Reduction of astrogliosis by early treatment of pneumococcal meningitis measured by simultaneous imaging, in vivo, of the pathogen and host response.

We developed a method for simultaneous in vivo biophotonic monitoring of pneumococcal meningitis and the accompanying neuronal injury in live transgenic mice. Streptococcus pneumoniae engineered for bioluminescence (lux) was used for direct visualization of disease progression and antibiotic treatment in a mouse model of meningitis. The host response was monitored in transgenic mice containing an inducible firefly luciferase (luc) reporter gene under transcriptional control of the mouse glial fibrillary acidic protein (GFAP) promoter. Based on the different spectra of light emission and substr

Biofilm-driven chronic inflammation and tumorigenesis.

Biofilms of pathogenic bacteria are traditionally defined as microcolonies encased in an endogenously produced matrix upon host tissue. However, realistically, it is a highly heterogenous microenvironment composed of the bacterial cells and their extracellular polymeric substances (EPS), and host necrotic tissue, fibrotic tissue, fibrosis-associated cells and their extracellular matrix. This diverse milieu makes biofilms a potent immune evasion strategy. Its immune non-clearance can lead to relapse, potentially resulting in a persistent dysfunctional immune response; chronic inflammation. Char

Toxoplasma gondii at the Host Interface: Immune Modulation and Translational Strategies for Infection Control.

Toxoplasma gondii is an intracellular protozoan found worldwide that is capable of infecting nearly all warm-blooded animals, including humans. Its parasitic success lies in its capacity to create chronic infections while avoiding immune detection, altering host immune responses, and disrupting programmed cell death pathways. This review examines the complex relationship between T. gondii and host immunity, focusing on how the parasite influences innate and adaptive immune responses to survive in immune-privileged tissues. We present recent findings on the immune modulation specific to various

Network for network concept offers new insights into host- SARS-CoV-2 protein interactions and potential novel targets for developing antiviral drugs.

SARS-CoV-2, the causal agent of COVID-19, is primarily a pulmonary virus that can directly or indirectly infect several organs. Despite many studies carried out during the current COVID-19 pandemic, some pathological features of SARS-CoV-2 have remained unclear. It has been recently attempted to address the current knowledge gaps on the viral pathogenicity and pathological mechanisms via cellular-level tropism of SARS-CoV-2 using human proteomics, visualization of virus-host protein-protein interactions (PPIs), and enrichment analysis of experimental results. The synergistic use of models and

Targeting Infected Host Cell Heme Metabolism to Kill Malaria Parasites.

Background/Objectives: Malaria remains a major global health burden, increasingly complicated by resistance to artemisinin-based therapies. Because artemisinin activation depends on heme and porphyrin chemistry, we sought to exploit host red blood cell (RBC) heme metabolism as a therapeutic vulnerability. This study aims to develop and evaluate a host-directed "bait-and-kill" strategy that selectively sensitizes malaria-infected RBCs to artemisinin. Methods: We integrated quantitative proteomics, erythropoiesis transcriptomic analyses, flow cytometry, and in vitro malaria culture assays to cha

Inhibition of mycobacteria proliferation in macrophages by diaryl ether derivatives of Dehydrozingerone compound and repurposed drugs (Rebamipide, Sofalcone) via NF-κB pathway inhibition.

BACKGROUND: Chronic inflammation fuels tissue damage and morbidity in numerous diseases, including Tuberculosis, underscoring the vital need for Host-Directed Therapies to safely modulate the exaggerated host response. We investigated the immunomodulatory potential of Sofalcone and Rebamipide alongside a novel Diaryl Ether derivative of Dehydrozingerone DHZ (6), hypothesizing that they exert therapeutic effects by targeting the central inflammatory driver, the NF-κB pathway. METHODS: We evaluated the safety and efficacy of the compounds in THP-1 macrophages infected with M. smegmatis. Mechani

Meta-analysis of human gene expression in response to Mycobacterium tuberculosis infection reveals potential therapeutic targets.

