Expression of the RET receptor tyrosine kinase and GDNFR-alpha in normal and leukemic human hematopoietic cells and stromal cells of the bone marrow microenvironment.

The RET proto-oncogene product is a receptor tyrosine kinase representing the signal-transducing molecule of a multisubunit surface receptor complex for the glial cell line-derived neurotrophic factor (GDNF), in which a novel glycosyl-phosphatidylinositol (PI)-linked protein (termed GDNFR-alpha) acts as the ligand-binding component. We have analyzed expression of RET and GDNFR-alpha in purified normal hematolymphopoietic cells, leukemia/lymphoma cell lines, and 154 primary samples from patients with hematopoietic malignancies encompassing different lineages and differentiation stages. Human Kaposi's sarcoma cell-mediated tumorigenesis in human immunodeficiency type 1 tat-expressing transgenic mice.

BACKGROUND: The human immunodeficiency virus type 1 (HIV-1) transactivator (Tat) protein has been linked to the development and course of Kaposi's sarcoma (KS) associated with acquired immunodeficiency disease syndrome (AIDS-KS). Tat is an 86-101 amino-acid protein encoded by two exons. To evaluate the growth-promoting effects of Tat in AIDS-KS in vivo, we developed transgenic mice expressing the one-exon-encoded 72 amino-acid protein (Tat(72)) and the two-exon-encoded 86 amino-acid protein (Tat(86)). METHODS: Human KS SLK cells were injected subcutaneously into CD4(+) T-cell-depleted male mi

Evaluation of folate receptor-alpha and other surface markers as potential targets for radionuclide therapy of ovarian cancer.

BACKGROUND: Epithelial ovarian cancer (EOC) has the highest mortality rate among gynecological malignancies, primarily due to frequent late-stage diagnosis and the development of resistance to chemotherapy. The aim of this study was to evaluate the expression of tumor-associated targets in a large cohort (n = 179) of various EOC subtypes, represented on two tissue microarrays (TMAs), to support the future development of radionuclide therapies. The study primarily assessed folate receptor-alpha and -beta isoforms (FRα and FRβ), but also somatostatin receptor-2 (SSTR2), prostate-specific membran

Artesunate alleviates kidney fibrosis by restoring klotho protein and suppressing Wnt/β-catenin signalling pathway.

Chronic kidney disease (CKD) is a global health concern that often progresses to renal failure and premature death. Regardless of etiology of CKD, kidney fibrosis is the main determinant of progressive CKD. Renal fibrosis is characterized by excessive collagen and extracellular-matrix (ECM) deposition, which impairs renal function with an irreversible loss of nephrons. Currently, there are no effective antifibrotic therapies to halt the progression of CKD to the end-stage-kidney-failure (ESKF). Artesunate has recently shown antifibrotic effects in various animal models, but its efficacy in ren

2-Styrylchromones Prevent IL-1β-Induced Pro-Inflammatory Activation of Fibroblast-like Synoviocytes while Increasing COX-2 Expression.

Rheumatoid arthritis (RA) is characterized by systemic immune and chronic inflammatory features, leading to the destruction of the joints. Presently, there are no effective drugs able to control synovitis and catabolism in the process of RA. 2-Styrylchromones (2-SC) are a small group of compounds characterized by the attachment of a styryl group to the chromone core that have already been associated to a wide range of biological activities, including antioxidant and anti-inflammatory activities. The present study investigated the effect of a set of six 2-SC on the interleukin-1β (IL-1β)-induce

Targeting carcinoma-associated mesothelial cells with antibody-drug conjugates in ovarian carcinomatosis.

