Identification of potential predictive biomarkers during JAK-inhibitor therapies in rheumatoid arthritis.

BACKGROUND: Targeted synthetic therapeutics, such as Janus kinase (JAK) inhibitors, have opened up a new platform for the treatment of various diseases, including rheumatoid arthritis (RA). As with all drugs, the efficacy of different medicine may vary from patient to patient, and there may be different side effects if the expected effect is not achieved. In addition to the uncertain success of treatments, they also place a heavy burden on the health care system, making the identification of potential predictive biomarkers for drug optimisation highly valuable. METHODS: In order to identify po

Explicating miRNA-mediated regulation of inflammatory pathways in COPD, MS, and lung cancer using explainable artificial intelligence: insights from peripheral blood profiles.

BACKGROUND: Chronic obstructive pulmonary disease (COPD), multiple sclerosis (MS), and lung cancer are linked by shared inflammatory pathways and immune dysregulation. miRNAs regulate these processes by influencing gene expression, yet their roles in the molecular mechanisms across neurological and respiratory systems are not fully understood. OBJECTIVE: This study aims to identify miRNAs and their target genes regulating inflammatory pathways, advancing the understanding of molecular genetics underlying COPD, MS, and lung cancer. METHODS: miRNA expression data (GSE61741) were analyzed using

EHP-101, an oral formulation of the cannabidiol aminoquinone VCE-004.8, alleviates bleomycin-induced skin and lung fibrosis.

Systemic sclerosis (SSc) or scleroderma is a chronic multi-organ autoimmune disease characterized by vascular, immunological, and fibrotic abnormalities. The etiology of SSc is unknown, but there is growing evidence that dysfunction of the endocannabinoid system (ECS) plays a critical role in its development. Since the semi-synthetic cannabinoquinoid VCE-004.8 could alleviate bleomycin (BLM)-induced skin fibrosis, we have investigated an oral lipid formulation (EHP-101) of this dual PPARγ/CB2 receptors activator for the prevention of skin- and lung fibrosis and of collagen accumulation in BLM

The correlation of neutrophil-to-lymphocyte ratio with the presence and activity of myasthenia gravis.

BACKGROUND: Glycyrrhiza, a perennial herbaceous medicinal plant, is extensively utilized in the pharmaceutical industry. The growth of Glycyrrhiza is frequently constrained by soil phosphorus availability, as a significant portion of arable land in China suffers from phosphorus deficiency. METHOD: This study utilized Ural Glycyrrhiza uralensis Fisch as the subject and examined the application of GR24, a synthetic strigolactone, under three phosphorus conditions: none (P1), low (P2), and high (P3). The research aimed to ascertain the optimal concentration of GR24 for promoting licorice growth

Glucocorticoid Receptor Translational Isoforms Generate Unique Glucocorticoid Responses in the Mouse Brain.

Glucocorticoids are primary stress hormones necessary for life that act on nearly every tissue in the body to maintain homeostasis. These hormones and their synthetic derivatives are widely used in the clinic to combat disease but are limited by serious adverse effects. The actions of glucocorticoids are mediated by the glucocorticoid receptor (GR). In addition to the classic full-length receptor (GR-A), seven highly conserved receptor isoforms with progressively shorter N-terminal transactivation domains (NTD) (GR-B, GR-C1, GR-C2, GR-C3, GR-D1, GR-D2, GR-D3) are produced from the single GR ge

[BRCA1 and BRCA2 - pathologists starting kit].

Dysfunction of tumor suppressor genes BRCA1 and BRCA2 is involved in the pathogenesis of malignant tumors, especially breast and ovarian carcinoma. BRCA1/2 genes may be inactivated by germinal and somatic mutations or epigenetic changes. Germinal mutations are responsible for the hereditary breast and ovarian carcinoma syndrome. Defects of BRCA1/2 genes lead to the failure of homologous recombination, the basic mechanism for DNA double strand break repair. The resultant genomic instability is associated with a high risk of malignant transformation of the cell, but it also results in a higher s

[Alternative splicing and the cure of spinal muscular atrophy].

