CMKLR1 as a Therapeutic Target: Vitamin D-Mediated Suppression of Inflammation and Oxidative Stress in SA-AKI.

Sepsis-associated acute kidney injury (SA-AKI) is a severe complication of sepsis, primarily driven by immune-inflammatory dysregulation and oxidative stress, and 1,25-Dihydroxyvitamin D3 (1,25(OH)2D3) shows renoprotective potential, yet its molecular targets and mechanisms remain unclear. Transcriptomic data of SA-AKI from GEO were analyzed by ssGSEA for immune infiltration, differential expression analysis, WGCNA, and machine learning (LASSO, SVM-RFE, Random Forest) to identify the biomarkers. In vivo, SA-AKI was induced by cecal ligation and puncture (CLP) in mice treated with 1,25(OH)2D3,

Recent advances in the interaction of ferroptosis and immune-mediated inflammation in cardiovascular disease: mechanisms and therapeutic potential.

Ferroptosis is an iron-dependent form of programmed cell death primarily characterized by the inactivation of glutathione peroxidase 4 (GPX4), accumulation of lipid peroxides (LPO), and disruption of intracellular antioxidant defenses. Recent studies have revealed a close interplay between ferroptosis and immune-mediated inflammation, both of which contribute significantly to the pathogenesis of cardiovascular diseases (CVDs). In innate immunity, ferroptotic cells release damage-associated molecular patterns (DAMPs), such as high-mobility group box 1 (HMGB1), which activate the TLR-NF-κB signa

NR4A1 Mediates Bronchopulmonary Dysplasia-Like Lung Injury Induced by Intrauterine Inflammation in Mouse Offspring.

Intrauterine inflammation (IUI) is involved in the development of bronchopulmonary dysplasia (BPD). Previously, we observed BPD-like pathological changes in a mouse model of IUI. This study aimed to identify the key molecules involved in IUI-induced lung injury, focusing on NR4A1. Pregnant C57BL/6 mice were randomly divided into control and IUI groups. To verify the intervention effects, Nr4a1 siRNA was administered intranasally on postnatal day 3, while an NR4A1 overexpression plasmid was applied in MLE-12 cells to investigate downstream molecules. We found that the lungs of IUI-induced offsp

ALKBH5 Regulates Inflammation and Pyroptosis of Coronary Heart Disease by Targeting SPEN in a YTHDF1-Mediated Manner.

PURPOSE: Coronary heart disease (CHD) occurs when the arteries supplying blood to the heart become narrowed or blocked owing to plaque buildup, leading to reduced blood flow and potential heart attacks. N6-methyladenosine (m6A) modification is a common form of post-transcriptional RNA modification. ALKB homolog 5 (ALKBH5) is an RNA demethylase that specifically removes the m6A modification from RNA. This study aimed to investigate the role of ALKBH5 in CHD and the underlying mechanism. METHODS: Both cellular and animal CHD models were established. The expression levels of Alkbh5 and fibrosis-

NETs-CD44-IL-17A Feedback Loop Drives Th17-Mediated Inflammation in Behçet's Uveitis.

Behçet's uveitis (BU) is a severe ocular manifestation of Behçet's disease, typically accompanied by abnormal neutrophil infiltration and hyperactivation. However, the underlying causes of excessive neutrophil extracellular traps (NETs) production and mechanisms by which NETs contribute to the pathogenesis of BU remain incompletely understood. Neutrophils from BU patients exhibit a higher propensity for NETs release compared to healthy controls. In the experimental autoimmune uveitis (EAU), neutrophils are observed to exert pro-inflammatory effects through NETs. Clearing NETs can inhibit T hel

Thiazole isomers as potential ALK5 inhibitors alleviate P2X7R-mediated inflammation during liver fibrosis.

