1. Rhubarb, rhizome of Rheum palmatum L. (RP), is an important herb in clinical Chinese medicine. 2. Cyclosporine (CSP) is an immunosuppressant with narrow therapeutic window. The oral bioavailability of CSP was associated with P-glycoprotein (P-gp) and CYP 3A4. CSP was used as a probe substrate to investigate the in vivo modulation effects of RP on P-gp and CYP 3A. 3. Rats were orally administered 2.5 mg/kg of CSP with and without 0.25 and 1.0 g/kg of RP. The blood CSP concentration was determined by a specific monoclonal fluorescence polarization immunoassay. 4. Both dosages of RP significan…

Modulation of the vitamin D receptor (VDR) with a ligand has the potential to be useful for the oral treatment of osteoporosis. One component of our lead generation strategy to identify synthetic ligands for VDR included a fragment based drug design approach. Screening of ligands in a VDR fluorescence polarization assay and a RXR/VDR conformation sensing assay resulted in the identification of multiple fragment hits (lean >0.30). These fragment scaffolds were subsequently evaluated for interaction with the VDR ligand binding domain using hydrogen-deuterium exchange (HDX) mass spectrometry. Sig…

A new rapid slide agglutination test (Amniostat-FLM/AFLM) for amniotic fluid phosphatidylglycerol has been developed. One hundred ninety-three samples of amniotic fluid were tested for lecithin/sphingomyelin (L/S) ratio and phosphatidylglycerol by thin-layer chromatography and fluorescence polarization and compared with the AFLM. All four tests agreed in 123/193 cases. Phosphatidylglycerol and AFLM were concordant in 176/193 cases. From these samples, data were available on 105 neonates, of whom 74 were delivered within 72 hours of amniocentesis. All positive AFLM tests were associated with ab…

BACKGROUND: Organellar viscosity is an important biomechanical parameter and is closely associated with multiple physiological and pathological processes. Push-pull molecular rotors can realize viscosity visualization via changes in fluorescence intensity, providing a promising noninvasive strategy for organellar viscosity detection. However, most such molecular rotors are susceptible to interference from environmental polarity, human serum albumin and other substances, and tend to induce the aggregation-caused quenching (ACQ) effect, which severely limits the accuracy and sensitivity of visco

Developing highly fluorescent biogenic carbon dots (CDs) as molecular probes and cellular imaging agents with excellent cellular biocompatibility and water dispersibility can advance the scope of CDs in various biomedical applications. The present work aims to produce CDs from biological precursors such as natural amino acids. Due to their molecular recognition properties, the resultant CDs exhibit biocompatibility, polar functional groups, and selective molecule sensing. Microwave (MW)-assisted carbonization of L-tryptophan, L-glutamic acid, and their mixture led to the formation of CDs. Howe

The Pgp (P-glycoprotein) multidrug transporter couples ATP hydrolysis at two cytoplasmic NBDs (nucleotide-binding domains) to the transport of hydrophobic compounds. Orthovanadate (V(i)) and fluoroaluminate (AlF(x)) trap nucleotide in one NBD by forming stable catalytically inactive complexes (Pgp-M2+-ADP-X), which are proposed to resemble the catalytic transition state, whereas the complex formed by beryllium fluoride (BeF(x)) is proposed to resemble the ground state. We studied the trapped complexes formed via incubation of Pgp with ATP (catalytically forward) or ADP (reverse) and V(i), BeF(

Circularly polarized luminescence (CPL) materials show great potential for applications in sensing, detection, information storage, 3D displays, and beyond. For practical use, mechanical robustness and long-term stability are essential. In this study, a porous cholesteric liquid crystal polymer network (porous CLCN) is fabricated as a chiral template. The porous structure allows for the infusion of fluorescent quantum dots (QDs). By selectively spraying a QD dispersion onto the template surface, arbitrary fluorescent patterns can be encoded into the CLCN film. The combination of the chiral pol

The improved solubility of cellulose, and the enlargement of its application is of importance. Herein, we have synthesized cellulose probes with aggregation-induced emission (AIE) effect, which demonstrates high efficiency and sensitivity in detecting Hg2+ /Hg+ ions. The cellulose underwent esterification with a 11‑carbon chain using pyridine as both catalyst and reaction medium, leading to break off H bonds and a significant decrease in crystallinity. This modification allowed the cellulose to be soluble in dichloromethane (DCM) and toluene. Additionally, long-chain oil-soluble BODIPY fluores

Lipid-lowering drugs are often taken with anticoagulant drugs in hyperlipidemia patients. Fenofibrate (FNBT) and warfarin (WAR) are common clinical lipid-lowering drugs and anticoagulant drugs, respectively. A study of binding affinity, binding force, binding distance, and binding sites was performed to determine the interaction mechanism between drugs and carrier proteins (bovine serum albumin, BSA), as well as their effects on BSA conformation. Both FNBT and WAR can form complexes with BSA by van der Waals force and hydrogen bonds. WAR had a stronger fluorescence quenching effect on BSA, a s

Interactions of human serum albumin (HSA) with two structurally similar quinazoline derivatives, S1 and S2 , which are potential anticancer drugs acting on PI3K/mTOR targets, were investigated in vitro utilizing multiple spectroscopy as well as molecular docking. The fluorescence quenching study demonstrated that HSA fluorescence could be statically quenched by S1 and S2 through the formation of an HSA-drug complex. Furthermore, the details of the binding site number, binding constant, as well as the thermodynamic parameters, were estimated at 298, 303, and 310 K. The results revealed that hyd

