Characterization of novel beta-galactosidase activity that contributes to glycoprotein degradation and virulence in Streptococcus pneumoniae.

The pneumococcus obtains its energy from the metabolism of host glycosides. Therefore, efficient degradation of host glycoproteins is integral to pneumococcal virulence. In search of novel pneumococcal glycosidases, we characterized the Streptococcus pneumoniae strain D39 protein encoded by SPD_0065 and found that this gene encodes a beta-galactosidase. The SPD_0065 recombinant protein released galactose from desialylated fetuin, which was used here as a model of glycoproteins found in vivo. A pneumococcal mutant with a mutation in SPD_0065 showed diminished beta-galactosidase activity, exhibi

Response to oxidative stress involves a novel peroxiredoxin gene in the unicellular cyanobacterium Synechocystis sp. PCC 6803.

Exposure to methyl viologen in the presence of light facilitates the production of superoxide that gives severe damage on photosynthetic apparatus as well as many cellular processes in cyanobacteria and plants. The effects of methyl viologen on global gene expression of a unicellular cyanobacterium Synechocystis sp. strain PCC 6803 were determined by DNA microarray. The ORFs sll1621, slr1738, slr0074, slr0075, and slr0589 were significantly induced by treatment of methyl viologen for 15 min commonly under conditions of normal and high light. One of these genes, slr1738, which encodes a ferric

Impaired oxidative stress resistance of Bacillus subtilis sigB mutants and the role of katA and katE.

Two catalases of B. subtilis have been studied which are subject to two different regulatory mechanisms. Whereas KatA belongs to the group of proteins specifically induced by oxidative stress, KatE is a general delta B-dependent stress protein, not induced by oxidative stress. There are two mechanisms of oxidative stress resistance, the adaptive resistance induced by low H2O2 concentrations and an unspecific resistance acquired in glucose-starved cells. Mutants lacking KatA are defective in the adaptive resistance and both exponentially growing and glucose-starved cells are 100-fold more sensi

The paired domain-containing factor Pax8 and the homeodomain-containing factor TTF-1 directly interact and synergistically activate transcription.

Pax genes encode for transcription factors essential for tissue development in many species. Pax8, the only member of the family expressed in the thyroid tissue, is involved in the morphogenesis of the gland and in the transcriptional regulation of thyroid-specific genes. TTF-1, a homeodomain-containing factor, is also expressed in the thyroid tissue and has been demonstrated to play a role in thyroid-specific gene expression. Despite the presence of Pax8 and TTF-1 also in a few other tissues, the simultaneous expression of the two transcription factors occurs only in the thyroid, supporting t

The failure of immune response evasion by linear plasmid 28-1-deficient Borrelia burgdorferi is attributable to persistent expression of an outer surface protein.

Infectivity and persistence by Borrelia burgdorferi, the etiologic agent of Lyme disease, rely stringently on regulatory events. Among these is the downregulation of lipoprotein antigen expression, exemplified by outer surface protein C (OspC), at the advent of specific immunity in the mammalian host. B. burgdorferi spirochetes that lack the linear plasmid 28-1 (lp28-1) succumb to the host's immune response. We thus explored the notion that these two phenomena were related--that lp28-1(-) organisms fail to downregulate ospC and thus are cleared following the appearance of anti-OspC antibody in

Bordetella bronchiseptica Diguanylate Cyclase BdcA Regulates Motility and Is Important for the Establishment of Respiratory Infection in Mice.

Bacteria can be motile and planktonic or, alternatively, sessile and participating in the biofilm mode of growth. The transition between these lifestyles can be regulated by a second messenger, cyclic dimeric GMP (c-di-GMP). High intracellular c-di-GMP concentration correlates with biofilm formation and motility inhibition in most bacteria, including Bordetella bronchiseptica, which causes respiratory tract infections in mammals and forms biofilms in infected mice. We previously described the diguanylate cyclase BdcA as involved in c-di-GMP synthesis and motility regulation in B. bronchiseptic

The WhiA and WhiB2 transcription factors mediate the Mycobacterium tuberculosis cell cycle by regulating DNA replication and cell division.

