Discovery of three toxin peptides with Kv1.3 channel and IL-2 cytokine-inhibiting activities from Non-Buthidae scorpions, Chaerilus tricostatus and Chaerilus tryznai.

Non-Buthidae venomous scorpions are huge natural sources of toxin peptides; however, only a few studies have been done to understand their toxin peptides. Herein, we describe three new potential immunomodulating toxin peptides, Ctri18, Ctry68 and Ctry2908, from two non-Buthidae scorpions, Chaerilus tricostatus and Chaerilus tryznai. Sequence alignment analyses showed that Ctri18, Ctry68 and Ctry2908 are three new members of the scorpion toxin α-KTx15 subfamily. Electrophysiological experiments showed that Ctri18, Ctry68 and Ctry2908 blocked the Kv1.3 channel at micromole to nanomole levels, bu…

Gonadal development in scorpion mud-turtles, Kinosternon scorpioides, in a controlled environment.

Research on gonadal development including sexual differentiation of gonads in chelonians is essential for understanding the mechanisms that determine offspring sex and, consequently, the sex ratio of egg clutches. This directly impacts population structure, as sex ratios influence reproductive dynamics and species' long-term viability. The aim of this study was to characterize the gonadal development of scorpion mud-turtle (Kinosternon scorpioides). A total of 85 embryos from stages 12-26 were analyzed, kept under digital temperature control (28.5 ± 0.5°C). The reproductive system was dissecte

State of the art on the development of a recombinant antivenom against Mexican scorpion stings.

Around 2,750 species of scorpions have been recorded worldwide and classified into 21 families and 208 genera. Of these, the family Buthidae stands out as one of the largest, comprising several genera including the genus Centruroides with 102 recorded species. This genus is home to the largest number of species dangerous to humans as described in Mexico, where there are 55 species of the genus Centruroides, of which more than 24 are of medical importance. Envenoming in humans is caused by the presence of peptides (toxins) in the venom that modify the gating mechanism of Na+ voltage dependent i…

In Vitro and In Vivo Anti-Candida albicans Activity of a Scorpion-Derived Peptide.

The increasing infection and drug resistance frequency has encouraged the exploration of new and effective anti-Candida albicans agents. In this study, CT-K3K7, a scorpion antimicrobial peptide derivative, effectively inhibit the growth of C. albicans. CT-K3K7 killed C. albicans cells in a dose-dependent manner, mainly by damaging the plasma membrane. CT-K3K7 could also disrupt the nucleus and interact with nucleic acid. Moreover, CT-K3K7 induced C. albicans cells necrosis via a reactive oxygen species (ROS)-related pathway. Furthermore, CT-K3K7 inhibited the hyphal and biofilm formation of C.

Mapping the interaction surface of scorpion β-toxins with an insect sodium channel.

The interaction of insect-selective scorpion depressant β-toxins (LqhIT2 and Lqh-dprIT3 from Leiurus quinquestriatus hebraeus) with the Blattella germanica sodium channel, BgNav1-1a, was investigated using site-directed mutagenesis, electrophysiological analyses, and structural modeling. Focusing on the pharmacologically defined binding site-4 of scorpion β-toxins at the voltage-sensing domain II (VSD-II), we found that charge neutralization of D802 in VSD-II greatly enhanced the channel sensitivity to Lqh-dprIT3. This was consistent with the high sensitivity of the splice variant BgNav2-1, be

1,3,4-Azadiphospholides as building blocks for scorpionate and bidentate ligands in multinuclear complexes.

Annulated oxy-substituted 1,3,4-azadiphospholides such as the anion in Na[1] are readily accessible phosphorus heterocycles made from the phosphaethynolate anion (OCP)- and 2-chloropyridines. The sodium salt Na[1] reacts with oxophilic element halides such as OPCl3, PhSiCl3, PhBCl2 and CpTiCl3 at room temperature to form exclusively the oxygen bound tris-substituted compounds E(1)3 (with E = OP, PhSi, PhB- or CpTi). Six equivalents of Na[1] with group four metal chlorides MCl4 (M = Ti, Zr, Hf) form cleanly the hexa-substituted dianions (Na2[M(1)6]) which are isolated in excellent yields. The t

Voltage-Gated K+/Na+ Channels and Scorpion Venom Toxins in Cancer.

Ion channels have recently been recognized as novel therapeutic targets in cancer research since they are overexpressed in different histological tissues, and their activity is linked to proliferation, tumor progression, angiogenesis, metastasis, and apoptosis. Voltage gated-potassium channels (VGKC) are involved in cell proliferation, cancer progression, cell cycle transition, and apoptosis. Moreover, voltage-dependent sodium channels (VGSC) contribute to decreases in extracellular pH, which, in turn, promotes cancer cell migration and invasion. Furthermore, VGSC and VGKC modulate voltage-sen

Scorpion Toxins Targeting Voltage-gated Sodium Channels Associated with Pain.

Pain affects approximately 30% of people and places a large economic and social burden on society. Despite the availability of a range of analgesics, complete alleviation of symptoms still rarely occurred. Effective and safe drugs for the treatment of pain are still an unmet clinical need. In recent years, the voltage-gated sodium channels (VGSCs) have been recognized as potential targets for analgesic development. VGSCs are major players in generating and propagating action potentials. They represent an appealing target for the development of new and safer drugs in the treatment of pain. The
