From opsin gene expression to visual performance: an investigation into the multiple spectral sensitivities of boxfishes (Ostracion spp.).

Colour vision arises from the comparison of spectral input across at least two differently tuned photoreceptors. In most vertebrates, colour discrimination is mediated by two to four differently tuned cone photoreceptors, providing sensitivity from ultraviolet (<400 nm) to red (∼600-700 nm) wavelengths. However, microspectrophotometry (MSP) indicated that the white-spotted boxfish, Ostracion meleagris, collected from Hawaiian reefs could possess five spectrally distinct cones. To explore the potential for increased cone diversity, we used a multidisciplinary approach to investigate colour visi

Local variations in L/M ratio influence the detection and color naming of small spots.

The distribution of long (L)-, middle (M)-, and short (S)-wavelength sensitive cones in the retina determines how different frequencies of incident light are sampled across space and has been hypothesized to influence spatial and color vision. We examined how the detection and color naming of small, short-duration increment stimuli (λ = 543 or 680 nm) depend on the local spectral topography of the cone mosaic. Stimuli were corrected for optical aberrations by an adaptive optics system and targeted to locations in the parafovea where cone spectral types were known. We found that sensitivity to

Zebrafish use spectral information to suppress the visual background.

Vision first evolved in the water, where the spectral content of light informs about viewing distance. However, whether and how aquatic visual systems exploit this "fact of physics" remains unknown. Here, we show that zebrafish use "color" information to suppress responses to the visual background. For this, zebrafish divide their intact ancestral cone complement into two opposing systems: PR1/4 ("red/UV cones") versus PR2/3 ("green/blue cones"). Of these, the achromatic PR1 and PR4, which are retained in mammals, are necessary and sufficient for vision. By contrast, the color-opponent PR2 and

Noninvasive Assessment of Ocular Structure in the Elderly 13-Lined Ground Squirrel.

The 13-lined ground squirrel (13-LGS) is an attractive model for vision research due to its cone-dominance, accessibility, and amenability to noninvasive retinal imaging. The ability to interpret results from longitudinal studies in this animal model is limited by a lack of normative data from aged animals. To address this, we used noninvasive imaging to characterize structural changes in the anterior segment and retina in 20 13-LGS aged 5-8 years of age. Our imaging revealed multiple age-related changes in the 13-LGS eye. Phenotypes include lens opacifications, loss of normal cone structure s

Age-Related Macular Degeneration, a Mathematically Tractable Disease.

A progression sequence for age-related macular degeneration onset may be determinable with consensus neuroanatomical nomenclature augmented by drusen biology and eye-tracked clinical imaging. This narrative review proposes to supplement the Early Treatment of Diabetic Retinopathy Study (sETDRS) grid with a ring to capture high rod densities. Published photoreceptor and retinal pigment epithelium (RPE) densities in flat mounted aged-normal donor eyes were recomputed for sETDRS rings including near-periphery rich in rods and cumulatively for circular fovea-centered regions. Longitudinal Assessment of OCT-Based Measures of Foveal Cone Structure in Achromatopsia.

PURPOSE: Achromatopsia (ACHM) is an autosomal recessive retinal disease associated with reduced or absent cone function. There is debate regarding the extent to which cone structure shows progressive degeneration in patients with ACHM. Here, we used optical coherence tomography (OCT) images to evaluate outer nuclear layer (ONL) thickness and ellipsoid zone (EZ) integrity over time in individuals with ACHM. METHODS: Sixty-three individuals with genetically confirmed ACHM with follow-up ranging from about 6 months to 10 years were imaged using either Bioptigen or Cirrus OCT. Foveal cone structu

Blue cone monochromacy and gene therapy.

Blue cone monochromacy (BCM) is a congenital vision disorder characterized by complete loss or severely reduced long- and middle-wavelength cone function, caused by mutations in the OPN1LW/OPN1MW gene cluster on the X-chromosome. BCM patients typically suffer from poor visual acuity, severely impaired color discrimination, myopia, and nystagmus. In this review, we cover the genetic causes of BCM, clinical features of BCM patients, genetic testing, and clinical outcome measurements for future BCM clinical trials. However, our emphasis is on detailing the animal models for BCM and gene therapy u

An atypical case of myopic strabismus fixus.

We present an atypical case of progressive fixed hypertropia in a patient with extreme myopia. Myopic strabismus fixus is typically characterized by progressive esotropia and hypotropia, which is caused by the protrusion of the posterior eyeball from the superotemporal quadrant of the extraocular muscle cone. In this case, the patient's right eye was fixed in a superomedial position, and the left eye was fixed and deviated upward. Magnetic resonance imaging revealed protrusion of a posterior scleral staphyloma inferotemporally in each eye. This case illustrates how the pattern of strabismus fi

On the physiological processes underlying optoretinography [Invited].

