Stage- and sex-specific associations of prenatal individual and mixed metals exposure with adiposity peak and adiposity rebound among children: a prospective birth cohort study.

BACKGROUND: Adiposity peak (AP) and adiposity rebound (AR) are key indicators of early childhood growth, which are closely related to obesity, premature puberty, and metabolic abnormalities. Epidemiological studies have shown that prenatal metal exposure is linked to obesity in children. However, studies concerning the association of metals during pregnancy with AP and AR in children are scarce. METHODS: We measured the concentration of 17 serum metal elements including aluminum (Al), vanadium, manganese (Mn), iron, cobalt (Co), nickel, copper, zinc, selenium, cadmium, barium (Ba), thallium (

Gestational organophosphate esters (OPEs) exposure in association with placental DNA methylation levels of peroxisome proliferator-activated receptors (PPARs) signaling pathway-related genes.

BACKGROUND: Organophosphate esters (OPEs) exposure could affect offspring health. However, the underlying mechanisms are not well documented. OBJECTIVES: Based on a birth cohort study, we aimed to investigate the associations among gestational OPEs exposure, placental DNA methylation levels of peroxisome proliferator-activated receptor (PPAR) signaling pathway-related genes, and fetal growth. METHODS: We measured the concentrations of eight OPE metabolites in maternal urine samples and neonatal anthropometric measurements in 733 mother-child pairs. In 327 placental samples, we assessed the D

Effect of in utero exposure to antiepileptic drugs on cortical networks and neurophysiological outcomes at 6 years.

OBJECTIVE: The human brain undergoes an activity-dependent organization during late gestation, making it very sensitive to all effects on the spontaneous neuronal activity. Pregnant mothers with epilepsy are treated with antiepileptic drugs (AEDs) that may reach the fetus and cause altered cortical network activity after birth. However, it is not known whether these functional effects of intrauterine AED exposure persist later in childhood. METHODS: We studied cortical activity networks computed from electroencephalographic recordings during sleep of 25, 6-year-old children with in utero expo

Pregnancy outcomes among female childhood, adolescent, and young adult cancer survivors assessed using internet-based nationwide questionnaire surveys in Japan.

OBJECTIVE: Female cancer survivors planning to become pregnant are concerned about the impact of cancer treatment on their ability to maintain normal pregnancy and the negative impact on their offspring. However, studies on the pregnancy outcomes of cancer survivors in Japan are limited. Therefore, this study aimed to investigate the pregnancy outcomes of female cancer survivors by comparing them with women without a history of malignant tumors in Japan. METHODS: This cross-sectional study included 3308 subjects, based on an internet-based questionnaire (self-reported) survey conducted in Jap

Trace element profiles in pregnant women's sera and umbilical cord sera and influencing factors: Repeated measurements.

In utero exposure to toxic heavy metals and deficient or excessive essential trace elements during pregnancy may have adverse effects on pregnant women and their offsprings, which are of great concern. The objective of the present study was to characterize serum concentrations of multiple trace elements at multiple time points during pregnancy in Chinese women. Three thousand four hundred and sixteen pregnant women in total were included from MABC (Ma'anshan Birth Cohort) study. Fasting sera in the morning and questionnaires were obtained at three separate follow-up visits. Nineteen trace elem

Time-Varying Exposure to Element Mixtures and Children's Cognition at 5 Years of Age: Findings from the New Hampshire Birth Cohort Study.

BACKGROUND: Exposure to metals and metalloids (hereafter referred to as 'metals') during gestation and early childhood may affect children's neurodevelopment. However, few studies have simultaneously evaluated the impact of exposure to both essential and nonessential metals across specific windows of exposure on children's neurodevelopmental outcomes. OBJECTIVE: To investigate whether levels of metals during gestation and the early postnatal period, individually and as a mixture, are related to child cognition at 5 years of age among participants in the New Hampshire Birth Cohort Study (NHBCS)

Maternal sleep disturbance during pregnancy and child intelligence quotient: A metabolome-wide association study in the Shanghai Birth Cohort.

BACKGROUND: The impact of maternal sleep disturbances during pregnancy on long-term neurodevelopment and the role of metabolites in this process are not well understood. In a prospective cohort study, we aimed to investigate the associations between maternal sleep disturbances during each trimester and child intelligence quotient (IQ) at the age of 4 years and to identify metabolites that might mediate these relationships. METHODS: This study included 1,870 mother-child pairs from the Shanghai Birth Cohort (SBC). Maternal sleep quality was assessed using the Pittsburgh sleep quality index (PS

Multiple pregnancies obtained by testicular spermatid injection in combination with intracytoplasmic sperm injection.

Recent studies have shown that the injection of spermatid cells into the human oocyte can result in normal fertilization, embryo development and even delivery of live, healthy offspring. In our study, 23 azoospermic cases with severe spermatogenetic defects in their testicular biopsy are presented. The serum follicle stimulating hormone (FSH) concentrations and histopathological results of these males have been documented and compared in terms of fertilization and embryo development. The mean FSH value of the azoospermic males was 15.8 +/- 2.3 mIU/l, ranging from 1.6 to 39 mIU/l. Elongated spe

Fecal progestin concentrations as an indicator of reproductive success in American Mink.

