BACKGROUND: Switchgrass (Panicum virgatum L.) is an important bioenergy and forage crop. The outcrossing nature of switchgrass makes it infeasible to maintain a genotype through sexual propagation. Current asexual propagation protocols in switchgrass have various limitations. An easy and highly-efficient vegetative propagation method is needed to propagate large natural collections of switchgrass genotypes for genome-wide association studies (GWAS).
RESULTS: Micropropagation by node culture was found to be a rapid method for vegetative propagation of switchgrass. Bacterial and fungal contamin

Hybrid wheat (Triticum spp.) has the potential to boost yields and enhance production under changing climates to feed the growing global population. Production of hybrid wheat seed relies on male sterility, the blocking of pollen production, to prevent self-pollination. One method of preventing self-pollination in the female plants is to apply a chemical hybridizing agent (CHA). However, some combinations of CHA and genotypes have lower levels of sterility, resulting in decreased hybrid purity. Differences in CHA efficacy are a challenge in producing hybrid wheat lines for commercial and exper

BACKGROUND: In 2006, an atypical U.S. case of bovine spongiform encephalopathy (BSE) was discovered in Alabama and later reported to be polymorphic for glutamate (E) and lysine (K) codons at position 211 in the bovine prion protein gene (Prnp) coding sequence. A bovine E211K mutation is important because it is analogous to the most common pathogenic mutation in humans (E200K) which causes hereditary Creutzfeldt - Jakob disease, an autosomal dominant form of prion disease. The present report describes a high-throughput matrix-associated laser desorption/ionization-time-of-flight mass spectromet

Balancing immunity and growth is a fundamental challenge for plants. Surprisingly, fungal cell-wall chitin elicits robust immune responses without causing growth inhibition, yet constitutive intracellular chitin signalling severely impairs growth. How plants resolve this paradox remains unclear. Here we identify two chitin-inducible, paralogous apoplastic chitinases in Arabidopsis thaliana, MIC1 and MIC2, both possessing direct antifungal activity through chitin hydrolysis. MIC2 generates immunogenic chitin oligomers that activate chitin-triggered immunity, whereas MIC1 ultimately degrades chi

BACKGROUND: Cytokinin oxidase/dehydrogenase (CKX) can irreversibly degrade cytokinin, regulating the growth and development of plants and helping them cope with environmental stress.
METHODS: To understand the expression characteristics and the biological functions of CKX in alfalfa (Medicago sativa), a novel gene, designated as MsCKX5 (GenBank: PV934228), was cloned, characterized and overexpressed in Arabidopsis thaliana.
RESULTS: qRT-PCR results showed that MsCKX5 gene expression was clearly tissue-specific and had the highest expression level in the stems. In addition, the expression lev

WHIRLY proteins, known for their DNA/RNA-binding capabilities conferred by a highly conserved WHIRLY domain, are integral to plant defense mechanisms. However, the role of OsWHY1 in rice immunity remains to be clarified. We examined OsWHY1 expression after treatment with exogenous hormones, PAMPs, and Magnaporthe oryzae. We generated OsWHY1-overexpressing rice plants and performed yeast two-hybrid (Y2H), bimolecular fluorescence complementation (BiFC), transcriptome, and RT-qPCR analyses. OsWHY1 expression was upregulated by exogenous hormones and PAMPs, with a significant increase at 24 h pos

Fusarium foot rot, caused by Fusarium solani (Mart.) Sacc., is a major threat to tomato production resulting in a tremendous loss of yield. The interaction between tomato and F. solani has not been well studied. Here we apply an integrative approach with physiological, transcriptomic, and metabolomic analysis to investigate the molecular basis of tomato defense responses to F. solani infection. Pathogen-infected plants have reduced leaf chlorophyll fluorescence, plant elongation, stem thickness and root growth compared to non-infected control. Transcriptome analysis reveals 5654 differentially

Plant evolution has been greatly influenced by polyploidization phenomena. Polyploid plants yield more and are more resistant to unfavorable environments than their diploid relatives. The triploid block, a postzygotic barrier that causes failure of endosperm development and thus seed arrest, often prevents polyploid breeding. Alterations in the parental dose in interploidy crosses alter endosperm development by changing the correct maternal: paternal ratio (2m:1p) that this tissue requires to properly fulfill its proliferation and cellularization. After many years of research, the study of epi

Tea (Camellia sinensis) is a well-known beverage crop rich in polyphenols with health benefits for humans. Understanding how tea polyphenols participate in plant resistance is beneficial to breeding resistant varieties and uncovering the resistance mechanisms. Here, we report that a Colletotrichum infection-induced 'pink ring' symptom appeared outside the lesion, which is highly likely to occur in resistant cultivars. By identifying morphological feature-specific metabolites in the pink ring and their association with disease resistance, and analysis of the association between metabolite and g

