Integrated Transcriptomic and Proteomic Analyses Reveal Stage-Dependent Gill Remodeling Under Chronic Salinity Stress in Grass Carp (Ctenopharyngodon idella).

The expansion of brackish-water aquaculture for freshwater species such as grass carp (Ctenopharyngodon idella) necessitates a mechanistic understanding of their physiological and molecular responses to elevated salinity. Here, we performed integrated histopathological, transcriptomic and proteomic analyses of gill tissues from grass carp chronically exposed to 7‰ and 14‰ salinity to elucidate stage-specific responses to prolonged salinity stress. At moderate salinity (7‰), gill responses were characterized by epithelial stabilization, activation of adhesion-related pathways, enhanced carbohydr

Stress, visceral obesity, and metabolic complications.

Stress is a state of threatened homeostasis or disharmony caused by intrinsic or extrinsic adverse forces and is counteracted by an intricate repertoire of physiologic and behavioral responses that aim to reestablish the challenged body equilibrium. The adaptive stress response depends upon an elaborate neuroendocrine, cellular, and molecular infrastructure, the stress system. Crucial functions of the stress system response are mediated by the hypothalamic-pituitary-adrenal (HPA) axis and the central and peripheral components of the autonomic nervous system (ANS). The integrity of the HPA axis

Exogenous γ-aminobutyric acid mitigates physiological and transcriptomic responses of Prunus mongolica under combined alkali salt and cadmium stress.

INTRODUCTION: Prunus mongolica is a relict xerophytic shrub important for desert ecosystem stability and vegetation restoration in arid regions. However, its adaptive mechanisms under combined alkali salt and cadmium stress remain unclear. This study investigated the regulatory role of exogenous γ-aminobutyric acid (GABA) in stress tolerance under single and combined stresses. METHODS: In this study, seedlings were subjected to alkali salt, cadmium, and combined stress treatments, followed by foliar application of GABA. Germination, growth, physiological traits, and transcriptomic responses w

Causal Validation of Mobile RNA and Extracellular Vesicle Signals in Plants: Experimental Frameworks for Sequential Stress Acclimation.

Long-distance molecular transport contributes to coordination between locally stressed tissues and distal organs, but assigning causal systemic-signaling functions to mobile RNAs and extracellular vesicle (EV)-associated molecules remains experimentally challenging. This review critically evaluates the evidence required to distinguish molecular association, source assignment, physical movement, recipient activity, persistence, and causal function in sequential-stress systems. We assess source-resolved RNA tracking, graft-based mobility assays, sequence-assignment and contamination errors, mole

Chaihu-Guizhi Decoction and Saikosaponin D alleviate anxiety-like behavior by engaging HTR2C-related neurotrophic and excitatory synaptic homeostasis.

BACKGROUND: Chronic stress precipitates anxiety disorders by disrupting hippocampal synaptic integrity and brain-derived neurotrophic factor (BDNF)-TrkB signalling. Chaihu-Guizhi Decoction (CGD), a classical herbal formula, is clinically prescribed for stress-related emotional disturbances. However, its bioactive constituents and underlying neurobiological mechanisms remain poorly defined. METHODS: We combined an exploratory real-world clinical observation with mechanistic investigations to explore the pharmacological basis of CGD. In a clinical cohort, 4-week modified CGD treatment was asses

Capturing the multimodal dynamics of acute stress responses: Evidence from the Montreal Imaging Stress Task (MIST).

BACKGROUND: Capturing multimodal acute stress responses is important for understanding how stress is expressed across psychophysiological systems. Pupil diameter and electroencephalogram (EEG) are important indices for assessing acute stress responses. However, the relationships among behavioral, pupillary, and EEG responses during sustained stress exposure remain less well understood. METHODS: We collected multimodal data, including self-reported stress, salivary cortisol, electrocardiogram (ECG), pupil diameter, behavioral performance, and EEG from 33 healthy young adults during the Montrea

Physiologic Responses to Dietary Sulfur Amino Acid Restriction in Mice Are Influenced by Atf4 Status and Biological Sex.