Ovarian carcinomatosis is characterized by the accumulation of carcinoma-associated mesothelial cells (CAMs) in the peritoneal stroma and mainly originates through a mesothelial-to-mesenchymal transition (MMT) process. MMT has been proposed as a therapeutic target for peritoneal metastasis. Most ovarian cancer (OC) patients present at diagnosis with peritoneal seeding, which makes tumor progression control difficult by MMT modulation. An alternative approach is to use antibody-drug conjugates (ADCs) targeted directly to attack CAMs. This strategy could represent the cornerstone of precision-ba

Potential role of fibroblast-like synoviocytes in joint damage induced by Brucella abortus infection through production and induction of matrix metalloproteinases.

Arthritis is one of the most common complications of human brucellosis, but its pathogenic mechanisms have not been elucidated. Fibroblast-like synoviocytes (FLS) are known to be central mediators of joint damage in inflammatory arthritides through the production of matrix metalloproteinases (MMPs) that degrade collagen and of cytokines and chemokines that mediate the recruitment and activation of leukocytes. In this study we show that Brucella abortus infects and replicates in human FLS (SW982 cell line) in vitro and that infection results in the production of MMP-2 and proinflammatory mediat

A Multi-Target Pharmacological Correction of a Lipoyltransferase LIPT1 Gene Mutation in Patient-Derived Cellular Models.

Mutations in the lipoyltransferase 1 (LIPT1) gene are rare inborn errors of metabolism leading to a fatal condition characterized by lipoylation defects of the 2-ketoacid dehydrogenase complexes causing early-onset seizures, psychomotor retardation, abnormal muscle tone, severe lactic acidosis, and increased urine lactate, ketoglutarate, and 2-oxoacid levels. In this article, we characterized the disease pathophysiology using fibroblasts and induced neurons derived from a patient bearing a compound heterozygous mutation in LIPT1. A Western blot analysis revealed a reduced expression of LIPT1 a

Expression of the integrin alpha8beta1 during pulmonary and hepatic fibrosis.

The fibrotic response after diverse forms of injury is characterized by the accumulation of extracellular matrix proteins, proliferation of myofibroblast-like cells, and organ contraction. Myofibroblasts are key effector cells in the development of the fibrotic response. They contribute to fibrosis through both increased cell number (proliferation) and enhanced matrix synthesis. Integrins, a class of cell adhesion molecules, are mediators of cell-extracellular matrix protein interactions that are important in the proliferative and migratory response of cells to matrix proteins. We have previou

Development and evaluation of 18F-labeled novel radiopharmaceuticals for PET imaging of fibroblast activation protein expressing tumors.

This study aimed to develop and evaluate two novel 18F-labeled radiopharmaceuticals, [18F]AlF-NOTA-De-FAPI and [18F]AlF-NOTA-Glu-FAPI, for Positron Emission Tomography (PET) imaging of tumors expressing fibroblast activation protein. Molecular docking simulations predicted the binding affinities of NOTA-De-FAPI and NOTA-Glu-FAPI with FAP. The radiotracers were synthesized and evaluated for radiochemical yield, purity, and molar activity. Surface plasmon resonance analysis measured binding kinetics. In vitro and in vivo stability and distribution coefficients were assessed. PET/CT imaging and e

Evaluation of [18F]FAPI-74 PET/CT in healthy dogs and in West Highland white terriers with canine idiopathic pulmonary fibrosis: a pilot study.

Canine idiopathic pulmonary fibrosis (CIPF) is a fatal disease affecting primarily West Highland white terriers (WHWT). CIPF remains challenging to diagnose and disease progression is difficult to predict. Recently, fibroblast activation protein (FAP) was identified as a cellular marker of active fibrosis in post-mortem lung biopsies from CIPF-affected WHWTs. Therefore, FAP-targeted imaging using FAP inhibitors (FAPI) may offer a noninvasive means of assessing active fibrosis in canine lungs in vivo. This study aimed to evaluate whether [18F]FAPI-74 positron emission tomography (PET) combined

Anti-Wrinkling and Anti-Melanogenic Effect of Pradosia mutisii Methanol Extract.

