HIV-related posts from a Chinese internet discussion forum: An exploratory study.

BACKGROUND: In China, the introduction of antiretroviral treatment has increased the number of people living with HIV/AIDS (PLWHA). New technologies, such as social media, might be useful for enhancing HIV surveillance, especially given the lack of Chinese research, which is related to stigma and discrimination. Thus, the relative anonymity of social media may make it useful for evaluating "hard to reach" PLWHA. SETTING: This study used social media data to assess whether it reflected the prevalence of HIV and to explore PLWHA' needs and online habits. METHODS: In 2017, the Baidu Tieba platf

[X-ray pulmonary manifestations in patients infected with the human immunodeficiency virus].

Altogether 155 patients with a newly detected positive reaction to HIV (a human immunodeficiency virus) were investigated in the Republic of Burundi. Chest x-ray was done in 80 of them. Pulmonary tuberculosis was diagnosed in 2 of them, pneumonia (chronic, interstitial and bronchial)--in 15. Enhancement and deformity of lung marking were detected in 45 patients (coincidence with clinical signs of acute bronchitis was found but in 5 of them). A conclusion has been made of interstitial pneumonias being typical of HIV-infected patients and of frequent enhancement of lung marking in the preclinica

Promoting male involvement to improve PMTCT uptake and reduce antenatal HIV infection: a cluster randomized controlled trial protocol.

BACKGROUND: Despite the availability of a dual therapy treatment protocol and infant feeding guidelines designed to prevent mother to child transmission (PMTCT) of HIV, of the over 1 million babies born in South Africa each year, only 70% of those born to HIV positive mothers receive dual therapy. Similar to other resource-poor nations facing the integration of PMTCT into routine pregnancy and infant care, efforts in South Africa to scale up PMTCT and reduce transmission to < 5% have fallen far short of the United Nation's goal of 50% reductions in paediatric HIV by 80% coverage of mothers. Mechanisms of HIV Neuropathogenesis: Role of Cellular Communication Systems.

BACKGROUND: One of the major complications of Human Immunodeficiency Virus (HIV) infection is the development of HIV-Associated Neurocognitive Disorders (HANDs) in approximately 50-60% of HIV infected individuals. Despite undetectable viral loads in the periphery owing to anti-retroviral therapy, neuroinflammation and neurocognitive impairment are still prevalent in HIV infected individuals. Several studies indicate that the central nervous system (CNS) abnormalities observed in HIV infected individuals are not a direct effect of viral replication in the CNS, rather these neurological abnormal

Activities of transmitted/founder and chronic clade B HIV-1 Vpu and a C-terminal polymorphism specifically affecting virion release.

UNLABELLED: Acute HIV-1 infection is characterized by a type I interferon response, resulting in the induction of host restriction factors. HIV-1 has evolved to counteract these factors, and one such adaptation, the ability of Vpu to counteract BST2/tetherin, is associated with the evolution of simian immunodeficiency virus (SIVcpz) into pandemic group M human immunodeficiency virus type 1 (HIV-1). During transmission between individuals, very few viruses or even a single virus, the "transmitted/founder" (T/F) virus, gives rise to the new infection, but in the new host the selective pressure o

A Peer-Mother Counseling Intervention Improves Early Infant HIV Testing in Rural Uganda.

BACKGROUND: Peer-led counseling interventions could improve early infant diagnosis of HIV by empowering mothers with knowledge and information on their role in preventing perinatal transmission. We hypothesized that a peer-led intervention would increase completion rates of infant HIV testing in rural Uganda. METHODS: From September 2019 to October 2021, we conducted the Enhanced viral load counseling with Standardized Peer-Support (ENHANCED-SPS) trial, which randomized 14 public health facilities to the intervention: peer-led counseling on HIV viral load and perinatal transmission, support f

LMP-420, a small-molecule inhibitor of TNF-alpha, reduces replication of HIV-1 and Mycobacterium tuberculosis in human cells.

