Interleukin-6 trans-signaling exacerbates inflammation and renal pathology in lupus-prone mice.

OBJECTIVE: Systemic lupus erythematosus (SLE) is a heterogeneous autoimmune disease that is characterized by the production of antinuclear antibodies (ANAs) and leads to immune complex deposition in the kidneys and nephritis. Lyn tyrosine kinase is a regulator of antibody-mediated autoimmune disease, as evidenced by studies in gene-targeted mice and as suggested in genome-wide association studies in SLE. Like SLE patients, Lyn-deficient mice have increased levels of interleukin-6 (IL-6). Deletion of IL-6 from Lyn-deficient mice abrogates levels of inflammation, pathogenic autoantibodies, and n

[Standards, Options and Recommendations: good practice for the management and shipment of histological and cytopathological cancer specimens].

CONTEXT: The "Standards, Options and Recommendations" (SOR) collaborative project was initiated in 1993 by the Federation of the French Cancer Centres (FNCLCC), with the 20 French Regional Cancer Centres, several French public university and general hospitals, as well as private clinics and medical specialty societies. Its main objective is the development of serviceable clinical practice guidelines in order to improve the quality of health care and the outcome of cancer patients. The methodology is based on a literature review, followed by critical appraisal by a multidisciplinary group of ex

Quantitative Partitioning of Photoautotrophic and Photoheterotrophic Contributions During Mixotrophic Growth of Scenedesmus obliquus.

Mixotrophic growth of microalgae involves the simultaneous assimilation of inorganic and organic carbon sources, but the quantitative contributions of photoautotrophic and photoheterotrophic metabolisms remain poorly resolved. In this study, a stoichiometric and data reconciliation framework was developed to partition mixotrophic metabolism of the microalga Scenedesmus obliquus into photoautotrophic and photoheterotrophic fluxes using macroscopic measurements of carbon, nitrogen and oxygen exchanges. Continuous cultures were conducted under photoautotrophic, photoheterotrophic and mixo-trophic

Molecular Imaging of Human Brain Organoids Using Mass Spectrometry.

Brain organoid models serve as a powerful tool for studying human brain development and function. Mass spectrometry imaging (MSI), a cutting-edge technology, allows us to map the spatial distribution of diverse molecules such as lipids, sugars, amino acids, drugs, and their metabolites within these organoids, all without the need for specific molecular probes. High-quality MSI data hinge on meticulous sample preparation. Fixatives play a pivotal role, but conventional options such as glutaraldehyde, paraformaldehyde, and cryopreserving such as sucrose may inadvertently impact tissue metabolite

A modified technique for the diagnosis of Hirschsprung disease from rectal biopsies.

BACKGROUND: A diagnosis of Hirschsprung disease requires the demonstration of acetylcholinesterase fibres on frozen sections obtained from snap frozen biopsies of the rectum. This histochemical technique is generally not available in laboratories in developing countries. We improvised on the methodology of tissue preservation to make the staining technique more user-friendly, economical and reliable in demonstrating acetylcholinesterase activity in fresh rectal mucosal biopsies for the diagnosis of Hirschsprung disease. METHODS: Between June 1999 and May 2002 fresh rectal biopsies from 40 sus

Ultrastructural studies and immunolocalization of enamel proteins in rodent secretory stage ameloblasts processed by various cryofixation methods.

BACKGROUND: Cryofixation rapidly immobilizes cell and tissue components in their native state, thereby resulting in an ultrastructural preservation very close to the living situation. We have applied this approach to examine the morphology of secretory stage ameloblasts and the distribution of enamel proteins in these cells. METHODS: Molar and incisor tooth germs from newborn mice and/or rats were quickly dissected and divided into segments. The segments were then rapidly frozen using slam, plunge or pressure freezing, freeze-substituted and embedded in Epon. In addition, incisors from older

Inoculation with Stenotrophomonas maltophilia LIMN and Enterobacter roggenkampii LCMG Enhances Maize (Zea mays L.) Yield and Reduces Nitrogen Fertilizer Dependence in Nutrient-Limited Soils of Semi-Arid Regions.

Excessive nitrogen (N) fertilization in maize (Zea mays L.) production causes environmental degradation, particularly in semi-arid regions where high chemical inputs are necessary to maintain crop production. Plant growth-promoting rhizobacteria (PGPR) could reduce the optimal N fertilization doses. This investigation aimed to isolate bacteria, screen them for plant growth-promoting traits, and evaluate their potential effects on maize grain under different chemical N fertilization doses. Isolates Stenotrophomonas maltophilia LIMN and Enterobacter roggenkampii LCMG demonstrated multifunctional

Transient acute kidney injury following femoral fracture in mice : the role of sympathetic activation.

AIMS: Femoral shaft fractures are commonly associated with high-energy trauma, and affect individuals of all ages. Kidney dysfunction frequently complicates fracture healing, and trauma-related acute kidney injury (TRAKI) elevates the risk of adverse outcomes. The mechanisms of post-fracture kidney injury, particularly the involvement of sympathetic activation, remain poorly understood. We aimed to define the impact of femoral fracture on the kidney and investigate mechanistically the potential contribution of sympathetic activation. METHODS: A total of 46 female C57BL/6J mice were subjected

Climate-Driven Changes in the Nutritional Value and Food Safety of Legume Seeds.

Background/Objectives: Leguminous plants (Fabaceae) are essential for global food and nutritional security due to their high protein content, bioactive compounds, and ecological role in nitrogen fixation. However, climate change poses significant threats to their productivity, quality, and safety. This review aims to summarize the nutritional, biochemical, and health-related importance of legumes, while highlighting the effects of climate change-particularly heat stress and pest pressure-on their nutritional value and public health implications. Methods: This review is based on an integrative …

Enhancement of Tomato Growth Through Rhizobacteria and Biocontrol of Associated Diseases.

The purpose of this study was to investigate the growth-promoting effects of four rhizobacterial isolates (RS60, RS65, RS46, and RP6) isolated from the tomato rhizosphere. These isolates were screened for key plant growth-promoting rhizobacteria (PGPR) mechanisms, including ammonia production, nitrogen fixation, phosphate solubilization, indole-3-acetic acid (IAA) production, and siderophore synthesis. Their potential to enhance seed germination and tomato plant growth was investigated in controlled and greenhouse conditions. Four isolates exhibited multiple PGPR attributes, notably IAA and am…

Comparative Proteomic Analysis Reveals the Regulatory Effects of H2S on Salt Tolerance of Mangrove Plant Kandelia obovata.

As a dominant mangrove species, Kandelia obovata is distributed in an intertidal marsh with an active H2S release. Whether H2S participates in the salt tolerance of mangrove plants is still ambiguous, although increasing evidence has demonstrated that H2S functions in plant responses to multiple abiotic stresses. In this study, NaHS was used as an H2S donor to investigate the regulatory mechanism of H2S on the salt tolerance of K. obovata seedlings by using a combined physiological and proteomic analysis. The results showed that the reduction in photosynthesis (Pn) caused by 400 mM of NaCl was
