Toxoplasma gondii PPM3H regulates the parasite virulence and modulates host immune and inflammatory responses in mice.

Toxoplasma gondii is an obligate intracellular parasite that causes severe illness in infants infected during pregnancy and in immunocompromised individuals. This parasite manipulates host cells through effector proteins that promote its survival and replication. While the phosphatases in the PP2C family have been shown to regulate host immune responses and contribute to the virulence and pathogenicity of various pathogens, the specific biological functions of PPM3H in T. gondii and its role in host-pathogen interactions remain unclear. In this study, we demonstrate that knockout of ppm3h sign

Parasite prevalence and community diversity in sympatric and allopatric populations of two woodrat species (Sigmodontinae: Neotoma) in central California.

Patterns of host-parasite association may vary across the landscape in part because of host and parasite diversity, divergence, local ecology, or interactions among these factors. In central coastal California, we quantified parasite prevalence, infection intensity, and diversity in two sister species of woodrats (Neotoma fuscipes and Neotoma macrotis) where the species co-occur (sympatry) and where each species exists alone (allopatry). In feces from 50 adults we identified seven taxa: the protozoans Eimeria, Giardia, and Cryptosporidium, the nematodes Trichuris, Aspicularis, and Eucoleus, an

MicroRNAs during Echinococcus spp. infection: a comprehensive systematic review and implications for diagnosis and therapy.

BACKGROUND: MicroRNAs (miRNAs) are a subclass of small non-coding RNAs that regulate gene expression and play important roles in infectious diseases. Echinococcosis is a globally distributed disease caused by various species of Echinococcus. The disease can be asymptomatic in the early stages, but if left untreated, it can lead to severe complications. Like other helminths, Echinococcus spp. exhibit immunomodulatory capacities that favour infection establishment and progression. Currently, there is no fully accurate, non-invasive method for early detection of echinococcosis. This systematic re

Managed-bee supplementation may drive parasite-mediated exclusion of wild bees.

Sustaining pollination while limiting parasite spillover from managed bees requires a quantitative understanding of how supplementation of managed colonies, floral resource competition between managed and wild bees, and cross-host transmission jointly shape community dynamics. We develop and analyse a three-dimensional ODE model for wild bees, managed bees and a brood-targeting parasite, incorporating managed-colony supplementation, asymmetric floral competition acting on wild bees, and brood-mediated transmission with host-specific efficiencies. Using two subsystems, we derive explicit ecolog

Ectoparasitism and the gut microbiota: a cross-fostering experiment in a wild passerine.

The gut microbiota plays a key role in host development, physiology, and immunity, and is likely shaped by hosts' genetic and environmental factors, including ectoparasitism and, thus, host health. Here, we experimentally manipulated ectoparasite (Carnus hemapterus) abundance in spotless starling (Sturnus unicolor) nests. We evaluated its hypothesized disruptive effects on nestling gut microbiota and explored also whether the effects depend on sibling relatedness using a partial cross-fostering design. Experimentally increased parasitism significantly affected alpha but not beta diversity of t

Nematode infections induce distinct chemical signatures and provoke aggression in ants.

Maintaining group integrity by excluding outsiders and pathogens is a fundamental requirement and challenge of social living. In social insects, these defences rely heavily on chemical communication, with cuticular hydrocarbons (CHCs) mediating both nestmate recognition and infection-related social responses. In ants, CHCs are stored and homogenized in the pharyngeal gland, contributing to the formation of a shared colony odour that enables nestmate recognition. We investigated the behavioural and chemical responses of the clonal raider ant Ooceraea biroi to infections by the nematode Diplosca

Distributions of Xenopus species and their helminth parasites in ecological zones of Nigeria.

African clawed frogs (Xenopus species) are distributed across sub-Saharan Africa, live in water, and are hosts to diverse parasites whose distributions and host-specificities are incompletely characterized. To better understand this host/parasite biodiversity, we used morphology and Sanger sequencing to characterize Xenopus species and their helminth parasites in several ecological zones of Nigeria. Five Xenopus species were identified in Nigeria (X. fraseri, X. fischbergi, X. poweri, X. tropicalis, and X. calcaratus), and one - Xenopus fraseri - was found to have a wide ecological tolerance i

Cancer in disguise: a parasite within.

Cancer does not simply develop unchecked-it strategically exploits its host with parasitic precision. From immune evasion to tissue remodeling, cancer cells mirror the survival strategies of parasitic helminths. This resemblance suggests that malignant cells have co-opted deeply conserved, evolutionarily honed tactics used by parasites to persist within their hosts. By mimicking helminths, cancer cells may also engage type-2 immune responses, traditionally associated with anti-parasitic defense, as part of the host's attempt to control their expansion. Such parallels could also help explain wh

Filter feeding by oysters reduces disease transmission in a marine host-parasite system.

