A pilot study of a non-invasive oral nitrate stable isotopic method suggests that arginine and citrulline supplementation increases whole-body NO production in Tanzanian children with sickle cell disease.

BACKGROUND: Low bioavailability of nitric oxide (NO) is implicated in the pathophysiology of sickle cell disease (SCD). We designed a nested pilot study to be conducted within a clinical trial testing the effects of a daily ready-to-use supplementary food (RUSF) fortified with arginine (Arg) and citrulline (Citr) vs. non-fortified RUSF in children with SCD. The pilot study evaluated 1) the feasibility of a non-invasive stable isotope method to measure whole-body NO production and 2) whether Arg+Citr supplementation was associated with increased whole-body NO production. SUBJECTS: Twenty-nine

Nuclear magnetic resonance analysis of the acetylation pattern of the neuronal Tau protein.

Lysine acetylation of the neuronal Tau protein was described as a novel mechanism of posttranslational regulation of Tau functions with important outcomes in microtubule binding and aggregation processes related to Alzheimer's disease. Here, we unravel at a per-residue resolution the acetylation pattern of full-length Tau by the Creb-binding protein (CBP) acetyltransferase using high-resolution nuclear magnetic resonance spectroscopy. Our study gives a quantitative overview of CBP-mediated acetylation and examines the catalytic proficiency because the nonenzymatic reaction with acetyl-coenzyme

Carbon dynamics in mycorrhizal symbioses is linked to carbon costs and phosphorus benefits.

The nutrient and carbon (C) allocation dynamics in mycorrhizal hyphal networks cause variation in costs and benefits for individual plants and fungi and influence the productivity, diversity and C cycling in ecosystems. We manipulated light and phosphorus (P) availability in a pot experiment with Trifolium subterraneum colonised by the arbuscular mycorrhizal (AM) fungus Glomus intraradices. Stable (13)C-labelling was used to trace assimilated CO(2) to the mycorrhizal fungus in roots and soil using compound-specific isotope ratio mass spectrometry. We used the neutral lipid fatty acid 16:1omega

Determination of cyanide in blood by isotope-dilution gas chromatography-mass spectrometry.

BACKGROUND: Cyanide (CN) is a lethal toxin. Quantification in blood is necessary to indicate exposure from many sources, including food, combustion byproducts, and terrorist activity. We describe an automated procedure based on isotope-dilution gas chromatography-mass spectrometry (ID GC/MS) for the accurate and rapid determination of CN in whole blood. METHODS: A known amount of isotopically labeled potassium cyanide (K13C15N) was added to 0.5 g of whole blood in a headspace vial. Hydrogen cyanide was generated through the addition of phosphoric acid, and after a 5-min incubation, 0.5 mL of

Molecular Engineering at Electrode Interface Enables Efficient Catalytic Nitrogen Reduction.

Molecular catalysts are promising for nitrogen (N2) fixation due to their well-defined sites and tunable coordination, yet most molecules in homogeneous systems reside in the bulk solution, leading to poor catalyst utilization. Here, we introduced a molecular engineering strategy by assembling a functionalized molecular molybdenum complex (PyMoBr3PNP) on graphdiyne (GDY) through π-π interactions. The resulting interface-assembled molecular electrode, PyMoBr3PNP/GDY/carbon paper (CP), catalyzed N2 conversion to ammonia (NH3) using 2,4,6-trimethylpyridinium triflate ([ColH][OTf]) as the proton s

Cooperative Supramolecular Polymerization in Both Polar and Non-Polar Solvents via Heteroatom-Driven Amphiphilicity.

Supramolecular polymers in water and non-polar solvents are extensively studied as model systems to mimic biological processes and as functional materials, respectively. The molecular designs to obtain supramolecular polymers in non-polar and polar solvents are typically orthogonal. Here, we disclose a molecular design in which we combine hydrogen-bonding amide units attached to alkyl chains for compatibility with non-polar solvents and heteroatom in the π-conjugated core for interaction with polar solvents, leading to supramolecular polymerization of the same monomeric unit in both non-polar

Turning the band alignment of carbon dots for visible-light-driven enzymatic asymmetric reduction of aromatic ketone.