Alternative splicing of the messenger RNA plays a fundamental role in the flow of genetic information from DNA to proteins by expanding the coding capacity of the genome. The regulation of alternative splicing is as important as the regulation of transcription to determine the specific characteristics of cells and tissues, the normal functioning of cells and the responses of eukaryotic cells to external signals. Basic knowledge of the pre-mRNA sequences and splicing factors that recognize them has allowed scientists to design a therapeutic synthetic oligonucleotide for spinal muscular atrophy.

Salirasib in the treatment of pancreatic cancer.

The Ras family of genes is involved in the cellular regulation of proliferation, differentiation, cell adhesion and apoptosis. The K-ras gene is mutated in over 90% of pancreatic cancer cases. Salirasib (S-trans,trans-farnesylthiosalycilic acid [FTS]) is a synthetic small molecule that acts as a potent Ras inhibitor. It is a farnesylcysteine mimetic that selectively disrupts the association of active RAS proteins with the plasma membrane. Animal studies demonstrated that salirasib inhibited tumor growth, downregulated gene expression in the cell cycle and Ras signaling pathways. In a clinical

High-level expression of the Japanese encephalitis virus E protein by recombinant vaccinia virus and enhancement of its extracellular release by the NS3 gene product.

Recombinant vaccinia viruses expressing the prM and E genes of the Japanese encephalitis virus (JEV) were constructed by use of synthetic promoters. While the recombinant virus mOJ6-SL, with an optimized vaccinia late-gene promoter, produced a 20-fold elevated level of E protein, as well as an 86-kDa precursor protein in infected cells, no significant quantitative difference was detected between the extracellular or cell-surface E protein produced by mOJ6-SL and those produced by mOJ6 with the 7.5-kDa promoter. However, when the cells were infected with Dengue 2 virus before infection with mOJ

Antisense Oligonucleotide Therapy for Ophthalmic Conditions.

Antisense oligonucleotides (AON) are synthetic single-stranded fragments of nucleic acids that bind to a specific complementary messenger RNA (mRNA) sequence and change the final gene product. AON were initially approved for treating cytomegalovirus retinitis and have shown promise in treating Mendelian systemic disease. AON are currently being investigated as a treatment modality for many ophthalmic diseases, including inherited retinal disorders (IRD), inflammatory response and wound healing after glaucoma surgery, and macular degeneration. They provide a possible solution to gene therapy fo

MicroRNA expression profiling of dibenzalacetone (DBA) treated intracellular amastigotes of Leishmania donovani.

BACKGROUND: Among different leishmanial infections, visceral leishmaniasis (VL) if not treated is the most severe form with high mortality rates. In India, it is caused by the protozoan parasite Leishmania donovani. The therapy of visceral leishmaniasis is limited due to high toxicity, resistance to existing drugs and increasing cases of Leishmania co-infections. Hence, there is a need to identify novel drug and targets to overcome these hindrances. MicroRNAs (miRNAs) are a class of small non-coding RNAs (∼22-24 nucleotide in length) that regulate gene expression in various biological processe

Precision genetic manipulation: CRISPR toolkits reshape plant biology.

The emergence of clustered regularly interspaced short palindromic repeats (CRISPR) technology has revolutionized plant genome engineering over the past decade. This review begins by highlighting four seminal early CRISPR studies published in Plant Physiology that laid critical foundations for crop genome editing and tool development. We then summarize major advances in optimizing CRISPR performance, expanding targeting scope, and developing strategies for transgene removal and transient editing. We also discuss a wide array of innovative applications beyond gene knockout, such as gene targeti

FXR mediates a chromatin looping in the GR promoter thus promoting the resolution of colitis in rodents.