Efforts to increase mink reproductive success (live births and litter sizes) can be partly assessed by measurement of blood progesterone levels. However, the stress of blood sampling increases the incidence of failed matings, aborted fetuses and death of the dam. We have therefore non-invasively measured fecal progesterone metabolite (progestin) concentrations during the reproductive cycle of mink. We tested the hypothesis that fecal progestin concentrations during the window of implantation (late March-early April) will, (1): be higher for whelping than non-whelping mink, and (2): be higher f

The clinical relevance of sex selection techniques.

Clinical and laboratory attempts to alter the sex ratio require more complete and thorough study. Improved identification of Y-bearing sperm through chromosome evaluation rather than by F-body identification is critical to provide a more precise definition. The tentative conclusions stated below are based on an assessment of literature from which it is generally difficult to draw conclusions: 1. The timing of intercourse in relation to ovulation and subsequent fertilization appears to influence the sex ratio. More females are conceived when coitus occurs relatively close to ovulation, and more

Profiles and maternal-to-fetal transfer of per- and polyfluoroalkyl substances in paired maternal-infant biological matrices: A birth cohort study in Chengdu, China.

Per- and polyfluoroalkyl substances (PFAS) have been associated with adverse maternal and developmental health outcomes. However, studies on PFAS profile in multiple matched mother-infant samples remain limited. In this study, we aimed to characterize exposure levels and maternal-to-fetal transfer patterns of both conventional and emerging PFAS in a birth cohort from Chengdu, China. Concentrations of 24 PFAS were quantitatively determined in 47 pairs of maternal and infant samples, including maternal serum, cord serum, amniotic fluid, maternal urine, infant urine, and breast milk, using high-p

Maternal Glucose Metabolism and Emotional and Behavioral Problems in Offspring: Modification by Erythrocyte Polyunsaturated Fatty Acids.

Background: Gestational diabetes mellitus (GDM) adversely affects offspring neurobehavioral outcomes, yet evidence regarding continuous markers of maternal glucose metabolism remains limited. Although polyunsaturated fatty acids (PUFAs) have been shown to affect associations between glucose metabolism and respiratory outcomes, their effects on children's emotional and behavioral problems remain unclear. This study investigated the association between maternal glucose metabolism and emotional and behavioral problems in children and the potential modifying effect of maternal erythrocyte PUFAs. M

Associations of antenatal micronutrient supplementation with adolescent blood pressure: evidence from a 14-year follow-up study of a randomized controlled trial.

BACKGROUND: This study directly examined the effects of antenatal micronutrient supplementation on adolescent blood pressure (BP). METHODS: This study involved adolescents from two rural counties in western China. Their mothers had previously participated in a cluster-randomized trial of antenatal micronutrient supplementation. All pregnant women were randomized to take a daily capsule of folic acid (FA) as control, folic acid plus iron (IFA), or multiple micronutrients (MMNs) until delivery. Adolescent BP was assessed using a validated electronic sphygmomanometer and converted into percentil

Gestational diabetes and childhood lung function at age 8 to 9 years in a diverse US cohort.

BACKGROUND: Gestational diabetes mellitus (GDM) is associated with systemic processes that may disrupt fetal development and contribute to suboptimal lung function in offspring. Distinct from respiratory conditions such as asthma, lung function is a broad indicator of respiratory health and is predictive of long-term health outcomes. GDM has previously been associated with childhood asthma, but studies on childhood lung function are sparse. OBJECTIVE: To evaluate the association between GDM and respiratory outcomes in children, including lung function, wheeze, and asthma. METHODS: We studied

Distinguishing syndromic and nonsyndromic cleft palate through analysis of protein-altering de novo variants in 816 trios.

De novo variants (DNs) are sporadically occurring variants that most commonly arise in the germline and are present in offspring but absent in both parents. As they are not under selective pressure, they may be enriched for disease-causing alleles and have been implicated in multiple rare genetic disorders. Cleft palate (CP) is a common craniofacial congenital anomaly occurring in ~1 in 1700 live births. Genome-wide association studies for CP have found fewer than a dozen loci, while exome and targeted sequencing studies in family-based and case-control cohorts often lack statistical power to

Timed fetal inflammation and postnatal hypoxia cause cortical white matter injury, interneuron imbalances, and behavioral deficits in a double-hit rat model of encephalopathy of prematurity.

Extreme preterm birth-associated adversities are a major risk factor for aberrant brain development, known as encephalopathy of prematurity (EoP), which can lead to long-term neurodevelopmental impairments. Although progress in clinical care for preterm infants has markedly improved perinatal outcomes, there are currently no curative treatment options available to combat EoP. EoP has a multifactorial etiology, including but not limited to pre- or postnatal immune activation and oxygen fluctuations. Elucidating the underlying mechanisms of EoP and determining the efficacy of potential therapies
