Methylenecyanamide (CH2═NCN) and (Z)- and (E)-Iminoacetonitriles (NC-CH═NH), Dimers of Hydrogen Cyanide.

(Z)- and (E)-iminoacetonitriles (NCCH═NH), two hydrogen cyanide dimers, are described as key compounds in prebiotic chemistry. Among the many possible dimers of HCN with covalent bonds, even the second on the scale of thermodynamic stability, methylenecyanamide (CH2═NCN), has been investigated little. We show that this compound can be isolated, is stable enough to give an adduct with a nucleophile or a diene, and can be easily generated under prebiotic conditions, highlighting a possible role in this medium. Comparison between isomers shows significant differences in formation and chemical rea

Dietary supplementation with Bacillus velezensis and Pichia guilliermondii improves growth performance through intestinal morphology and functionality enhancement in weaning piglets.

The post-weaning period in piglets is often associated with significant changes in intestinal structure and function, which can negatively affect growth and performance. This study aimed to evaluate the effects of dietary supplementations with prebiotics and/or probiotics on intestinal morphology and functionality during the post-weaning phase. Piglets were supplemented with different diets, including combinations of Pichia guilliermondii (prebiotic, PRE) and Bacillus velezensis (probiotic, PRO), and their growth performance, intestinal histology, and functionality were assessed. Growth perfor…

Effect of beta-glucan supplementation on cystic fibrosis colonic microbiota: an in vitro study.

BACKGROUND: Children with cystic fibrosis (CF) present with gut dysbiosis, and current evidence impedes robust recommendations on the use of prebiotics. This study aimed at establishing the prebiotic potential of a commercial beta-glucan on the in vitro colonic microbiota of a child with CF compared to a healthy counterpart (H). METHODS: A dynamic simulator of colonic fermentation (twin-SHIME® model) was set up including the simulation of the proximal (PC) and distal colon (DC) of the CF and the H subjects by colonizing the bioreactors with faecal microbiota. During two weeks the system was s…

Can modulation of gut microbiota affect anthropometric indices in patients with non-alcoholic fatty liver disease? An umbrella meta-analysis of randomized controlled trials.

BACKGROUND AND AIM: Modulating the gut microbiota population by administration of probiotics, prebiotics, and synbiotics has shown to have a variety of health benefits in different populations, particularly those with metabolic disorders. Although the promising effects of these compounds have been observed in the management of patients with non-alcoholic fatty liver disease (NAFLD), the exact effects and the mechanisms of action are yet to be understood. In the present study, we aimed to evaluate how gut microbiota modulation affects anthropometric indices of NAFLD patients to achieve a compre…

Gut microbiome dynamics of patients on dialysis: implications for complications and treatment.

The gut microbiome plays a significant role in dialysis. As disease progresses, the choice of dialysis method and dietary habits change, and the diversity and richness of the gut microbiome in patients on dialysis change as well. The uremic toxins produced exacerbate inflammatory responses and oxidative stress, leading to markedly different incidence rates of complications such as cardiovascular disease and dialysis-associated peritonitis among patients on dialysis. The intake of probiotics, prebiotics, synbiotics, and natural medicines during daily life can regulate the gut microbiome, reduce…

Relationship among Parkinson's disease, constipation, microbes, and microbiological therapy.

This comprehensive review elucidates the complex interplay between gut microbiota and constipation in Parkinson's disease (PD), a prevalent non-motor symptom contributing significantly to patients' morbidity. A marked alteration in the gut microbiota, predominantly an increase in the abundance of Proteobacteria and Bacteroidetes, is observed in PD-related constipation. Conventional treatments, although safe, have failed to effectively alleviate symptoms, thereby necessitating the development of novel therapeutic strategies. Microbiological interventions such as prebiotics, probiotics, and feca…

A Combination of Inulin and Long-Chain Fructooligosaccharides Enhances in Vitro Iron Bioavailability and Supports a Favorable Infant Gut Microbiome.

Iron deficiency is common during early life. Prebiotics may enhance iron bioavailability (IB) in the small intestine and in the colon. To study this, IMF was supplemented with inulin, lcFOS, their combinations (1:1 or 2:1), or scGOS:lcFOS (9:1). IMFs were digested in vitro, or fermented ex vivo yielding short-chain fatty acids (SCFA) and altering microbiota composition. Digested IMF containing either prebiotics or SCFA were incubated on Caco-2 cells to assess IB under simulated small-intestinal or colonic conditions respectively. Fermentation of inulin:lcFOS and scGOS:lcFOS increased Bifidobac…

One-Pot Formation of Pairing Proto-RNA Nucleotides and Their Supramolecular Assemblies.

