PKMζ-PKCι/λ double-knockout demonstrates atypical PKC is crucial for the persistence of hippocampal LTP and spatial memory.

PKMζ is a persistently active atypical PKC (aPKC) isoform thought to maintain late-phase long-term potentiation (late-LTP) and long-term memory. PKMζ-knockout mice, however, still exhibit hippocampal LTP and spatial memory while lacking neocortical LTP, questioning whether this kinase is fundamental to enduring synaptic potentiation and memory. Tsokas et al. (2016) suggested that the other aPKC, PKCι/λ, may compensate for PKMζ during maintenance in the hippocampus of PKMζ-null mice. In wild-type mice, PKCι/λ drives early-LTP and short-term memory, whereas in PKCι/λ-knockout mice, PKMζ compensa

Revisiting and Revising Memory Consolidation: Personal Reflections on the Research Legacy of Ivan Izquierdo.

Two important themes in Ivan Izquierdo's research each offered both answers and questions about the topic of memory formation and maintenance. The first theme provided evidence supporting the view that short- and long-term memory were distinct processes and could be selectively modulated by several treatments, with some affecting only short-term, others only affecting long-term memory, and still others affecting both. Over many years, Izquierdo's laboratory documented molecular responses across time after training obtaining results that showed differences as well as similarities in the biochem

Novel Roles for the Insulin-Regulated Glucose Transporter-4 in Hippocampally Dependent Memory.

UNLABELLED: The insulin-regulated glucose transporter-4 (GluT4) is critical for insulin- and contractile-mediated glucose uptake in skeletal muscle. GluT4 is also expressed in some hippocampal neurons, but its functional role in the brain is unclear. Several established molecular modulators of memory processing regulate hippocampal GluT4 trafficking and hippocampal memory formation is limited by both glucose metabolism and insulin signaling. Therefore, we hypothesized that hippocampal GluT4 might be involved in memory processes. Here, we show that, in male rats, hippocampal GluT4 translocates

Long-lasting improvement of memory after targeted cognitive remediation in temporal epilepsy: A randomized control study.

BACKGROUND: Memory deficits are a major burden in temporal lobe epilepsy (TLE), contributing to the impaired quality of life of patients. Short- and long-term efficacy of memory remediation in TLE is still debated. The aim of our study was to assess the short- and long-term efficacy of a cognitive remediation program, COMETE (COgnitive Rehabilitation of MEmory in Temporal Epilepsy), targeting specifically episodic memory in patients with TLE. METHODS: In this prospective randomized single-blinded trial, patients from different French epilepsy centers were randomly assigned in a 2:4 ratio, via

Memory consolidation drives the enhancement of remote cocaine memory via prefrontal circuit.

Remote memory usually decreases over time, whereas remote drug-cue associated memory exhibits enhancement, increasing the risk of relapse during abstinence. Memory system consolidation is a prerequisite for remote memory formation, but neurobiological underpinnings of the role of consolidation in the enhancement of remote drug memory are unclear. Here, we found that remote cocaine-cue associated memory was enhanced in rats that underwent self-administration training, together with a progressive increase in the response of prelimbic cortex (PrL) CaMKII neurons to cues. System consolidation was

Autobiographical memory in mild cognitive impairment and Alzheimer's disease: a comparison between the Levine and Kopelman interview methodologies.

Previous studies have produced inconsistent results concerning the two components of autobiographical memory--personal semantic memory and episodic memory. Results in subjects with mild cognitive impairment (MCI) and dementia of Alzheimer's type (DAT) have varied concerning the existence of a temporal gradient in retrograde amnesia. These results have important theoretical implications regarding multiple trace theory versus standard consolidation models of long-term memory (LTM). We investigated whether this variability arises from differences in the methods used in assessing autobiographical

Memory dysfunction in panic disorder: an investigation of the role of chronic benzodiazepine use.

BACKGROUND: Studies of the neurocognitive effects of long-term benzodiazepine use have been confounded by the presence of neurocognitive deficits characterizing the clinical conditions for which these medications are taken. Similarly, studies of the neurocognitive effects of anxiety disorders have been confounded by the inclusion of chronically benzodiazepine-medicated patients. This study was designed to tease apart the potentially confounding effects of long-term benzodiazepine use and panic disorder (PD) on memory and visuoconstructive abilities. METHODS: Twenty chronically benzodiazepine-

ChABC infusions into medial prefrontal cortex, but not posterior parietal cortex, improve the performance of rats tested on a novel, challenging delay in the touchscreen TUNL task.

Perineuronal nets (PNNs) are specialized extracellular matrix structures that surround subsets of neurons throughout the central nervous system (CNS). They are made up of chondroitin sulfate proteoglycans (CSPGs), hyaluronan, tenascin-R, and many other link proteins that together make up their rigid and lattice-like structure. Modulation of PNNs can alter synaptic plasticity and thereby affect learning, memory, and cognition. In the present study, we degraded PNNs in the medial prefrontal (mPFC) and posterior parietal (PPC) cortices of Long-Evans rats using the enzyme chondroitinase ABC (ChABC

Netrin-1 improves the amyloid-β-mediated suppression of memory and synaptic plasticity.