Optoretinography (ORG) is a label-free imaging of the light-evoked changes in the retina associated with alterations in the cell shape and refractive indices. Its most precise version, the phase-resolved optical coherence tomography (pOCT), exhibited sensitivity of about 10 nm in vivo, limited by the signal-to-noise ratio and accuracy of the tissue registration. While it is yet insufficient for the detection of single action potentials, which are about 1 nm in amplitude, it enables monitoring slower and larger deformations in other retinal layers. In response to a single flash delivered to the

Slowly progressive changes of the retina and retinal pigment epithelium in Briard dogs with hereditary retinal dystrophy. A morphological study.

Seven eyes from 2 generations of Briard dogs (5 weeks--7 years old) with congenital night blindness and (in the second generation) impairment of day vision to varying degrees, were examined by light and electron microscopy. Specimens from 4 locations were studied: the central area, the midperiphery of the tapetal area, the upper periphery and the lower periphery. Disorientation of rod outer segment disc membranes was seen in the 5-week-old dog. Large electron-lucent inclusions were found in the RPE at 3.5 months of age. These inclusions occurred most frequently in the central and midperipheral

RPE65 knockout Xenopus laevis have a compromised but detectable electroretinogram and altered visual responses, without retinal degeneration or altered melanophore dispersion.

The isomerohydrolase RPE65 is a critical element of the visual cycle, the series of enzymatic reactions by which the chromophore of the visual pigments is regenerated following light exposure. In humans, mutations in the RPE65 gene cause a severe form of blindness called Leber's congenital amaurosis. Studies of Rpe65-/- mice have shown dramatic depletion of 11-cis-retinal in the retina, resulting in a slow retinal degeneration. However, a number of studies suggest that RPE65 may not be necessary for the regeneration of photopigment in all photoreceptor types. Using CRISPR/Cas9 technology, we p

Postprenylation processing of phosphodiesterase is critical for robust cone photoreceptor response.

In mammalian photoreceptors, several phototransduction proteins undergo prenylation, a posttranslational lipid modification. These prenylated proteins are then processed by RAS-converting enzyme 1 (RCE1), which cleaves the final three amino acids. RCE1-mediated endoproteolysis is critical for maintaining rod photoreceptor structure and function; its early loss in the retina causes rapid degeneration and disrupts the trafficking of rod phosphodiesterase 6 (PDE6). However, the early and rapid degeneration has limited our understanding of RCE1's role in cones. To address this challenge, we develo

Macular function in tilted disc syndrome.

Tilted disc syndrome can cause visual field defects due to an optic disc anomaly. Recent electrophysiological findings demonstrate reduced central outer retinal function with ophthalmoscopically normal maculae. We measured macular sensitivity with the microperimeter and performed psychophysical assessment of mesopic rod and cone luminance temporal sensitivity (critical fusion frequency) in a 52-year-old male patient with tilted disc syndrome and ophthalmoscopically normal maculae. We found a marked reduction of sensitivity in the central 20 and reduced rod- and cone-mediated mesopic visual fun

Visual field and electrophysiological abnormalities due to vigabatrin.

The purpose of this study was to determine the electrophysiological changes in patients using the anti epileptic drug vigabatrin and to correlate these findings with the previously reported risk for visual field loss in these patients. In 1998 the neurologists of both involved hospitals referred all patients on vigabatrin medication for ophthalmological examination to the outpatients clinics. Of the 33 patients whom were referred to our outpatient clinics, four had to be dropped from the study because of disability to perform the examinations the remaining 29 patients were included in the stud

Whole-mount Retinal Organoid Visualization with Cellular Resolution.

Retinal organoids derived from human induced pluripotent stem cells are intricate 3D structures that mimic the human retina, offering a powerful platform for studying retinal development, disease mechanisms, and potential therapeutic strategies. Moreover, as they are derived from patients, they are becoming increasingly popular as they hold great promise as a tool for personalized medicine. Unlike conventional 2D cell cultures, retinal organoids preserve the native 3D architecture of the retina, allowing for a more realistic representation and enabling more physiologically relevant studies. Mammalian Retinal Bipolar Cells: Morphological Identification and Systematic Classification in Rabbit Retina With a Comparative Perspective.

Retinal bipolar cells (BCs) convey visual signals from photoreceptors to more than 50 types of rabbit retinal ganglion cells (Famiglietti, 2020). More than 40 years ago, 10-11 types of bipolar cells were recognized in rabbit and cat retinas (Famiglietti, 1981). Twenty years later, 10 were identified in mouse, rat, and monkey, while recent molecular genetic studies indicate that there are 15 types of bipolar cell in mouse retina (Shekhar et al., 2016). The present detailed study of more than 800 bipolar cells in 11 Golgi-impregnated rabbit retinas indicates that there are 14-16 types of cone bi

In vitro biomechanical modulation--retinal detachment in a box.

BACKGROUND: To illustrate the importance of biomechanical impact on tissue health within the central nervous system (CNS), we herein describe an in vitro model of rhegmatogenous retinal detachment (RRD) in which disruption and restoration of physical tissue support can be studied in isolation. METHODS: Adult retinal porcine explants were kept in culture for 3 or 12 hours without any tissue support, simulating clinical RRD, after which they were either maintained in this state or reattached to the culture membrane for an additional 48 hours. RESULTS: In vitro detachment resulted in gliosis an