Ultraviolet (UV) exposure causes skin photoaging leading to skin wrinkling and sagging via production of reactive oxygen species (ROS). For this reason, protection from photoaging is an important feature in cosmeceutical and dermatological products. Natural product-derived biomaterials are highly desired as future possible ingredients, because these biomaterials are often safe and effective. In this study, we aimed to characterize the skin protective activity of Pradosia mutisii, traditionally used to treat sunburn and erythema. We determined the free radical scavenging, anti-melanogenic, and

[18F]F-FAPI PET/CT is superior to [18F]F-FDG PET/CT for lymph node staging in lung cancer: a pathologically validated study.

PURPOSE: Accurate lymph node staging in lung cancer remains challenging. This study evaluates the diagnostic efficacy of fibroblast activation protein inhibitor (FAPI) versus fluorodeoxyglucose (FDG) positron emission tomography/computed tomography (PET/CT) using histopathology as the reference standard. METHODS: Thirty-two lung cancer patients with FDG-positive lymph nodes underwent [¹⁸F]F-FAPI PET/CT. Pathological confirmation of 71 lymph nodes (23 metastatic, 48 benign) was obtained via surgery or endobronchial ultrasound-guided transbronchial needle aspiration (EBUS-TBNA). Diagnostic effi

Myeloid-lineage CAR knockin mice enable allogeneic immunotherapy for liver and lung fibrosis.

The preparation of chimeric antigen receptor immune cells (CAR-ICs) typically involves a costly and time-intensive process. Moreover, the transduction of CAR genes into ICs, particularly macrophages, presents inherent challenges. Here, we report a novel strategy to obtain CAR-modified macrophages (CAR-Ms) directly from living animals to treat pulmonary and hepatic fibrosis. We first validated adenovirus-transduced CAR-Ms targeting fibroblast activation protein (FAP) in vitro, as well as in mouse models of pulmonary and hepatic fibrosis. Using a Cre/LoxP strategy, we generated mice with myeloid

Inflammatory 68 Ga FAPI-46 Uptake Mimicking a Sister Mary Joseph's Nodule in a Case of Gastric Signet Ring Cell Carcinoma.

Fibroblast activation protein (FAP) is a type II transmembrane protease expressed in cancer-associated fibroblasts (CAF) that constitute a part of the tumor stroma and influence the tumor microenvironment. Although FAP is not expected to be physiologically expressed in noncancer-associated cells, it can be observed in wound healing, arthritis, atherosclerotic plaques, and fibrosis. In this case, we would like to present early-stage 68 Ga FAPI-46 PET/CT inflammation findings at the trocar entry site mimicking the Sister Mary Joseph's nodule in a patient diagnosed with gastric signet ring cell c

Fibroblast Activation Protein Acts as a Biomarker for Monitoring ECM Remodeling During Aortic Aneurysm via 68Ga-FAPI-04 PET Imaging.

Traditional imaging modalities used to monitor the diameter of aortic aneurysms (AAs) often fail to follow pathological progression. Fibroblast activation protein (FAP), a key regulator of extracellular matrix (ECM) remodeling, plays a pivotal role in aortic disease. However, its expression in the aortic wall during aneurysm progression and its potential correlation with disease severity remains unexplored. Here, utilizing histology the levels of FAP are higher in the aortic wall of patients with AA compared to healthy controls. In three distinct animal models of AA, a progressive increase in

Harnessing the Potential of FAP-IL12mut TMEkine for Targeted and Enhanced Antitumor Responses.

Although cancer immunotherapy has yielded encouraging outcomes in hematologic malignancies, it has faced challenges in achieving the same level of effectiveness in numerous solid tumors, primarily because of the presence of immunosuppressive tumor microenvironments (TME). The immunosuppressive qualities of the TME have generated considerable interest, making it a focal point for treatments aimed at enhancing immune responses and inhibiting tumor progression. Fibroblast activation protein (FAP), an attractive candidate for targeted immunotherapy, is prominently expressed in the TME of various s
