Prenatal diagnosis of parvovirus B19-induced hydrops fetalis by chemiluminescence in situ hybridization.

Parvovirus B19 can be transmitted transplacentally from the infected mother to the fetus during pregnancy, and hydrops fetalis, abortion, or stillbirth can result. In our study we explored the use of chemiluminescence in situ hybridization to detect B19 DNA on cord blood cells, amniotic fluid cells, and pleuric fluid cells from several cases of hydrops fetalis. B19 DNA was detected by using digoxigenin-labeled probes immunoenzymatically visualized with the chemiluminescent adamantil-1,2-dioxetane phenyl phosphate substrate for alkaline phosphatase. The luminescent signal emitted from the hybri…

Prospective evaluation of 618 pregnant women exposed to parvovirus B19: risks and symptoms.

OBJECTIVE: To assess the risk of maternal parvovirus B19 infection from exposure to various sources and the fetal morbidity of those infections. METHODS: We obtained demographic and occupational information about pregnant women exposed to sources of B19 and about the nature and duration of the exposures. We performed serologic testing 10-14 days after exposure using an indirect capture enzyme-linked immunosorbent assay. Women with immunoglobulin (Ig) M were examined with weekly ultrasound until 12 weeks after exposure, and the outcome of the pregnancy was ascertained from interviews with pati…

Life-threatening anaemia caused by B19 parvovirus infection in a non-immunocompromised patient.

A 21-year-old Surinam boy suffering from a combined heterozygous sickle-cell beta-thalassaemia presented with an extreme anaemia (haemoglobin concentration 0.7 mmol/l) and severe haemolysis without signs of increased erythropoiesis. An acute B19 parvovirus infection was diagnosed by a transient increase of IgM anti-B19 parvovirus titres and a positive polymerase chain reaction for B19 parvovirus. The patient recovered spontaneously after a short period of supportive care with red cell transfusions and mechanical ventilation.

Spontaneous resolution of non-immune hydrops fetalis secondary to transplacental parvovirus B19 infection.

We report a case of hydrops fetalis secondary to transplacental parvovirus B19 infection, in which spontaneous resolution of the hydrops occurred 4 weeks after the initial presentation. This case emphasizes the role of conservative management based on ultrasound evidence of continued resolution of the hydrops and intrauterine growth retardation.

Persistence of parvovirus H-1 DNA in human B- and T-lymphoma cells.

Persisting DNA of parvovirus H-1 could be demonstrated in cells of two human lymphoma cell lines, the Burkitt lymphoma cell line BL2 and the T-cell leukemia cell line Jurkat which survived infection with parvovirus H-1. Persistence of H-1 DNA rendered the cells resistant to a second H-1 infection. This resistance to H-1 superinfection persisted even after loss of H-1 DNA occurring after approximately 150-200 cell generations. Resistance to H-1 superinfection was accompanied by reduced uptake of infectious particles and by a block of H-1 DNA replication. This suggests that persistent H-1 infect…

Aplastic crisis in sickle cell disorders: bone marrow necrosis and human parvovirus infection.

Aplastic crisis in patients with sickle cell disease who develop a parvovirus infection may be associated with extensive bone marrow necrosis as well as acute selective erythroblastopenia. This illness may be manifested by pyrexia, lymphadenopathy, bone tenderness and significant hypoxemia with minimal roentgenographic findings in the lungs. It is uncertain whether the hypoxemia is caused by the effects of the viral infection on the lungs or is secondary to sickling of red blood cells in the pulmonary vasculature or both. The hypoxia may be sufficiently severe to require treatment with both ox…

Human parvovirus-associated red cell aplasia in the absence of underlying hemolytic anemia.

Human parvovirus (HPV) infection has recently been implicated as the cause of aplastic crisis in patients with hemolytic anemias such as congenital spherocytosis and sickle cell anemia. The virus causes a transient red cell aplasia which, in patients with a shortened red cell life span, is manifested as a rapid worsening of the anemia and an absence of peripheral reticulocytosis. Recovery is associated with the presence of giant pronormoblasts in the bone marrow, and several days later, a brisk peripheral reticulocytosis. In normal subjects, HPV causes erythema infectiosum (fifth disease) but …

Prevalence of parvovirus B19 infection in patients with hematological diseases in the Western Brazilian Amazon.

INTRODUCTION: Parvovirus B19 (B19V) infects erythroid precursor cells, compromising red blood cell production. This can trigger severe anemia and other hematological diseases in susceptible patients. However, data on B19V epidemiology in patients with hematological diseases are scarce, especially in the northern region of Brazil. The aim of this study was to estimate the prevalence of IgG and IgM antibodies against B19V, and to detect B19 DNA, in patients with hematological diseases undergoing METHODS: Plasma samples from 421 patients were subjected to enzyme immunoassay for the detection of a…

Clinical and virological impacts of human parvovirus B19 epidemics on fulminant myocarditis in childhood.