Vertebrate oocyte polarity has been observed for two centuries and is essential for embryonic axis formation and germline specification, yet its underlying mechanisms remain unknown. In oocyte polarization, critical RNA-protein (RNP) granules delivered to the oocyte's vegetal pole are stored by the Balbiani body (Bb), a membraneless organelle conserved across species from insects to humans. However, the mechanisms of Bb formation are still unclear. Here, we elucidate mechanisms of Bb formation in zebrafish through developmental biomolecular condensation. Using super-resolution microscopy, live

The unique solvatochromic attitude of an analyte owing to its coordination with metal ions in solvents of different polarities is challenging. Herein, we introduce two new solvatochromic 4-(pentan-3-yl) benzaldehyde-based triazolyl silatrane probes (5 and 6). The solvatochromic behavior of both probes 5 and 6 was studied using Reichardt's E (30) and the Kamlet-Taft empirical scale by UV-visible spectra in 14 solvents (hydrogen-bond donor (HBD) and non-HBD), and the results show that probes 5 and 6 exhibit reverse solvatochromism. Probe 5 witnessed an enhancement in this behavior upon coordinat

Hepatocellular mitochondrial depolarization (mtDepo) after ethanol (EtOH) increases respiration to stimulate EtOH detoxification. mtDepo also triggers mitophagy, which may contribute to alcohol-associated liver disease. This study characterized sublobular respiration and distribution of mtDepo and mitophagy after acute EtOH. C57BL/6J and GFP-LC3 transgenic mice were gavaged with 6 g/kg EtOH or vehicle and administered MitoTracker Red (MTR). Hepatocytes were zonally sorted by MTR fluorescence for assessment of oxygen consumption rates (OCRs). Cytochrome P4502E1 (CYP2E1) immunolabeling identifie

Condition-dependent merging of blue, green, and red light emission to produce white light during cellular imaging from twisted intramolecular charge transfer (TICT)-based molecule has been targeted in this study. Instead of exploiting the typical white light emissive organic molecule (WLE-OM) as a WLE biomarker, our experimental and computational studies focused on donor-π-acceptor (D-π-A)-type iminocoumarin NET-N as the TICT fluorescent probe to monitor cancer cell apoptosis via bioimaging. Steady-state fluorescence studies of NET-N showed blue, yellow, and orange-red emissions in hexane, tol

Merulinic acid (heptadecenylresorcinolic acid, resorcinolic acid) is one of the members of resorcinolic lipids, the natural amphiphilic long … ipid bilayer contained glycolipids (MGDG and DGDG) and sphingomyelin. Changes of TMA-DPH and NBD-PE fluorescence polarization show that the degree of merulinic acid incorporation into liposomal membrane is not very high. The polar "heads" of the molecules of investigated compounds are localized on the level of fatty acid's ester bonds in phospholipid molecules. Merulinic a …

The photophysics of 3-benzoxazol-2-yl-chromen-2-one was studied in different solvents. High molar absorptivities, between 14,800 and 22,900 dm3/mol cm, were observed for the absorption peak related to the S0 --> S1 transition which suggests a pi --> pi* character. This compound presents a limited solvatochromism, attributed to the benzoxazole group, and high fluorescence quantum yields, phi(f). The fluorescence quantum yield is lowered with the increase of solvent polarity, favouring the participation of internal conversion as deactivation path of the S1 state. The Stokes shift shows that the

BACKGROUND: The reliance of clear cell renal cell carcinoma (ccRCC) on exogenous cholesterol import implies a metabolic susceptibility. This susceptibility represents a potential avenue that can be exploited as a novel therapeutic approach for ccRCC. Circular RNAs (circRNAs) are emerging regulators in cancer, yet their roles in ccRCC lipid metabolism and tumor microenvironment remodeling remain unclear. This study investigates the tumor-promoting role of circABCA1 in ccRCC cholesterol homeostasis and M2 macrophage polarization.
METHODS: The expression levels of circABCA1, IGF2BP3, SCARB1, aut

Dysbiosis of the oral microbiota is related to chronic inflammation and carcinogenesis. Fusobacterium nucleatum (Fn), a significant component of the oral microbiota, can perturb the immune system and form an inflammatory microenvironment for promoting the occurrence and progression of oral squamous cell carcinoma (OSCC). However, the underlying mechanisms remain elusive. Here, we investigated the impacts of Fn on OSCC cells and the crosstalk between OSCC cells and macrophages. 16 s rDNA sequencing and fluorescence in situ hybridization verified that Fn was notably enriched in clinical OSCC tis

BACKGROUND: Activated microglia play a key role in initiating the inflammatory cascade following ischemic stroke and exert proinflammatory or anti-inflammatory effects, depending on whether they are polarized toward the M1 or M2 phenotype. The present study investigated the regulatory effect of icaritin (ICT) on microglial polarization in rats after cerebral ischemia/reperfusion injury (CI/RI) and explored the possible anti-inflammatory mechanisms of ICT.
METHODS: A rat model of transient middle cerebral artery occlusion (tMCAO) was established. Following treatment with ICT, a G protein-coupl

Radiation-induced fibrosis (RIF) is a common delayed effect of acute ionizing radiation exposure (DEARE) affecting diverse tissues including the heart, lungs, liver and skin, leading to reduced tissue function and increased morbidity. Monocytes, which may be classified into classical (CD14++, CD16-), intermediate (CD14++, CD16+) and non-classical (CD14+/low, CD16++) subtypes in humans and non-human primates (NHPs), and monocyte-derived macrophages may play an integral role in the pathogenesis of RIF. We tested the hypothesis that moderate to high levels of total-body exposure to radiation woul