Keto reductases are crucial NAD(P)H-dependent enzymes used for the enantioselective synthesis of alcohols from prochiral ketones. Typically, the NADPH cofactor is regenerated through a second enzyme and/or substrate. However, photocatalytic cofactor regeneration using water as a sacrificial electron and hydrogen donor presents a promising alternative, albeit a challenging one. Herein we fabricated several nitrogen-doped carbon dots (CDs) with visible light absorption properties, good water solubility and biocompatibility for photocatalytic regeneration of NADPH. The CD with a smaller size and

Impact of haze on potential pathogens in surface bioaerosol in urban environments.

Air quality considerably affects bioaerosol dynamics within the atmosphere. Frequent haze events, with their associated alterations in bioaerosol composition, may pose potential health risks. This study investigated the microbial diversity, community structure, and factors of PM2.5 within an urban environment. We further examined the impact of haze on potentially pathogenic bacteria in bioaerosols, and analyzed the sources of haze pollution. Key findings revealed that the highest levels of microbial richness and diversity were associated with lightly polluted air conditions. While the overall

Distributions of Total Mercury and Methylmercury in Dragonflies from a Large, Abandoned Mercury Mining Region in China.

Dragonflies (Order Odonata) often are considered to be biosentinels of environmental contamination, e.g., heavy metals and/or persistent organic pollutants (POPs). Dragonflies (n = 439) belonging to 15 species of 8 genera were collected from an abandoned mercury (Hg) mining region in China to investigate the bioaccumulation of total Hg (THg) and methylmercury (MeHg). THg and MeHg concentrations in dragonflies varied widely within ranges of 0.06-19 mg/kg (average: 1.5 ± 2.2 mg/kg) and 0.02-5.7 mg/kg (average: 0.75 ± 0.65 mg/kg), respectively. THg and MeHg were positively correlated with bodywei

Methylmercury biomagnification in aquatic food webs of Poyang Lake, China: Insights from amino acid signatures.

As the dominant mercury species in fish, methylmercury (MeHg) biomagnifies during its trophic transfer through aquatic food webs. MeHg is known to bind to cysteine, forming the complex of MeHg-cysteine. However, relationship between MeHg and cysteine in large-scale food webs has not been explored and contrasted with MeHg biomagnification models. Here, we quantified the compound-specific nitrogen isotopic analysis of amino acids (CSIA-AA), MeHg, and amino acid composition in aquatic organisms of Poyang Lake, the largest freshwater lake in China. The trophic positions (TPAA) of organisms ranged

Indole: A Promising Scavenging Agent for Methylglyoxal and Related Carbonyls in Tryptophan Containing Maillard Model Systems.

In situ generation of efficient carbonyl trapping agents from amino acids during food processing can be considered a useful approach to control the accumulation of harmful Maillard reaction products in food. Tryptophan is one such amino acid that can be used to generate carbonyl trapping agents. Indole, the main thermal degradation product of tryptophan, is known to react with simple aldehydes through electrophilic aromatic substitution type reactions mainly at carbon positions 2 and 3 in addition to the ring nitrogen. The ability of indole to scavenge three moles of reactive aldehydes per mol

Long-term ferrocyanide application via deicing salts promotes the establishment of Actinomycetales assimilating ferrocyanide-derived carbon in soil.

Cyanides are highly toxic and produced by various microorganisms as defence strategy or to increase their competitiveness. As degradation is the most efficient way of detoxification, some microbes developed the capability to use cyanides as carbon and nitrogen source. However, it is not clear if this potential also helps to lower cyanide concentrations in roadside soils where deicing salt application leads to significant inputs of ferrocyanide. The question remains if biodegradation in soils can occur without previous photolysis. By conducting a microcosm experiment using soils with/without pr

Isotopic Fingerprints of Iron-Cyanide Complexes in the Environment.

