Effects of pentobarbital anesthesia on nociceptive processing in the medial and lateral pain pathways in rats.

OBJECTIVE: To investigate the effects of pentobarbital anesthesia on nociceptive processing in the medial and lateral pain pathways. METHODS: Laser stimulation was employed to evoke nociceptive responses in rats under awake or anesthetic conditions. Pain-related neuronal activities were simultaneously recorded from the primary somatosensory cortex (SI), ventral posterolateral thalamus (VPL), anterior cingulate cortex (ACC), and medial dorsal thalamus (MD) with 4 eight-wire microelectrode arrays. RESULTS: Compared with the awake state, pentobarbital anesthesia significantly suppressed the neu

Altered dorsal premotor-motor interhemispheric pathway activity in focal arm dystonia.

Given the possible role of dorsal premotor cortex (PMd) in the pathophysiology of dystonia, we used transcranial magnetic stimulation (TMS) methods to study PMd and PMd-primary motor cortex (M1) interactions in patients with focal arm dystonia. Here, we tested the connectivity between left PMd and right M1 as well as the intracortical excitability of PMd in 11 right-handed patients with focal arm/hand dystonia and nine age-matched healthy controls. The results showed that excitability of the inhibitory connection between PMd and M1 was reduced in patients, but there was no significant differen

Coherence and recurrency: maintenance, control and integration in working memory.

Working memory (WM), including a 'central executive', is used to guide behavior by internal goals or intentions. We suggest that WM is best described as a set of three interdependent functions which are implemented in the prefrontal cortex (PFC). These functions are maintenance, control of attention and integration. A model for the maintenance function is presented, and we will argue that this model can be extended to incorporate the other functions as well. Maintenance is the capacity to briefly maintain information in the absence of corresponding input, and even in the face of distracting in

Excitation of marginal and substantia gelatinosa neurons in the primate spinal cord: indications of their place in dorsal horn functional organization.

Electrophysiological recordings were made from superficial parts of the spinal dorsal horn in monkeys, using dye-filled micropipette electrodes to permit iontophoretic marking of the recording sites for subsequent histological recovery. Focal field potentials and unitary activity evoked by dorsal root volleys including slowly-conducting components (both myelinated and unmyelinated) were found in the posteromarginal zone and the substantia gelatinosa (SG). Unitary potentials identified as being of the type recorded from cellular regions were separated into categories according to which group of

An explanation of eye movements seen in internuclear ophthalmoplegia.

Recent findings in experiments on monkeys show that a horizontal saccade is made by a pulse-step of neural activity, part of which rises to medial rectus (MR) motoneurons on the fibers of the medial longitudinal fasciculus (MLF). If inhibitory as well as excitatory fibers run in the MLF, then the data show that each MLF must carry both the excitatory and inhibitory activity required for contralateral, horizontal saccades. Therefore, interruption of these fibers removes not only excitation from the ipsilateral MR but inhibition from the contralateral MR. Such a lesion also disturbs the correct

Parameter-dependent effects of transcranial ultrasound stimulation on corticospinal excitability and inhibitory control.

BACKGROUND: Transcranial ultrasound stimulation (TUS) with various parameters can modulate cortical excitability, but its neuromodulatory effects on excitability and behavior have not been consistently replicated across independent labs or compared within subjects. OBJECTIVE: To examine the neuromodulatory effects of four TUS protocols against sham on motor cortical excitability and inhibitory control. METHODS: Twenty-six healthy adults were enrolled to complete up to five sessions (four active TUS protocols and one sham); 24 completed all the five sessions, separated by at least one week. M

Synaptic inhibitory dynamics drive benzodiazepine response in pediatric status epilepticus.