Most contemporary theories for the chemical origins of life include the prebiotic synthesis of informational polymers, including strong interpretations of the RNA World hypothesis. Existing challenges to the prebiotic emergence of RNA have encouraged exploration of the possibility that RNA was preceded by an ancestral informational polymer, or proto-RNA, that formed more easily on the early Earth. We have proposed that the proto-nucleobases of proto-RNA would have readily formed glycosides with ribose and that these proto-nucleosides would have formed base pairs as monomers in aqueous solution

Synbiotic laden with Faecalibacterium prausnitzii, fructooligosaccharides, and galactooligosaccharides mitigates quinpirole-induced OCD-like symptoms in rats.

Obsessive-compulsive disorder (OCD) is a mental illness of unwanted recurrent thoughts and compulsive behaviors. The significance of the microbiota-gut-brain axis has been increasingly documented in the manifestation of neuropsychiatric disorders. Herein, we investigated the impact of synbiotic containing Faecalibacterium prausnitzii ATCC 27,766 (F. prausnitzii) and prebiotics [fructooligosaccharides (FOS) and galactooligosaccharides (GOS)] on quinpirole-induced OCD-like symptoms in rats. Repeated quinpirole injections led to compulsive- and anxiety-like behaviors, as rats showed reduced head-…

Cometary Delivery of Hydrogen Cyanide to the Early Earth.

Delivery of water and organics by asteroid and comet impacts may have influenced prebiotic chemistry on the early Earth. Some recent prebiotic chemistry experiments emphasize hydrogen cyanide (HCN) as a feedstock molecule for the formation of sugars, ribonucleotides, amino acids, and lipid precursors. Here, we assess how much HCN originally contained in a comet would survive impact, using parametric temperature and pressure profiles together with a time-dependent chemistry model. We find that HCN survival mainly depends on whether the impact is hot enough to thermally decompose H2O into reacti

On the origins of life's homochirality: Inducing enantiomeric excess with spin-polarized electrons.

Life as we know it is homochiral, but the origins of biological homochirality on early Earth remain elusive. Shallow closed-basin lakes are a plausible prebiotic environment on early Earth, and most are expected to have significant sedimentary magnetite deposits. We hypothesize that ultraviolet (200- to 300-nm) irradiation of magnetite deposits could generate hydrated spin-polarized electrons sufficient to induce enantioselective prebiotic chemistry. Such electrons are potent reducing agents that drive reduction reactions where the spin polarization direction can enantioselectively alter the r

Solid-State Fermentation of Riceberry Rice with Mushroom Mycelium for Enhanced Beta-Glucan Production and Health Applications.

Beta-glucans (β-glucans), polysaccharides found in cereals and fungi, are recognized for their prebiotic and potential anti-cancer activities, particularly in the colorectal area. This study aims to optimize the production of β-glucan through the solid-state fermentation of germinated Riceberry rice with Pleurotus ostreatus and evaluate the bioactivities of the resulting extract. The crude β-glucan extract, obtained with a recovery rate of 54.95% and 79.98% purity, demonstrated an effective extraction process, as confirmed by thermogravimetric analysis (TGA). Fourier-transform infrared spectro

Arabinoxylan and Pectin Metabolism in Crohn's Disease Microbiota: An In Silico Study.

Inflammatory bowel disease is a chronic disorder including ulcerative colitis and Crohn's disease (CD). Gut dysbiosis is often associated with CD, and metagenomics allows a better understanding of the microbial communities involved. The objective of this study was to reconstruct in silico carbohydrate metabolic capabilities from metagenome-assembled genomes (MAGs) obtained from healthy and CD individuals. This computational method was developed as a mean to aid rationally designed prebiotic interventions to rebalance CD dysbiosis, with a focus on metabolism of emergent prebiotics derived from …

Production of tartrates by cyanide-mediated dimerization of glyoxylate: a potential abiotic pathway to the citric acid cycle.

An abiotic formation of meso- and DL-tartrates in 80% yield via the cyanide-catalyzed dimerization of glyoxylate under alkaline conditions is demonstrated. A detailed mechanism for this conversion is proposed, supported by NMR evidence and (13)C-labeled reactions. Simple dehydration of tartrates to oxaloacetate and an ensuing decarboxylation to form pyruvate are known processes that provide a ready feedstock for entry into the citric acid cycle. While glyoxylate and high hydroxide concentration are atypical in the prebiotic literature, there is evidence for natural, abiotic availability of eac