Tracing the origin of iron-cyanide complexes in the environment is important because these compounds are potentially toxic. We determined the stable isotopic compositions of cyanide-carbon (CCN) and cyanide-nitrogen (NCN) in 127 contaminated solids and 11 samples of contaminated groundwater from coal carbonization sites, blast furnace operations, and commercial cyanide applications. Coal-carbonization-related cyanides had unique high mean δ(13)CCN values of -10.5 ± 3.5‰ for the solids and -16.1 ± 1.2‰ for the groundwater samples, while the values for blast furnace sludge (-26.9 ± 1.5‰), commer

The biosynthesis of papaverine proceeds via (S)-reticuline.

Papaverine is one of the earliest opium alkaloids for which a biosynthetic hypothesis was developed on theoretical grounds. Norlaudanosoline (=tetrahydropapaveroline) was claimed as the immediate precursor alkaloid for a multitude of nitrogen containing plant metabolites. This tetrahydroxylated compound was proposed to be fully O-methylated. The resulting tetrahydropapaverine should then aromatize to papaverine. In view of experimental data, this pathway has to be revised. Precursor administration to 8-day-old seedlings of Papaver followed by direct examination of the metabolic fate of the sta

[Water use efficiency and leaf nutrient characteristics of five major tree species in broadleaved Korean pine forest in Changbai Mountains, China].

Water use efficiency (WUE) of five dominant tree species (Pinus koraiensis, Fraxinus mandshurica, Acer mono, Quercus mongolica, and Tilia amurensis) was estimated using the stable carbon isotope method in a broadleaved Korean pine forest in Changbai Mountains. Leaf carbon (C), nitrogen (N), and phosphorus (P) contents were measured to analyze nutrient utilization of the dominant species. The relationship between WUE and leaf nutrient contents was systematically assessed. WUE was different due to the variations of micrometeorological factors at different locations in the canopy. The four broadl

Extrusion of lupines with or without addition of reducing sugars: Effects on the formation of Maillard reaction compounds, partition of nitrogen and Nε-carboxymethyl-lysine, and performance of dairy cows.

The extrusion of leguminous seeds induces the formation of Maillard reaction compounds (MRC) as a product of protein advanced glycation and oxidation, which lowers protein degradability in the rumen. However, the quantitative relationship between the parameters of pretreatment (i.e., addition of reducing sugars) and extrusion, and the formation of MRC has not been established yet. Moreover, the fate of the main stable MRC, Nε-carboxymethyl-lysine (CML), in the excretory routes has never been investigated in ruminants. We aimed to test the effects of the temperature of extrusion of white lupine

Metabolic Performance and Fate of Electrons during Nitrate-Reducing Fe(II) Oxidation by the Autotrophic Enrichment Culture KS Grown at Different Initial Fe/N Ratios.

Autotrophic nitrate-reducing Fe(II)-oxidizing (NRFeOx) microorganisms fix CO2 and oxidize Fe(II) coupled to denitrification, influencing carbon, iron, and nitrogen cycles in pH-neutral, anoxic environments. However, the distribution of electrons from Fe(II) oxidation to either biomass production (CO2 fixation) or energy generation (nitrate reduction) in autotrophic NRFeOx microorganisms has not been quantified. We therefore cultivated the autotrophic NRFeOx culture KS at different initial Fe/N ratios, followed geochemical parameters, identified minerals, analyzed N isotopes, and applied numeri

Differential changes to splanchnic and peripheral protein metabolism during the diet-induced development of metabolic syndrome in rats.

Little is known about the effects of the development of metabolic syndrome (MS) on protein and amino acid (AA) metabolism. During this study, we took advantage of the variability in interindividual susceptibility to high fat diet-induced MS to study the relationships between MS, protein synthesis, and AA catabolism in multiple tissues in rats. After 4 mo of high-fat feeding, an MS score (ZMS) was calculated as the average of the z-scores for individual MS components [weight, adiposities, homeostasis model for the assessment of insulin resistance (HOMA-IR), and triglycerides].