BACKGROUND: Co-infections of human immunodeficiency virus (HIV) and Mycobacterium tuberculosis (M. Tb) are steadily increasing and represent a major health crisis in many developing countries. Both pathogens individually stimulate tumor necrosis factor-alpha (TNF) release from infected cells and TNF, in turn, enhances the replication of each. A recent report on a Phase I clinical trial suggested that etanercept (soluble TNF receptor) might be beneficial in treating HIV/M. Tb co-infected patients. We sought to determine if a small molecule inhibitor of TNF synthesis and activity could block rep

The Intergenerational Impact of a Slow Pandemic: HIV and Children.

Human immunodeficiency virus (HIV) has, over the last four decades, infected millions of young women and their children. Interventions developed in parallel with the spread of the virus have been able to reduce rates of vertical transmission from mother to child. The impact of HIV in children can be direct in children living with HIV (CLHIV) and exposed to HIV and uninfected, or indirect through impacts on their parents, caregivers, and family. In 2018, the United Nations joint programme on AIDS (UNAIDS) estimated that 1.7 million children were living with HIV, 160,000 were newly infected with

HIV risk inside U.S. prisons: a systematic review of risk reduction interventions conducted in U.S. prisons.

HIV prevalence in correctional populations is approximately five times that of the general adult population. This systematic review examines the broad question of HIV prevention and interventions to reduce inmate HIV-related risk behaviors in U.S. federal and state prisons. We conducted a systematic review of multiple databases and Google Scholar to identify behavioral, biomedical, social, and policy studies related to HIV among U.S. prison populations from 1980-2014. Studies were excluded if they did not focus on HIV, prison inmates, if they were conducted outside of the U.S., if they involve

Using program evaluation to improve the performance of a TB-HIV project in Banteay Meanchey, Cambodia.

SETTING: Cambodia has the highest human immunodeficiency virus (HIV) prevalence (1.9%) and tuberculosis (TB) incidence (508/100000) in Asia. Banteay Meanchey, a province with high HIV prevalence of 1.9%, established a pilot project in 2003 to enhance TB-HIV activities. We evaluated this project to improve performance. METHODS: In March 2005, we analyzed 17 months of data on all persons diagnosed with HIV or TB at 11 participating clinics. We determined barriers to HIV testing and TB screening, modified the program to reduce these barriers and assessed whether our interventions improved testin

Extracellular Vesicle-Liposome-Darunavir Formulation for the Treatment of HIV Neuropathogenesis.

This study evaluates the efficacy of an extracellular vesicles-liposome-darunavir (EV-Lip-DRV) formulation for the treatment of HIV neuropathogenesis, including neurocognitive disorders. The EV-Lip-DRV formulation was developed through a process involving thin-film hydration and extrusion, followed by ultrafiltration to remove unloaded DRV. The encapsulation efficiency was found to be 41.75 ± 2.19%, with a particle size of ∼189 nm and zeta potential of ∼-7.8 mV. The hemocompatibility test confirmed the safety of the formulation for red blood cells, while drug release profiles demonstrated a su

Broad synergistic antiviral efficacy between a novel elite controller-derived dipeptide and antiretrovirals against drug-resistant HIV-1.

INTRODUCTION: The naturally occurring dipeptide Tryptophylglycine (WG) is enhanced in human immunodeficiency virus (HIV-1) infected Elite Controllers (EC). We have shown that this dipeptide has an anti-HIV-1 effect and evaluated now its synergistic antiretroviral activity, in combination with current antiretrovirals against multi-drug resistant HIV-1 isolates. METHODS: Drug selectivity assay with WG-am and ARVs agains HIV-1 resistant isolates were carried out. Subsequently, two methods, Chou-Talalay's Combination Index (CI) and ZIP synergy score (SS), were used to quantify the synergism. RES

Loss of Nef-mediated CD3 down-regulation in the HIV-1 lineage increases viral infectivity and spread.

Nef is an accessory protein of primate lentiviruses that is essential for efficient replication and pathogenesis of HIV-1. A conserved feature of Nef proteins from different lentiviral lineages is the ability to modulate host protein trafficking and down-regulate a number of cell surface receptors to enhance replication and promote immune evasion. Notably, the inability of Nef to down-regulate CD3 from infected T cells distinguishes HIV-1 Nef and its direct simian precursors from other primate lentiviruses. Why HIV-1 does not employ this potential immune evasion strategy is not fully understoo

The impact and mechanism of HIV infection on tuberculous granuloma formation.