OBJECTIVE: Pediatric status epilepticus (SE) is a medical emergency associated with significant morbidity. Benzodiazepines (BZPs) are the current first-line treatment, but do not work in more than one third of children presenting with SE. Animal studies have shown that SE can cause changes in synaptic inhibition signaling that can ultimately lead to BZPs becoming ineffective. However, the relevance of these mechanisms in pediatric patients with SE remains unknown. METHODS: To test this hypothesis, we combine clinical electroencephalographic (EEG) recordings with dynamic causal modeling (DCM).

Ultrastructural characterization of hippocampal inhibitory synapses under resting and stimulated conditions.

The present study uses electron microscopy to document ultrastructural characteristics of hippocampal GABAergic inhibitory synapses under resting and stimulated conditions in three experimental systems. Synaptic profiles were sampled from stratum pyramidale and radiatum of the CA1 region from (1) perfusion fixed mouse brains, (2) immersion fixed rat organotypic slice cultures, and from (3) rat dissociated hippocampal cultures of mixed cell types. Synapses were stimulated in the brain by a 5 min delay in perfusion fixation to trigger an ischemia-like excitatory condition, and by treating the tw

Fluctuation in cortical excitation/inhibition modulates capability of attention across time scales ranging from hours to seconds.

Sustained attention, as the basis of general cognitive ability, naturally varies across different time scales, spanning from hours, e.g. from wakefulness to drowsiness state, to seconds, e.g. trial-by-trail fluctuation in a task session. Whether there is a unified mechanism underneath such trans-scale variability remains unclear. Here we show that fluctuation of cortical excitation/inhibition (E/I) is a strong modulator to sustained attention in humans across time scales. First, we observed the ability to attend varied across different brain states (wakefulness, postprandial somnolence, sleep

Interhemispheric Inhibition Between Primary Motor Cortices is Not Altered in Individuals With Chronic Lateral Epicondylalgia.

Lateral epicondylalgia (LE), commonly referred to as tennis elbow, is a musculoskeletal condition characterized by pain and sensorimotor dysfunction. In some individuals with chronic unilateral LE, sensorimotor symptoms develop on the unaffected side despite no evidence of tissue damage. Altered interhemispheric inhibition (IHI) is one mechanism that could underpin this phenomenon. The aim of this cross-sectional study was to examine IHI between the primary motor cortices (M1) in individuals with chronic LE and healthy controls. In 20 individuals with chronic LE and 20 healthy participants, tr

Direct evidence for decreased presynaptic inhibition evoked by PBSt group I muscle afferents after chronic SCI and recovery with step-training in rats.

KEY POINTS: Presynaptic inhibition is modulated by supraspinal centres and primary afferents in order to filter sensory information, adjust spinal reflex excitability, and ensure smooth movement. After spinal cord injury (SCI), the supraspinal control of primary afferent depolarization (PAD) interneurons is disengaged, suggesting an increased role for sensory afferents. While increased H-reflex excitability in spastic individuals indicates a possible decrease in presynaptic inhibition, it remains unclear whether a decrease in sensory-evoked PAD contributes to this effect. We investigated wheth

Parietal conditioning enhances motor surround inhibition.

BACKGROUND: Motor surround inhibition (mSI) is a phenomenon supportive for executing selective finger movements, wherein synergist muscles are selectively facilitated while surround muscles are inhibited. Previous studies of conditioning inputs to several intracortical and cortico-cortical inhibitory networks did not show an influence on mSI. The inhibitory posterior parietal-motor network, which is crucial for executing fine movements, however, has not been studied. OBJECTIVE/HYPOTHESIS: To investigate the role of inhibitory posterior parietal-motor network in mSI. We hypothesized that condi

Inhibition increases response variability and reduces stimulus discrimination in random networks of cortical neurons.

Much of what is known about the contribution of inhibition to stimulus discrimination is due to extensively studied sensory systems, which are highly structured neural circuits. The effect of inhibition on stimulus representation in less structured networks is not as clear. Here we exercise a biosynthetic approach in order to study the impacts of inhibition on stimulus representation in non-specialized network anatomy. Combining pharmacological manipulation, multisite electrical stimulation and recording from ex-vivo randomly rewired networks of cortical neurons, we quantified the effects of i

Pain in cervical dystonia: Evidence of abnormal inhibitory control.

