Autofluorescence (AF) in formalin-fixed and paraffin-embedded tissues limit their use in immunofluorescence staining techniques. INTRODUCTION: Pemphigus comprises of a group of autoimmune bullous disorders with intraepithelial lesions involving the skin and mucous membranes. Pemphigus is characterized histologically by an intraepidermal blister and immunopathologically by the finding of in vivo bound immunoglobulin G (IgG) antibodies against desmosomal adhesion proteins on the surface of keratinocytes. Indirect immunofluorescence for IgG is considered as a gold standard method for diagnosis of this group of bullous disorders on the condition that fresh frozen tissue is accessible.

BACKGROUND: Necrotizing fasciitis is a soft tissue gangrenous infection that is optimally treated by early diagnosis, radical surgical debridement of all involved necrotic tissue, broad spectrum antibiotics, and aggressive nutritional support. This chapter describes the rationale behind and means of construction of inexpensive, low to moderate throughput protein arrays. The method of construction is based on injection of analytes into a block of frozen optimum cutting temperature (OCT), the gel media used for frozen sections, and sectioned on a cryostat. BACKGROUND: Adjunctive retroperitoneal lymphadenectomy (RPLND) plays an important role in the management of patients with metastatic nonseminomatous germ cell tumors (NSGCT). Currently, a bilateral RPLND is recommended for residual disease after chemotherapy.
METHODS: The authors systematically have removed all residual masses and used intraoperative frozen section analysis to dictate the extent of surgery in the patients with NSGCT: If frozen section revealed necrosis, then a limited RPLND was performed; otherwise, a bilateral RPLND was attempted.

Beta-adrenergic receptors were localized and quantified with autoradiographic methods in frozen sections of eyes of albino rabbits using [125I]cyanopindolol and [3H]dihydroalprenolol. Quantification in albino rabbits revealed highest beta 2-adrenoceptor densities in the ciliary body, the corneal epithelium and the corneal endothelium. Moreover, the iris showed significant amounts of specific binding, whereas the retina and sclera are nearly free of these receptors.

T-lymphocytes were identified in frozen brain sections derived from patients with chronic inflammatory disorders of the CNS by using a specific heteroantiserum and the unlabelled antibody enzyme method. Clusters of T-cells were found in post-mortem material of cases with multiple sclerosis (MS) and subacute sclerosing panencephalitis (SSPE). The results suggest that T-lymphocytes are involved in the pathogenesis of both MS and SSPE.
