Peroxidase-mimicking nanozymes have emerged as powerful catalytic alternatives to natural enzymes for a wide range of applications. Among them, iridium (Ir)-based nanozymes exhibit exceptional intrinsic peroxidase-like activity. However, the high cost and scarcity of noble-metal Ir severely limit practical and large-scale use. Herein, we address this challenge by engineering a core-shell Ni-Ir nanozyme in which inexpensive nickel (Ni) serves as the core and catalytically active Ir forms a thin surface shell, resulting in a low Ir content of only 4.9 at.% (denoted as Ir4.9%Ni). This architectur…

Reactive oxygen species exacerbate nonalcoholic steatohepatitis (NASH) by oxidizing macromolecules; yet how they promote NASH remains poorly understood. Here, we show that peroxidase activity of global hepatic peroxiredoxin (PRDX) is significantly decreased in NASH, and palmitic acid (PA) binds to PRDX1 and inhibits its peroxidase activity. Using three genetic models, we demonstrate that hepatic PRDX1 protects against NASH in male mice. Mechanistically, PRDX1 suppresses STAT signaling and protects mitochondrial function by scavenging hydrogen peroxide, and mitigating the oxidation of protein t…

Gastric cancer (GC) is one of the most lethal malignancies due to high metastatic rate, making the identification of new therapeutic targets critical for developing effective anti-GC treatments. Glutathione peroxidase 4 (GPx4), a key regulator of ferroptosis and redox homeostasis, contributes to progression and influences patient survival. However, the molecular mechanism by which GPx4 drives GC progression has not been fully illuminated. In this study, we found that GPx4 was overexpressed and negatively associated with poor prognosis and distant metastasis, as confirmed by single-cell RNA seq…

Flower-like Fe3S4 nanosheets were synthesized via a one-step solvothermal method and employed as peroxidase-mimicking nanozymes in a colorimetric immunoassay for the sensitive detection of carcinoembryonic antigen (CEA). Comprehensive structural and chemical characterizations confirmed the successful synthesis and surface modification of the Fe3S4 nanosheets, enabling efficient antibody conjugation. The nanozymes demonstrated significant peroxidase-mimicking behavior by facilitating the hydrogen peroxide (H2O2)-mediated oxidation of 3,3',5,5'-tetramethylbenzidine (TMB), where hydroxyl radicals…

This study discloses the noncovalent immobilization of a bienzyme cascade composed of glucose oxidase (GOx) and horseradish peroxidase (HRP) onto magnetically responsive polyamide microparticles (PA MPs). Porous PA6, PA4, and PA12 MPs containing iron fillers were synthesized via activated anionic ring-opening polymerization in suspension, alongside neat PA6 MPs used as a reference. Four hybrid catalytic systems (GOx/HRP@PA) were prepared through sequential adsorption of HRP and GOx onto the various PA MP supports. The initial morphologies of the supports and the hybrid biocatalysts were charac…

Efficiently enhancing the activity and selectivity of targeted nanozymes is a challenging task, primarily due to the inherent structural stability and heterogeneous atomic composition of traditional nanozymes. Herein, theoretical design is carried out to select Fe-based oxides (FO) nanozymes with high peroxidase (POD)-like activity by incorporating different nonmetallic atoms (N, P, S, and B). Among these dopants, B emerged as a superior candidate because it could effectively tune the adsorption energies of *OH intermediates and *H2O2, thereby endowing the nanozymes with superior POD-like perf…

Pseudomonas sp. AU10 is an Antarctic psychrotolerant bacterium that produces a dye-decolorizing peroxidase (DyP-AU10). The recombinant enzyme (rDyP-AU10) is a heme-peroxidase that decolors dyes and modifies kraft lignin. In this work, we report the best activity parameters for lignin modification (at 45 °C and pH 4) and show that the enzyme increases the number of aldehydes, ketones, and phenolic compounds. The analyses of the HPLC profile of samples also support that rDyP-AU10 induces the chemical change of kraft lignin. The enzyme also acts as a biobleaching agent on cellulose pulps, as show…

Peroxidase-mimicking nanozymes that can generate toxic hydroxyl radicals (. OH) hold great promise as antibacterial alternatives. However, most of them display optimal performance under strongly acidic conditions (pH 3-4), and are thus not feasible for many medical uses, including burn infections with a wound pH close to neutral. Herein, we report a copper-based nanozyme (CuCo2 S4 ) that exhibits intrinsic peroxidase-like activity and can convert H2 O2 into . OH at neutral pH. In particular, bimetallic CuCo2 S4 nanoparticles (NPs) exhibited enhanced peroxidase-like activity and antibacterial c…

The organization of nanoparticles (NPs) with controlled chemical composition and size distribution into well-defined sheets will find many practical applications, but the chemistry remains problematic. Therefore, we report a facile method to assemble NPs to free-floating two-dimensional (2D) nanosheets with a superlattice and thicknesses reaching 22.8 nm. The ligand oleic acid is critical in the formation of nanosheets. As assembled, these free-floating 2D nanosheets remain intact in both polar and nonpolar solvents, e.g., deionized water, ethanol, N,N-dimethylformamide, dimethyl sulfoxide, to

