The effect of global warming on rising aquatic temperatures is producing ever-steeper thermoclines. Fish encountering these sharp changes in water temperature might experience an acute-warming stress. Temperature is the most dominant environmental factor affecting heart function in fish, and without compensatory mechanisms as temperatures rise (e.g. higher heart rate), it could imperil cardiovascular performance. To enhance heart function during acute warming, fish release adrenaline to boost Ca2+ influx in heart cells (cardiomyocytes). However, the relationship between acute warming, elevated

This study was conducted to determine whether early dietary supplementation with an intermediate vitamin D metabolite could improve metabolic efficiency and liver function in rainbow trout. Over a 20-week feeding trial from first feeding, rainbow trout were fed either a control diet containing vitamin D3 alone or a diet additionally supplemented with 80 µg/kg 25-OH-D3. Plasma analysis confirmed that 25-OH-D3 was efficiently absorbed, with detectable levels of 25-OH-D3 and its epimer, indicating enhanced vitamin D status. While growth was similar between the experimental groups, fish receiving

Intensive rearing of farmed fish may risk disease spillover into free-living populations. This study concerns the blood-feeding gill monogenean of salmonids, Discocotyle sagitatta, on the Isle of Man, UK. Heavy infections in 2 fish farms have led to severe disease with periodic mass mortality. Infection levels in rainbow trout (Oncorhynchus mykiss, overall n = 556) increased with age (i.e. years exposed): by year 3, prevalence was 100%, mean intensity c. 100 (maximum 1150) worms/host. Output from farms of many millions of parasite eggs/day has the potential for transmission to downstream popul…

Urban stormwater runoff frequently contains the car tire transformation product 6PPD-quinone, which is highly toxic to juvenile and adult coho salmon (Onchorychus kisutch). However, it is currently unclear if embryonic stages are impacted. We addressed this by exposing developing coho salmon embryos starting at the eyed stage to three concentrations of 6PPD-quinone twice weekly until hatch. Impacts on survival and growth were assessed. Further, whole-transcriptome sequencing was performed on recently hatched alevin to address the potential mechanism of 6PPD-quinone-induced toxicity. Acute mort

The life cycle of Flavobacterium psychrophilum (Fp), the causative agent of bacterial coldwater disease (BCWD) and rainbow trout fry syndrome (RTFS), appears to involve interactions with spleen and head kidney macrophages. To develop an in vitro model for studying this, F. psychrophilum was incubated with a rainbow trout splenic monocyte/macrophage-like cell line (RTS11) and fundamental macrophage functions evaluated. The animal cell basal medium, L15, supplemented with bovine serum (FBS) supports RTS11 maintenance, and surprisingly, L15 with 2% FBS (L15/FBS) also supported F. psychrophilum gr

Multiple species and stocks of Pacific salmon Oncorhynchus spp. have experienced large declines in the number of returning adults over a wide region of the Pacific Northwest due to poor marine survival (low smolt-to-adult survival rates). One possible explanation for reduced survival is thiamine deficiency. Thiamine (vitamin B1 ) is an essential vitamin with an integral role in many metabolic processes, and thiamine deficiency is an important cause of salmonid mortality in the Baltic Sea and in the Laurentian Great Lakes. To assess this possibility, we (1) compared muscle thiamine content over…

Infectious diseases are a constant threat to industrialised farming, which is characterised by high densities of farms and farm animals. Several mathematical and statistical models on spatio-temporal dynamics of infectious diseases in various farmed host populations have been developed during the last decades. Here we present a spatio-temporal stochastic model for the spread of a disease between and within aquaculture farms. The spread between farms is divided into several transmission pathways, including (i) distance related spread and (ii) other types of contagious contacts. The within-farm

Pesticides are frequently detected in urban streams and are believed to be primarily the result of homeowner use. Although concentrations in most cases are low (<1 µg/L), there is concern that pesticide inputs threaten efforts to restore and enhance salmon habitat. The authors exposed early life stages of coho salmon (Oncorhynchus kisutch) to a pesticide mixture ("cocktail") representative of those pesticides most frequently reported in urban streams in western Washington State, USA. Life stages were continuously exposed to pulses of the cocktail simulating those in urban streams in fall and w…

A magnetic cell separation technique (MACS) was developed for isolating and characterizing peanut lectin agglutinin positive (PNA(+)) cells from rainbow trout gills. Percoll density separated mitochondria-rich (MR) cells were serially labeled with PNA-FITC and an anti-FITC antibody covalently coupled to a 50-nm iron particle and then applied to a magnetic column. PNA(+) MR cells were enriched to >95% purity. Transmission electron microscopy analysis of both the PNA(+) and PNA negative (PNA(-)) fraction showed that PNA binds to MR chloride cells while the PNA(-) cell fraction is comprised of MR

International biodiversity assessments often overlook the role of emerging infectious pathogens in the decline of freshwater fish population … orldwide and also suggest that the origin of the US strains is to be found among contaminated Asian Oncorhynchus tshawytscha living in sympatry with native Asian P. parva population. Finally, several important preventative steps are suggested as a way to manage the impact of S. destruens on local fish communities.

As a consequence of global climate change, we will be likely facing an increasing frequency and intensity of flood events. Thus, the ecotoxicological relevance of sediment re-suspension is of growing concern. It is vital to understand contaminant uptake from suspended sediments and relate it to effects in aquatic biota. Here we report on a computational study that utilizes a physiologically based toxicokinetic model to predict uptake, metabolism and excretion of sediment-borne pyrene in rainbow trout (Oncorhynchus mykiss). To this end, data from two experimental studies were compared with the …

Parasite prevalence in predatory mammals is influenced by numerous factors including diet, sex, season, and habitat. We examined the effect of such factors on the prevalence of gastrointestinal helminths in North American river otters ( Lontra canadensis ) in Yellowstone Lake, Yellowstone National Park. Otters in this ecosystem have recently experienced a decline in their main prey, Yellowstone cutthroat trout ( Oncorhynchus clarkii bouvieri), and have, in turn, increased consumption of alternative foods. Helminths were found in 13.2% of otter fecal samples. The dominant parasite was a Diphyll…

Freshwater ecosystems are experiencing significant biodiversity loss due to multiple anthropogenic stressors, including temperature fluctuations and chemical pollutants such as 6PPD-quinone (6PPD-Q), a toxic byproduct of tire wear. 6PPD-Q is known for its lethal effects on species like coho salmon (Oncorhynchus kisutch) and could pose considerable risks even at environmentally relevant concentrations. However, little is known about how 6PPD-Q interacts with warming to affect cold-water fish, especially during sensitive early life stages. Arctic charr (Salvelinus alpinus), a species endemic to …

High salmonid toxicity of 6PPD-quinone, a transformation product of the tire antioxidant 6PPD, has accelerated the search for safer tire alt … s indicate that early-life zebrafish are far less sensitive to these PPD-quinones than coho salmon (Oncorhynchus kisutch). Given the high observed effect concentrations, which approached or exceeded the predicted water solubility limits, developmental risk of 7PPD-Q and 77PD-Q to zebrafish appears low.