Tuberculosis is caused by Mycobacterium tuberculosis (Mtb), a pathogen with a remarkable ability to survive within its human host by balancing replication and persistence throughout the course of infection. The success of Mtb depends on its ability to adapt to diverse and hostile environments imposed by the host. This adaptation is primarily mediated through transcriptional regulation of gene expression. While Mtb encodes several transcription factors essential for growth, it remains unclear which transcription factors directly regulate the bacterial cell cycle. In this work, we characterized

Identification of a novel gene in ROD9 island of Salmonella Enteritidis involved in the alteration of virulence-associated genes expression.

Salmonella enterica subsp. I serovar Enteritidis (S. Enteritidis), one of the causative agents for non-typhoidal gastrointestinal diseases in humans is an intracellular bacterium and mechanism for its invasion into host cells is critical to cause infection. The virulence of the pathogen is explained by the expression of genes located on its pathogenicity islands, mostly encoded under SPI-1 and SPI-2. However, S. Typhimurium SL1344, despite sharing ∼98% of its genome with S. Enteritidis P125109, lacks few regions of differences (ROD) that are hypothesized to impart virulence potential to S. Ent

Investigating the role of Bacillus subtilis type II toxin-antitoxin system in drought stress survival.

Toxin-antitoxin (TA) systems, present in plasmids and bacterial chromosomes, are widespread in bacteria such as Bacillus subtilis and are known to be involved in growth regulation, bacterial tolerance to environmental stress conditions as well as biofilm formation. The aim of the current study was to investigate the role of TA systems in drought condition stress in B. subtilis isolates. The presence of TA systems including mazF/mazE and yobQ/yobR in B. subtilis (strain 168) was investigated using the polymerase chain reaction (PCR) method. TA system expression at 438 and 548 g/L of ethylene gl

A cumate-inducible promoter system for tuned gene expression and magnetosome overproduction in magnetotactic bacteria.

Magnetosomes are membrane-bound iron-rich organelles synthesized by magnetotactic bacteria as sensors for navigation in the Earth's magnetic field. In the well-studied model Magnetospirillum gryphiswaldense, their multistep biosynthesis is tightly controlled by > 30 specific genes clustered within a genomic magnetosome island. The lack of versatile genetic tools for strong, tunable, and orthogonal transcriptional control has impeded the analysis, engineering, and transplantation of this complex biosynthetic pathway. In addition, recent studies revealed an intricate transcriptional architecture

MtvS1 and MtvS2 Interact with RNA polymerase to regulate the Francisella type V-A CRISPR-Cas system.

Bacteria and archaea often harbor multiple CRISPR-Cas loci to defend against mobile genetic elements. Little is known, however, about whether and how different CRISPR-Cas systems are differentially regulated, in many instances due to the impossibility of studying CRISPR immunity in native hosts. Here we investigated the regulation of the endogenous type II-B and type V-A CRISPR-Cas systems present in the opportunistic human pathogen Francisella novicida U112. We found that while the type II-B system is constitutively expressed, the type V-A system is differentially expressed at stationary phas

Spatiotemporal Regulation of N2 Fixation and Photosynthesis in Trichodesmium: New Insights From mRNA CARD-FISH and nanoSIMS.

To protect the O2-sensitive nitrogenase enzyme, diazotrophic cyanobacteria separate N2 fixation from photosynthesis in space or time. However, in the filamentous Trichodesmium, technical limitations and conflicting results have so far prevented a consensus on the separation mechanism. Here, we specifically adapted single-cell methods for simultaneously capturing both photosynthesis and N2 fixation at the levels of gene expression, fixation and transport of carbon and nitrogen. Testing expression of nifH (nitrogenase) and psbA (photosystem II), we found that about half of the cells exclusively

Alternative ribosomal protein RpmE2 is produced under zinc limitation in Neisseria gonorrhoeae and slows translation and bacterial growth.