The co-infection of Mycobacterium tuberculosis (MTB) and HIV continues to pose a major challenge to healthcare systems. Currently, the effects of HIV infection on tuberculous granulomas are not fully understood. This review discusses the impact of HIV infection on the formation and function of tuberculous granulomas, highlighting key immunological mechanisms and the interactions between HIV and MTB infections. The co-infection results in atypical granulomas with weakened immune defenses, which facilitate the dissemination of MTB and accelerate the progression of tuberculosis. Additionally, thi

Differential risk factors for HIV drug and sex risk-taking among non-treatment-seeking hospitalized injection drug users.

Injection drug users (IDUs) are at increased risk of contracting HIV. From a clinical trial assessing an intervention to enhance the linkage of hospitalized patients to opioid treatment after discharge, we conducted multivariate analysis of baseline data from hospitalized IDUs with a history of opioid dependence (n = 104) to identify differences in factors predicting HIV drug and sex risk behaviors. Factors significantly associated with HIV drug risk were being non-Hispanic Caucasian and recent cocaine use. Being female, binge drinking, and poorer mental health were significantly associated wi

Short communication: activating transcription factor 4 (ATF4) promotes HIV type 1 activation.

Activating transcription factor 4 (ATF4) is a central factor in the cellular response to multiple stresses, including altered metabolic conditions, anoxia and hypoxia, and redox stress. ATF4 is triggered by endoplasmic reticulum stress and consequent unfolded protein response. This report identifies for the first time ATF4 as a transcription factor upregulated by HIV-1 infection. Upregulation of ATF4 enhances HIV replication, by synergistic interactions with HIV Tat. Moreover, in specific cell lines ATF4 has a direct transactivating potential on the LTR, even in the absence of Tat. We also pro

Toll-Like Receptor (TLR) Signaling Enables Cyclic GMP-AMP Synthase (cGAS) Sensing of HIV-1 Infection in Macrophages.

HIV-1 replicates in cells that express a wide array of innate immune sensors and may do so simultaneously with other pathogens. How a coexisting innate immune stimulus influences the outcome of HIV-1 sensing, however, remains poorly understood. Here, we demonstrate that the activation of a second signaling pathway enables a cyclic GMP-AMP synthase (cGAS)-dependent type I interferon (IFN-I) response to HIV-1 infection. We used RNA sequencing to determine that HIV-1 alone induced few or no signs of an IFN-I response in THP-1 cells. In contrast, when supplemented with suboptimal levels of bacteri

The interaction between HIV-1 central polypurine tract and host SAMHD1 dNTPase during HIV-1 vector transduction in human primary nondividing monocyte-derived macrophages.

Sterile alpha motif and histidine-aspartate domain-containing protein 1 (SAMHD1) is a dNTPase that depletes intracellular dNTP pools in nondividing cells such as human monocyte-derived macrophages (MDMs). These low dNTP levels suppress HIV-1 reverse transcription kinetics in MDMs. In contrast, viral protein X (Vpx), an accessory protein encoded by human immunodeficiency virus type 2 (HIV-2) and some simian immunodeficiency viruses (SIVs), induces degradation of SAMHD1, thereby increasing cellular dNTP pools and promoting a more permissive infection in MDMs. Our previous study demonstrated that

Leading with HOPE: A Clinical Communication Campaign to Promote Partner HIV Testing in Assisted Partner Notification and Antenatal Care Programs in Uganda.

Among heterosexual couples in sub-Saharan Africa, fears of relationship dissolution and inability to bear healthy children following an HIV-positive result remain barriers to HIV testing. Recently expanded availability of pre-exposure prophylaxis (PrEP) affords a critical opportunity to promote HIV testing with reassuring communication that highlights use for relationship preservation and healthy childbearing. We aimed to develop a theoretically-grounded communication intervention to promote partner HIV testing within assisted partner notification (APN) and antenatal care (ANC) programs in Uga