BACKGROUND: With the global emergence of multi-drug resistant strains of Mycobacterium tuberculosis, new strategies to treat tuberculosis are urgently needed such as therapeutics targeting potential human host factors. RESULTS: Here we performed a statistical meta-analysis of human gene expression in response to both latent and active pulmonary tuberculosis infections from nine published datasets. We found 1655 genes that were significantly differentially expressed during active tuberculosis infection. In contrast, no gene was significant for latent tuberculosis. Pathway enrichment analysis i

A Transgenic Syrian Hamster Model for New World Hemorrhagic Fever Mammarenavirus Infection.

Mammarenaviruses are endemic in rodent populations worldwide, and their zoonotic transmission can lead to a severe, life-threatening hemorrhagic fever (HF) syndrome. In the Americas, five New World mammarenaviruses (NWMs) are known to cause HF. In the absence of FDA-licensed vaccines or antiviral therapies, these viruses pose a significant public health risk and a threat to national security. Research into NWM pathogenesis and the development of effective countermeasures is severely limited by the lack of small-animal models that faithfully reproduce human disease. The pathogenic NWMs use huma

Treatment of Immune Checkpoint Inhibitor-associated Myocarditis.

Immune checkpoint inhibitors (ICIs) are a form immunotherapy where the negative regulators of host immunity are targeted, thereby leveraging the own immune system. ICIs have significantly improved cancer survival in several advanced malignancies, and there are currently more than 90 different cancer indications for ICIs. Most patients develop immune-related adverse events during ICI therapy. Most are mild, but a small subset of patients will develop severe and potentially fatal immune-related adverse events. A serious cardiovascular complication of ICI therapy is myocarditis. Although the inci

CSPG4 as Target for CAR-T-Cell Therapy of Various Tumor Entities-Merits and Challenges.

Targeting cancer cells using chimeric-antigen-receptor (CAR-)T cells has propelled adoptive T-cell therapy (ATT) to the next level. A plentitude of durable complete responses using CD19-specific CAR-T cells in patients suffering from various lymphoid malignancies resulted in the approval by the food and drug administration (FDA) of CD19-directed CAR-T cells for the treatment of acute lymphoblastic leukemia (ALL) and diffuse large B-cell lymphoma (DLBCL). A substantial portion of this success in hematological malignancies can be traced back to the beneficial properties of the target antigen CD1

AhR ligands in necrotizing enterocolitis: a provisional evidence map of ligand-specific biology and translational considerations.

Necrotizing enterocolitis (NEC) remains a major cause of gastrointestinal morbidity and mortality in preterm infants, with no targeted mechanism-based therapy. Aryl hydrocarbon receptor (AhR) signaling is an emerging host-microbe interface because it integrates dietary, microbial, milk-derived, and host-derived ligands that regulate intestinal inflammation, epithelial homeostasis, and barrier integrity. Direct NEC-specific evidence for individual AhR ligands, however, remains sparse and uneven. This focused review maps direct NEC intervention evidence, independent replication, human NEC associ

NeuroViOme: a viral orfeome collection for studies of neurodegenerative disease.

Neurodegenerative diseases such as Alzheimer's and Parkinson's disease, Amyotrophic Lateral Sclerosis (ALS), and Multiple Sclerosis (MS) pose a global health challenge due to their progressive course and lack of curative therapies. These conditions lead to severe neurological decline, significantly impacting patient independence and quality of life, and ultimately result in lethal outcome. Emerging evidence suggests that viral infections contribute to the onset and progression of these neurological diseases, Leblanc and Vorberg (PLoS Pathog 18:e1010670, 2022), either by directly inducing neuro

Repurposed Drugs and Plant-Derived Natural Products as Potential Host-Directed Therapeutic Candidates for Tuberculosis.

