Fetal exposure to maternal and paternal smoking and the risks of wheezing in preschool children: the Generation R Study.

BACKGROUND: Previous studies have suggested that fetal smoke exposure is associated with increased risks of wheezing during childhood. The underlying pathways are unknown. We examined the associations of parental smoking during pregnancy with wheezing in preschool children and whether these associations are explained by postnatal smoke exposure or small for gestational age at birth. METHODS: This study was embedded in a population-based prospective cohort study. Parental smoking was prospectively assessed by questionnaires. Wheezing was reported at 1 to 4 years. Small for gestational age at b

Prenatal exposure to maternal and paternal smoking on attention deficit hyperactivity disorders symptoms and diagnosis in offspring.

The study examined the effect of maternal and paternal smoking during pregnancy on the child's inattention and hyperactivity/impulsivity symptoms, and the risk for attention deficit hyperactivity disorder (ADHD) and oppositional defiant disorder (ODD). Generalized estimating equations, incorporating data from multiple informants (parents and teachers), was used to evaluate levels of ADHD as a function of parental smoking. The risk for ADHD, ODD, and comorbid ADHD and ODD was evaluated using polytomous logistic regression. We found that maternal, but not paternal, smoking was significantly asso

Umbilical cord monitoring of in utero drug exposure to buprenorphine and correlation with maternal dose and neonatal outcomes.

Buprenorphine is under investigation in the U.S. as pharmacotherapy for opioid-dependent pregnant women. Buprenorphine and metabolites were quantified in umbilical cord specimens from women receiving daily buprenorphine doses. Correlations between maternal buprenorphine dose, buprenorphine and metabolite umbilical cord concentrations, and neonatal outcomes were investigated, as well as the ability to identify heroin and cocaine relapse during pregnancy. Umbilical cord concentrations were compared to those of matched umbilical cord plasma and meconium. Buprenorphine metabolites were detected in

Gestational ethanol and nicotine exposure: effects on maternal behavior, oxytocin, and offspring ethanol intake in the rat.

Alcohol consumption and smoking during pregnancy is common, despite the known adverse effects of these drugs on fetal development. Though studies on the effects of each drug separately are published, little is known about the effect of concurrent use of alcohol and nicotine in humans or in preclinical models. In this report, we examined the impact of continuous gestational exposure to both ethanol via liquid diet and nicotine via an osmotic minipump on maternal behavior, offspring ethanol intake, and oxytocin levels in a rat model. Dams were tested for the onset of maternal behavior with litte

Genetic susceptibilities in the association between maternal exposure to tobacco smoke and the risk of nonsyndromic oral cleft.

Maternal tobacco consumption is considered as a risk factor for nonsyndromic oral clefts. However, this risk is moderate and may be modulate … risk in this population, but we observed statistically significant increased risks associated with maternal exposure to environmental tobacco smoke (ETS). No variant of any of the three developmental genes was significantly associated with oral cleft. The fetal CYP1A1*2C variant allele was associated with a statistically significant decreased risk, compared with t …

Maternal exposure to 2,3,7,8-tetrachlorodibenzo-p-dioxin and the body burden in offspring of long-evans rats.

OBJECTIVES: In utero and lactational exposure to 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) results in a wide variety of developmental effects in pups at doses much lower than those causing overt toxicity in adult animals. We investigated the relationship between tissue concentrations of TCDD in dams and fetuses and developmental effects on pups. MATERIALS AND METHODS: Pregnant Long-Evans rats were given TCDD at a single oral dose of 12.5, 50, 200, or 800 ng of TCDD or [(3)H]-TCDD/kg bw on gestation day (GD) 15. Dams were sacrificed on GD16 and GD21, and the tissue concentrations of TCDD were

Interactive effects of prenatal cocaine and nicotine exposure on maternal toxicity, postnatal development and behavior in the rat.