INTRODUCTION: Several observations would suggest that dystonic pain is not simply muscular in origin. While ascending nociceptive pathways are normal in cervical dystonia, it is unknown whether descending inhibitory pain pathways are also normal. METHODS: We applied a conditioned pain modulation protocol and concomitantly recorded laser evoked potentials in patients with cervical dystonia (n = 15), blepharospasm (n = 15) and healthy volunteers (n = 15). RESULTS: During the application of a heterotopic noxious conditioning stimulation, patients with cervical dystonia, but not with blepharospa

The Hippocampus and Neocortical Inhibitory Engrams Protect against Memory Interference.

Our experiences often overlap with each other, yet we are able to selectively recall individual memories to guide decisions and future actions. The neural mechanisms that support such precise memory recall remain unclear. Here, using ultra-high field 7T MRI we reveal two distinct mechanisms that protect memories from interference. The first mechanism involves the hippocampus, where the blood-oxygen-level-dependent (BOLD) signal predicts behavioral measures of memory interference, and representations of context-dependent memories are pattern separated according to their relational overlap. The

Maturation of Breathing-Related Inhibitory Neurotransmission in the Medulla Oblongata of the Embryonic and Perinatal Zebra Finch (Taeniopygia guttata).

The medullary portion of the embryonic zebra finch hindbrain was isolated and superfused with physiologically relevant artificial cerebral spinal fluid. This in vitro preparation produced uninterrupted rhythmic episodes of neural activity via cranial nerve IX (glossopharyngeal) from embryonic day 4 (E4) through hatching on E14. Cranial nerve IX carries motor activity to the glottis during the inspiratory phase of breathing, and we focused on the role of synaptic inhibition during the embryonic and perinatal maturation of this branchiomotor outflow. We show that spontaneous neural activity (SNA

Comparison of Bladder Inhibitory Effects of Patterned Spinal Nerve Stimulation With Conventional Neuromodulation in the Rat.

OBJECTIVES: The present study compared the effectiveness of patterned frequency of spinal nerve stimulation (SNS) with continuous, fixed-frequency nerve stimulation in an animal model of the bladder reflex contraction (BRC). MATERIALS AND METHODS: In anesthetized female rats, wire electrodes were placed under each of the L6 spinal nerve to produce bilateral SNS. A cannula was placed into the bladder via the urethra, and the urethra was ligated to ensure an isovolumetric bladder. RESULTS: Using motor threshold intensity, continuous stimulation at fixed frequencies of 4 Hz (n = 5) and 10 Hz (n

Two common psychophysical measures of surround suppression reflect independent neuronal mechanisms.

Psychophysical surround suppression is believed to reflect inhibitory neuronal mechanisms in visual cortex. In recent years, two psychophysical measures of surround suppression have been much studied: (i) duration thresholds on a motion-discrimination task (which are worse for larger than for smaller stimuli) and (ii) contrast thresholds on a contrast-detection task (which are worse when grating stimuli are surrounded by a stimulus of the same orientation than when they are presented in isolation or surrounded by a stimulus of orthogonal orientation). Changes in both metrics have been linked t

Virtual lesions of the inferior parietal cortex induce fast changes of implicit religiousness/spirituality.

Religiousness and spirituality (RS) are two ubiquitous aspects of human experience typically considered impervious to scientific investigation. Nevertheless, associations between RS and frontoparietal neural activity have been recently reported. However, much less is known about whether such activity is causally involved in modulating RS or just epiphenomenal to them. Here we combined two-pulse (10 Hz) Transcranial Magnetic Stimulation (TMS) with a novel, ad-hoc developed RS-related, Implicit Association Test (IAT) to investigate whether implicit RS representations, although supposedly rather