Triticum timopheevii Zhuk. is a tetraploid wheat that is utilized worldwide as a valuable breeding source for wheat improvement. Gene-based biotechnologies can contribute to this field; however, T. timopheevii exhibits recalcitrance and albinism in tissue cultures, making this species of little use for manipulation through genetic engineering and genome editing. This study tested various approaches to increasing in vitro somatic embryogenesis and plant regeneration, while reducing the portion of albinos in cultures derived from immature embryos (IEs) of T. timopheevii. They included (i) adjust

Currently, molecular breeding is regarded as an important tool for the improvement of many crop species. However, in sorghum, recently heralded as an important bioenergy crop, progress in this field has been relatively slow and limited. In this review, we present existing efforts targeted at genetic characterization of sorghum mutants. We also comprehensively review the different attempts made toward the isolation of genes involved in agronomically important traits, including the dissection of some sorghum quantitative trait loci (QTLs). We also explore the current status of the use of transge

Grey mould is a global disease caused by fungi that seriously threatens the production of many economically important crops, including strawberries. Although long non-coding RNAs (lncRNAs) have been confirmed to be involved in regulating various physiological processes in plants, elucidating the mechanisms underlying their role in contributing to plant resistance against Botrytis cinerea infection requires comprehensive investigation. In this study, lncRNA10007 was identified by lncRNA sequencing and observed to be downregulated following the induction of B. cinerea. Western blotting confirmed

Fruit size is not only a crucial fitness trait in plant evolution but also an important agronomic target in crop domestication and improvement, yet its regulatory mechanisms remain poorly characterized. Here, we identified an F-box E3 ubiquitin ligase gene, GhEBS1 (ENLARGED BOLL SIZE 1), as a positive regulator of cotton boll size via map-based cloning. An 845-bp structural variant in the promoter-proximal intronic region of GhEBS1 alters local chromatin accessibility and histone modifications, specifically H3K27 and H3K9 methylation levels and acetylation, thereby modulating GhEBS1 expression

Paclobutrazol (PBZ), a triazole-based plant growth retardant, is widely employed in agriculture and horticulture to control plant height and promote root growth. However, its persistence and mobility in the environment raise concerns regarding its bioavailability and toxicity in aquatic ecosystems. In this study, we investigated the reproductive toxicity of PBZ in zebrafish (Danio rerio), a well-established model for aquatic toxicology and vertebrate reproductive research. Adult zebrafish were exposed to PBZ (0, 0.1, 1 ppm) for 2, 4, 6, or 8 weeks. After exposure, breeding was conducted using

The molecular dialogue between rice and the brown planthopper (BPH; Nilaparvata lugens) is mediated by insect salivary effectors, yet the molecular mechanisms underlying plant perception and immune activation remain largely unclear. Here, we characterized the BPH salivary protein NlSP2, which is secreted into rice tissues during feeding. Silencing of NlSP2 in BPH impairs salivary sheath formation, feeding ability, and survival, whereas its ectopic expression in rice enhances resistance to BPH via MAPK activation, H2O2 accumulation, and induction of defense-related genes. We identify the rice c

Meiosis is central to sexual reproduction and the main source of genetic diversity in plants. Understanding how meiotic processes are regulated has direct relevance to agriculture. As meiotic recombination is the vehicle of plant breeding, gaining the ability to influence recombination patterns can accelerate crop improvement. Maize is a powerful model for studying plant meiosis, thanks to its large chromosomes, well-developed genetics, and the availability of diverse cytogenetic and molecular tools. Insights gained from maize studies can extend to other species. In this review, we describe a …

The MYB transcription factors family constitutes one of the largest and most functionally diversified regulatory gene families in plants. Beyond their established roles in growth and development, MYB TFs function as central integrators of metabolic reprogramming and environmental adaptation by coordinating developmental processes, secondary metabolism, hormone signaling, and stress-responsive pathways. Despite substantial progress in elucidating the functions of individual MYB members, a systematic synthesis linking their structural diversification, evolutionary expansion, and functional speci

Plant-microbe interactions are essential for plant health, yet the genetic basis of host control over root-associated microbiome assembly in Nicotiana tabacum remains unclear. We integrated whole-genome resequencing, amplicon sequencing, and metabolomics across 50 tobacco cultivars, together with microbial genome-wide association and mediation analyses, to investigate host-microbiome covariation. Host genotype significantly influenced the diversity and composition of root-associated microbial communities, particularly in the rhizosphere. Several host loci were associated with heritable microbi