The aim of this study was to investigate the effects of netrin-1 (NT-1) on amyloid-beta (Aβ)-induced impairments in learning-memory and synaptic plasticity. The NT-1 or its vehicle was administered four times into the Aβ+NT or Aβ+V groups, respectively. The Aβ+SNT group received a single dose of NT-1. The Aβ+HNT group received heat-inactivated NT-1. For the learning-memory and synaptic plasticity assessment, field potentials recording and behavioral experiment were used. Bilateral injection of Aβ1-42 inhibits induction of long-term potentiation (LTP) and decreased memory performance in all the

Memory performance in a sample of very low birth weight adolescents.

Prematurely born participants with very low birth weight (VLBW) are at high risk of brain injury in the perinatal period and of later cognitive impairment. Studies of long-term memory sequelae in VLBW participants are scarce and focus on verbal and visual memory assessed by standard clinical memory tests. There is even less research into everyday memory, and the results obtained are contradictory. This study explores long-term memory deficits in VLBW adolescents using 2 standard clinical memory tests and 1 everyday memory test. Results show impairment only in everyday memory. These memory defi

Effects of antipsychotic drugs on memory functions of schizophrenic patients.

In this research we investigated the effects of 4 antipsychotic drugs with different anticholinergic components on different memory functions of schizophrenic patients. Drugs were administered in cumulative doses and memory was tested 90 min after each drug was administered. The results show that chlorpromazine and thioridazine impaired short-term verbal memory after 6 h of sequential administration. Trifluoperazine and haloperidol improved short-term verbal memory from the third to the fifth administration. Immediate memory, long-term memory and visual short-term memory were not impaired by a…

[Memory training: an important constituent of milieu therapy in senile dementia].

At the Memory Clinic in Basel we provide long-term memory training as part of our "milieu therapy". Mildly demented elderly out-patients attend weekly group sessions of 60 min for a special semistructured memory training. The goal is to relearn and practice strategies which help to preserve or extend the phase of autonomy and thus life quality and self-esteem. In several studies we tested the efficiency of the training on mood, global mental status, short-term memory, cognitive flexibility, and quality of life by comparing the performance of groups of patients attending the memory training to

Freezing of Gait in Parkinson's Disease: A Scoping Review on the Path Towards Real-Time Therapies.

BACKGROUND: Freezing of gait (FoG) is a common symptom of Parkinson's disease, especially in its later stages of progression. Characterized by involuntary stopping during normal gait patterns, FoG greatly increases fall risk, reducing quality of life. Given the complex presentation and etiology of FoG, current treatments have proven ineffective in managing episodes. In recent years, machine learning algorithms have been leveraged to derive actionable clinical insights from biomedical datasets. As a manifestation of neuromechanical dysfunction, impending FoG episodes may be characterized throug

Facilitating the spread prediction of public health emergencies based on spatio-temporal neural network.

To overcome the shortcomings of traditional models in capturing the spatiotemporal dynamics of epidemic spread, this study introduces an innovative model, hybrid spatio-temporal neural network architecture (MI-GCN-LSTM), which integrates mutual information (MI), long short term memory (LSTM), and graph convolutional network (GCN) to collaboratively predict epidemic trends across multiple regions during public health emergencies. First, an adjacency matrix is constructed using mutual information theory to quantify the influence of complex behaviors such as interregional population movement, thu

Cattle lameness detection using depth image and deep learning.

Lameness in cattle is a significant welfare and economic concern. To address this, we developed an end-to-end deep learning framework for 24/7 lameness monitoring using top-down depth images of cattle. The framework integrates three key stages: instance segmentation for detection, a custom multi-object tracking algorithm for identity preservation, and a spatio-temporal model for classification. We compared multiple instance segmentation models (Mask R-CNN, YOLOv8m-seg, YOLOv11m-seg) and evaluated three proposed tracking algorithms version1, 2 and 3 (PTAV1, PTAV2, and PTAV3). For classification

Cardiac activity monitoring for the heart rate classification with enhanced BiLSTM model.

Cardiac activity monitoring is important for assessing cardiovascular status and supporting computational analysis of heart-rate pattern variations from wearable PPG signals. The development of wearable devices and public datasets has made continuous and noninvasive monitoring more viable. However, there are still problems that may impair the effectiveness of the technique such as motion artifact, signal noises, changes in the physiological states, and differences in sensor quality. Moreover, it is hard to create algorithms that would be highly accurate in the real-life scenario and would be a

Deep Learning-Based Sleep Apnea Classification with Hybrid 1D-CNN-LSTM and 1D-CNN-GRU Models.

Sleep apnea is a common sleep disorder characterized by recurrent interruptions in breathing during sleep, leading to severe health complications such as cardiovascular diseases and reduced quality of life. Early and accurate diagnosis is essential for effectively managing and preventing these risks. This study proposes two hybrid deep learning architectures, 1D-CNN-LSTM and 1D-CNN-GRU, for sleep apnea classification using ECG signals from the PhysioNet Apnea-ECG database. The models leverage R-R intervals and R-wave amplitudes as primary input features, where 1D-CNN extracts spatial features

Propofol improves learning and memory in post-traumatic stress disorder (PTSD) mice via recovering hippocampus synaptic plasticity.

AIMS: Propofol, the most commonly used intravenous anesthetic, is known for its protective effect in various human and animal disease models such as post-traumatic stress disease (PTSD). However, it still needs efforts to clarify the effect of propofol on fear memory extinction and the relevant mechanisms. METHODS: Fear memory extinction was examined in PTSD mice model. Thirty-six mice were randomly divided into three groups: a shock + propofol group (sh + Pro), shock + normal saline group (sh + NS), and sham group. The mice were treated with propofol (150 mg/kg) or normal saline (of the same