BACKGROUND: Dietary sulfur amino acid restriction (SAAR) improves body composition and metabolic health across several model organisms in part through induction of the integrated stress response (ISR). OBJECTIVE: We investigate the hypothesis that activating transcription factor 4 (ATF4) acts as a converging point in the ISR during SAAR. METHODS: Using liver-specific or global gene ablation strategies, in both female and male mice, we address the role of ATF4 during dietary SAAR. RESULTS: We show that ATF4 is dispensable in the chronic induction of the hepatokine fibroblast growth factor 21…

Plant-specific and conserved mechanisms of the polymerase-associated factor 1 complex in replication stress responses.

DNA replication stress threatens genome stability, but how plants respond to this challenge remains unclear. Here we show that the evolutionarily conserved polymerase-associated factor 1 complex (PAF1C) is required for the replication stress response in Arabidopsis. Plants lacking functional PAF1C are hypersensitive to replication stress-inducing agents. Mechanistically, we reveal a plant-specific pathway in which the stress-activated kinase WEE1 phosphorylates PAF1 to prevent its protein degradation, a regulatory mechanism absent in yeast. By contrast, we uncover a conserved pathway in which

In Defense of Homeostasis: Innate Immunity in Cardiac Injury and Repair.

The ability of the heart to compensate for and adapt to a superimposed environmental stress determines whether the heart will maintain preserved structure and function or decompensate and fail. In mammalian tissues, sterile tissue injury activates a suite of stress-activated programs that integrate cytoprotective stress responses that are tightly coupled with carefully regulated and precisely timed inflammatory signaling, thereby promoting tissue repair while simultaneously limiting collateral damage. Although the terminology for this phenomenon is still evolving, the recently described concep

Changes in gene expression in rat thymocytes identified by cDNA array support the occurrence of oxidative stress in early magnesium deficiency.

Magnesium deficiency in experimental animals leads to inflammation, exacerbated immune stress response and a decrease of specific immune response. It also results in a significant increase in free radical species and subsequent tissue injury. An accelerated thymus involution was observed in Mg-deficient rats in relation to enhanced apoptosis and enhanced susceptibility to oxidative stress. To examine the stress-inducing effects of low Mg status on thymocytes, cDNA arrays were used to evaluate changes in gene expression in weaning rats submitted to Mg deficiency of short duration (2 days). Wheat microbiome interactions under climate change: Mechanisms of abiotic stress tolerance and sustainable crop resilience.

Climate change-induced stresses, including drought, heat, and salinity, are increasingly constraining wheat productivity globally and pose a significant threat to global food security. Although beneficial rhizosphere microorganisms are known to enhance wheat stress tolerance, the mechanisms underlying wheat-microbiome interactions under climate-stress conditions remain poorly understood. This review highlights that wheat actively recruits and reshapes stress-resilient microbial communities, particularly members of Bacillus and Pseudomonas, which promote stress adaptation by regulating reactive

Overcoming methicillin resistance in Staphylococcus aureus via metal ion disruption of bacterial metabolism.

Methicillin-resistant Staphylococcus aureus (MRSA) is a significant public health threat due to both extensive antimicrobial resistance and immune evasion capabilities, necessitating alternative therapeutic strategies. Disruption of bacterial metal ion homeostasis, a process already leveraged by host nutritional immunity, represents a promising therapeutic approach. The synthetic ionophore PBT2 delivers zinc (Zn) directly into the bacterial cytosol, where it can dysregulate cellular processes and restore the efficacy of conventional antibiotics. Here, we use PBT2 and Zn (PZ) to study the cellu

The acute transcriptional responses to dietary methionine restriction are triggered by inhibition of ternary complex formation and linked to Erk1/2, mTOR, and ATF4.

The initial sensing of dietary methionine restriction (MR) occurs in the liver where it activates an integrated stress response (ISR) that quickly reduces methionine utilization. The ISR program is regulated in part by ATF4, but ATF4's prototypical upstream regulator, eIF2α, is not acutely activated by MR. Bioinformatic analysis of RNAseq and metabolomics data from liver samples harvested 3 h and 6 h after initiating MR shows that general translation is inhibited at the level of ternary complex formation by an acute 50% reduction of hepatic methionine that limits formation of initiator methion…

Targeting the kinase insert loop of PERK selectively modulates PERK signaling without systemic toxicity in mice.

