PCPDTBT Conjugated Polymer Nanoparticles for Photothermal Inactivation of Multidrug-Resistant mcr‑1-Positive Escherichia coli.

Antimicrobial resistance (AMR) poses a global health challenge, threatening the effectiveness of the current treatment of bacterial infections. The emergence of plasmid-mediated resistance, notably the mcr-1 gene in Escherichia coli (E. coli), further complicates therapeutic options by conferring resistance to colistin, a last-resort antibiotic. This study explores the potential of conjugated polymer nanoparticles made of poly-[2,6-(4,4-bis-(2-ethylhexyl)-4H-cyclopenta-[2,1-b;3,4-b']-dithiophene)-alt-4,7-(2,1,3-benzothiadiazole)] (PCPDTBT NPs) for antimicrobial photothermal therapy (PPT) again

Emerging threat to antibiotic resistance: Impact of mycotoxin deoxynivalenol on gut microbiota and clonal expansion of extensively drug-resistant enterococci.

Mycotoxins, antibiotic-resistant bacteria (ARB), and antibiotic resistance genes (ARGs) are significant environmental pollutants that pose considerable threats to environmental health and human safety through the food chain. This study is the first to investigate the impact of deoxynivalenol (DON), the most common mycotoxin, on antibiotic resistance dynamics in gut microbiota, demonstrating that DON exposure significantly selects for ARB and ARGs. Results indicated that 80.69 % of the ARGs with the highest increase in the DON group were exclusive to gram-positive bacteria, particularly those r

Global distribution of antimicrobial resistance genes in Escherichia coli isolated from wild animals using genomes available in public databases.

Antimicrobial resistance (AMR) is a pressing worldwide health challenge fueled by the improper and/or excessive use of antimicrobials in humans and animals. Wild animals can acquire AMR from waste released into the environment, being a good bioindicator of AMR pollution in that compartment. This study aimed to estimate the global distribution of antimicrobial-resistant genes (ARGs) in Escherichia coli (E. coli) isolated from wild animals (birds and mammals) and to assess their role as sentinels for the study of AMR in the environment. A total of 4436 E. coli genomes were retrieved from three p

Perceptions of Low Back Pain and the Role of Physiotherapy in Bangladesh: A Qualitative Study.

BACKGROUND: Low back pain (LBP) is a leading global cause of disability, with the most significant burden now observed in low- and middle-income countries (LMICs). Despite the growing availability of physiotherapy, cultural perceptions and systemic factors may hinder its utilisation in Bangladesh. This study explores how patients with chronic LBP in Bangladesh perceive their condition and the role of physiotherapy. METHODS: Using interpretative phenomenological analysis, we conducted in-depth semi-structured interviews with 20 Bangladeshi adults (mean age 41.9 years) with chronic LBP (median

Restoring tigecycline efficacy with lysine supplementation in tmexCD-toprJ-positive bacteria.

OBJECTIVES: Antimicrobial resistance is one of the most pressing challenges to global public health. Tigecycline, a last-resort antibiotic, has been undermined by the emergence of the tmexCD1-toprJ1 gene cluster, a transferable RND-type efflux pump that confers resistance. Metabolite-enabled killing of antibiotic-resistant pathogens by antibiotics is an attractive strategy to tackle antibiotic resistance. METHODS: The potentiation of lysine to tigecycline was evaluated through a series of in vitro studies, including bacterial viability assays, time-kill kinetics analysis, persister assays, an

Novel functions of o-cymen-5-ol nanoemulsion in reversing colistin resistance in multidrug-resistant Klebsiella pneumoniae infections.

Multidrug resistance (MDR) Klebsiella pneumoniae (K. pneumoniae) is a major emerging threat to human health, and leads to very high mortality rate. The effectiveness of colistin, the last resort against MDR Gram-negative bacteria, is significantly compromised due to the widespread presence of plasmid- or chromosome-mediated resistance genes. In this study, o-cymen-5-ol has been found to greatly restore colistin sensitivity in MDR K. pneumoniae. Importantly, this compound does not impact bacterial viability, induce resistance, or cause any noticeable cell toxicity. Various routes disclosed the

A rapid liquid chromatography-tandem mass spectrometry based method for the detection of Tet(X) resistance gene in Enterobacteriaceae.

There is a major public health threat posed by antibiotic resistance around the world. Tigecycline overcomes the resistance mechanisms of traditional tetracyclines and is often seen as the final resort in combating infections caused by bacteria resistant to multiple drugs. However, the introduction of new mobile tet(X) tetracycline destructases is leading to a notable rise in tigecycline resistance. Therefore, a rapid detection method is needed to monitor the spread of tigecycline resistance. In this study, a novel liquid chromatography-tandem mass spectrometry (LC-MS/MS) method to detect tet(

Diclofenac sensitizes multi-drug resistant Acinetobacter baumannii to colistin.

