Expression of His-tagged Shigella IpaC in Arabidopsis.

Although the expression of histidine (His)-tagged proteins in bacteria is routine, few His-tagged proteins have been expressed in plants, and no His-tagged proteins from bacterial pathogens have been expressed in plants, to our knowledge. Here, we demonstrate expression of the Shigella flexneri invasion plasmid antigen, IpaC, in Arabidopsis thaliana. S. flexneri is the causitive trigger for bacillary dysentery, and IpaC is essential for bacterial entry into epithelial cells. IpaC, attached to a 5' leader containing six tandem His codons, was cloned into a pBI121 vector. This clone was introduc

Knockout of the S-acyltransferase Gene, PbPAT14, Confers the Dwarf Yellowing Phenotype in First Generation Pear by ABA Accumulation.

The development of dwarf fruit trees with smaller and compact characteristics leads to significantly increased fruit production, which is a major objective of pear (Pyrus bretschneideri) breeding. We identified the S-acylation activity of PbPAT14, an S-acyltransferase gene related to plant development, using a yeast (Saccharomyces cerevisiae) complementation assay, and also PbPAT14 could rescue the growth defect of the Arabidopsis mutant atpat14. We further studied the function of PbPAT14 by designing three guide RNAs for PbPAT14 to use in the CRISPR/Cas9 system. We obtained 22 positive transg

SlTWD1 Affects Leaf Growth Primarily via Cell Expansion and Contributes to Leaf and Fruit Size Control in Tomato.

Leaf growth is a major determinant of plant architecture and productivity. TWISTED DWARF1 (TWD1/FKBP42) is a conserved immunophilin involved in plant growth and development, but its function in tomato remains unclear. Here, we identified and characterized SlTWD1, the tomato homolog of TWD1. SlTWD1 is highly conserved among land plants and functionally complemented the developmental defects of an Arabidopsis thaliana cr-twd1 mutant. SlTWD1 was broadly expressed in vegetative and reproductive tissues. CRISPR/Cas9-mediated disruption of SlTWD1 caused severe dwarfism and reduced leaf and fruit siz

The m6A-YTH regulatory system in plants: A status.

Plants use mRNA methylation to regulate gene expression. As in other eukaryotes, the only abundant methylated nucleotide in plant mRNA bodies is N6-methyladenosine (m6A). The conserved core components of m6A-based genetic control are a multi-subunit nuclear methyltransferase, and a set of nuclear and cytoplasmic RNA-binding proteins consisting of an m6A recognition module, the YT521-B homology (YTH) domain, and long intrinsically disordered regions (IDRs). In plants, this system is essential for growth during embryonic and post-embryonic development, but emerging evidence also points to key fu

Interaction between pathogenesis-related (PR) proteins and phytohormone signaling pathways in conferring disease tolerance in plants.

Pathogenesis-related (PR) proteins are critical defense signaling molecules induced by phytopathogens. They play a vital role in plant's defense signaling pathways and innate immunity, particularly in systemic acquired resistance (SAR) and serve as key molecular markers of plant defense. Overexpressing PR genes, such as chitinase, thaumatin, glucanase, thionin and defensin, either individually or in combination, have significantly boosted plants' defense responses against various pathogens. However, signaling pathways regulating the expression of these versatile proteins remain only partially

Gene networks underlying the early regulation of Paraburkholderia phytofirmans PsJN induced systemic resistance in Arabidopsis.

Plant defense responses to biotic stresses are complex biological processes, all governed by sophisticated molecular regulations. Induced systemic resistance (ISR) is one of these defense mechanisms where beneficial bacteria or fungi prime plants to resist pathogens or pest attacks. In ISR, the defense arsenal in plants remains dormant and it is only triggered by an infection, allowing a better allocation of plant resources. Our group recently described that the well-known beneficial bacterium Paraburkholderia phytofirmans PsJN is able to induce Arabidopsis thaliana resistance to Pseudomonas s

Genome-wide identification and expression pattern analysis of the COBRA-like gene family of Triticum aestivum (L.) under drought and heat stresses.