BACKGROUND: Outbreaks of pediatric myocarditis and increased activity of human parvovirus B19 (B19V) have been reported worldwide following the coronavirus disease 2019 (COVID-19) pandemic. However, the direct association between prevalent erythema infectiosum and fulminant myocarditis (FM) remains elusive because of the lack of surveillance data. This study aimed to evaluate the incidence of pediatric FM during the prolonged period and to characterize B19V genotypes and clinical outcomes of the positive cases for improved disease control. METHODS: Pediatric patients with FM retrospectively un…

Establishment of a TaqMan-based quantitative real-time PCR for the detection of porcine parvovirus.

Porcine Parvovirus (PPV) is a non-enveloped DNA virus that predominantly induces reproductive disorders in swine. The ongoing emergence of novel PPV variants and the frequent co-infections with other viruses have led to significant economic losses within the swine industry. This study, utilizing 31 previously reported complete PPV genome sequences, identified a conserved fragment of the PPV-NS1 gene through homology analysis. A TaqMan-based real-time quantitative PCR (TaqMan-qPCR) detection method was developed targeting this specific fragment. Sensitivity assessments determined a detection li…

Genomic surveillance and evolution of co-circulating goose parvovirus and waterfowl circovirus in China.

As the world's largest waterfowl producer, China faces economic losses from Goose Parvovirus (GPV), worsened by waterfowl circovirus co-infections that increase pathogenicity and immunosuppression. However, current surveillance systems for these viruses lack systematization. This study conducted a comprehensive genetic analysis of GPV and waterfowl circovirus to identify genomic characteristics and recombination events. Surveillance analysis of GPV revealed a significant host-associated genotypic divergence from 2018 to 2024 in China. Goose isolates were predominantly the Mutated GPV (MGPV, 87…

Generalized Petechial-Purpuric Eruption in a Child: A Rare Presentation of Parvovirus B19 Infection.

While parvovirus B19 is classically associated with erythema infectiosum and papular purpuric gloves and socks syndrome, multiple additional cutaneous manifestations have been reported in the literature. This paper describes a 10-year-old girl with a generalized petechial and purpuric eruption involving the trunk, extremities, and face with distinct perioral involvement. Diagnostic evaluation, including histopathology and polymerase chain reaction testing, confirmed acute parvovirus B19 infection with spontaneous resolution over several weeks. This case highlights an atypical presentation that…

Structural and functional studies of the main replication protein NS1 of human parvovirus B19.

Parvovirus B19 (B19V) is a ubiquitous virus that can infect the majority of human population and cause erythema infectiosum, acute arthropathy, and many other diseases. The main replication protein NS1 plays a critical role in cell cycle arrest, transactivation of viral and host genes, and replication and package of B19V genome. Both DNA nicking and unwinding activities are required for the in vivo function of NS1, but the underlying basis is poorly understood. Here, we report extensive structural and biochemical studies of NS1, showing that NS1 can unwind various types of DNA substrates. The …

Protein Myristoylation Plays a Role in the Nuclear Entry of the Parvovirus Minute Virus of Mice.

Being nonpathogenic to humans, rodent parvoviruses (PVs) are naturally oncolytic viruses with great potential as anti-cancer agents. As these viruses replicate in the host cell nucleus, they must gain access to the nucleus during infection. The PV minute virus of mice (MVM) and several other PVs transiently disrupt the nuclear envelope (NE) and enter the nucleus through the resulting breaks. However, the molecular basis of this unique nuclear entry pathway remains uncharacterized. In this study, we used MVM as a model to investigate the molecular mechanism by which PVs induce NE disruption dur…

Short Beak and Dwarfism Syndrome in Ducks in Poland Caused by Novel Goose Parvovirus.

Short beak and dwarfism syndrome (SBDS), which was previously identified only in mule ducks, is now an emerging disease of Pekin ducks in China and Egypt. The disease is caused by the infection of ducks with a genetic variant of goose parvovirus-novel goose parvovirus (nGPV). In 2019, SBDS was observed for the first time in Poland in eight farms of Pekin ducks. Birds in the affected flock were found to show growth retardation and beak atrophy with tongue protrusions. Morbidity ranged between 15% and 40% (in one flock), while the mortality rate was 4-6%. Co-infection with duck circovirus, a kno…

Pathological lesions in the immune organs of ducklings following experimental infection with goose parvovirus.

Goose parvovirus (GPV) is the etiological agent of Derzsy's disease, with a natural reservoir consisting only of geese and Muscovy ducks. However, the pathological changes in the immune organs of ducklings experimentally infected with GPV remain unknown. In this study, 2-day-old Cherry Valley ducklings were intramuscularly injected with GPV. Immune organs (e.g., thymus, bursa of Fabricius, spleen, Harderian gland, cecal tonsil, bone marrow, and peripheral blood lymphocytes [PBLs]) were collected 1, 3, 5, 7, 10, and 14 days post-infection (dpi). Pathological lesions were assessed by histology a…