Chronic activation of the unfolded protein response (UPR), notably the branch comprising the kinase PERK and the translation initiation factor eIF2α, is a pathological feature of many neurodegenerative diseases caused by protein misfolding. Partial reduction of UPR signaling at the level of phosphorylated eIF2α is neuroprotective and avoids the pancreatic toxicity caused by full inhibition of PERK kinase activity. However, other stress pathways besides the UPR converge on phosphorylated eIF2α in the integrated stress response (ISR), which is critical to normal cellular function. We explored wh…

Starvation and antimetabolic therapy promote cytokine release and recruitment of immune cells.

Cellular starvation is typically a consequence of tissue injury that disrupts the local blood supply but can also occur where cell populatio … Most cytokines were up-regulated in a manner dependent on NF-κB and the transcription factor of the integrated stress response ATF4, which bound directly to the IL-8 promoter. Furthermore, glutamine deprivation, as well as the antimetabolic drugs 2-deoxyglucose and metformin, also promoted the release of IL-6 and IL-8. Finally, some of the factors released from starved cell …

Similar patterns of activation in stress-sensitive brain regions during acute and chronic stress in adult male and female rats.

Exposure to stress is a risk factor for many psychiatric disorders, with females often exhibiting greater vulnerability than males. Chronic stress produces sex-specific changes in the structure and function of rat medial prefrontal cortex (mPFC). A possible explanation for these sex-dependent effects is that activation of the hypothalamic-pituitary-adrenal axis and of stress-sensitive brain regions during restraint is different in males and females. Here, we examined serum corticosterone concentrations and c-Fos expression in the mPFC, basolateral amygdala (BLA), and paraventricular nucleus (P

Glucose as fuel for HPA axis regulation: A narrative review on energy metabolism, stress reactivity, and early life adversity.

Stress physiology and energy metabolism are inherently linked. While the physiological stress response mobilizes energy, early experimental work by Clemens Kirschbaum and colleagues demonstrated that glucose availability in turn shapes the stress response: Fasting attenuates cortisol reactivity, whereas glucose administration restores or amplifies it. Since then, an active line of research on this topic evolved, yet the role of glucose availability in shaping stress regulation still remains incompletely understood. In this targeted, narrative review, we first synthesize evidence from the last

Physiological amplitude: a systems-level framework for adaptive capacity in aging and metabolism.

BACKGROUND: Aging is characterized by progressive loss of physiological complexity, inter-system coordination, and adaptive capacity. While mitochondrial dysfunction, metabolic inflexibility, impaired stress responses, and circadian dysregulation are well established, an integrative systems-level construct linking these processes remains lacking. OBJECTIVE: To introduce physiological amplitude as a unifying framework describing the bounded dynamic range of coordinated variability across interacting physiological systems. METHODS: We develop a conceptual, hypothesis-generating framework groun

Peptidoglycan Remodeling Enables Escherichia coli To Survive Severe Outer Membrane Assembly Defect.

Gram-negative bacteria have a tripartite cell envelope with the cytoplasmic membrane (CM), a stress-bearing peptidoglycan (PG) layer, and the asymmetric outer membrane (OM) containing lipopolysaccharide (LPS) in the outer leaflet. Cells must tightly coordinate the growth of their complex envelope to maintain cellular integrity and OM permeability barrier function. The biogenesis of PG and LPS relies on specialized macromolecular complexes that span the entire envelope. In this work, we show that Escherichia coli cells are capable of avoiding lysis when the transport of LPS to the OM is comprom

Use of Microalgae-Derived Astaxanthin to Improve Cytoprotective Capacity in the Ileum of Heat-Induced Oxidative Stressed Broilers.

The gastrointestinal tract has a pivotal role in nutrient absorption, immune function, and overall homeostasis. The ileum segment of the small intestine plays respective roles in nutrient breakdown and absorption. The purpose of this study was to investigate the impact of heat-induced oxidative stress and the potential mitigating effects of an astaxanthin antioxidant treatment on the ileum of broilers. By comparing the growth performance and gene expression profiles among three groups-thermal neutral, heat stress, and heat stress with astaxanthin-thermal neutral temperature conditions of 21-22
