Wss1 and Ddi1 DNA-Protein crosslink repair proteases protect Saccharomyces cerevisiae and Candida albicans against oxidative stress.

Oxidative stress is a major microbicidal mechanism of phagocytes causing molecular damages, including DNA-protein crosslinks (DPCs), toxic DNA lesions that can lead to genomic instability. DPC repair depends on Wss1 and Ddi1 proteases, so they may be critical for fungal survival in hosts, yet their roles have not been fully elucidated. In this study, the roles of Wss1 and Ddi1 under oxidative stress were examined in the model yeast Saccharomyces cerevisiae and Candida albicans, an important human fungal pathogen. Direct measurements of DPCs using SDS/KCl precipitation showed that oxidative str…

Effect of Non-Saccharomyces Yeasts Derived from Traditional Fermented Foods on Beer Fermentation Characteristics and Flavor Profiles.

In recent years, numerous studies have demonstrated that non-Saccharomyces yeasts hold potential for industrial application and aroma generation during fermentation. Non-Saccharomyces wild yeasts can be important tools in the development of new products, and the objective of this work was to obtain and characterize novel yeast isolates for their ability to produce beer. Traditional fermented beverages serve as a vital source of yeast strains that can exhibit unique characteristics during the brewing process. Thus, 22 strains of Saccharomycopsis fibuligera were isolated from traditional ferment…

Harnessing native nitrogen in lignocellulosic biomass for cellulosic ethanol production by ancestral xylose isomerase-engineered Saccharomyces cerevisiae.

Efficient xylose-utilizing Saccharomyces cerevisiae, straightforward pretreatment, elimination of detoxification steps, reduced cellulase dosage, and cost-effective nutrients are critical for the commercialization of lignocellulosic ethanol production. In this study, three highly efficient xylose-utilizing S. cerevisiae, which were capable of consuming 40 g/L xylose within 14 h and consuming a mixture of 80 g/L glucose and 40 g/L xylose within 18 h, were developed by integrating artificial ancestral xylose isomerases into diploid S. cerevisiae genome, followed by laboratory evolution and colon…

Identifying promoters to enhance heterologous gene expression in recombinant Saccharomyces cerevisiae strains cultivated on non-native substrates.

Efficient bioconversion of lignocellulosic biomass (LCB) to ethanol by Saccharomyces cerevisiae requires its engineering to express heterologous enzymes at titres high enough to make significant impacts on industrial consolidated bioprocessing (CBP). Promoters are required for this purpose, but are reportedly influenced by various environmental factors as well as the protein specific nature of expression, warranting the need for assessment under the conditions for which they are intended. Heterologous xylosidase- and xylanase-encoding genes (xln43_SED1 and xyn2) were individually cloned under …

Novel function of ribosomal protein RPL14B: regulation of CdSe quantum dots biosynthesis in living Saccharomyces cerevisiae cells.

Biosynthesis of CdSe quantum dots is the process of converting metal ions into semiconductor nanomaterials. Studies have shown that CdSe quantum dots synthesized by Saccharomyces cerevisiae are rich in ribosomal proteins, but the role of ribosomal proteins in the synthesis of CdSe quantum dots remains unclear. In this paper, ribosomal proteins enriched during the synthesis of CdSe quantum dots by S. cerevisiae were screened, and their effects on the synthesis of quantum dots were detected by gene knockout and over-expression. The results reveal that ribosomal protein RPL14B is involved in the …

Impact of complex inhibitors in straw hydrolysates on xylose-fermenting Saccharomyces cerevisiae and inhibitor-tolerant strain construction.

Enhancing the ability of Saccharomyces cerevisiae strains to ferment mixed sugars under complex inhibitors is crucial for boosting the economic feasibility of cellulosic ethanol production. Using comparative transcriptomics, this study explored the impact of combined inhibitors (acetic acid, formic acid, furfural, and vanillin) on the xylose-fermenting S. cerevisiae strain s6 during mixed-sugar fermentation. The results revealed significant disruptions in carbon metabolism, amino acid metabolism, nucleic acid metabolism, energy metabolism, and signal transduction pathways, suggesting that cell…

Characterization of polysulfides in Saccharomyces cerevisiae cells and finished wine from a cysteine-supplemented model grape medium.

