Behavioral Experimental Paradigms for the Evaluation of Drug's Influence on Cognitive Functions: Interpretation of Associative, Spatial/Nonspatial and Working Memory.

BACKGROUND: Currently, a large number of people throughout the world are affected by neurodegenerative disorders such as Alzheimer's disease, Parkinson's disease and Huntington's disease which appear with a lapse in recall, attention and altered cognitive functions. Learning and memory, the fundamental indices defining cognitive functions, are the complex psychological processes governing acquisition, consolidation, and retrieval of stored information. These processes are synchronized by the coordination of various parts of the brain including hippocampus, striatum and amygdala. OBJECTIVE: Th

[Mechanisms of intrinsically photosensitive retinal ganglion cells regulating learning and memory: a review].

Intrinsically photosensitive retinal ganglion cells (ipRGCs) regulate multiple memory-related brain regions via subtype-specific projections … 40 Hz rhythmic blue-light flicker can strengthen visual and auditory working memory and facilitate memory consolidation by activating ipRGC-related pathways, accompanied by prefrontal cortex activation and enhanced hippocampal-thalamic functional connectivity, which reveals promising non-pharmacological potential for memory modulation. Beyond healthy subjects, blue-l …

Recovering object-location memories after sleep deprivation-induced amnesia.

It is well established that sleep deprivation after learning impairs hippocampal memory processes and can cause amnesia. It is unknown, however, whether sleep deprivation leads to the loss of information or merely the suboptimal storage of information that is difficult to retrieve. Here, we show that hippocampal object-location memories formed under sleep deprivation conditions can be successfully retrieved multiple days following training, using optogenetic dentate gyrus (DG) memory engram activation or treatment with the clinically approved phosphodiesterase 4 (PDE4) inhibitor roflumilast. Glucocorticoid receptor antagonism disrupts the reconsolidation of social reward-related memories in rats.

Reconsolidation is the process whereby consolidated memories are destabilized upon retrieval and restabilized to persist for later use. Although the neurobiology of the reconsolidation of both appetitive and aversive memories has been intensively investigated, reconsolidation of memories of physiologically relevant social rewards has received little attention. Social play, the most characteristic social behaviour displayed by young mammals, is highly rewarding, illustrated by the fact that it can induce conditioned place preference (CPP). Here, we investigated the role of signalling mechanisms

Gut-derived memory T cells: key mediators of neuroinflammation via the gut-brain axis.

The gut-brain axis constitutes a bidirectional communication network that links the gut microbiota with the central nervous system (CNS) and plays a pivotal role in regulating neuroinflammation. Memory T cells, as central orchestrators of adaptive immunity, undergo differentiation, trafficking, and functional reprogramming within the gut microenvironment. This process establishes a novel framework for understanding the pathogenesis of neuroinflammatory disorders. In this review, we summarize the physiological crosstalk within the gut-brain axis and outline the functional characteristics of mem

The relationship between sleep quality and cognitive performance in a sample of Colombian adults.

Sleep plays a vital role in cognitive regulation, memory consolidation and brain health. In Colombia, sleep patterns may be influenced by cultural and socioeconomic factors such as co-sleeping, occupational stress and limited healthcare access, which can exacerbate sleep-related disorders like sleep apnoea. This study examined the relationship between objectively measured sleep quality and cognitive performance in Colombian adults with sleep apnoea. We hypothesised that poorer sleep quality, which is characterised by lower efficiency and more apnoea (hypopnoea events), would be associated with…

The impact of the BDNF Val66Met genotype on intrusive memories following trauma exposure and in PTSD is moderated by sex and timing of trauma exposure.

Intrusive memories are a key symptom of Post-Traumatic Stress Disorder (PTSD). Brain Derived Neurotrophic Factor (BDNF) has been proposed as a possible mechanism influencing intrusive memories in PTSD given its role in synaptic plasticity and memory consolidation. The BDNF Val66Met polymorphism has been linked PTSD susceptibility and episodic memory disturbances however previous research outcomes have been variable, potentially due to a failure to control for important confounds such as sex, ethnicity, BMI, developmental stage and extent of previous trauma experiences. This study explored the …

Accelerated long-term forgetting in patients with acquired brain injury.

OBJECTIVE: Recent research suggests that patients with neurological disorders without overt seizures may also experience accelerated long-term forgetting (ALF). This term describes unimpaired learning and memory performance after standard retention intervals, but an excessive rate of forgetting over delays of days or weeks. The objective of this retrospective study was to investigate ALF in patients with an acquired brain injury (ABI) and to associate memory performance with executive functions. METHODS: Verbal memory performance (short-term recall, 30-min recall, 1-week recall) was assessed

Silent Synapses in Cocaine-Associated Memory and Beyond.