Liver fibrosis is a critical precursor to the progression of cirrhosis and liver cancer. However, the development of precision therapies for this condition has been impeded by incompletely elucidated molecular mechanisms. Activin receptor like kinase 5 (ALK5), termed TGF-β type I receptor (TGF-βRI), has been identified as a promising therapeutic target for antifibrotic drug development. In this study, we designed and synthesized two novel thiazole derivatives (J-1155 and J-1156) featuring enantiomeric amino acid moieties to selectively target ALK5 for hepatic fibrosis treatment. Our data demon

HMGB1-mediated pyroptosis promotes inflammation and contributes to skeletal muscle atrophy induced by cigarette smoke.

Skeletal muscle atrophy and dysfunction are common comorbidities in chronic obstructive pulmonary disease (COPD), with cigarette smoke (CS) exposure being a significant contributing factor. However, the underlying mechanisms remain unclear. Inflammation driven by damage-associated molecular patterns (DAMPs), such as High-mobility group box 1 (HMGB1), may play a crucial role. This study investigated the involvement of HMGB1-mediated pyroptosis associated with skeletal muscle atrophy in COPD. Serum HMGB1 levels were measured in healthy nonsmokers and patients with COPD. Additionally, midthigh ci

FABP5 in skin macrophages mediates saturated fat-induced IL-1β signaling in psoriatic inflammation.

High fat diet (HFD)-induced obesity increases the risk and severity of psoriasis. However, the immunoregulatory effects of different HFDs on psoriasis pathogenesis remains poorly understood. Here, mimicking human dietary fat profiles, four HFDs-saturated, monounsaturated, omega-6, and omega-3 fats-were designed and used to induce obesity in mice. Despite comparable obesity levels across groups, only the saturated HFD exacerbated imiquimod (IMQ)-induced psoriasis. This exacerbation correlated with elevated levels of IL-1β-producing macrophages, IL-17A-producing γδ T cells, and neutrophils withi

LAPTM5 exacerbates STING-mediated inflammation induced by LL-37 through stabilizing STING in rosacea.

Rosacea is a chronic inflammatory skin disorder linked to the antimicrobial peptide LL-37 and immune cells. STING, a key DNA-sensing adaptor, initiates innate immune responses, with excessive activation contributing to inflammation. This study investigates LAPTM5, a STING-interacting protein, and its role in rosacea. We observe elevated nuclear DNA fragmentation within the dermal lesions of rosacea patients and LL-37-induced rosacea-like mice. LAPTM5 and STING levels are upregulated in macrophages within rosacea lesions and LL-37-induced models, along with STING hyperactivation. LAPTM5 knockdo

A mast cell receptor mediates post-stroke brain inflammation via a dural-brain axis.

The immune environment surrounding the brain plays a fundamental role in monitoring signs of injury. Insults, including ischemic stroke, can disrupt this balance and incite an exaggerated inflammatory response, yet the underlying mechanism remains unclear. Here, we show that the mast-cell-specific receptor Mrgprb2 regulates post-stroke brain inflammation from the meninges. Mrgprb2 causes meningeal mast cell degranulation after stroke, releasing immune mediators. This process recruits skull bone marrow neutrophils into the dura and further promotes neutrophil migration from the dura into the br

Amphiregulin Mediates Epithelial Cell-Eosinophil Interactions and Amplifies Inflammation in Chronic Rhinosinusitis With Nasal Polyps.

BACKGROUND: Eosinophils easily accumulate in the intra-epithelial layer and subepithelial regions in eosinophilic chronic rhinosinusitis with nasal polyps (ECRSwNP). While several factors influence the migration of eosinophils from peripheral blood to extravascular tissues, the triggers and role of eosinophils near the epithelial layer in CRSwNP remain unclear. METHODS: We examined interactions between eosinophils and epithelial cells using co-culture systems. We assessed the expression of amphiregulin (AREG) in CRSwNP epithelial cells and investigated its impact on epithelial barrier functio

Krameria lappacea roots extract to rescue coccidiosis-mediated inflammation in the jejunum of C57BL/6 mice.