Polysulfide degradation in wine can result in hydrogen sulfide (H2S) release, imparting a rotten-egg smell that is detrimental to wine quality. Although the presence of wine polysulfides has been demonstrated, their biogenesis remains unclear. This study investigated the role of Saccharomyces cerevisiae in polysulfide formation during fermentation, with and without 5 mM cysteine supplementation as an H2S source. Using an established liquid chromatography-tandem mass spectrometry method, monobromobimane derivatives of hydropolysulfides, including CysSSSH, CysSSSSH and GSSSSH, and two oxidized p…

Saccharomyces boulardii, a yeast probiotic, inhibits gut motility through upregulating intestinal serotonin transporter and modulating gut microbiota.

Saccharomyces boulardii (Sb) is a widely used fungal probiotic in treating various digestive diseases, including irritable bowel syndrome (IBS). However, the specific mechanisms of Sb relieving IBS remain unclear. The abnormal serotonin transporter (SERT) / 5-hydroxytryptamine (5-HT) system could cause disordered gastrointestinal sensation and motility, which closely related to IBS pathogenesis. The aim of this study was to explore the effects and mechanisms of Sb on regulating gut motility. Sb supernatant (SbS) was administered to intestinal epithelial cells and mice. SbS upregulated SERT exp…

Quantitative Physiology of Non-Energy-Limited Retentostat Cultures of Saccharomyces cerevisiae at Near-Zero Specific Growth Rates.

So far, the physiology of Saccharomyces cerevisiae at near-zero growth rates has been studied in retentostat cultures with a growth-limiting supply of the carbon and energy source. Despite its relevance in nature and industry, the near-zero growth physiology of S. cerevisiae under conditions where growth is limited by the supply of non-energy substrates remains largely unexplored. This study analyzes the physiology of S. cerevisiae in aerobic chemostat and retentostat cultures grown under either ammonium or phosphate limitation. To compensate for loss of extracellular nitrogen- or phosphorus-c…

Surface display of synthetic phytochelatins on Saccharomyces cerevisiae for enhanced ethanol production in heavy metal-contaminated substrates.

The aim of this work was to study the feasibility of surface displaying synthetic phytochelatin (EC) on Saccharomyces cerevisiae to overcome the inhibitory effect of heavy metals on ethanol production. Via the fusion of a gene encoding EC to an α-agglutinin gene, the engineered S. cerevisiae was able to successfully display EC on its surface. This surface engineered yeast strain exhibited an efficient cadmium adsorption capability and a remarkably enhanced cadmium tolerance. Moreover, its ethanol production efficiency was significantly improved as compared to a control strain in the presence o…

Quaternary ammonium salt N-(dodecyloxycarboxymethyl)-N,N,N-trimethyl ammonium chloride induced alterations in Saccharomyces cerevisiae physiology.

We investigated the influence of the quaternary ammonium salt (QAS) called IM (N-(dodecyloxycarboxymethyl)- N,N,N-trimethyl ammonium chloride) on yeast cells of the parental strain and the IM-resistant mutant (EO25 IMR) growth. The phenotype of this mutant was pleiotropic. The IMR mutant exhibited resistance to ethanol, osmotic shock and oxidative stress, as well as increased sensitivity to UV. Moreover, it was noted that mutant EO25 appears to have an increased resistance to clotrimazole, ketoconazole, fluconazole, nystatin and cycloheximide. It also tolerated growth in the presence of crysta

A physiological, biochemical and proteomic characterization of Saccharomyces cerevisiae trk1,2 transport mutants grown under limiting potassium conditions.

Saccharomyces cerevisiae mutants lacking both isoforms of the main plasma membrane potassium transporter display impaired potassium transport and defective growth at limiting concentrations of the cation. Moreover, they are hyperpolarized and have a lower intracellular pH than wild-type. In order to unravel global physiological processes altered in trk1,2 mutants, we have established conditions at which both wild-type and mutants can grow at different rates. Using a combination of physiological, biochemical and proteomic approaches, we show that during growth at suboptimal potassium concentrat…

Virus-like particles for enterovirus 71 produced from Saccharomyces cerevisiae potently elicits protective immune responses in mice.

Human Enterovirus 71 (EV71) is recognized as the leading causative agent of hand-foot-and-mouth disease (HFMD) in the Asia-Pacific region in recent years. There are still no approved antiviral drugs or vaccines against EV71 infection yet. In this study, we have developed an advanced platform for production of the virus-like particles (VLPs) for EV71 in Saccharomyces Cerevisiae by co-expressing P1 and 3CD genes of EV71. These VLPs exhibited similar morphology and protein composition as EV71 empty particles produced from EV71-infected cells. Immunization with VLPs in mice elicited robust neutral…
