Soybean Meal Promote Endothelium-Independent Vasorelaxation and ACE Inhibition.

Fermentation of soybean is a process in which soy proteins are broken down into small peptides to exert various physiological functions beyond their nutritional value and to improve food source bioactive components responsible for health benefits. Enzymatic hydrolysis could speed up the degradation of proteins during fermentation of soybean, thus resulting in higher peptide production. In the present study, fermented soy meal (fermented with Bacillus subtilis from Douchi) was hydrolyzed by thermolysin, and the water extraction was then separated into four fractions using ultrafiltration membra

Beneficial effect of oat-based postbiotics produced through capsule or free-cell fermentation on lipopolysaccharide activated intestinal cells.

The aim of this study was to design an innovative oat fermentation process using capsules to produce postbiotics with anti-inflammatory and antioxidant potential. Capsule fermentation led to higher bacterial growth (3.30*109 CFU/mL) and lactic acid production (20.49 mg/mL), compared to traditional free-cell fermentation (4.73*108 CFU/mL and 9.08 mg/mL, respectively), along with increased cell and product yields. The dried postbiotics were also subjected to in vitro digestion, mimicking the gastrointestinal one. A functional characterization of the four postbiotics, undigested and digested post

Enhanced functionality and reduced allergenicity of silkworm pupae protein through Lactiplantibacillus plantarum fermentation.

With rising demand for sustainable protein sources, insect proteins have gained attention for their nutritional value and environmental advantages. Silkworm pupae protein (SPP) is rich in essential amino acids and bioactives but has limited application due to poor solubility and high allergenicity. This study employed Lactiplantibacillus plantarum A2 (A2) to ferment SPP and evaluated structural, functional, and allergenic changes in vitro and in mice. Fermentation increased α-helix content, surface hydrophobicity, and sulfhydryl groups while reducing β-sheet content and fluorescence. Hydrolysi

Salt-dependent osmoadaptation and metabolic responses of Lactiplantibacillus plantarum BS3-1 in fermented bamboo shoots.

Salt concentration shapes the growth and metabolism of lactic acid bacteria in vegetable fermentations, yet the effects of salt gradients on the fermentation performance of suansun-derived Lactiplantibacillus plantarum BS3-1, together with associated temporal transcriptional changes in BS3-1 and metabolite variation in the fermentation broth, remain insufficiently characterized. Here, BS3-1 was evaluated across a 0-7% (w/w) salt gradient during suansun fermentation. The 0%, 3%, and 5% treatments were selected for time-series transcriptomic analysis of fermentation-derived cell pellets and unta
