Specimens of DMBA-induced mammary tumour, DMBA-induced subcutaneous sarcomas, and virus induced sarcomas in the rat were incubated for 24-72 hours as organ cultures. The effect of various concentrations of testosterone, progesterone, and 17-beta-oestradiol on cell survival and multiplication was measured as changes in incorporation of H3-TdR into DNA. All three types of tumour reacted to the hormones. Statistical analysis of data demonstrated possible influence of factors other than hormone treatment. The specificity of hormone sensitivity assays in vitro is questioned.

Tuberous sclerosis (TSC) is an autosomal dominant genetic disorder in which benign hamartomas develop in multiple organs, caused by mutations in either TSC1 or TSC2. We developed a murine model of Tsc2 disease using a gene targeting approach. Tsc2-null embryos die at embryonic days 9.5-12.5 from hepatic hypoplasia. A previous report was made on the carcinogenicity of 1-(2-hydroxyethyl)-1-nitrosourea [(HENU) CAS: 13743-07-2] in rats. Because the cyclic nitrosocarbamate 3-nitroso-2-oxazolidinone (NOZ) is readily produced during the synthesis of HENU and can be confused with HENU, HENU was retested and NOZ was tested for carcinogenicity. Analysis of DNA histograms obtained from rat renal cancer cells stained with propidium iodide and submitted to flow cytometry revealed a tumor cell population with prominent 2C and 4C peaks and with usually less than 10% each of S-phase or hyper-4C cells. It has been shown in animal experiments that injections of large amounts of cadmium cause sarcoma at injection sites or testicular damage and eventually testicular tumors. Long-term exposure with small doses of cadmium has not caused testicular or prostatic tumors in experimental animals.