Glutamatergic synapses are key cellular sites where cocaine experience creates memory traces that subsequently promote cocaine craving and seeking. In addition to making across-the-board synaptic adaptations, cocaine experience also generates a discrete population of new synapses that selectively encode cocaine memories. These new synapses are glutamatergic synapses that lack functionally stable AMPARs, often referred to as AMPAR-silent synapses or, simply, silent synapses. They are generated de novo in the NAc by cocaine experience. After drug withdrawal, some of these synapses mature by recr

The role of pregnenolone sulphate in spatial orientation-acquisition and retention: an interplay between cognitive potentiation and mood regulation.

Neurosteroids can alter neuronal excitability interacting with specific neurotransmitter receptors, thus affecting several functions such as cognition and emotionality. In this study, we investigated, in adult male rats, the effects of the acute administration of pregnenolone-sulfate (PREGS) (10 mg/Kg, s.c.) on cognitive processes using the Can test, a non aversive spatial/visual task which allows the assessment of spatial information-acquisition during the baseline training, and of memory retention in the longitudinal study. Furthermore, on the basis of PREGS pharmacological profile, the modu

Hippocampus and contextual fear conditioning: recent controversies and advances.

Dorsal hippocampal (DH) lesions produce a severe deficit in recently, but not remotely, acquired contextual fear without impairing memory of discrete training stimuli, i.e., DH lesions produce an anterograde and time-limited retrograde amnesia specific to contextual memory. These data are consistent with the standard model which posits temporary involvement of the hippocampus in recent memory maintenance. However, three recent controversies apparently weaken the case for a selective mnemonic role for the hippocampus in contextual fear. First, although retrograde amnesia (from posttraining lesi

Treatment of trauma with lithium to forestall the development of posttraumatic stress disorder by pharmacological induction of a mild transient amnesia.

Posttraumatic stress disorder (PTSD) is a severe anxiety disorder that develops after exposure to trauma. Symptoms include persistent reexpe … ted by endogenous stress hormones released with trauma and after trauma. The molecular processes of memory consolidation in neurons are mediated by intracellular signaling pathways. One secondary messenger signaling pathway with a putative role in long-term potentiation (LTP) is the inositol 1,4,5-trisphosphate (IP3) and diacylglycerol (DAG) secondary messenger system …

Posterior Hippocampal Spindle Ripples Co-occur with Neocortical Theta Bursts and Downstates-Upstates, and Phase-Lock with Parietal Spindles during NREM Sleep in Humans.

Human anterior and posterior hippocampus (aHC, pHC) differ in connectivity and behavioral correlates. Here we report physiological differences in humans of both sexes. During NREM sleep, the human hippocampus generates sharpwave ripples (SWRs) similar to those which in rodents mark memory replay. We show that while pHC generates SWRs, it also generates approximately as many spindle ripples (SSR: ripples phase-locked to local spindles). In contrast, SSRs are rare in aHC. Like SWRs, SSRs often co-occur with neocortical theta bursts (TBs), downstates (DSs), sleep spindles (SSs), and upstates (USs

Glucocorticoid enhancement of dorsolateral striatum-dependent habit memory requires concurrent noradrenergic activity.

Previous findings indicate that post-training administration of glucocorticoid stress hormones can interact with the noradrenergic system to enhance consolidation of hippocampus- or amygdala-dependent cognitive/emotional memory. The present experiments were designed to extend these findings by examining the potential interaction of glucocorticoid and noradrenergic mechanisms in enhancement of dorsolateral striatum (DLS)-dependent habit memory. In experiment 1, different groups of adult male Long-Evans rats received training in two DLS-dependent memory tasks. In a cued water maze task, rats wer

Mineralocorticoid and glucocorticoid receptors and their differential effects on memory performance in people with Addison's disease.

By using a repeated measures design with nine people with Addison's disease (mean age, 37.9 years), the effects following activation of the MRs only, GRs only, and a combination of MRs/GRs, on working memory and the episodic and seman …

D-cycloserine administered directly to infralimbic medial prefrontal cortex enhances extinction memory in sucrose-seeking animals.

Neural substrates of propranolol-induced impairments in the reconsolidation of nicotine-associated memories in smokers.

The majority of smokers relapse even after successfully quitting because of the craving to smoking after unexpectedly re-exposed to smoking-related cues. This conditioned craving is mediated by reward memories that are frequently experienced and stubbornly resistant to treatment. Reconsolidation theory posits that well-consolidated memories are destabilized after retrieval, and this process renders memories labile and vulnerable to amnestic intervention. This study tests the retrieval reconsolidation procedure to decrease nicotine craving among people who smoke. In this study, 52 male smokers

Negative allosteric modulation of mGlu7 disrupts fear memory reconsolidation and glutamatergic signaling in rat and human brain tissue.

Anxiety- and stress-related disorders are amongst the most frequent neurological disorders, and efficient treatment is lacking. Metabotropic glutamate receptors (mGlu) have emerged as promising targets for intervention. Of particular interest is mGlu7, because of its expression in the lateral amygdala (LA), a region critical for fear learning. In the present study we examined the effects of the highly specific negative allosteric modulator of mGlu7 (ADX71743) on fear memory reconsolidation. Our investigation unveils that infusion in rats of ADX71743 in the LA or subcutaneously disrupts the rec