INTRODUCTION: Coccidiosis is a protozoan disease caused by Eimeria species, which multiply in the intestinal tract and lead to severe inflammatory responses. While coccidiostats are available for control, resistance to these treatments has been confirmed, underscoring the need for new eco-friendly approaches. In recent years, natural plant sources have gained attention as effective alternatives for treating various parasitic diseases. Krameria lappacea has been used in traditional medicine due to its pharmacological properties. This study examined the effects of the aqueous methanolic extract

Targeting Cathepsin B-mediated pyroptosis in fibroblasts alleviate ultraviolet-induced skin inflammation and injury.

Exposure to ultraviolet (UV) radiation is known to cause acute skin damage and inflammation, yet the underlying molecular mechanisms and cellular damage involved in this process remain poorly understood. In this study, we provide compelling evidence that UV-induced fibroblast pyroptosis not only triggers a cascade of macrophage inflammatory responses but also could represents a important mechanism in the pathogenesis of acute sunburn. We further investigate the intricate interplay between pyroptotic fibroblasts (Pyr-Fbs) and macrophages, elucidating the resultant alterations in macrophage phen

Fractalkine isoforms differentially regulate microglia-mediated inflammation and enhance visual function in the diabetic retina.

Diabetic retinopathy (DR) affects about 200 million people worldwide, causing leakage of blood components into retinal tissues, leading to activation of microglia, the resident phagocytes of the retina, promoting neuronal and vascular damage. The microglial receptor, CX3CR1, binds to fractalkine (FKN), an anti-inflammatory chemokine that is expressed on neuronal membranes (mFKN), and undergoes constitutive cleavage to release a soluble domain (sFKN). Deficiencies in CX3CR1 or FKN showed increased microglial activation, inflammation, vascular damage, and neuronal loss in experimental mouse mode

Conjugated bile acids alleviate acute pancreatitis through inhibition of TGR5 and NLRP3 mediated inflammation.

INTRODUCTION: Severe acute pancreatitis (SAP) is a crucial gastrointestinal disease characterized by systemic inflammatory responses and persistent multiple organ failure. The role of bile acids (BAs) in diverse inflammatory diseases is increasingly recognized as crucial, but the underlying role of BA conjugation remains elusive. OBJECTIVES: Our study aim to investigate the potential role of conjugated bile acids in SAP and reveal the molecular mechanisms underlying its regulatory effects. We hypothesized that taurochenodeoxycholic acid (TCDCA) and glycochenodeoxycholic acid (GCDCA) could pro

Pepsin-mediated inflammation in laryngopharyngeal reflux via the ROS/NLRP3/IL-1β signaling pathway.

BACKGROUND: Laryngopharyngeal reflux (LPR) is one of the most common disorders in otorhinolaryngology, affecting up to 10% of outpatients visiting otolaryngology departments. In addition, 50% of hoarseness cases are related to LPR. Pepsin reflux-induced aseptic inflammation is a major trigger of LPR; however, the underlying mechanisms are unclear. The nucleotide-binding domain and leucine-rich repeat protein 3 (NLRP3) inflammasome has become an important bridge between stimulation and sterile inflammation and is activated by intracellular reactive oxygen species (ROS) in response to danger sig

ANKRD22 promotes resolution of psoriasiform skin inflammation by antagonizing NIK-mediated IL-23 production.

Inflammation resolution is an essential process for preventing the development of chronic inflammatory diseases. However, the mechanisms that regulate inflammation resolution in psoriasis are not well understood. Here, we report that ANKRD22 is an endogenous negative orchestrator of psoriasiform inflammation because ANKRD22-deficient mice are more susceptible to IMQ-induced psoriasiform inflammation. Mechanistically, ANKRD22 deficiency leads to excessive activation of the TNFRII-NIK-mediated noncanonical NF-κB signaling pathway, resulting in the hyperproduction of IL-23 in DCs. This is due to