UNLABELLED: To enhance its pathogenic potential, Neisseria gonorrhoeae (Gc) pirates zinc from human metal sequestration proteins using TonB-dependent outer membrane transport systems. However, cytoplasmic mechanisms by which Gc adapts to zinc limitation are still uncharacterized. rpmE2 and rpmJ2 transcripts, encoding alternative L31 and L36 ribosomal proteins, respectively, which are not predicted to bind zinc, are significantly more abundant in zinc-limited Gc. In other bacterial species, alternative ribosomal proteins replace canonical zinc-binding ribosomal proteins when zinc availability i

Biochemistry and transcriptional regulation of bacterial sulfur oxidation.

Sulfur is one of the most important and versatile elements in biology and forms the basis of energy metabolism in many prokaryotes. Its extensive large-scale redox transformations drive the global biogeochemical sulfur cycle and profoundly influence environmental chemistry. In dissimilatory sulfur oxidizers, the oxidation of reduced sulfur compounds is directly coupled to energy conservation via photosynthetic or respiratory electron transport chains. Sulfur-oxidizing bacteria and archaea are remarkably diverse, reflecting both the wide range of habitats they inhabit and the variety of metabol

CliPME: the clinical pathogenic bacteria mutation and expression database.

The ongoing battle between humans and pathogenic bacteria has fueled rapid microbial evolution. Although whole-genome sequencing (WGS) has transformed the ability to track genomic mutations, existing tools lack comprehensive solutions for analyzing mutational patterns and their functional consequences in pathogenic bacteria. Here, we present CliPME, an innovative platform that bridges this critical gap by combining mutation detection, mutation effect prediction, and regulatory network analysis, specifically designed for bacterial genomics. We develop qMut, a high-performance R package designed

MneA and TerC exhibit distinct roles in manganese detoxification and virulence in Vibrio parahaemolyticus.

Vibrio parahaemolyticus is a significant zoonotic pathogen that severely threatens global public health and aquaculture. Manganese (Mn) acts as a cofactor for numerous enzymes and is crucial for bacterial oxidative stress resistance and virulence. However, excessive Mn is toxic, prompting bacteria to develop diverse Mn detoxification mechanisms. Herein, two genes (mneA and terC) located downstream of the Mn-sensing yybP-ykoY riboswitch were identified in V. parahaemolyticus. The expression of mneA and terC is specifically induced by Mn. The mneA deletion mutant (ΔmneA) exhibited impaired growt

Sucrose improve Lactiplantibacillus plantarum LIP-1's tolerance to heat by increasing biofilm production.

Optimizing the carbon source to increase biofilm production and thus boost the heat tolerance of strains is a promising strategy. However, related research is scarce. This study investigated the effects of varying glucose and sucrose amounts added to MRS medium on biofilm production and heat tolerance by Lactiplantibacillus plantarum LIP-1. Transcriptomic, proteomic, and metabolomic approaches were combined to analyze the intrinsic mechanism underlying the sucrose-induced increase in biofilm production. We then investigated the protective role of the biofilm for the strain. Compared with the c

DNA adenine methylation influences gene expression and biofilm formation in Streptococcus mutans.

UNLABELLED: Streptococcus mutans, a key oral pathogen, utilizes quorum sensing to regulate biofilm formation-a major virulence factor in the development of dental caries (tooth decay). Our recent research uncovered a complex interplay between the CSP-ComDE quorum sensing pathway and the Type II DpnII restriction-modification (R-M) system in S. mutans. The DpnII R-M system methylates adenine at 5'-GATC-3' sites and cleaves unmethylated DNA, significantly influencing foreign DNA acquisition and gene expression. In this study, we investigated the impact of a ΔRM mutant, which lacks adenine methyl

Loss of LasR function leads to decreased repression of Pseudomonas aeruginosa PhoB activity at physiological phosphate concentrations.

The Pseudomonas aeruginosa LasR transcription factor plays a role in quorum sensing (QS) across phylogenetically distinct lineages. However, isolates with loss-of-function mutations in lasR (LasR- strains) are commonly found in diverse settings, including infections where they are associated with worse clinical outcomes. In LasR- strains, the LasR-regulated transcription factor RhlR can also be stimulated by the activity of the two-component system PhoR-PhoB in low-inorganic phosphate (Pi) conditions. Here, we demonstrate a novel link between LasR and PhoB in which the absence of LasR increase
