Inhibition of soluble epoxide hydrolase rescues cognitive deficits by preserving neurovascular integrity and attenuating glial and neuropathology in diabetic-related dementia.

Diabetes mellitus (DM) is a major risk factor contributing to the development of Alzheimer's disease-related dementias (ADRD). While one of the early symptoms of both Alzheimer's disease (AD) and DM-related ADRD is a reduction in cerebral blood flow, the underlying biological mechanisms driving this decline remain to be fully elucidated. Genome-wide association studies have linked AD/ADRD to single-nucleotide polymorphisms in the gene encoding soluble epoxide hydrolase (sEH), an enzyme we previously reported to be upregulated in the brains of an AD rat model. Our previous studies also demonstr

Beyond the brain-centric view: A systems physiology framework for exercise-induced neuroprotection in Alzheimer's disease.

Alzheimer's disease (AD) is regarded as a multifactorial disease that combines central pathological changes and peripheral organ dysfunction. Its core features include Aβ protein deposition, tau protein pathology, synaptic dysfunction, and neuroinflammation. Exercise has been proven to be one of the most effective non-pharmacological intervention measures. However, its mechanism of action in AD has not been fully elucidated from the traditional brain-centered research perspective. Moreover, most existing studies focus on isolated organ systems or a single signaling pathway, failing to fully re

fMRI BOLD turnover (TBOLD) as a proxy for neuroinflammation.

Inflammation is associated with detrimental health outcomes, including deleterious effects on the brain. We have recently introduced a new measure of brain function reflecting the moment-to-moment change, or turnover, in the resting-state blood oxygen level-dependent (BOLD) signal, or TBOLD. In previous studies, an increase of TBOLD with age was found to be associated with a concomitant decrease in brain volume, whereas both changes were absent in individuals carrying the neuroprotective human leukocyte antigen (HLA) allele DRB1*13:02. Given that TBOLD reflects neurovascular coupling and that

Moving Beyond Outcome-Based Exercise Studies in Cognitive Aging: A Mechanism-Informed Narrative Review of Cerebrovascular Anchors and Supportive Biological Endpoints.

Exercise training is frequently examined in relation to cognitive and brain health in aging populations. Yet many exercise-cognition studies remain difficult to interpret mechanistically because cognitive outcomes are often assessed without sufficient alignment with exercise dose, biological endpoints, or participant vulnerability. This mechanism-informed narrative review argues that future research in cognitive aging should move beyond outcome-based interpretation alone and more explicitly connect the exercise stimulus with cerebrovascular endpoints, supportive biological markers, baseline vu

Muscle and mind: rewiring cognitive-motor recovery through exercise-responsive neurophysiology in neurological populations.

Exercise in neurological rehabilitation is often prescribed to improve mobility, yet its influence extends to cognition through interacting peripheral, central, and behavioral pathways. This narrative review synthesizes evidence on how skeletal muscle activity can engage peripheral biomarkers and system-level interactions that become clinically meaningful only when translated into behaviorally relevant cognitive-motor integration (CMI). We examine neurotrophic, metabolic, immune, vascular, and endocrine relays, then consider central translation through synaptic plasticity, metabolic adaptation

Unveiling the silent link: Normal-tension glaucoma's enigmatic bond with cardiac blood flow.

This comprehensive review embarks on a captivating journey into the complex relationship between cardiology and normal-tension glaucoma (NTG), a condition that continues to baffle clinicians and researchers alike. NTG, characterized by optic nerve damage and visual field loss despite normal intraocular pressure, has long puzzled clinicians. One emerging perspective suggests that alterations in ocular blood flow, particularly within the optic nerve head, may play a pivotal role in its pathogenesis. While NTG shares commonalities with its high-tension counterpart, its unique pathogenesis and pot

Multifocal Cerebral Microinfarcts Modulate Early Alzheimer's Disease Pathology in a Sex-Dependent Manner.

Alzheimer's disease (AD) constitutes a major cause of dementia, affecting more women than men. It is characterized by amyloid-β (Aβ) deposition and neurofibrillary tangles (NFTs) formation, associated with a progressive cognitive decline. Evidence indicates that AD onset increases the prevalence of cerebral microinfarcts caused by vascular pathologies, which occur in approximately in half of AD patients. In this project, we postulated that multifocal cerebral microinfarcts decisively influence early AD-like pathology progression in a sex dependent manner in young APP/PS1 mice. For this purpose

Sepsis modulates cortical excitability and alters the local and systemic hemodynamic response to seizures.

Non-convulsive seizures and status epilepticus are frequent and associated with increased mortality in septic patients. However, the mechanism through which seizures impact outcome in these patients is unclear. As previous studies yielded an alteration of neurovascular coupling (NVC) during sepsis, we hypothesized that non-convulsive seizures, might further impair NVC, leading to brain tissue hypoxia. We used a previously developed ovine model of sepsis. Animals were allocated to sham procedure or sepsis; septic animals were studied either during the hyperdynamic phase (sepsis group) or after

Spreading depolarization: A phenomenon in the brain.

In 1944, the physiologist Leão while studying epilepsy in the rabbit noticed a sudden temporary cessation of electrocorticographic (ECoG) activity accompanied with a large negative slow potential change recorded by extracellular electrodes, that is later known as spreading depolarizations (SDs). The depression of the brain electrical activity was slowly propagating through the cerebral cortex. The mechanism of propagation is still controversial. SDs and seizures are following each other interchangeably, yet the puzzle needs more investigation to be clarified. SDs have an obvious effect on bloo

Cocaine's cerebrovascular vasoconstriction is associated with astrocytic Ca2+ increase in mice.