BACKGROUND: The use of automated immunometric methods for the detection of anti-thyroid peroxidase antibodies (TPOAb), the main serological marker of autoimmune thyroid diseases (AITD), has expanded in recent years. However, it is not known whether these new automated platforms have improved the diagnostic performance of TPOAb assays. The aim of this study was to evaluate the potential improvement of the inter-method agreement of current automated third generation systems, 12 years after a previous study, which had assessed the analytical variability between semi-automated second generation me…

Based on degradation of sphingomyelin/cholesterol liposomes containing entrapped horseradish peroxidase, we evaluated the Sphingomyelinase-D (SMase-D) activity of scorpion, spider and snake venoms by monitoring spectrophotometrically the product of oxidation of HRP released. The results indicate that Loxosceles crude venoms (Loxosceles intermedia, Loxosceles laeta, Loxosceles gaucho and Loxosceles similis) displayed SMase-D activity in a concentration-dependent manner. Furthermore, this activity was blocked by the anti-loxoscelic antivenom. However, Tityus serrulatus scorpion venom, Phoneutria…

Current drug therapies against Trypanosoma cruzi, the causative agent of Chagas disease, have limited effectiveness and are highly toxic. T. cruzi-specific metabolic pathways that utilize trypanothione for the reduction of peroxides are being explored as potential novel therapeutic targets. In the present study we solved the X-ray crystal structure of one of the T. cruzi enzymes involved in peroxide reduction, the glutathione peroxidase-like enzyme TcGPXI (T. cruzi glutathione peroxidase-like enzyme I). We also characterized the wild-type, C48G and C96G variants of TcGPXI by NMR spectroscopy a…

Glutathione peroxidase 1 (Gpx1) plays an important role in cellular defense by converting hydrogen peroxide and organic hydroperoxides to nonreactive products, and Gpx1(-/-) mice, which are characterized by reduced tissue glutathione peroxidase activity, are known to exhibit enhanced oxidative stress. Peroxides participate in tissue injury, as well as the hypertrophy of cultured cells, yet the role of Gpx1 to prevent end organ damage in cardiovascular tissue is not clear. We postulated that Gpx1 deletion would potentiate both aortic and cardiac hypertrophy, as well as mean arterial blood press…

A selenium dependent glutathione peroxidase (Se-GPx) cDNA was cloned from haemocyte by a reverse-transcription polymerase chain reaction (RT-PCR) and rapid amplification of cDNA (RACE). The 913 bp cDNA contained an open reading frame (ORF) of 558 bp encoded a deduced amino acid sequence of 186 amino acids. The prawn Se-GPx sequence contains a selenocysteine (Sec) residue which is encoded by the unusual stop codon, (115)TGA(117). According to the molecular modeling analysis, the active site Sec residue, located in the loop between beta3 and alpha2 in a pocket on the protein surface, and hydroge…

Recent evidences have shown that the in vivo antioxidant chemistry of organoselenium compounds such as diphenyl diselenide (DPDS) is complex and it is not completely understood. The complexity is partly due to the fact that DPDS is generally thought to exert its antioxidant action by mimicking glutathione peroxidase (GPx) with concomitant utilization of glutathione (GSH) in vitro. In contrast to in vitro data, we recently observed that DPDS increases rather than diminish GSH levels in diabetic models. The present study therefore sought to investigate a possible change in the antioxidant mechan…

Catalase-peroxidases (KatGs) are unique bifunctional heme peroxidases that exhibit peroxidase and substantial catalase activities. Nevertheless, the reaction pathway of hydrogen peroxide dismutation, including the electronic structure of the redox intermediate that actually oxidizes H(2)O(2), is not clearly defined. Several mutant proteins with diminished overall catalase but wild-type-like peroxidase activity have been described in the last years. However, understanding of decrease in overall catalatic activity needs discrimination between reduction and oxidation reactions of hydrogen peroxid…

Phospholipid hydroperoxide glutathione peroxidase (PHGPx) is an ubiquitous antioxidant enzyme, but the exact expression pattern in mammalian tissues is still unknown. The expression and cellular localization of PHGPx mRNA were examined in male mice using real time-polymerase chain reaction and in situ hybridization techniques. The rank order of PHGPx mRNA expression across tissues exhibiting substantial levels of expression was:testes >> heart > cerebrum > or = ileum > stomach = liver = jejunum > or = epididymis. In testes, PHGPx mRNA was highly expressed in spermiogenic cells and Leydig cells…

Previous studies indicate that noise induced hearing loss (NIHL) involves a decrease in glutathione peroxidase (GPx) activity and a subsequent loss of outer hair cells (OHC). However, the cellular localization of this GPx decrease and the link to OHC loss are still poorly understood. In this report, we examined the cellular localization of GPx (GPx1, GPx 3 and GPx 4) in F-344 rat before and after noise exposure and after oral treatment with ebselen, a small molecule mimic of GPx activity. Results indicate that GPx1 is the major isoform within the cochlea and is highly expressed in cells of the…