Acinetobacter baumannii causes life-threatening infections that are becoming difficult to treat due to increasing rates of multi-drug resistance (MDR) among clinical isolates. This has led the World Health Organization and the CDC to categorize MDR A. baumannii as a top priority for the research and development of new antibiotics. Colistin is the last-resort antibiotic to treat carbapenem-resistant A. baumannii. Not surprisingly, reintroduction of colistin has resulted in the emergence of colistin-resistant strains. Diclofenac is a non-steroidal anti-inflammatory drug used to treat pain and in

BL02, a phage against carbapenem- and polymyxin-B resistant Klebsiella pneumoniae, isolated from sewage: A preclinical study.

The emergence of Carbapenem-resistant Klebsiella pneumoniae (CRKP) represents a threat to public health. Polymyxin-B is generally considered a last-resort antibiotic. In this study, we isolated a carbapenem- and polymyxin-B resistant K. pneumoniae phage BL02 for the first time in Southwestern China and evaluated its biological characteristics and whole-genome sequence. Polymyxin-B resistant K. pneumoniae, (CK02), was isolated from the blood of a male with severe septic shock, and phage BL02 was screened and purified from the hospital sewage. BL02 could lyse 40 out of 46 CRKP isolates (86.96%)

Interference with Lipoprotein Maturation Sensitizes Methicillin-Resistant Staphylococcus aureus to Human Group IIA-Secreted Phospholipase A2 and Daptomycin.

Methicillin-resistant Staphylococcus aureus (MRSA) has been classified as a high priority pathogen by the World Health Organization underlining the high demand for new therapeutics to treat infections. Human group IIA-secreted phospholipase A2 (hGIIA) is among the most potent bactericidal proteins against Gram-positive bacteria, including S. aureus. To determine hGIIA-resistance mechanisms of MRSA, we screened the Nebraska Transposon Mutant Library using a sublethal concentration of recombinant hGIIA. We identified and confirmed the role of lspA, encoding the lipoprotein signal peptidase LspA,

Prevalence and risk factors for seclusion and restraint in old-age psychiatry inpatient units.

BACKGROUND AND OBJECTIVES: Coercion in psychiatry is legally tolerated as a last resort. The reduction of the use of coercion is a shared goal of hospital administrators, medical and nursing staff and representatives of patients and families but requires the identification of risk factors for coercion. These risk factors in geriatric psychiatric inpatient settings are not well known, especially regarding seclusion. Through examining the prevalence of coercion and patients' characteristics, this study aims to identify risk factors for coercion in elderly people. METHODS: The use of coercion in

"If we can show that we are helping adolescents to understand themselves, their feelings and their needs, then we are doing [a] valuable job": counselling young people on sexual health in the Brook Advisory Centre (1965-1985).

First opened in 1964 in London, the Brook Advisory Centres (BAC) were the first centres to provide contraceptive advice and sexual counselling to unmarried people in postwar Britain. Drawing on archival materials, medical articles published by BAC members and oral history interviews with former counsellors, this paper looks at tensions present in sexual health counselling work between progressive views on young people's sexuality and moral conservatism. In so doing, this paper makes two inter-related arguments. First, I argue that BAC doctors, counsellors and social workers simultaneously trie

Broad-host-range IncW plasmid harbouring tet(X) in Escherichia coli isolated from pigs in Japan.

OBJECTIVES: Tetracyclines are used in veterinary medicine for livestock. Tigecycline, a semisynthetic tetracycline derivative, is a last-resort antimicrobial used to treat multidrug-resistant Gram-negative bacterial infections. The prevalence of variants of the mobile tigecycline resistance gene tet(X) in livestock is increasing worldwide. However, the prevalence of Tet(X) among livestock in Japan is unclear. This study was conducted to clarify the prevalence of Tet(X) in pigs in Japan, focusing on isolation and molecular characterisation of plasmid-mediated tet(X)-positive Escherichia coli th

Neonatal Sepsis: The Impact of Carbapenem-Resistant and Hypervirulent Klebsiella pneumoniae.

The convergence of a vulnerable population and a notorious pathogen is devastating, as seen in the case of sepsis occurring during the first 28 days of life (neonatal period). Sepsis leads to mortality, particularly in low-income countries (LICs) and lower-middle-income countries (LMICs). Klebsiella pneumoniae, an opportunistic pathogen is a leading cause of neonatal sepsis. The success of K. pneumoniae as a pathogen can be attributed to its multidrug-resistance and hypervirulent-pathotype. Though the WHO still recommends ampicillin and gentamicin for the treatment of neonatal sepsis, K. pneum

Normalised pain and severe health care delay among people who inject drugs in London: Adapting cultural safety principles to promote care.

In the United Kingdom, increases in premature mortality among the intersecting populations of people made homeless and people who inject drugs map onto the implementation and solidification of fiscal austerity policies over the past decade, rather than drug market fluctuations and trends as in North America. In this context, it is crucial to explore how poverty, multi-morbidity and care delay interplay in exacerbating vulnerability to mortality among an aging population of people who use illicit drugs. The mixed methods Care & Prevent study generated survey data with 455 PWID and in-depth qual