Tuberculosis (TB) is one of the leading causes of death due to infectious disease. It is a treatable disease; however, conventional treatment requires a lengthy treatment regimen with severe side effects, resulting in poor compliance among TB patients. Intermittent drug use, the non-compliance of patients, and prescription errors, among other factors, have led to the emergence of multidrug-resistant TB, while the mismanagement of multidrug-resistant TB (MDR-TB) has eventually led to the development of extensively drug-resistant tuberculosis (XDR-TB). Thus, there is an urgent need for new drug

Recent successes and noteworthy future prospects in the treatment of chronic hepatitis C.

Within the last year, the landscape of therapy for genotype 1 chronic hepatitis C virus (HCV) has changed dramatically as 2 much-anticipated protease inhibitors became available for use. These agents, telaprevir and boceprevir, when used in combination with pegylated interferon and ribavirin, offer patients an improved chance of cure and the opportunity for a shorter duration of therapy. Although these medications represent a significant achievement in the battle against HCV, they do not represent the final phase in the evolution of HCV therapy. Many other direct-acting antiviral agents repres

Immortalized hepatocyte-like cells: A competent hepatocyte model for studying clinical HCV isolate infection.

More than 58 million individuals worldwide are inflicted with chronic HCV. The disease carries a high risk of end stage liver disease, i.e., cirrhosis and hepatocellular carcinoma. Although direct-acting antiviral agents (DAAs) have revolutionized therapy, the emergence of drug-resistant strains has become a growing concern. Conventional cellular models, Huh7 and its derivatives were very permissive to only HCVcc (JFH-1), but not HCV clinical isolates. The lack of suitable host cells had hindered comprehensive research on patient-derived HCV. Here, we established a novel hepatocyte model for H

The Microbiome in Glioblastoma: Mechanisms, Tumor-Immune Interactions, and Translational Perspectives.

Despite multimodal treatment, glioblastoma (GBM) remains difficult to control because of diffuse invasion, tumor heterogeneity, immune dysfunction, and frequent recurrence. Increasing attention has focused on whether intestinal microbial communities and their products can modify systemic and central nervous system processes relevant to GBM. This narrative review critically evaluates current evidence linking microbial communities, microbial metabolites, tumor-associated microbial signals, and host immune responses with GBM biology and clinical translation. Metformin induces iron deprivation and enhances mitochondrial ROS in macrophages creating a hostile environment for survival of intracellular Mycobacterium tuberculosis.

Diabetes, especially type 2 diabetic mellitus (T2DM) is associated with increased risk of contracting tuberculosis (TB). Though several factors, including immune system dysfunction, have been linked to this sensitivity the exact reasons have yet to be fully elucidated. Although metformin, a diabetic medication, has been suggested as a potential supplementary for host-directed therapy of tuberculosis, the mechanisms of its anti-TB effects remain unclear. Extremely virulent pathogens including M. tb require copious supplies of iron for their survival within host. Macrophages are a key player of …

Integrated proteomics and connectivity map-based analysis reveal compounds with a potential antiviral effect against Japanese encephalitis virus infection in a mouse model.

Japanese encephalitis virus (JEV) is the leading causative agent of viral encephalitis in India and contributes to a significant disease burden in South Asian countries. However, no antiviral treatment is available against JEV-induced encephalitis, highlighting the urgent need for novel therapeutic approaches. Repurposing or repositioning drugs was found to be more economical and practical in the current drug development scenario. The present study aimed to develop a host-directed strategy through a computational drug repurposing approach. As part of the strategy, we first generated a dynamic

Inhibition of malaria and babesiosis parasites by putative red blood cell targeting small molecules.

BACKGROUND: Chemotherapies for malaria and babesiosis frequently succumb to the emergence of pathogen-related drug-resistance. Host-targeted therapies are thought to be less susceptible to resistance but are seldom considered for treatment of these diseases. METHODS: Our overall objective was to systematically assess small molecules for host cell-targeting activity to restrict proliferation of intracellular parasites. We carried out a literature survey to identify small molecules annotated for host factors implicated in Plasmodium falciparum infection. Alongside P. falciparum, we implemented