Glucocorticoids (GCs) are important endocrine regulators of a wide range of physiological processes ranging from immune function to glucose and lipid metabolism. For decades, synthetic glucocorticoids such as dexamethasone have been the cornerstone for the clinical treatment of inflammatory bowel diseases (IBD). A previous study has shown that farnesoid X receptor (FXR) enhances the transcription of NR3C1 gene, which encodes for human GR, by binding to a conserved FXR response element (FXRE) in the distal promoter of this gene. In the present study we demonstrate that FXR promotes the resoluti

[Expression of the gene for tick-borne viral encephalitis virus NS3 protein in Escherichia coli cells].

On the base of two overlapping cDNA-clones of tick-borne encephalitis virus (TBEV) genome and synthetic DNA fragments full DNA-copy of the TBEV NS3 protein gene was constructed and expressed in the E. coli cells. It was demonstrated that the relatively low biosynthesis level of full-length NS3 protein in the bacteria was due to the toxicity of the N-terminal region of the protein, consisting of it's first 180 amino acid residues. A form of the gene with deletion of nucleotides coding for the toxic region (called NS3*) was constructed and effective bacterial product of NS3* protein was obtained

Nitrogen removal and microbial community responses in anammox-enhanced biofilters across diverse urban waters.

Biofilters offer decentralized nitrogen treatment but often show variable performance across influent types. This study evaluated anammox-enhanced biofilters (A-BF, inoculated with enriched anammox biomass) and control biofilters (C-BF) treating wastewater (WW), stormwater (SW), and greywater (GW) under synthetic and real conditions for 54 weeks. Under synthetic conditions, A-BF outperformed C-BF, with the greatest improvement in ammonium-rich WW, where TN removal reached 65.7±5.1% compared with 17.2±2.4% in C-BF. This was supported by enrichment of Candidatus Brocadia (1.19%) and high anammox

Localization of dopamine D2 receptor mRNA in glomus cells of the rabbit carotid body by in situ hybridization.

The localization of mRNA coding for the dopamine D2 receptor was studied in the rabbit carotid body using in situ hybridization with synthetic 35S-labelled oligodeoxynucleotides. Using autoradiography on cryostat or semi-thin sections, labelling was observed over the cytoplasm of glomus cells, but not over sustentacular cells. A quantitative study showed that labelling intensity (silver grain density) was increased by haloperidol treatment. These results suggest that glomus cells express the dopamine D2 receptor gene and that this expression is regulated.

Selective Cre-mediated gene deletion identifies connexin 43 as the main connexin channel supporting olfactory ensheathing cell networks.

Many functions of glial cells depend on the formation of selective glial networks mediated by gap junctions formed by members of the connexin family. Olfactory ensheathing cells (OECs) are specialized glia associated with olfactory sensory neuron axons. Like other glia, they form selective networks, however, the connexins that support OEC connectivity in vivo have not been identified. We used an in vivo mouse model to selectively delete candidate connexin genes with temporal control from OECs and address the physiological consequences. Using this model, we effectively abolished the expression

RIG-I Stimulation Enhances the Effector Function and Proliferation of Primary Human CD8+ T Cells.

Cytotoxic CD8 T lymphocytes are crucial in antiviral immune responses. However, their recruitment to infection sites renders them at risk of viral infection, which could affect their effector activity. CD8 T lymphocytes express RIG-I, which detects cytosolic viral RNA and subsequently induces antiviral gene expression. We investigated how Influenza A virus infection and synthetic triphosphorylated double-stranded RNA, a specific RIG-I ligand, influence TCR-dependent effector responses in primary human CD8 T cells. Cells were isolated from healthy donors and either infected with the reassortant

The effect of Hydrocortisone on implantation through upregulation of tight junction genes: A lab trial study.

OBJECTIVE: Emotional stress leading to cortisol release is an important infertility factor in females. Increased cortisol levels can significantly affect tight junction proteins and dysregulate the implantation process. This study investigated the effect of exogenous synthetic Cortisol (Hydrocortisone) on the expression of specific genes encoding several junctional proteins in the endometrial epithelial cells. METHODS: Following endometrium sampling from 25 participants, the biopsied tissue was digested and filtered through a cell strainer to prepare endometrial epithelial cells. After conflu