BACKGROUND: T. aestivum (Triticum aestivum) (L.), rich in protein, carbohydrates, and minerals, is one of the world’s most important cereal grain crops, serving as the staple food source for 30% of the human population, underscoring its critical role in global food security. However, adverse environmental conditions such as high temperature and drought can severely restrict its growth, making the improvement of abiotic stress tolerance in T. aestivum a key research priority. The COBRA-like (COBL) gene family is key to regulating plant cell wall formation and abiotic stress responses. So far, a

Ectopic Expression of AeNAC83, a NAC Transcription Factor from Abelmoschus esculentus, Inhibits Growth and Confers Tolerance to Salt Stress in Arabidopsis.

NAC transcription factors play crucial roles in plant growth, development and stress responses. Previously, we preliminarily identified that the transcription factor AeNAC83 gene was significantly up-regulated under salt stress in okra (Abelmoschus esculentus). Herein, we cloned the nuclear-localized AeNAC83 from okra and identified its possible role in salt stress response and plant growth. The down-regulation of AeNAC83 caused by virus-induced gene silencing enhanced plant sensitivity to salt stress and increased the biomass accumulation of okra seedlings. Meanwhile, AeNAC83-overexpression A

SPIRAL: A versatile online single time-point circadian analysis platform for rice.

The circadian clock synchronises plant physiology with environmental oscillations to promote plant fitness. The commonly-used methods for rhythm monitoring in dicots include rhythmic leaf movement tracking and luciferase-based imaging. For monocots, however, the leaf erectness makes these methods ineffective. Leveraging over 11,000 transcriptome samples, the circadian time-course profiling, the simulation-based algorithm optimisation, and the experimental validation, we developed SPIRAL, an online single time-point circadian analysis platform for rice and unexpectedly revealed a ∼28-hour endog

Comparative Genomics of the Tea XTH Gene Family Reveals the Involvement of CsXTH27 in Internode Elongation.

Xyloglucan endotransglucosylase/hydrolases (XTHs) are key cell-wall-remodeling enzymes that mediate xyloglucan modification, cell expansion, and organ elongation, yet the composition and variation in the XTH gene family across tea accessions remain unclear. In this study, we systematically identified and comparatively analyzed the XTH gene family across the genomes of nine tea accessions, yielding 477 putative XTH genes. Phylogenetic, conserved-motif, protein-domain, and conserved catalytic-motif analyses indicated that the major structural features of tea XTH proteins are broadly conserved, w

Dynamics of floral volatile emission and transcriptional regulation during flowering in Malus 'Prairie Rose'.

Malus 'Praire Rose' (MPR) stands out for its unique rose-like appearance and floral scent (FS) with potential applications in the fragrance industry. Despite limited exploration at the molecular level, this study investigated the FSs production mechanism in MPR across four flower developmental stages using metabolomics and transcriptomics. Results revealed that the predominant FSs in MPR flowers were terpenoids (à-Ionone, trans-á-Ionone, geranylacetone, and alpha-farnesene, etc.) and phenolic compounds (phenylethyl alcohol, 2-(4-Methoxyphenyl)ethanol, etc.). The highest concentration and diver

Integrated transcriptomic and proteomic analyses provide insights into potential molecular mechanisms underlying globulin content variation in maize embryos.

BACKGROUND: Globulin in maize kernels is a high-quality protein, which is mainly enriched in the embryo. However, academic research focusing on the molecular mechanism underlying globulin synthesis remains relatively limited to date. Therefore, unveiling the molecular mechanism of globulin synthesis in maize embryos is of great significance for the breeding of high-protein maize varieties. To gain better insights into this mechanism, we integrated transcriptomic and proteomic analysis to identify differentially expressed genes (DEGs) and differentially expressed proteins (DEPs) between two typ

Dynamic transcriptome profiling revealed a key gene ZmJMJ20 and pathways associated with cadmium stress in maize.

Cadmium (Cd) pollution in soil poses a global concern due to its serious impacts on human health and ecological security. In plants, tremendous efforts have been made to identify some key genes and pathways in Cd stress responses. However, studies on the roles of epigenetic factors in response to Cd stress were still limited. In the study, we first gain insight into the gene expression dynamics for maize seedlings under 0 h, 12 h, and 72 h Cd stress. As a result, six distinct groups of genes were identified by hierarchical clustering and principal component analysis. The key pathways associate