Two experiments were performed to investigate the interactive effects of prenatal coadministration of cocaine hydrochloride (C) and nicotine tartrate (N). Experiment I was designed to determine doses of C and N that could be coadministered without altering maternal gestational parameters and/or fetal viability. Exposure of Sprague-Dawley rats to combined high-dose C (20 mg/kg) and high-dose N (5.0 mg/kg) on gestation days 8-21 was not more toxic to dam or fetus that that of exposure to C alone. Experiment II investigated pregnancy outcome, postnatal development, and behavior of the offspring f

Maternal alcohol exposure before and during pregnancy: effect on development of neurons and glial cells in culture.

The effect of maternal alcohol exposure on nerve cell development was investigated in neurons and glial cells cultured from foetal rat brain. Neurons were grown for one week from two-week-old cortical brain cells and glial cells were cultured for four weeks from newborn cortical brain cells. Two types of maternal alcohol treatment were performed; either before and during pregnancy or only until the beginning of pregnancy. In both situations, we found a delayed nerve cell maturation assessed by microscopic observations and determination of enzymatic markers of nerve cell development (non-neuron

Cytologic problems in diagnosis of endocervical atypia in young females with and without maternal history of diethylstilbestrol exposure.

Sixteen cytologic endocervical atypias are reported with corroborative tissue analysis. Maternal diethylstilbestrol (DES) history was positive in three patients, one with clear cell adenocarcinoma, one with colposcopy and biopsy proven benign vaginal adenosis, and one with squamous carcinoma in situ of the cervix. Colposcopy was performed in two cases, one with adenosis and one with clear cell adenocarcinoma. In three patients, aged 19, 17, and 25, respectively, clear cell adenocarcinomas were found with unknown DES history and positive cytology. In five other cytologically positive cases, endo

Maternal exposure to ozone during pregnancy and risk of gestational diabetes in a Beijing pregnancy cohort study.

BACKGROUND: Gestational diabetes mellitus (GDM) exerts profound and lasting impacts on both maternal and offspring health, and the prevalence of GDM has been increasing. Several studies have demonstrated a close association between air pollution and GDM. Ozone, recognized as an emerging contaminant, has exhibited a rising trajectory in exposure levels over the past few years. While ozone exposure levels are rising, studies have reported inconsistent findings regarding its association with the risk of GDM. More importantly, identifying possible exposure windows is essential for informing target

Maternal exposure to the thyroperoxidase-inhibiting pesticide amitrole induces hypothyroidism and developmental neurotoxicity in the rat brain.

Thyroid hormone (TH) system-disrupting chemicals pose significant risks to human health and the environment, highlighting the urgent need for improved toxicological testing methods. A major concern is that environmental chemicals may induce developmental neurotoxicity by compromising TH signaling during critical life stages. In this study, we evaluated the thyroidal and neurotoxic effects of two compounds that interfere with TH signaling by inhibiting TH synthesis in the thyroid gland. Methimazole (MMI) is a pharmaceutical specifically designed to treat hyperthyroidism by inhibiting thyroperox

Resilience paths: maternal experiences in coping with syphilis exposure or congenital infection and the role of support networks.

BACKGROUND: Congenital syphilis is a significant public health problem and can lead to several unfavorable perinatal outcomes and severe sequelae in children. In Brazil, the number of cases of congenital and gestational syphilis continues to increase, highlighting the need for timely diagnosis, treatment, and prevention. A lack of adequate follow-up during prenatal care may result in unwanted outcomes for the neonate, such as prolonged hospitalization and impacts on neurodevelopment. Although many studies have focused on the clinical aspects of congenital syphilis, there is a gap in understand

Paternal and Maternal Exposures to Per- and Polyfluoroalkyl Substances and Child Behavioral Difficulties: A Parental Comparison Study.

Per- and polyfluoroalkyl substances (PFAS) are widespread environmental pollutants with documented developmental toxicity. Prior research of prenatal PFAS exposure and offspring neurodevelopment did not consider the possible influence from paternal exposure. Using the INUENDO cohort, we studied 334 father-mother-singleton triads enrolled from antenatal clinics in Greenland, Poland, and Ukraine. We measured five PFAS in parental serum samples collected around the 31 weeks of gestation. We assessed child behavioral difficulties at ages 5-9 years by the parent-rated Strength and Difficulties Ques

Maternal exposure to trace elements and perfluoroalkyl acids and their associations with alterations of the hypothalamic-pituitary-adrenal axis.

