The effects of morphine, cyclazocine and naloxone on penicillin- and strychnine-induced epileptic foci were studied in rabbits. The intracortical injection of penicillin (75, 150 and 300 units) elicited isolated spikes followed by repeated ictal events. Thirty healthy women in active labour received an intrathecal injection of morphine 0.5 mg (n = 12) or 1 mg (n = 18) in 7.5% dextrose. Both doses provided excellent analgesia for labour, 93% of patients obtaining at least 50% pain relief. Analgesia began 15-60 min after injection and did not decrease until 6-8 h after injection. Morphine was administered intravenously to normal and to Brattleboro (genetically deficient in antidiuretic hormone) rats. The injection caused a significant decrease in mean arterial pressure and urine flow in all animals, including those homozygous for diabetes insipidus. It is suggested that the antidiuretic effect of morphine may occur independently of ADH release.

Large doses of morphine sulfate have been reported to cause myocardial lactate production and reduction in coronary blood flow in animals. In the mouse hot-plate test (50 degrees C), muscimol produced analgesia which was blocked by bicuculline but not by picrotoxin. We wanted to determine if naloxone antagonizes the direct effects of morphine transferred from the mother to her fetus and if this antagonism resembles fetal narcotic withdrawal. After continuous infusions of morphine into pregnant ewes, injections of naloxone into their fetuses produced increases in fetal arterial blood presure; decreases in fetal heart rate, arterial blood pH, and PO2; and defecation of meconium.