Human and animal studies have reported widespread reductions in cerebral blood flow associated with chronic cocaine exposures. However, the molecular and cellular mechanisms underlying cerebral blood flow reductions are not well understood. Here, by combining a multimodal imaging platform with a genetically encoded calcium indicator, we simultaneously measured the effects of acute cocaine on neuronal and astrocytic activity, tissue oxygenation, hemodynamics and vascular diameter changes in the mouse cerebral cortex. Our results showed that cocaine constricted blood vessels (measured by vessel

Retinal Vascular Reactivity as Assessed by Optical Coherence Tomography Angiography.

The vascular supply to the retina has been shown to dynamically adapt through vasoconstriction and vasodilation to accommodate the metabolic demands of the retina. This process, referred to as retinal vascular reactivity (RVR), is mediated by neurovascular coupling, which is impaired very early in retinal vascular diseases such as diabetic retinopathy. Therefore, a clinically feasible method of assessing vascular function may be of significant interest in both research and clinical settings. Recently, in vivo imaging of the retinal vasculature at the capillary level has been made possible by t

Co-localized impaired regional cerebrovascular reactivity in chronic concussion is associated with BOLD activation differences during a working memory task.

The purpose of this study was to quantify differences in blood oxygen level dependent (BOLD) activation on a working memory task, baseline cerebral blood flow (CBF0), and cerebrovascular reactivity (CVR) between participants with and without a history of concussion. A dual-echo pseudo-continuous arterial spin labelling (pCASL) sequence was performed on a group of 10 subjects with a previous concussion (126 ± 15 days prior) and on a control group (n = 10) during a visual working memory protocol. A separate dual-echo pCASL sequence was used to derive CVR and CBF0 measurements from a boxcar hyper

Abnormal oxygen homeostasis in the nucleus tractus solitarii of the spontaneously hypertensive rat.

What is the central question of this study? Arterial hypertension is associated with impaired neurovascular coupling in the somatosensory cortex. Abnormalities in activity-dependent oxygen consumption in brainstem regions involved in the control of cardiovascular reflexes have not been explored previously. What is the main finding and its importance? Using fast-cyclic voltammetry, we found that changes in local tissue PO2 in the nucleus tractus solitarii induced by electrical stimulation of the vagus nerve are significantly impaired in spontaneously hypertensive rats. This is consistent with p

Resting-state functional MR imaging: a new window to the brain.

Resting-state (RS) functional magnetic resonance (MR) imaging constitutes a novel paradigm that examines spontaneous brain function by using blood oxygen level-dependent contrast in the absence of a task. Spatially distributed networks of temporal synchronization can be detected that can characterize RS networks (RSNs). With a short acquisition time of less than 10 minutes, RS functional MR imaging can be applied in special populations such as children and patients with dementia. Some RSNs are already present in utero, while others mature in childhood. Around 10 major RSNs are consistently fou

The primary vascular dysregulation syndrome: implications for eye diseases.

Vascular dysregulation refers to the regulation of blood flow that is not adapted to the needs of the respective tissue. We distinguish primary vascular dysregulation (PVD, formerly called vasospastic syndrome) and secondary vascular dysregulation (SVD). Subjects with PVD tend to have cold extremities, low blood pressure, reduced feeling of thirst, altered drug sensitivity, increased pain sensitivity, prolonged sleep onset time, altered gene expression in the lymphocytes, signs of oxidative stress, slightly increased endothelin-1 plasma level, low body mass index and often diffuse and fluctuat

Astrocyte-Microglia Crosstalk in Post-Hemorrhagic Neurovascular Microenvironment: Mechanistic Nodes, Cross-Stroke Comparisons, and Therapeutic Reprogramming.

BACKGROUND: Intracerebral hemorrhage (ICH) produces a rapidly evolving and spatially heterogeneous neurovascular microenvironment in which secondary injury is shaped not only by hematoma volume and location, but also by the interaction of blood-derived toxins, blood-brain barrier disruption, edema, oxidative stress, protease activity, and glial responses. Increasing evidence suggests that these processes are better understood as dynamic network events rather than isolated inflammatory pathways. MAIN BODY: This review applies a network-centered framework to astrocyte-microglia coupling, viewin

Beyond the vision: central serous chorioretinopathy, anxiety and depression-a systematic review.

BACKGROUND: Central Serous Chorioretinopathy (CSCR) is an exudative maculopathy characterized by serous retinal detachment, choroidal hyperpermeability, and retinal pigment epithelium dysfunction. Beyond its ocular features, CSCR has long been associated with stress-related mechanisms and emotional dysregulation. Psychological factors such as anxiety and depression may influence disease onset, recurrence, and recovery; however, no previous review has systematically synthesized quantitative evidence on these variables in CSCR. METHODS: This systematic review followed the PRISMA 2020 guidelines

The Occlusal Contact Area Is Associated with the Magnitude but Not Peak Timing of Changes in Chewing-Induced Brain Blood Flow in Mandibular Prognathism.

Background/Objectives: Jaw deformities affect not only facial aesthetics but also various oral functions. While previous studies have demonstrated that mandibular prognathism (MP) alters masticatory-induced brain blood flow (BBF), the temporal characteristics of these hemodynamic changes have remained unclear. In this cross-sectional observational study, we investigated the following two specific objectives: (1) whether food hardness affects not only the magnitude but also the temporal patterns of BBF changes during mastication and (2) how malocclusion is associated with these temporal hemodyn