Environmental exposures to trace elements and perfluoroalkyl acids (PFAAs) could alter hypothalamic-pituitary-adrenal (HPA) function during pregnancy. Concentrations of trace elements measured in maternal red blood cells and PFAAs measured in maternal plasma, as well as maternal salivary cortisol were quantified from second trimester samples in 243 women from the Alberta Pregnancy Outcomes and Nutrition (APrON) cohort. Regression analyses examine associations between trace elements and PFAAs, and cortisol awakening response (CAR), daytime cortisol and diurnal cortisol slope. Bayesian kernel ma

Effect of maternal bisphenol exposure on adverse pregnancy and neonatal outcomes: The Japan Environment and Children's study.

Maternal exposure to bisphenols, including bisphenol A (BPA), reportedly results in adverse pregnancy and offspring health outcomes. However, large birth cohort studies on the risk of BPA exposure during pregnancy remain limited. Here, we examined bisphenols' effects on adverse pregnancy and neonatal outcomes using data from the Japan Environment and Children's Study, a nationwide birth cohort study. We selected 4523 women with pregnancies resulting in singleton live births. First-trimester urine concentrations of bisphenols A, F, S, and AF were determined via high-performance liquid chromatog…

The maternal exposure of digoxin and flecainide in relation to the safety and effectiveness in the treatment of non-hydropic fetal tachycardia.

BACKGROUND: Sustained fetal tachycardia requires transplacental anti-arrhythmic therapy. Little is known about the dose-concentration-effect correlation and safety in the mother, fetus, and newborn. OBJECTIVE: This study evaluates the relationship between maternal dose of digoxin and flecainide therapy for fetal tachycardia, maternal and umbilical cord concentrations, and side effects. METHODS: This was a retrospective case series. We included 28 pregnant women initially treated with digoxin monotherapy for fetal tachycardia between June 2007 and January 2023. The main end point was the corr

Exposure to Human Immunodeficiency Virus Is Associated With Altered Composition of Maternal Microchimeric T Cells in Infants.

Human immunodeficiency virus-exposed but uninfected infants (iHEU) display altered immunity and are at increased risk of infection. We previously reported that iHEU have decreased maternal microchimerism (MMc)-maternal cells transferred to the offspring in utero/during breastfeeding. We quantified MMc in T-cell subpopulations in iHEU and HIV-unexposed infants (iHU) to determine whether a selective deficiency in MMc contributes to altered cellular immunity. Across all infants, MMc levels were highest in CD8+ T cells; however, the level of CD8+ T-cell MMc was lower in iHEU versus iHU. In limited

Prenatal exposure to air pollution and maternal and fetal thyroid function: a systematic review of the epidemiological evidence.

BACKGROUND: Exposure to ambient air pollution is a top risk factor contributing to the global burden of disease. Pregnant persons and their developing fetuses are particularly susceptible to adverse health outcomes associated with air pollution exposures. During pregnancy, the thyroid plays a critical role in fetal development, producing thyroid hormones that are associated with brain development. Our objective is to systematically review recent literature that investigates how prenatal exposure to air pollution affects maternal and fetal thyroid function. METHODS: Following the Navigation Gu

A pilot study on in-utero exposure to psychotropic drugs: A comparison of pharmacological classes on neonatal and maternal outcomes.

OBJECTIVE: In this retrospective cohort study, we compared neonatal and maternal outcomes after exposure of different psychopharmacological classes of drugs. Both psychiatric diseases and pharmacological treatment of these are associated with lower birth weights, lower APGAR scores, and NICU admission. Therefore, we tried to rule out the role of psychotropics as if no differences were found between pharmacological classes, the lower birthweights might not be attributable to these. METHOD: We divided our groups in exposed to atypical antipsychotic drugs, Selective Serotonin Reuptake Inhibitors