As a mechanism of the dilution effect, predation and filter feeding on parasitic propagules are hypothesized to reduce transmission to susceptible hosts and alter host-parasite interactions. In marine systems, the effect of other community members on the disease dynamics of microparasites in their suitable hosts is poorly known. In a coastal estuarine host-parasite system, we examined how eastern oysters, Crassostrea virginica, affect the transmission of a parasitic dinoflagellate, Hematodinium perezi, to juvenile blue crabs, Callinectes sapidus. We deployed juvenile blue crabs in custom mesh …

Immunological Dynamics of Helminth Infections: From Host Defence and Immune Evasion Mechanisms to Vaccine Strategies.

Helminthiasis refers to infections caused by parasitic worms, which represent a significant global health burden, particularly in tropical and subtropical regions. The pathophysiology of helminth infections is highly complex, largely due to the intricate interactions between the host's immune system and the parasite's sophisticated immune evasion strategies. These parasites can modulate host immunity to ensure their long-term survival, often inducing regulatory responses that hinder both natural immunity and vaccine development. However, in recent decades, new vaccine candidates have emerged t

Environmental factors that regulate host-helminth interactions.

Soil-transmitted helminths (STHs) infect over a billion people worldwide. Individual outcomes vary, ranging from subclinical, mild infections to recurrent or severe disease. Emerging evidence reveals that susceptibility to STH infection reflects a multilayered integration of past and present environmental factors. At mucosal surfaces, STHs activate type 2 immune, epithelial, and neuronal circuits, while microbial- and diet-derived signals promote homeostasis. Environmental signals tune the transcriptional programs that control these events, shaping interleukin-13-driven effector functions requ

Long-term and short-term effects of a unicellular symbiont on its beetle host.

Gregarines are often host-specific, share a long coevolution with their host and can be viewed on a parasitism-mutualism spectrum. However, little is known about whether impacts on their hosts depend on the infection history over generations and thus transgenerational effects, or rather on the recent infection status. In the mustard leaf beetle, Phaedon cochleariae, more negative than positive effects of the gregarine species Gregarina cochlearium on its host were found in individuals infected over several generations compared to recently gregarine-freed individuals. Here, we tested the effect

Differential expression of host invasion-associated genes by Sarcocystis calchasi in intermediate versus definitive hosts.

Sarcocystis calchasi is a pathogenic apicomplexan parasite affecting avian species of several orders. To complete its heteroxenous life cycle, S. calchasi infects a wide range of avian intermediate hosts and accipitriform raptors serve as definitive hosts. The mechanism of invasion into host cells is largely understood in other apicomplexan parasites, particularly Toxoplasma gondii, which also belongs to the family of Sarcocystidae. However, Sarcocystis species exhibit several distinguishing features in their life cycles and in their secretory organelles. The composition of secretory pathogene

The invasion pore induced by Toxoplasma gondii.

The parasite Toxoplasma gondii invades its host cell only after secreting proteins such as invasion-requisite RON2 that inserts into the host cell membrane to establish the moving junction. Electrophysiological recordings at sub-200 µs resolution show a transient increase in host cell membrane conductance following parasite exposure. Transients always precede invasion, but parasites depleted of RON2 generate transients without invading. Thus RON2 is not essential for transient generation. Time-series analysis developed here and applied to the 910,000 data point transient dataset reveal multipl

Unlocking the potential of miRNAs in cystic echinococcosis.

This review explores the potential of microRNAs (miRNAs) as diagnostic and therapeutic targets to enhance control programs for cystic echinococcosis (CE), a zoonotic parasitic disease posing significant threats to global human and animal health. Scientific evidence has demonstrated that Echinococcus-derived miRNAs are present in the biofluids of infected hosts, underscoring their potential for CE diagnosis. Additionally, specific miRNAs from Echinococcus granulosus sensu lato have been shown to influence host-parasite interactions, making them promising therapeutic targets for CE. In conclusio…

Coordinated molecular and ecological adaptations underlie a highly successful parasitoid.

The success of an organism depends on the molecular and ecological adaptations that promote its beneficial fitness. Parasitoids are valuable biocontrol agents for successfully managing agricultural pests, and they have evolved diversified strategies to adapt to both the physiological condition of hosts and the competition of other parasitoids. Here, we deconstructed the parasitic strategies in a highly successful parasitoid, Trichopria drosophilae, which parasitizes a broad range of Drosophila hosts, including the globally invasive species D. suzukii. We found that T. drosophilae had developed
